Objective

Taking the odor as the research object,this paper revealed the scientific connotation of the qualitative standard that“the drugwith strong odor has good quality”of Tianma(Gastrodiae Rhizoma).

Methods

The contents of9monomer components and polyphenols in 53 batches of Tianma(Gastrodiae Rhizoma)were determined by high performance liquid chromatography and Folin-Ciocalteu colorimetricmethod.The odor of the samples was quantified by electronic nose technology,and the correlation between the two was analyzed by bivariate correlation method.Taking the quantitative characteristic value of odor as the index,the qualitative identificationmodel of gas-based quality evaluation of differentgrades of Tianma(Gastrodiae Rhizoma)was established by using chemometrics method.

Results

Gastrodin,protocatechuic acid,4-hydroxybenzaldehyde,vanillin,parishin B and polyphenols were all positively correlated with the odor.Differentgrades of Tianma(Gastrodiae Rhizoma)can be distinguished by PCA and DFA models.

Conclusion

The stronger the odor of Tianma(Gastrodiae Rhizoma)is,the higher the contents of the above-mentioned pharmacodynamic components are,which reveals the traditional mechanism that“the drug with strong odor has good quality”of Tianma(Gastrodiae Rhizoma).The establishment of a qualitative identification model based on objective quantification of odor can be used for the identification of differentgrades of Tianma(Gastrodiae Rhizoma),which lays a foundation for the quality evaluation of Tianma(Gastrodiae Rhizoma).

目的

以气味为研究对象,揭示天麻“气浓者为佳”的辨状论质标准的科学内涵。

方法

采用高效液相色谱法与福林酚比色法分别测定53批天麻中9个单体成分及多酚的含量,并借助电子鼻技术定量样品气味值,采用双变量相关法分析二者之间的相关性。以气味的量化特征值为指标,采用化学计量学方法,建立不同等级天麻基于气的品质评价定性识别模型。

结果

天麻素、原儿茶酸、对羟基苯甲醛、香兰素、巴利森苷B、多酚均与天麻气味呈正相关,不同等级的天麻能够用PCA和DFA模型区分开。

结论

天麻的“气”越浓,则含有的上述药效成分的含量越高,从而揭示了天麻“气浓者为佳”的辨状论质机制,建立了基于气味客观量化识别不同等级天麻的定性模型,为天麻药材的质量评价奠定基础。

目的

以气味为研究对象,揭示天麻“气浓者为佳”的辨状论质标准的科学内涵。

方法

采用高效液相色谱法与福林酚比色法分别测定53批天麻中9个单体成分及多酚的含量,并借助电子鼻技术定量样品气味值,采用双变量相关法分析二者之间的相关性。以气味的量化特征值为指标,采用化学计量学方法,建立不同等级天麻基于气的品质评价定性识别模型。

结果

天麻素、原儿茶酸、对羟基苯甲醛、香兰素、巴利森苷B、多酚均与天麻气味呈正相关,不同等级的天麻能够用PCA和DFA模型区分开。

结论

天麻的“气”越浓,则含有的上述药效成分的含量越高,从而揭示了天麻“气浓者为佳”的辨状论质机制,建立了基于气味客观量化识别不同等级天麻的定性模型,为天麻药材的质量评价奠定基础。

Objective

Taking the odor as the research object,this paper revealed the scientific connotation of the qualitative standard that“the drugwith strong odor has good quality”of Tianma(Gastrodiae Rhizoma).

Methods

The contents of9monomer components and polyphenols in 53 batches of Tianma(Gastrodiae Rhizoma)were determined by high performance liquid chromatography and Folin-Ciocalteu colorimetricmethod.The odor of the samples was quantified by electronic nose technology,and the correlation between the two was analyzed by bivariate correlation method.Taking the quantitative characteristic value of odor as the index,the qualitative identificationmodel of gas-based quality evaluation of differentgrades of Tianma(Gastrodiae Rhizoma)was established by using chemometrics method.

Results

Gastrodin,protocatechuic acid,4-hydroxybenzaldehyde,vanillin,parishin B and polyphenols were all positively correlated with the odor.Differentgrades of Tianma(Gastrodiae Rhizoma)can be distinguished by PCA and DFA models.

Conclusion

The stronger the odor of Tianma(Gastrodiae Rhizoma)is,the higher the contents of the above-mentioned pharmacodynamic components are,which reveals the traditional mechanism that“the drug with strong odor has good quality”of Tianma(Gastrodiae Rhizoma).The establishment of a qualitative identification model based on objective quantification of odor can be used for the identification of differentgrades of Tianma(Gastrodiae Rhizoma),which lays a foundation for the quality evaluation of Tianma(Gastrodiae Rhizoma).

