Objective

To study the effect of Huangqi (Astragali Radix) on Vitamin D receptor (VDR) expression of neutrophils in the psoriatic mice and the effect of neutrophil extrallular trapping network formation.

Methods

Healthy male BALB/c mice were selected, and the imiquimod cream was used to construct a mouse model of psoriasis. After successful modeling, the mice were divided into model group, Huangqi (Astragali Radix) low-dose, medium-dose and high-dose groups and vitamin D group according to the random number table method, and a blank control group was set up. HE staining was used to observe the histopathology of mouse skin lesions. Enzyme-linked immunosorbent assay (ELISA) was used to detect the expression levels of 25 (OH) D3and myeloperoxidase (MPO) in serum. Real-Time qPCR was used to detect the expression of VDR mRNA in peripheral blood neutrophils. Western blot was used to detect the expression of VDR protein in peripheral blood neutrophils. Immunofluorescence was used to detect the expression of MPO protein in skin lesions.

Results

Compared with the blank control group, the model group showed obvious pathological changes of psoriasis-like tissues. The levels of 25 (OH) D3in serum, VDR mRNA and VDR protein expressions in peripheral blood neutrophils were significantly decreased in the model group (P<0.05), and the levels of MPO in serum and MPO protein expression in skin lesions were significantly increased (P<0.05). Compared with those of the model group, the psoriasis-like histopathology of each drug intervention group was improved. The expression levels of 25 (OH) D3in serum of Huangqi (Astragali Radix) high-dose group and Vitamin D group and the expression levels of VDR mRNA and VDR protein in peripheral blood neutrophils of Huangqi (Astragali Radix) medium-dose group, Huangqi (Astragali Radix) high-dose group and Vitamin D group were significantly increased (P<0.05). The expression levels of MPO in serum and MPO protein in skin lesions of Huangqi (Astragali Radix) medium-dose group, Huangqi (Astragali Radix) high-dose group and Vitamin D group were significantly decreased (P<0.05).

Conclusion

Huangqi (Astragali Radix) may inhibit the formation of NETs by regulating VDR, thereby improving the inflammatory state and keratinocyte proliferation in psoriasis mice.

目的

研究黄芪对银屑病小鼠中性粒细胞维生素D受体（Vitamin D receptor，VDR）表达及中性粒细胞胞外诱捕网（Neutrophil extracellular traps，NETs）形成的影响。

方法

选取健康雄性BALB/c小鼠，选用咪喹莫特乳膏构建银屑病小鼠模型，造模成功后按照随机数字表法分为模型组、黄芪低剂量组、黄芪中剂量组、黄芪高剂量组、维生素D组，另设空白对照组。苏木素-伊红（HE）染色观察小鼠皮损组织病理，酶联免疫吸附试验（Enzyme-linked immunosorbent assay，ELISA）检测血清中25-羟基维生素D[25-Hydroxyvitamin D3，25（OH）D3]、髓过氧化物酶（Myeloperoxidase，MPO）表达水平，实时荧光聚合酶链反应（Real-time quantitative polymerase chain reaction，Real-Time qPCR）检测外周血中性粒细胞VDR mRNA表达，蛋白免疫印迹法（Western blot，WB）检测外周血中性粒细胞VDR蛋白表达，免疫荧光检测皮损组织MPO蛋白表达。

结果

与空白对照组相比，模型组可见明显银屑病样组织病理改变；模型组血清中25（OH）D3、外周血中性粒细胞VDR mRNA、VDR蛋白表达水平显著降低（P<0.05），血清中MPO及皮损组织中MPO蛋白表达水平显著升高（P<0.05）。与模型组相比，各药物干预组银屑病样组织病理改善；黄芪高剂量组、维生素D组血清中25（OH）D3与黄芪中剂量组、黄芪高剂量组、维生素D组外周血中性粒细胞VDR mRNA、VDR蛋白表达水平显著升高（P<0.05），黄芪中剂量组、黄芪高剂量组、维生素D组血清中MPO及皮损组织中MPO蛋白表达水平显著降低（P<0.05）。

