Objective:

To optimize the extraction process of total flavonoids from Mayaqi and study its antioxidant activity and stability.

Method:

Key factors were screened by Plackett-Burman experiment, and the optimal process was obtained by Box-Behnken Design surface design; The antioxidant activity of total flavonoids of Mayaqi was evaluated by measuring the scavenging effects on hydroxyl radicals, DPPH radicals, ABTS radicals and the ability to reduce ferric ions. The correlation between the concentration of total flavonoids extract and different antioxidant activities was by SPSS software, and the effects of temperature, light and pH on the stability of antioxidant activity were investigated.

Result:

The optimal extraction technology for total flavonoids from Mayaqi was as follows: extraction 140 W, ethanol concentration 65%, temperature 70 ℃, extraction time 30 min, and the extraction rate of total flavoids was 4.608 5 ±0.031 7 mg/g. The scavenging effects of the total flavonoids extract on Mayaqi DPPH radicals, and ABTS radicals, and the ability to reduce ferric ions were 64.16%、55.67%、95.97% and 50.49%, of the same concentration of Vc, respectively. Correlation showed that the concentration of the total flavonoids extract from Mayaqi was significantly correlated with the scavenging rate of xyl radicals. The antioxidant stability test showed that the total flavonoids extract from Mayaqi should be stored at low (below 25 ℃), away from light, and in neutral conditions.

Conclusion:

This technology is stable and feasible, and the total flavonoids extract Mayaqi has high antioxidant activity.
