Objective

To investigate the role and mechanism of FABP5 in pulmonary inflammation associated with silicosis by analyzing multi-omics data in conjunction with a silicosis mouse model.

Methods

Single-cell sequencing was performed to analyze the potential role of FABP5 in occupational respiratory diseases; a silicosis mouse model was used to validate the relationship between FABP5 and pulmonary inflammation in silicosis; intra-abdominal injection of FABP5 inhibitors to modulate FABP5 function; and employed hematoxylin-eosin staining, immunohistochemistry, Western blotting, real-time quantitative PCR, and transcriptomic sequencing to investigate the role and potential mechanisms of FABP5 in silica-induced lung tissue inflammation in mice.

Results

Single-cell sequencing revealed that FABP5 expression was elevated by 3.10-fold (P<0.01) and 4.71-fold (P<0.05) in lung tissue from patients with pneumoconiosis and in a mouse model of silicosis, respectively, and was highly enriched in macrophages; Transcriptomic sequencing revealed that, compared to the silicosis model group, inhibition of FABP5 downregulated the expression of inflammatory genes such as TNF, IL-1α, and IL-1β, with the relative protein expression levels of TNF-α and IL-1β decreasing by 4.67-fold (P<0.001) and 0.25-fold (P<0.01), respectively.

Conclusion

FABP5 is a key pathogenic factor in silicosis-induced pulmonary inflammation. In vivo inhibition of FABP5 alleviates pulmonary tissue inflammation in silicosis, providing insights for research into the mechanisms and clinical management of silicosis.