结论

黄芪可能通过调控VDR，抑制NETs的形成，从而改善银屑病小鼠炎症状态及角质细胞增生。

目的

研究黄芪对银屑病小鼠中性粒细胞维生素D受体（Vitamin D receptor，VDR）表达及中性粒细胞胞外诱捕网（Neutrophil extracellular traps，NETs）形成的影响。

方法

选取健康雄性BALB/c小鼠，选用咪喹莫特乳膏构建银屑病小鼠模型，造模成功后按照随机数字表法分为模型组、黄芪低剂量组、黄芪中剂量组、黄芪高剂量组、维生素D组，另设空白对照组。苏木素-伊红（HE）染色观察小鼠皮损组织病理，酶联免疫吸附试验（Enzyme-linked immunosorbent assay，ELISA）检测血清中25-羟基维生素D[25-Hydroxyvitamin D3，25（OH）D3]、髓过氧化物酶（Myeloperoxidase，MPO）表达水平，实时荧光聚合酶链反应（Real-time quantitative polymerase chain reaction，Real-Time qPCR）检测外周血中性粒细胞VDR mRNA表达，蛋白免疫印迹法（Western blot，WB）检测外周血中性粒细胞VDR蛋白表达，免疫荧光检测皮损组织MPO蛋白表达。

结果

与空白对照组相比，模型组可见明显银屑病样组织病理改变；模型组血清中25（OH）D3、外周血中性粒细胞VDR mRNA、VDR蛋白表达水平显著降低（P<0.05），血清中MPO及皮损组织中MPO蛋白表达水平显著升高（P<0.05）。与模型组相比，各药物干预组银屑病样组织病理改善；黄芪高剂量组、维生素D组血清中25（OH）D3与黄芪中剂量组、黄芪高剂量组、维生素D组外周血中性粒细胞VDR mRNA、VDR蛋白表达水平显著升高（P<0.05），黄芪中剂量组、黄芪高剂量组、维生素D组血清中MPO及皮损组织中MPO蛋白表达水平显著降低（P<0.05）。

结论

黄芪可能通过调控VDR，抑制NETs的形成，从而改善银屑病小鼠炎症状态及角质细胞增生。

Objective

To study the effect of Huangqi (Astragali Radix) on Vitamin D receptor (VDR) expression of neutrophils in the psoriatic mice and the effect of neutrophil extrallular trapping network formation.

Methods

Healthy male BALB/c mice were selected, and the imiquimod cream was used to construct a mouse model of psoriasis. After successful modeling, the mice were divided into model group, Huangqi (Astragali Radix) low-dose, medium-dose and high-dose groups and vitamin D group according to the random number table method, and a blank control group was set up. HE staining was used to observe the histopathology of mouse skin lesions. Enzyme-linked immunosorbent assay (ELISA) was used to detect the expression levels of 25 (OH) D3and myeloperoxidase (MPO) in serum. Real-Time qPCR was used to detect the expression of VDR mRNA in peripheral blood neutrophils. Western blot was used to detect the expression of VDR protein in peripheral blood neutrophils. Immunofluorescence was used to detect the expression of MPO protein in skin lesions.

Results

Compared with the blank control group, the model group showed obvious pathological changes of psoriasis-like tissues. The levels of 25 (OH) D3in serum, VDR mRNA and VDR protein expressions in peripheral blood neutrophils were significantly decreased in the model group (P<0.05), and the levels of MPO in serum and MPO protein expression in skin lesions were significantly increased (P<0.05). Compared with those of the model group, the psoriasis-like histopathology of each drug intervention group was improved. The expression levels of 25 (OH) D3in serum of Huangqi (Astragali Radix) high-dose group and Vitamin D group and the expression levels of VDR mRNA and VDR protein in peripheral blood neutrophils of Huangqi (Astragali Radix) medium-dose group, Huangqi (Astragali Radix) high-dose group and Vitamin D group were significantly increased (P<0.05). The expression levels of MPO in serum and MPO protein in skin lesions of Huangqi (Astragali Radix) medium-dose group, Huangqi (Astragali Radix) high-dose group and Vitamin D group were significantly decreased (P<0.05).

Conclusion

Huangqi (Astragali Radix) may inhibit the formation of NETs by regulating VDR, thereby improving the inflammatory state and keratinocyte proliferation in psoriasis mice.
