ArchiveBased on the theory of traditional Chinese medicine,to develop a functional food with homology between medicine and food, composed of Hericium erinaceus,Astragalus radix,Paeonia radix alba,Citri reticulatae pericarpium,Amomi fructus,Magrarita pulveratum, etc.,and to evaluate its auxiliary protective function against gastric mucosal injury.
The anti-Helicobacter pylori activity was evaluated through in vitro experiments. The rat model of alcohol-induced gastric mucosal injury was adopted to evaluate its protective effect on gastric mucosa. The health care function of assisting in protecting gastric mucosa was evaluated through human clinical trials.
In vitro experiments indicated that this functional food had inhibitory and killing effects on the standard strain of Helicobacter pylori (ATCC 700392),with a minimum inhibitory concentration (MIC) of 75 mg·mL-1 and a minimum bactericidal concentration (MBC) of 150 mg·mL-1. Animal experiments showed that the total lesion score of the gastric mucosa histopathological examination in the 4.50 g·kg-1 BW dose group was significantly lower than that in the model control group (P<0.05). The gastric mucosal injury score indices in the 1.50 g·kg-1 BW and 4.50 g·kg-1 BW dose groups were also significantly lower than those in the model control group, indicating an auxiliary protective effect on mucosal injury. Human clinical trials revealed that, compared with their own baseline before the trial, the clinical symptom and sign scores related to gastric mucosal injury in the trial group were significantly reduced (P<0.05).
This study successfully develops a functional food with a clear auxiliary protective function against gastric mucosal injury. Its mechanism of action may be related to the inhibition of Helicobacter pylori and the enhancement of gastric mucosal defense, and it has a good application prospect.
This study aimed to investigate the anti-lung cancer effects of Yiqi Chutan formula (YQCTF) and explore its remodeling effects on the gut and tumor microecology.
Mice were subcutaneously implanted with LLC cells to establish a xenograft model and were intragastrically administered different extracts of YQCTF. Tumor volume and weight were recorded and the peripheral blood cell analysis was performed. 16S rRNA sequencing was used to analyze the composition and abundance of the gut and tumor microbiota.
Different YQCTF extracts inhibited the tumor growth in LLC-bearing mice, reduced the spleen index, and reduced the proportion of peripheral blood mononuclear cells. Furthermore,YQCTF intervention modulated the microbial structure by suppressing the abundance of Prevotella, Oscillibacter, and Alistipes in the gut, while increasing the abundance of Prevotella in tumor tissue. It also inhibited the enrichment of Clostridium_XIVa in both intestinal contents and tumor tissues.
YQCTF attenuates lung cancer growth in mice by modulating the microbiota, including restoring short-chain fatty acid-producing bacterial communities, and suppressing pro-inflammatory bacteria.
Patients with EGFR-mutated lung adenocarcinoma often develop acquired resistance to targeted drugs such as Osimertinib, leading to poor prognosis. Tumor stem cell properties are an important mechanism for drug resistance, and the role of the stemness gene SOX-2 in Osimertinib resistance remains unclear. This study aimed to investigate the function of SOX-2 in Osimertinib resistance in EGFR-mutated lung adenocarcinoma.
This study first analyzed the expression of SOX-2 in lung adenocarcinoma and its correlation with prognosis using the public databases GEPIA,TIMER,and HPA. Additionally,SOX-2 was overexpressed in lung adenocarcinoma cell lines H1975 and PC9,and cellular sensitivity to Osimertinib was assessed using the CCK-8 assay. Sphere-forming capacity was evaluated via 3D Matrigel culture assay.
Overexpression of SOX-2 significantly enhanced the resistance of H1975 and PC9 cells to Osimertinib and enhanced their clonogenic capacity and proliferative ability under three-dimensional culture conditions.
High SOX-2 expression promotes Osimertinib resistance in EGFR-mutated lung adenocarcinoma, and this study provides experimental evidence for the prognosis assessment of EGFR-mutated lung adenocarcinoma.

To mine and analyze adverse drug event (ADE) signals associated with the use of three tetracycline antibiotics (doxycycline, minocycline, and tigecycline) in children, providing evidence for the safe use of tetracyclines in pediatric populations.
ADE reports of doxycycline, minocycline, and tigecycline in children from Q1 2004 to Q2 2025 were extracted from the FDA Adverse Event Reporting System (FAERS) database. Data mining and analysis were performed using the Reporting Odds Ratio (ROR) method and the Bayesian Confidence Propagation Neural Network (BCPNN).
