Objective

To study the function of expelling water retention with drastic purgation of Jingdaji(Euphorbiae Pekinensis Radix,EPR)before and after being processed with vinegar on cancerous ascitesmodel rats.

Methods

Seventy-two SD male rats were random ly divided into 9 groups:the normal group,model group,positive group(furosemide),EPR and vinegar product groups at low,medium and high dose.Except the normal group,all other groups had intraperitoneal injection with Walker-256 ratascites cancer cells to replicate cancerous ascitesmodel rats.Positive group and each administration groups were orally administered with the corresponding powder,respectively.Normal group and model group were given the same amount of sodium carboxymethyl cellulose(0.5%)for 7 d.1 hour after the lastadministration,the amount of urine and ascites,and the contents of the renin-angiotensin-aldosterone system(RAAS)and antidiuretic hormones(ADH)in the serum were measured.Liver and kidney function indicators,oxidative damage indicators and pathological morphological changes in liver,stomach,intestine and kidney tissues were observed.

Results

Compared with the normal group,the model group showed a significant decrease in urine output and a significant increase in ascites volume(P<0.01),which indicated themodelwas successfully set up.The levels of plasma renin activity(PRA),angiotensinⅡ(AngⅡ),aldosterone(ALD),ADH,creatinine(CRE),blood urea nitrogen(BUN)and the activities of aspartate aminotransferase(AST),alanine aminotransferase(ALT)and malondialdehyde(MDA)in the serum and tissues all significantly increased(P<0.01),while the contents of superoxide dismutase(SOD)and glutathione(GSH)were significantly decreased(P<0.01),also there were obvious tissue damage in liver,stomach,intestine and kidney.Compared with those in themodel group,the urine volume in each administration groups increased,ascites volume,the levels of PRA,AngⅡ,ALD,ADH,CRE and BUN,the activities of AST and ALT and the content of MDA in the serum and tissues decreased,and the contents of SOD and GSH increased.The injury of each administration groups reduced,and there was no significant difference between EPR raw and vinegar-processed product powders.

Conclusion

EPR before and after processing with vinegar have the effects of expelling water retention with drastic purgation,by stimulating the RAAS to eliminate ascites and alleviate oxidative damage of cancerous as cites model rats.

目的

研究京大戟醋制前后对癌性腹水模型大鼠泻水逐饮功效及毒性的差异。

方法

72只雄性SD大鼠被随机分为9组:正常组、模型组、阳性组(呋塞米)、京大戟和京大戟醋品低、中、高剂量组。除正常组外,其他组均腹腔注射Walker-256大鼠腹水癌细胞株,建立癌性腹水大鼠模型。阳性组与京大戟各给药组每天进行粉末灌胃,正常组和模型组给予等量的羧甲基纤维素钠(0.5%),连续7 d。末次给药后1 h,测定其尿量和腹水量、血清中肾素-血管紧张素II-醛固酮系统(renin-angiotensin-aldosterone system,RAAS)和抗利尿激素(antidiuretic hormone,ADH)的含量,检测其肝肾功能指标和氧化损伤指标,观察肝、胃、肠、肾组织病理形态学变化。

结果

与正常组相比,模型组尿量显著减少、腹水量显著增多(P<0.01),表明造模成功;血清中血清肾素(plasma renin activity,PRA)、血管紧张素Ⅱ(angiotensinⅡ,AngⅡ)、醛固酮(aldosterone,ALD)、ADH、肌酐(creatinine,CRE)、尿素氮(blood urea nitrogen,BUN)的含量与血清和组织中的谷草氨酶(aspartate transaminase,AST)、谷丙转氨酶(alanine aminotransferase,ALT)活力,丙二醛(malondialdehyde,MDA)的含量均显著增加(P<0.01),超氧化物歧化酶(superoxide dismutase,SOD)、谷胱甘肽(glutathione,GSH)的含量显著减少(P<0.01),且肝、胃、肠、肾病理切片均有明显组织损伤。与模型组相比,各给药组尿量均有所提高,腹水量、血清中的PRA、AngⅡ、ALD、ADH、CRE、BUN含量与血清和组织中的AST、ALT活力,MDA含量均下降,SOD、GSH含量升高,各给药组大鼠组织损伤情况均有改善,且生、醋间无显著性差异。

