ArchiveTo investigate the effects of active liquid probiotics on the laxative efficacy and promotion of lead excretion in zebrafish.
Zebrafish at 5 dpf (days post fertilization) were stained with Nile Red and then divided into 3 groups, treated at 28 °C for 1 day, and their intestinal fluorescence intensity was analyzed. Zebrafish at 6 hpf (hours post fertilization) were used to establish a lead poisoning model, treated at 28 ℃ for 4 days, and then the lead content in vivo was determined.
The maximum tested concentration (MTC) of active liquid probiotics for laxative effect in zebrafish was 15.6 µL·mL-1. At this concentration, the intestinal fluorescence intensity of the probiotic group was significantly reduced (compared with the normal control group, P < 0.001). Similarly, for its effect on promoting lead excretion in zebrafish, the MTC was 7.81 µL·mL-1, and the lead content in the probiotic group at this concentration decreased by approximately 50% compared with the model group (P < 0.001).
Active liquid probiotics can rapidly colonize the zebrafish intestine and promote the proliferation of beneficial flora, thereby achieving laxative effects and regulating intestinal vitality. Active liquid probiotics can utilize their unique ion-binding capacity to specifically adsorb heavy metal lead for excretion, enhancing lead excretion.
To establish an isoelectric focusing (IEF) analysis method for the physicochemical testing of human basic fibroblast growth factor (hbFGF).
By comparing different IEF test conditions, the optimal method for determining the isoelectric point of hbFGF protein was screened, and the specificity, precision and durability of this method were methodologically verified.
The optimal system was the ampholyte system of Bio-Lyte and Ampholyte (2∶1). PBS buffer showed no interference with this method. The average pI value for repeatability was 10.0, with an RSD of 0.82% (≤3.0%). The average pI value for intermediate precision was 10.0, with a total RSD of 0.98% (≤6.0%). In terms of durability under different voltages (460, 480 and 500 V), the average pI value was 10.0, with an RSD of 1.3% (≤3.0%). In terms of durability under different electrophoresis times (70 min, 90 min, and 110 min), the average pI value was 10.0, with an RSD of 2.0% (≤3.0%).
The IEF method established for detecting the isoelectric point of hbFGF protein is accurate, durable and highly specific. It has high stability and can be used for the routine daily isoelectric point detection of hbFGF protein.
To predict the freezing point of the NaCl-NH4Cl-H2O ternary system using the semi-empirical hydration model (SEHM).
The freezing point of the ternary system was computed from the solution concentration based on the SEHM model, and compared with values obtained from air humidity cryoscopy and cooling curve cryoscopy (FPD-4A cryoscope).
The freezing points calculated by the SEHM model showed good agreement with those measured by air humidity cryoscopy and cooling curve cryoscopy. However, the cooling curve method was limited to dilute solutions due to the limited measurement range of the instrument.
The SEHM model provides a reliable approach for calculating the freezing point of the NaCl-NH4Cl-H2O ternary system based on solution concentration.

The aim of this study is to explore the bioinformatics-based screening of natural active ingredients for chronic atrophic gastritis (CAG) from the perspective of ferroptosis using machine learning and network pharmacology, thereby opening a new horizon for the treatment of CAG.
Datasets related to CAG were retrieved from the GEO database, and ferroptosis-related targets were collected from the FerrDb database. The R package GEO-query was used to obtain the expression profiles and clinical information of the CAG-related dataset GSE27411, followed by screening of differential targets and CAG differentially expressed targets were used to construct a protein-protein interaction (PPI) network, and the phenotypes of the key targets were analyzed using the VarElect tool. Support vector machine (SVM), a machine learning algorithm, was employed to further screen disease target genes. These natural active ingredients take the overlapping targets of ferroptosis and CAG as their potential action targets. Gene Ontology (GO) functional enrichment analysis and WikiPathwayss enrichment analysis were performed on the potential targets using the R package clusterProfiler. LASSO regression was utilized to screen natural active ingredients related to ferroptosis and CAG. The Coremine database was used to identify small-molecule drugs, and the TCMSP database was employed to find the natural drugs from which these small-molecule natural products are derived as well as their corresponding targets, followed by molecular docking analysis.
The GSE27411 dataset was selected for analysis. After differential expression analysis, 993 CAG-related differential targets were identified, along with 455 ferroptosis-related targets. A total of 29 overlapping targets were obtained through intersection analysis. Functional enrichment analysis revealed that these overlapping targets were associated with lipids and therosclerosis, reactive oxygen species (ROS), the RIG-I-like receptor signaling pathway, the FoxO signaling pathway, among others. Seven diagnostic genes were identified via LASSO-COX regression analysis: JUN, EGFR, BRD4, ATG5, BACH1, QSOX1, and NR1D2. Coremine identified several small-molecule drugs, including Apabetalone, cobaltiprotoporphyrin, sulmazole, 4-(4-fluorophenyl)-2-(4-hydroxyphenyl)-5-(4-pyridyl) imidazole, tin protoporphyrin IX, Chloroquine and Gefitinib, all of which exhibited favorable molecular docking result. Among these, cobaltiprotoporphyrin showed the optimal docking performance, suggesting its potential significance for CAG treatment.
Through bioinformatics approaches and drug regularity analysis, this study provides basic theoretical and data support for the treatment of CAG from the perspective of ferroptosis, and offers a new entry point for the search of therapeutic drugs targeting CAG.

To investigate a potential relationship between metabolic parameters and steady-state serum concentration of olanzapine (OLA) in patients treated with OLA combined with or without VAL in patients with mental disorders.
A retrospective analysis was performed on 225 inpatients with mental disorders who received OLA monotherapy or OLA-VAL combination therapy. The serum concentration of OLA was measured, and fasting blood samples were analyzed for insulin, glucose, total cholesterol (TC), triglycerides (TG), high-density lipoprotein cholesterol (HDL-c), low-density lipoprotein cholesterol (LDL-c), and uric acid (UA). The effects of OLA serum concentration, with or without VAL, on the aforementioned parameters and the correlation with these parameters were investigated.
VAL co-administration decreased OLA concentration and C/D ratio by 19.57% (P < 0.01) and 32.76%(P < 0.001), respectively.Patients taking OLA combined with VAL had higher weight (4.83%, P <0.01), BMI (4.07%, P < 0.05) and UA level (8.96%, P < 0.05), but lower glucose level (13.83%, P < 0.05), than those taking OLA alone. VAL co-administration had no significant effect on the levels of TC, HDL-c, LDL-c, TG and INS.The serum OLA concentration was negatively correlated with body weight in the OLA-VAL combination group (r =-0.199, P < 0.05), and there exists no correlation with other metabolic parameters.
There is no correlation between the serum OLA concentration and glycolipid metabolic parameters in the OLA monotherapy group, however, a negative correlation is found between serum OLA concentration and body weight in the OLA-VAL combination group; meanwhile, significantly higher body weight, BMI and UA levels are observed in this group.