A total of 1 653 pediatric ADE reports were identified, involving doxycycline (664 reports),minocycline (926 reports),and tigecycline (63 reports). Significant signals were detected across 19 important System Organ Classes (SOCs). In children under 2 years old, only doxycycline was found to be associated with adverse reactions, primarily affecting the Blood/Lymphatic System,Metabolism/Nutrition,and Injury/Poisoning categories. In the 2-7 years age group,doxycycline-related events concentrated in Nervous System and Eye disorders, while tigecycline-related events concentrated in Gastrointestinal System disorders. For children aged 8-17 years old, signals were detected for all three drugs: doxycycline was primarily associated with Psychiatric and Gastrointestinal disorders; minocycline was mainly associated with nervous system, eye, and skin and subcutaneous tissue disorders. Tigecycline was primarily associated with injury and poisoning.
Adverse drug reactions (ADRs) of the three tetracyclines are varied across different pediatric age groups. Doxycycline is associated is neurological damage,Horner syndrome, and psychiatric adverse reactions. Minocycline will induce long-term autoimmune disorders, increasing the risk of type 1 diabetes. Tigecycline is found to induce unlabeled hypertriglyceridemia not stated in the drug labeling, necessitating enhanced lipid monitoring during clinical use.
Off-label drug use serves as the primary therapeutic approach for rare diseases, yet systematic evaluation of the risks of its associated adverse drug reactions (ADRs) is lacking. Conventional pharmacovigilance systems are often inadequate due to data sparsity, underreporting, and challenges in signal detection. Artificial intelligence (AI) enables the development of proactive, precise, and real-time ADR surveillance and early-warning frameworks. This review summarizes recent advances in applying AI for active surveillance of off-label ADRs in rare diseases. Key applications include identification of concealed patients, intelligent risk early warning based on multi-source data, and clinical decision support systems powered by AI,such as DrugGPT,“Xiehe-TaiChu” and “Nezha-LingTong”. Despite their promising prospects, significant challenges persist, including data scarcity and heterogeneity, limited model interpretability, privacy and ethical concerns, and the absence of a mature regulatory framework. Future efforts require multidisciplinary collaboration among clinicians, pharmacists, data scientists, and policymakers to establish a patient-centered,data-driven,trustworthy and controllable intelligent monitoring ecosystem for the medication safety of rare diseases.
To summarize the clinical therapeutic experience in managing Acinetobacter baumannii infection originating from a kidney donor.
A single-case analysis was conducted on the treatment of Acinetobacter baumannii infection derived from a kidney donor. During the ex vivo kidney preservation phase, the organ was continuously perfused with a tigecycline-containing solution at low temperature for approximately 10 hours, followed by transplantation. Postoperatively, systemic therapy with tigecycline combined with other antibiotics was continued for 6 days. During treatment, hepatic and renal function, serum amylase, lipase, and complete blood count (CBC) were monitored dynamically. Microbiological cultures of urine and drainage fluid were regularly repeated.
From postoperative day (POD) 3 onward, urine and drainage fluid cultures were repeatedly negative. Throughout the treatment period, the patient′s liver function tests, serum amylase, and lipase levels remained within normal limits. Blood urea nitrogen (BUN) and creatinine levels gradually returned to near-normal ranges. No significant drug-related adverse reactions were observed. The patient was successfully discharged on postoperative day 22.
The use of a tigecycline-containing perfusion solution during ex vivo kidney cold preservation, combined with a short-course of postoperative systemic anti-infective therapy, can safely and effectively eradicate donor-derived Acinetobacter baumannii.
This study aims to systematically analyze the current situation and causes of drug shortages in multi-level medical institutions in a city in Southern China,as well as their impact on clinical services,so as to provide a scientific basis and policy recommendations for optimizing drug supply chain management and establishing an early warning system for shortages.
Based on 1 272 drug shortage records reported by 109 medical institutions in the city between January 2023 and December 2024,descriptive statistical methods were employed to categorize and organize the data from the dimensions of institution level and pharmacological category. Data were independently collated and verified by two researchers, and Microsoft Excel was used for statistical analysis.
During the research period,a total of 1 272 drug shortage records were reported, involving 54 specific product specifications. Tertiary hospitals ranked the highest in all three aspects: proportion of institutions (34.87%),number of shortage reports (52.67%),and number of drug specifications involved (39). In terms of pharmacological categories, cardiovascular system drugs (31.45%),respiratory system drugs (16.04%),and nervous system drugs (15.96%) accounted for the highest proportions of shortage reports. The causes of drug shortages, with "other reasons" (28.4%),price factors (22.1%),and production discontinuation, output reduction or insufficient production capacity (13.8%) identified as the main contributing factors.
Drug shortages result from the combined effects of market failures, supply chain vulnerabilities, and policy constraints. It is urgent to establish a multi-party collaborative and systematically linked intervention system to improve the stability and accessibility of drug supply.