结论

京大戟醋制前后都有泻水逐饮的作用,可通过激发肾素-血管紧张素Ⅱ-醛固酮系统消除腹水,醋制后可缓解癌性腹水模型大鼠的氧化损伤。

72只雄性SD大鼠被随机分为9组:正常组、模型组、阳性组(呋塞米)、京大戟和京大戟醋品低、中、高剂量组。除正常组外,其他组均腹腔注射Walker-256大鼠腹水癌细胞株,建立癌性腹水大鼠模型。阳性组与京大戟各给药组每天进行粉末灌胃,正常组和模型组给予等量的羧甲基纤维素钠(0.5%),连续7 d。末次给药后1 h,测定其尿量和腹水量、血清中肾素-血管紧张素II-醛固酮系统(renin-angiotensin-aldosterone system,RAAS)和抗利尿激素(antidiuretic hormone,ADH)的含量,检测其肝肾功能指标和氧化损伤指标,观察肝、胃、肠、肾组织病理形态学变化。

结果

与正常组相比,模型组尿量显著减少、腹水量显著增多(P<0.01),表明造模成功;血清中血清肾素(plasma renin activity,PRA)、血管紧张素Ⅱ(angiotensinⅡ,AngⅡ)、醛固酮(aldosterone,ALD)、ADH、肌酐(creatinine,CRE)、尿素氮(blood urea nitrogen,BUN)的含量与血清和组织中的谷草氨酶(aspartate transaminase,AST)、谷丙转氨酶(alanine aminotransferase,ALT)活力,丙二醛(malondialdehyde,MDA)的含量均显著增加(P<0.01),超氧化物歧化酶(superoxide dismutase,SOD)、谷胱甘肽(glutathione,GSH)的含量显著减少(P<0.01),且肝、胃、肠、肾病理切片均有明显组织损伤。与模型组相比,各给药组尿量均有所提高,腹水量、血清中的PRA、AngⅡ、ALD、ADH、CRE、BUN含量与血清和组织中的AST、ALT活力,MDA含量均下降,SOD、GSH含量升高,各给药组大鼠组织损伤情况均有改善,且生、醋间无显著性差异。

结论

京大戟醋制前后都有泻水逐饮的作用,可通过激发肾素-血管紧张素Ⅱ-醛固酮系统消除腹水,醋制后可缓解癌性腹水模型大鼠的氧化损伤。

Objective

To study the function of expelling water retention with drastic purgation of Jingdaji(Euphorbiae Pekinensis Radix,EPR)before and after being processed with vinegar on cancerous ascitesmodel rats.

Methods

Seventy-two SD male rats were random ly divided into 9 groups:the normal group,model group,positive group(furosemide),EPR and vinegar product groups at low,medium and high dose.Except the normal group,all other groups had intraperitoneal injection with Walker-256 ratascites cancer cells to replicate cancerous ascitesmodel rats.Positive group and each administration groups were orally administered with the corresponding powder,respectively.Normal group and model group were given the same amount of sodium carboxymethyl cellulose(0.5%)for 7 d.1 hour after the lastadministration,the amount of urine and ascites,and the contents of the renin-angiotensin-aldosterone system(RAAS)and antidiuretic hormones(ADH)in the serum were measured.Liver and kidney function indicators,oxidative damage indicators and pathological morphological changes in liver,stomach,intestine and kidney tissues were observed.

Results

Compared with the normal group,the model group showed a significant decrease in urine output and a significant increase in ascites volume(P<0.01),which indicated themodelwas successfully set up.The levels of plasma renin activity(PRA),angiotensinⅡ(AngⅡ),aldosterone(ALD),ADH,creatinine(CRE),blood urea nitrogen(BUN)and the activities of aspartate aminotransferase(AST),alanine aminotransferase(ALT)and malondialdehyde(MDA)in the serum and tissues all significantly increased(P<0.01),while the contents of superoxide dismutase(SOD)and glutathione(GSH)were significantly decreased(P<0.01),also there were obvious tissue damage in liver,stomach,intestine and kidney.Compared with those in themodel group,the urine volume in each administration groups increased,ascites volume,the levels of PRA,AngⅡ,ALD,ADH,CRE and BUN,the activities of AST and ALT and the content of MDA in the serum and tissues decreased,and the contents of SOD and GSH increased.The injury of each administration groups reduced,and there was no significant difference between EPR raw and vinegar-processed product powders.

