    Objective

To observe the effect of acupuncture treatment on coexpression of neuronal nitric oxide synthase(nNOS)and postsynaptic dense-95(PSD-95)in the hippocampus of depression model rats.

    Method

Fifty SPF grade rats were randomly divided into six groups:normal group, model group(intraperitoneal injection of 0.2 mg·kg-1·d-1 of reserpine),acupuncture group,ZL006 group, acupuncture combined with ANA-12 group,8-OH-DPAT group. There were 9 rats in the normal group and model group, and 8 rats in each of the remaining groups. The changes in body weight and anal temperature were observed. Open field test, sucrose preference test and forced swimming test were applied to observe depression-like behaviors in rats. The morphological changes of hippocampal neurons were observe by HE staining. Immunofluorescence double staining was used to observe the positive expression of nNOSand PSD-95,as well as tyrosine kinase receptor B(TrkB)and PSD-95 in neurons of the hippocampal CA1 and CA3 regions.

    Result

There were coexpressions of nNOSand PSD-95 as well as TrkB and PSD-95 in neurons of the hippocampal CA1 and CA3 regions. Compared with those in the model group,depression-like behaviors were significantly improved in the acupuncture group and ZL006 group(P<0.05,P<0.01,P<0.001). The coexpression levels of nNOSand PSD-95 in the CA3 region were significantly down-regulated(P<0.05,P<0.01),and the coexpression levels of TrkB and PSD-95 in the CA3 region were significantly up-regulated(P<0.05). The reduction and damage of hippocampal neurons were significantly improved. The expression levels of nNOSin the hippocampal CA1 region were significantly down regulated(P<0.01)and the expression levels of TrkB in the CA1 region were significantly up-regulated(P<0.01)in the acupuncture group. The expression level of nNOSin the CA1 region of the 8-OH-DPAT group was significantly down-regulated(P<0.01). Compared with those in the acupuncture group, the coexpression levels of nNOSand PSD-95 in the CA3 region of the ANA-12 group were significantly up-regulated(P<0.05),and the above differences were statistically significant.

    Conclusion

Acupuncture regulates Shaoyang pivot, which may participate in the antidepressant effect by up-regulating the coexpression levels of nNOSand PSD-95 in hippocampal CA3 neurons, and this mechanism of action may be related to TrkB receptors in the CA3 region.

    Objective

To explore the potential molecular mechanisms underlying the occurrence and development of liver cancer based on bioinformaticsand to screen and validate the mechanisms and targets of Biejiajian Pill(鳖甲煎丸,BJJP)on liver cancer.

    Methods

Single-cell data of hepatocellular carcinoma(HCC)was downloaded from the GEO database, processedand subjected to communication analysis, enrichment and metabolic analysis, and reanalysis of cellular subpopulations. Subsequently, the components and action targets of BJJP were collected, and in conjunction with transcriptomic results, the intersection targets between BJJP and HCC were identified and analyzed. Finally, protein and small molecule information was obtained from the PDB and PubChem databases to simulate drug-target molecular docking. A computational dynamics simulation environment was constructed to assess the interactions.

    Results

Transcriptomic analysis revealed that intercellular communication was more active in the peritumoral group of HCC. The cells were predominantly enriched in pathways related to cell cycle checkpoints and immune responses. The hepatocyte metabolism was active, while immune cell metabolism was suppressed. The natural killer(NK)cells gradually evolved from possessing normal cytotoxic capabilities to lower capabilities, and hepatocytes gradually transformed from normal to cancerous cells. Network pharmacology results showed that BJJP potentially regulated 124 targets in HCC,mostly enriched in pathways such as immune response, inflammatory response, glycolysisand interleukin-17. Molecular docking was performed with key proteins and potential drug action targets, and the top three with the strongest binding affinity were subjected to dynamic simulation, which showed that these three receptor-ligand complexes were relatively stable.

    Conclusion

HCCcells may promote malignant proliferation and immune escape by disrupting cell cycle and immune functions. BJJP may achieve antitumor effects by regulating pathways such as anti-inflammatory,immune responseand cell division. The key drug-target relationships were validated with good results in molecular docking and molecular dynamics, demonstrating their favorable binding affinity.

