Objective

To investigate the effects of Ginkgo biloba extract on the inflammatory response in the rats with ulcerative colitis (UC) based on the Toll-like receptor 4 (TLR4) /nuclear factor-κB (NF-κB) signaling pathway.

Methods

Forty SD rats were divided model group, Ginkgo biloba extract low-dose group, Ginkgo biloba extract high-dose group and normal group, each with 10 rats. The levels of interleukin (IL) -1 β, IL-6 and tumor necrosis factor (TNF) -α were measured using the Enzyme linked immunosorbent assay (ELISA) method. The expressions of TLR4 and NF-κB proteins in colon tissue was assessed via Western blot (WB). The Disease Activity Index (DAI), colon length, colon mucosal damage index (CMDI), inflammatory factors, TLR4 and NF-κB protein expressions were compared among the groups.

Result

The DAI score, colon mucosal damage index (CMDI) score, the protein gray values of IL-1β, IL-6, TNF-α, TLR4 and NF-κB in the model group, Ginkgo biloba extract low-dose group and Ginkgo biloba extract high-dose group were higher than those of the normal group, while the colon length was shorter than that of the normal group (P<0.05). The DAI score, CMDI score, the protein gray values of IL-1β, IL-6,TNF-α, TLR4 and NF-κB of the Ginkgo biloba extract low-dose and high-dose groups were lower than those of the model group, while the colon length was longer than that of the model group (P<0.05). The DAI score, CMDI score, the protein gray values of IL-1β, IL-6, TNF-α, TLR4 and NF-κB of the Ginkgo biloba extract high-dose group were lower than those of the Ginkgo biloba extract low-dose group, while the colon length was longer than that of the low-dose Ginkgo biloba extract group (P<0.05).

Conclusion

Ginkgo biloba extract can alleviate the inflammatory response in the UC rats, and its mechanism may be related to the regulation of TLR4/NF-κB signaling pathway expression.