Conclusion

EPR before and after processing with vinegar have the effects of expelling water retention with drastic purgation,by stimulating the RAAS to eliminate ascites and alleviate oxidative damage of cancerous as cites model rats.

癌性腹水归属于中医“臌胀”之范畴,病源是水运行受阻、蓄积于胸腹[1],具有量大顽固、反复出现的特点,且治疗困难、预后差,临床上虽已使用各种治疗策略,但均未取得满意效果。近年来研究发现甘遂[2]、京大戟[3]等单味中药和十枣汤[4]等中药复方在治疗腹水和改善患者生存状况方面确有疗效。京大戟作为峻下逐水代表药物,临床上用于治疗急慢性肾炎水肿、肝硬化腹水,尤其在治疗腹水方面有一定的优势[5]。但由于其具有峻烈的毒性,中医临床根据“有毒宜制”原则,内服常用醋京大戟[6]入药来减轻不良反应,并以《内经》中“有故无殒”指导思想进行临床应用。目前对京大戟的研究多集中在正常动物,发现其有明显的刺激性毒性,醋制后毒性均下降[57-8],但对其在病理状态下产生的泻水逐饮功效与毒性差异缺少研究,未能体现有毒中药临床应用“有故无殒”理论的内涵。故本实验采用Walker-256大鼠腹水癌细胞造模的癌性腹水模型大鼠作为研究对象,按照临床1、2、4倍剂量灌胃给予大鼠京大戟和醋品粉末,从肾素-血管紧张素Ⅱ-醛固酮系统(renin-angiotensin-aldosterone system,RAAS)、肝肾功能、组织病理形态学以及氧化损伤等方面探讨京大戟醋制前后对癌性腹水模型大鼠的泻水逐饮功效及肝胃肠肾毒性的差异,以期为京大戟临床的合理应用提供依据。
LLC-WRC 256(Walker 256大鼠腹水癌细胞株)(SXC0280)购自上海拜力生物科技有限公司。无特定病原体(SPF)的雄性健康SD大鼠、体质量180~220 g,共72只,购于南京市江宁区青龙山动物繁殖场,动物许可证号SCXK(苏)2020-0010。饲养期间保持空气流通和足够的光照,动物自由饮水及进食,实验前适应性饲养5 d。本实验获得南京中医药大学实验动物伦理委员会批准,编号:2019A015。
京大戟购于广西,经南京中医药大学乐巍副教授鉴定为大戟科植物大戟Euphorbia pekinensis Rupr的根,批号20170211。羧甲基纤维素钠(CMC-Na,国药集团化学试剂有限公司,批号:20101222),水合氯醛(德思特生物,批号:DST201108-212),呋塞米片(江苏亚邦爱普森药业有限公司,批号1912107),氯化钠(国药集团化学试剂有限公司,批号:20200811),4%多聚甲醛溶液(biosharp,批号:20206277),谷丙转氨酶(alanine aminotransferase,ALT)、谷草氨酶(aspartate transaminase,AST)、超氧化物歧化酶(superoxide dismutase,SOD)、丙二醛(malondialdehyde,MDA)、谷胱甘肽(glutathione,GSH)测定试剂盒(批号:20200518)均购自南京建成生物工程研究所。肾素(plasma renin activity,PRA)ELISA试剂盒,血管紧张素Ⅱ(angiotensinⅡ,Ang-Ⅱ)ELISA试剂盒,醛固酮(aldosterone,ALD)ELISA试剂盒,抗利尿激素(antidiuretic hormone,ADH)ELISA试剂盒(批号:202001110)均购自南京金益柏生物科技有限公司。
