To investigate the difference of expression levels of IL-4 and IL-18 in the brain tissue of insomnia model rats with 'kidney failing to store willpower' Treated with the Processed Polygala tenuifolia.
A total of 40 male SD rats were randomly divided into normal group, model group, Esazolam group, Processed Polygala tenuifolia low-dose group and Processed Polygala tenuifolia high-dose group with 8 rats in each group. D-galactose (D-gal) combined with p-chlorophenylalanine (PCPA) was used to prepare the rats with 'kidney failing to store willpower'. After successful modeling, the western medicine group was given Esazolam 0.2mg/kg/d by gavage, and the low dose Chinese medicine group was given Processed Polygala tenuifolia 2.1g/kg/d by gavage, the high dose group was given 4.2g/kg/d gavage, and the normal group and model group were given equal volume normal saline gavage for 7 days. Morris water maze test was used to evaluate the learning and memory ability of rats. HE and Nissl staining were used to observe the cell morphology and structure of rat brain. The expression levels of IL-4 and IL-18 in rat brain were detected by IHC and Western blot.
Compared with normal group, incubation period and total motor distance of rats in model group were significantly increased (P <0.01), the percentage of exploration time in the target quadrant was significantly reduced (P <0.01). The neuronal nucleus was dislocated, the nucleolus was enlarged, the inflammatory cells were infiltrated, and the number of nissl was significantly decreased (P < 0.01). The intercellular spaces of neuron cells are widened, and the synaptic structure is incomplete, and the expression level of IL-4 was significantly decreased (P <0.01), the expression level of IL-18 was significantly increased (P < 0.01). Compared with model group, latency and total distance of movement of rats in low dose group and high dose group were significantly decreased (P < 0.01), the percentage of exploration time in the target quadrant significantly increased (P < 0.01). The neuronal damage and inflammatory cell infiltration were alleviated to varying degrees, and the number of nissl was increased to varying degrees (P < 0.01). The morphology and structure of neuronal nucleosomes were improved to varying degrees, and the expression level of IL-4 was significantly increased (P < 0.01), the expression level of IL-18 was significantly decreased (P <0.01).
The Processed Polygala tenuifolia can significantly improve the sleep, learning and memory ability of insomnia model rats with 'kidney failing to store willpower', and alleviate the inflammatory damage state of the neurons in rats with 'kidney failing to store willpower'. The mechanism of improving the sleep of the rats with insomnia model rats with 'kidney failing to store willpower' may be related to the increase of IL-4 and the decrease of IL-18 expression level in the brain tissue.