Aging is a biological process in which cells continue to lose and tissue and organ functions gradually undergo degenerative changes. In this process,a class of diseases whose incidence rate increases exponentially with age are called age-related diseases(ARDs). In middle age,the gradual decline of Yangqi, spleen deficiency, kidney deficiency and coldness are the key pathological mechanisms for the occurrence of ARDS. Ferroptosis belongs to iron dependent non apoptotic programmed death. Age related iron homeostasis and redox imbalance make it a key link in the occurrence and development of ARDs. It is an important part of the modern biological connotation of treating ARDs from spleen and kidney, including the correlation between“deficiency of spleen Yang, lack of transportation”and mitochondrial iron homeostasis imbalance, and the correlation between“deficiency of kidney Yang, loss of vital Qi”and redox imbalance. Taking ferroptosis as the breakthrough point, this article enriched the understanding of traditional Chinese medicine theory on ARDs,and summarized the progress of Fuzi(Aconiti Lateralis Radix Praeparata)in clinical and basic research in recent years, aiming to provide new thinking for the study of the mechanism of traditional Chinese medicine and western medicine of ARDs.

Lung cancer is one of the most prevalent malignant tumors in clinical practice with high morbidity and mortality, which poses a major threat to human life and health. Even though great progress has been made on therapeutic strategy, the outcome is still unsatisfactory. Traditional Chinese medicine(TCM)has significant superiority in improving quality of life, delaying disease progression and improving chemoresistance. For malignant diseases,TCM has unique advantages to some extent. Zeqi Decoction(泽漆汤,ZQD),is scattered from Synopsis of the Golden Chamber and possesses the effect of dissipating phlegm and clearing heat. It is frequently used in the treatment of lung cancer in TCM. Basic experiments demonstrated that ZQD inhibited the growth and progression of lung cancer through various pathways, such as inducing cell apoptosis, blocking cell cycle, inhibiting tumor cell migration and invasion, reducing the number of myeloid-derived suppressor cells(MDSCs)and inhibiting their immunosuppressive activity, enhancing the killing powder of natural killer(NK)cells and so on. In addition, main active ingredients, such as luteolin, quercetin, kaempferol, gallic acid,beta-sitosterol,ursolic acid, baicalein and baicalin, also play various roles in anti-tumor,anti-oxidant,anti-angiogenesis,regulating inflammation and immunity. This review aimed to summary clinical application and basic mechanism of ZQD in the treatment of lung cancer. Meanwhile, furthermore researches are needed to explore the mechanism of ZQD in lung cancer.

    Objective

To explore whether ethyl acetate extract of Liujunzi Decoction(EAELD)can regulate the energy metabolism of TE-1 of esophageal cancer cells via the microRNA-1(miR-1)/hypoxia inducible factor-1α(HIF-1α)axis.

    Methods

Stable transfection blocking miR-1 expression cell lines(miR-1 inhibitor,IN)and blank controls(miR-1 NC,NC)have been established in the preliminary experiments. The half maximal inhibitory concentration(IC50)and 30% inhibitory concentration(IC30)of EAELD on NC were obtained using MTT assay, and were used as the Liujunzi Decoction high and low concentrations to compare their effects on IN and NC,respectively. The effect of EAELD on miR-1 and HIF-1αmRNA in TE-1 cells was measured by quantitative real-time PCR(RT-qPCR). Dual-luciferase gene reporter assay was used to validate the targeting relationship of miR-1 and HIF-1α. Then the experiments were divided into NC,Liujunzi Decoction low concentration,Liujunzi Decoction high concentration,IN+Liujunzi Decoction low concentration and IN+Liujunzi Decoction high concentration groups. Seahorse energy metabolism analysis system detected the mitochondrial pressure and glycolysis rate, and the protein expression level was detected by Western Blot.