Allegra 64R Centrifuge超高速台式离心机(美国贝克曼库尔特公司),HH-S恒温水浴锅(江苏金怡仪器科技有限公司),FW80微型高速万能试样粉碎机(天津市泰斯特仪器有限公司),全自动酶标仪(美国Bio-Red公司),FA1104N电子分析天平(上海精密仪器厂),快速研磨仪器(上海净信实业发展有限公司),呼吸麻醉机(深圳瑞沃德生命科技有限公司),DM光学显微镜(德国Leica公司),GHP-9050恒温箱(上海精宏实验设备有限公司),HI1210摊片机、RM2245石蜡切片机、HI1220烘片机、ASP200s脱水机、EG1150包埋机(德国Leica公司),MILLI-Q DIRECT 8水纯化系统(美国Millipore公司)。
京大戟和京大戟醋品制备同课题组前期的研究[9],打粉过4号筛备用,临用前加0.5% CMC-Na溶解。
按体质量随机分为正常组和造模组,造模组参考有关文献研究复制癌性腹水模型[2],分为模型组、阳性组、京大戟低、中、高剂量组(简称生低组、生中组、生高组),京大戟醋品低、中、高剂量组(简称醋低组、醋中组、醋高组),每组8只,造模第2天开始给药。正常组与模型组给予0.5% CMC-Na,阳性组(呋塞米片研碎后,加0.5% CMC-Na制成0.42 mg·mL-1药液),低、中、高(25.3 mg·m L-1、50.6 mg·mL-1、101.2 mg·mL-1,按临床剂量的1、2、4倍换算)各剂量组均按1 mL/100 g灌胃给药京大戟和京大戟醋品粉末,连续灌胃7 d,1次/d。
实验期间观察各组大鼠毛色、活动状况及精神状态等一般生活状况。第4天给药后,将正常对照组、模型组以及模型给药组的大鼠分别置于代谢笼中,禁食不禁水。于第7天取血后称重,腹水抽取完毕后再次称重,通过计算体质量的差值间接测量腹水量。
第7天给药1 h后,各组大鼠使用呼吸麻醉机麻醉,将大鼠麻醉后开始颈总动脉取血,室温静置30 min后,3500 r/min离心10 min,取上清液即得大鼠血清,-80℃冻存,备用。照相应ELISA试剂盒说明书方法测定各组大鼠血清中PRA、AngⅡ、ALD、ADH的含量。
第7天各组大鼠最后一次给药1 h后麻醉,颈总动脉取血。静置30 min,待血清析出后,3500 r/min离心10 min,取上清液备用,按照试剂盒使用说明书,进行ALT与AST活力、CRE与BUN含量的测定。
取各组大鼠肝、胃、肠、肾组织0.2 g,加入预冷生理盐水匀浆,将匀浆液以3500 r/min离心10 min,取上清液即得10%各组织匀浆。取各组大鼠的血清及匀浆液,按照相应试剂盒的说明书测定肝、胃、肠、肾组织及血清中MDA、GSH的含量及SOD活力;取2%各组织匀浆按考马斯亮蓝(BCA)试剂盒说明书方法测定各组大鼠肝、胃、肠、肾组织中蛋白的含量。
取新鲜的肝、胃、肠、肾组织,用0.9%的生理盐水冲洗,并迅速投入固定于4%多聚甲醛溶液中。石蜡包埋、切片厚5 μm,苏木素(HE)染色,在光学显微镜下观察肝、胃、肠、肾的组织形态。
所测实验数据均以表示,采用GraphPad prism 7.0软件对所得数据进行分析,两组比较采用t检验,多组间比较采用单因素方差分析,数据方差不齐,不满足t检验条件时,采用Wilcoxon秩和检验,P<0.05表示两组数据之间的差异具有统计学意义。
正常组大鼠进食饮水正常,毛发光滑润泽,行动迅捷灵活;与正常组相比,模型组大鼠的进食量与饮水量显著减少,腹围逐渐增大,毛发暗淡粗糙伴有毛发直立,行动迟缓萎靡不振,部分大鼠还有便溏尿血的现象;与模型组比较,各给药组大鼠进食量有所增加,行动精神状态有所改善。
表1所示,与正常组比较,模型组的尿量显著减少,腹水量显著增多(P<0.01),表明造模成功。与模型组相比,生高组和醋高组的尿量有明显增多、腹水量明显减少(P<0.05),其余各组的尿量有所增多、腹水量有所减少,但生、醋组间差异均不具有统计学意义;这表明京大戟醋制前后可不同程度抑制腹水产生。
