Latest ArticlesTo investigate the effect of Mulberry twig (Sangzhi) alkaloids (SZ-A), which is a new traditional Chinese medicinal, on myocardial fibrosis in diabetes mellitus (DM) and its underlying molecular mechanism.
The diabetic mouse model was established using high-fat diet combined with streptozotocin injections. Mice were randomly divided into non-diabetic (ND), diabetic (DM), and SZ-A low- (50 mg/kg/d) or high-dose (100 mg/kg/d) groups. Myocardial fibrosis-related proteins (Collagen Ⅰ, TGF-β1, Smad2/3, and their phosphorylated forms) were analyzed by Western blot, while mRNA levels of fibrosis markers (Collagen Ⅰ/Ⅲ, α-SMA, Fn1, Postn) were quantified via qRT-PCR. Molecular docking simulations (AlphaFold2/Autodock) were performed to validate interactions between SZ-A components (DNJ, FGM, DAB) and TGF-β1.
SZ-A treatment significantly reduced myocardial Collagen deposition in diabetic mice, downregulated mRNA expression of Collagen Ⅰ/Ⅲ, α-SMA, Fn1, and Postn (P < 0.05), and suppressed TGF-β1 expression and Smad2/3 phosphorylation. Molecular docking revealed stable binding affinities between DNJ/FGM/DAB and TGF-β1. Exogenous TGF-β1 administration reversed SZ-A's anti-fibrotic effects, restoring Collagen Ⅰ, TGF-β1, and p-Smad2/3 levels (P <0.05).
SZ-A ameliorates diabetic myocardial fibrosis by targeting the TGF-β1/Smad signaling pathway, highlighting its therapeutic potential for diabetic cardiomyopathy.
To explore the intervention mechanism of icaritin (ICT) on myocardial injury in sepsis.
Mice were divided into control group, model group, AMPK activator (MET) group, icaritin (ICT) group and icaritin+AMPK inhibitor compound C (ICT+CC) group, with 12 mice in each group. Except for the control group, the other mice were intraperitoneally injected with LPS. ICT group was intraperitoneally injected with ICT 30 minutes before modeling, and ICT+CC group was injected with AMPK inhibitor Compound C at the same time. After 24 hours of modeling, cardiac function, myocardial pathological changes, AMPK pathway and pyroptosis-related protein expression, serum inflammatory factors and myocardial injury markers were detected.
The intervention of ICT and AMPK activators significantly improved the survival rate of mice, increased cardiac LVEF and LVFS, and decreased LVIDD and LVIDS. Tissue damage was reduced and inflammatory cell infiltration was reduced. The levels of serum TNF-α, IL-1β, IL-18, CK-MB and LDH were decreased. The ratio of p-AMPK/AMPK was increased, and the expression of NLRP3, ASC, Cleaved Caspase-1, GSDMD-NT and Cleaved IL-1β was decreased (P < 0.05). Compound C could reverse the improvement effect of ICT on myocardial injury, increase inflammation index and pyroptosis protein expression (P <0.05).
ICT can improve myocardial injury and inflammatory response in sepsis mice, and its mechanism may be related to the activation of AMPK to inhibit pyroptosis mediated by NLRP3 signaling pathway.
The theory of blood turbidity holds great significance in the syndrome differentiation and treatment of pulmonary nodules (PNs). The state of blood turbidity is a unique interpretation by traditional Chinese medicine of the microenvironment of PNs. Based on the theory of blood turbidity and combined with the dynamic evolution law of PNs, this article conducts an in-depth analysis of the syndrome differentiation and treatment of PNs. It is proposed that "the injury of collaterals by turbid blood" is the core pathogenesis of the occurrence and development of PNs. It is pointed out that the stagnation in collaterals is the pathological basis for the occurrence of PNs. The damage of collaterals by turbid blood is the pathological key to the progression of PNs. The blockage of collaterals by turbid toxin is the premise for the canceration of PNs. Therefore, in the treatment, it is advocated to start from the perspective of "the injury of collaterals by turbid blood", with the general principle of clearing and resolving blood turbidity and restoring the function of collaterals. Combining syndrome differentiation with stage differentiation, integrating the methods of relieving depression and regulating qi, assisting yang and restraining yin, resolving phlegm, promoting blood circulation, removing blood stasis, detoxifying, dredging collaterals, nourishing collaterals, and opening collaterals, which provides ideas for the treatment of PNs.