    Results

MTT results showed that EAELD could significantly inhibit the proliferation of NCin a concentration gradient dependent manner. The low and high concentration groups were 20 μg/mL(IC30)and 40 μg/mL(IC50),respectively. The RT-qPCR results showed that the EAELD high concentration group significantly promoted the gene expression of miR-1(P<0.05)and HIF-1αshowed a downward trend, without statistically significant difference. Dual-luciferase gene reporter assay showed that the fluorescence intensity of wild type(WT)+mimics group was significantly lower than that of WT+negative control(mimics NC)group(P<0.05),and there was no significant difference in the fluorescence intensity of mutant(MUT)+mimics group compared with that of MUT+mimics NCgroup. When high and low concentrations of EAELD were applied to IN,the MTT results showed that compared with that of NCgroup, the inhibitory effect of EAELD on IN group was significantly reduced(P<0.05). Seahorse energy metabolism results showed that compared with those of NC group, the ratio of mitochondrial oxygen consumption to glycolysis(P<0.01,P<0.05),glycolytic potential(P<0.01),basal respiration value, proton leakage oxygen consumption(P<0.01,P<0.05),proton leakage oxygen consumption(P<0.01,P<0.05),mitochondrial maximum oxygen consumption capacity and spare respiratory capacity(P<0.01)were inhibited in the high and low concentration groups of Liujunzi Decoction. The high concentration group could inhibit basal glycolysis, compensate glycolysis,non-mitochondrial oxygen consumption and ATPsynthesis oxygen consumption(P<0.01). Compared with those of NC+Liujunzi Decoction low concentration group, basal glycolysis, mitochondrial to basal glycolysis ratio, glycolytic potential(P<0.05)and oxygen consumption for ATPsynthesis(P<0.01)were significantly increased in IN+Liujunzi Decoction low concentration group. Compared with those of NC+Liujunzi Decoction high concentration group, basal glycolysis, compensatory glycolysis(P<0.05),basal respiratory capacity, proton leakage oxygen consumption, maximum mitochondrial oxygen consumption and non-mitochondrial oxygen consumption(P<0.01)were significantly increased in IN+Liujunzi Decoction high concentration group. WB results showed that compared with NCgroup,EAELD high and low concentration groups significantly decreased the protein relative expression levels of HK2(P<0.01),GLUT1(P<0.05,P<0.01)and MCT1(P<0.01,P<0.05). The relative protein expressions of PKM2(P<0.05),MCT2 and MCT4(P<0.01)in the high concentration of EAELD group were significantly decreased. Compared with those of NC+Liujunzi Decoction low concentration group, the expressions of HK2,GLUT1,PKM2(P<0.01),p-PKM2,MCT1 and MCT2(P<0.05)in IN+Liujunzi Decoction low concentration group were significantly increased. Compared with those of NC+Liujunzi Decoction high concentration group, the expressions of HK2,GLUT1,PKM2(P<0.01),p-PKM2,MCT1 and MCT2(P<0.05)in IN+Liujunzi Decoction low concentration group were significantly increased. IN+Liujunzi Decoction high concentration group could significantly promote the expressions of HK2,GLUT1,PKM2 and MCT4(P<0.05).

    Conclusion

EAELD can inhibit the energy metabolism of esophageal cancer cell line TE-1 by down-regulating the expressions of GLUT1,PKM2,p-PKM2,HK2,MCT1,MCT2 and MCT4,and the mechanism is partly related to miR-1/HIF-1αaxis.

Chronic pulmonary heart disease(CPHD)is the result of the development of a variety of pulmonary diseases to the end-stage stage, with a mixture of deficiency and excess, prolonged course, and complex and changeable conditions. The treatment in remission determines the prognosis of the disease and is the key to the treatment of the disease. The theory of“integration of essence from the skin-hair and the vessels”comes from Inner Canon of Huangdi, which refers to the combination of Qi and blood. Lung governs Qi and heart governs blood. The correlation of Qi and blood under physiological conditions is the mutual use of heart and lung. Under the pathological conditions, the skin-hair and the vessels are disharmonious,Qi and blood are not smooth, the righteous Qi is deficient, and the disease is gradually serious. Over time,it causes cardiopulmonary remodeling, showing a series of symptoms of cardiopulmonary dysfunction. Based on the core pathogenesis of CPHD in remission period,it is advisable to treat it with strengthening body resistance, resolving phlegm and removing blood stasis. The treatment should supplement Qi and nourish blood, benefit Qi and nourish Yin,and cultivate righteous Qi to strengthen the foundation;resolve phlegm and dredge collaterals, promote blood circulation and dispel blood stasis to dispel evils. In this way,if the hair veins are regulated,Qi and blood will be harmonious, the heart and lung will be normal, and the disease can be relieved.