表2所示,与正常组相比,模型组各个激素含量均有显著性增多(P<0.01)。与模型组相比,各给药组的PRA、AngⅡ、ALD、ADH含量均明显减少(P<0.05,P<0.01),其中高剂量组作用较为显著,但生、醋组间差异均不具有统计学意义。以上结果说明京大戟醋制前后对血清肾素-血管紧张素Ⅱ-醛固酮系统和抗利尿激素都有一定调节作用,具有泻水逐饮的功效。
与正常组相比,模型组AST、ALT活力和CRE、BUN含量均有显著性增加(P<0.01),表明癌性腹水模型对大鼠肝肾损伤明显,见表3。与模型组相比,各给药组的AST活力均有所升高,ALT活力(除醋高组)均有所升高,但生、醋组间差异均不具有统计学意义;各给药组的CRE、BUN含量均有降低,且具有统计学差异(P<0.05,P<0.01)。此说明京大戟醋制前后对癌性腹水模型大鼠的肝肾损伤均有一定的缓解作用,对肾损伤的缓解作用更好。
表4所示,与正常组相比,模型组的血清和肝、胃、肠、肾组织中GSH含量显著减少(P<0.01)。与模型组相比,各给药组的血清和组织中的GSH含量均显著增加(P<0.05,P<0.01),但生、醋组间差异不具显著性统计学意义;且京大戟醋品各剂量组高于相应的京大戟各剂量组。
与正常组相比,模型组的血清和肠、肾中MDA含量显著性增加(P<0.01),肝和胃中MDA含量明显增加(P<0.05),见表5。与模型组相比,醋中/高组肠组织MDA含量有明显减少(P<0.05,P<0.01),其余各给药组的MDA含量均有所下降,但生、醋组间差异不具显著性统计学意义;且京大戟醋品各剂量组均低于相应的京大戟各剂量组。
表6所示,与正常对照组相比,模型组的血清和肝、肠、肾中SOD活力显著降低(P<0.01),胃中SOD活力明显降低(P<0.05)。与模型组相比,各给药组的SOD活力均升高(P<0.01,P<0.05),胃组织中差异不具有显著性统计学意义;且京大戟醋品各剂量组高于相应的京大戟各剂量组。
综合以上结果,模型组的SOD、GSH含量显著性下降,MDA含量显著性上升,且均具有统计学差异,说明腹水癌细胞对大鼠的组织器官产生了氧化损伤;与模型组相比,各给药组的SOD、GSH与MDA含量都有所下降,说明京大戟醋制前后均能改善Walker-256癌细胞所致大鼠的氧化损伤的作用。
如插页ⅩLⅣ图1所示,显微镜下观察各组大鼠的肝组织HE染色切片,正常组大鼠显微镜下可见中心静脉,肝细胞排列较规则,汇管区可见小胆管,未见明显异常;模型组大鼠可见肝细胞损伤缺失,炎症细胞浸润,肝血窦中可见红细胞;与模型组相比,各模型给药组大鼠的肝细胞及组织结构尚可,排列较整齐,生中、生高组局部可见炎症细胞浸润。
如插页ⅩLⅤ图2所示,在显微镜下观察各组大鼠胃组织HE染色切片,正常组大鼠胃黏膜结构完整,腺体排列紧密,未见明显异常;模型组大鼠胃黏膜上皮细胞严重脱落,胃腔内可见渗出。与模型组相比,各模型给药组的大鼠胃黏膜损伤情况均有改善。
如插页ⅩLⅥ图3所示,显微镜下观察各组大鼠肠组织HE染色切片,正常组大鼠肠黏膜绒毛结构完整,肠黏膜皱褶清晰可见;模型组大鼠可见炎性细胞全层浸润,黏膜完整性明显受损,炎症累积肌层使其疏松化,绒毛间隙增宽,部分黏膜出现糜烂;与模型组相比,各模型给药组大鼠的肠道损伤情况均有改善。
如插页ⅩLⅦ图4所示,显微镜下观察各组大鼠的HE染色切片,正常组大鼠肾小球及周围肾小管结构正常且清晰,无损伤现象;而模型组中肾小球囊腔萎缩,肾间质水肿,间质内炎细胞浸润;与模型组相比,各模型给药组大鼠肾损伤情况均有改善。
中医临床对于有毒中药的应用,常以《内经》中“有故无殒”思想作为指导,其强调的辨证施治是中医临床合理用药的指导原则。根据“有故无殒”的思想,没有病邪的存在,药性作用于机体就表现出毒性;而同样的药物临床用于病人,药性作用于病邪,表现出来的是治疗作用[10]。因此本研究采用制作方法简单、性质稳定周期短、造模成功率高的Walker-256乳腺癌细胞复制癌性腹水模型[11],该模型符合中医“痰饮”范畴,可模拟临床肿瘤腹水患者。
2020年版《中华人民共和国药典》记载京大戟以丸散入药,内服需醋制[6]。中药饮片粉末是中药饮片以粉末形态能够直接口服给药的重要形式[12],故本实验按临床1、2、4倍剂量[13]灌胃给予模型大鼠京大戟和醋品粉末,测定泻水逐饮效应相关指标,从整体水平研究京大戟醋制前后药效作用差异;观察其行为活动及组织病理形态学,检测肝肾功能指标,比较京大戟醋制前后对模型大鼠肝、胃、肠、肾毒性的影响;检测SOD、MDA和GSH指标,以探究京大戟醋制前后对癌性腹水模型大鼠氧化损伤的影响。
腹水增多会引起体内水盐代谢紊乱[14],肾素-血管紧张素-醛固酮系统(RAAS)作为调节水液代谢的重要内分泌系统则被激活[15],导致血液中PRA、AngⅡ、ALD含量增高,在RAAS系统中PRA是其引发物,PRA能够将血液中的血管紧张素原转化为Ang I,Ang I继转化为AngⅡ,后者能够促进ALD的合成与分泌,ALD能够调节水钠重吸收以维持水盐代谢的平衡,因此ALD升高会使肾脏对水的重吸收增多,尿液排出量减少,水液停留而生成腹水[216-17]。此外,AngⅡ还可以增加近端小管对钠的重吸收,并刺激ADH的释放[18],从而抑制尿液的生成排泄[16]。通过对实验结果分析可以发现,大鼠的尿量与腹水量呈负相关,相对于正常组,模型组的尿量显著性减少,腹水量显著性增加,差异均具有统计学意义,说明癌性腹水模型造模成功。相对于模型组,各给药组的尿量有所增加,腹水量、PRA、AngⅡ、ALD、ADH含量有所减少,高剂量比低剂量作用明显,且各给药组间差异无统计学意义,说明京大戟醋制前后均有一定的利尿作用且抑制腹水的生成,对RAAS系统和ADH有调节作用。
AST和ALT是肝细胞损害的敏感性指标[19],中药致肝细胞损伤时,ALT、AST转移进入血液[17]引起血清活性升高,激起炎症反应,进而使得体内清除自由基的酶系活力降低[20]。CRE和BUN是评价肾功能的重要指标[21],当肾脏功能受到损伤时,CRE和BUN不能及时排泄,导致水平增高[22]。众多研究表明,脂质过氧化反应参与了肝、胃、肠、肾损伤的致病机制[23-26],MDA作为氧化损伤的标志物,可以反映出组织中氧化损伤的程度[27],而SOD与GSH作为抗氧化酶,其活性水平则体现机体抵御氧化损伤的能力[28]。本实验结果显示与正常组相比,模型组的AST、ALT活力,MDA、CRE、BUN含量均有显著提高,SOD活力、GSH含量显著下降,且具有统计学差异,结合组织病理切片说明腹水模型对大鼠的肝胃肠肾组织器官产生了损伤。与模型组相比,各模型给药组的AST、ALT活力大都有所上升,SOD活力、GSH含量有所上升,MDA、CRE、BUN含量均下降。京大戟在给药后,能引起胃肠蠕动增加,刺激黏膜,分泌亢进,引起大便泻泄,再导致水泻,消除腹水,这种现象与临床基本相像。与模型组相比,各模型给药组血清中的CRE、BUN含量降低,表明其可影响肾脏排尿功能而引起水泻。结合组织病理切片可见京大戟各给药组对癌性腹水模型大鼠的器官损伤都有一定的缓解作用,且醋品的缓解作用优于京大戟,但两者之间差异无统计学意义。
本课题组前期研究发现,京大戟对正常大鼠肝、胃、肠毒性明显,对肾则无显著毒性,醋制后通过减轻其氧化损伤缓解毒性[29]。结合本文对模型动物的研究发现,京大戟在正常大鼠上表现出毒性作用,而在癌性腹水模型大鼠上的毒性作用并没有加重,反而改善了氧化损伤,且醋制前后都有泻水逐饮的作用,可通过激发肾素-血管紧张素Ⅱ-醛固酮系统消除腹水,表现出治疗作用。