天麻为兰科植物天麻Gastrodia elata BL.的干燥块茎,具有祛风镇静、抗惊厥等功效[1-2],主产于云南、贵州、四川等地[3]。临床上常用于高血压病、心脑血管疾病的治疗[4-6]。天麻含有多种化学成分,其中酚类成分是天麻重要的药效成分且含量较高[7]。《七十六种药材商品规格标准》将商品天麻划分为以下等级:一等品为每千克26支以内,二等品为每千克46支以内,三等品为每千克90支以内,四等品为每千克90支以外[8],等级越高质量越差。目前药材市场存在各种规格等级较为混乱、质量参差不齐的问题,特别是市场流通的天麻粉末,采用性状等鉴别存在一定难度,为此亟待建立快速鉴别药材质量的方法。
谢宗万先生1994年首次提出中药“辨状论质”的概念[9],中药传统辨状论质标准“以……者为佳”“以……次之”是目前评价中药材质量的经典方法[10]。《中药商品学》中记载天麻的辨状论质标准为“以个大、色黄白、质坚实沉重、有鹦哥嘴、断面明亮、气浓者为佳”[11],天麻“以马尿味浓者为佳”是目前药材流通领域鉴定药材质量的常用方法,但迄今为止,依据气味判断质量的辨气论质机制尚未明确。
2024年,康廷国教授首次提出了“中药性状学”新概念[12],提出“性状学-化学-生物学”三维质量评价体系,为揭示中药辨状论质机制提供新的研究思路。基于此,本文针对天麻药材市场质量良莠不齐问题,采用人工智能感官定量方法——电子鼻技术,对53批不同等级天麻的气味进行客观量化,鉴定气味成分,并采用双变量相关法,将天麻气味的量化值与其化学成分进行相关性分析,从而揭示天麻“气浓者为佳”的科学内涵,同时建立不同等级天麻的定性识别模型。为天麻的质量评价奠定一定的基础。
AR2140型电子分析天平(上海奥豪斯公司);HeraclesⅡ型超快速气相色谱电子鼻(法国阿尔法莫斯公司);Agilent 1100高效液相色谱仪;UV-1750型紫外分光光度计(日本岛津);KQ-250D型数控超声波清洗器(昆山市超声仪器有限公司)。
没食子酸(批号Y19M8C36143)购于上海源叶生物科技有限公司;福林酚(北京索莱宝科技有限公司);乙腈、甲醇为色谱级,购于瑞典欧普森公司;甲酸、碳酸钙均购于天津市科密欧化学试剂有限公司;水,购于杭州娃哈哈集团有限公司。
共收集53批天麻药材,经辽宁中医药大学张慧教授鉴定为兰科植物天麻(Gastrodia elata BL.)的干燥块茎,具体见表1
称样量为1.5 g,检测器温度为270℃;样品在80℃下孵育30 min,以500μL/s的速度进样,进样口的温度为200℃,进样体积5000μL,样品采集时间为160 s;柱温为45℃,随后以0.5℃/s的速度升到70℃再以2℃/s的速度升到250℃;捕集阱温度为40℃,捕集20 s。
每个批次天麻样品平行测试3次。在超快速气相色谱电子鼻Heracles II上按照检测程序进行测定分析,样品通过弱极性和中等极性的色谱柱进行分离,得到气相色谱图。使用Alpha Soft V14.2软件分析,叠加天麻的气味指纹峰,见插页ⅩⅩⅥ图1。将不同等级天麻的气味指纹峰进行比较分析,其中云南不同等级天麻间色谱峰峰面积存在差异,且随等级的递增呈现递增的趋势。
53批天麻样品在极性和非极性色谱柱上的气味指纹峰较多,且具有一定的差异,利用Alpha Soft V14.2软件对其结果进行主成分分析(PCA),筛选出9个可区分53批样品的差异气味成分。用AroChemBase数据库进行定性分析,按气味量化值由小到大依次为庚醛、异丁醇、乙醇、2-甲基-1-戊醇、丙烯醛、正丙醇、(E)-3-己烯醛、(E,Z)-2,6-壬二烯、甲酸甲酯。9种差异气味成分的相对含量见表2
天麻素、对羟基苯甲醛、原儿茶酸、巴利森苷E、对羟基苯甲醇、香兰素、巴利森苷B、巴利森苷C、巴利森苷A为天麻的药效成分,具有神经保护、镇静镇痛、抗血小板聚集等作用。现采用本实验室前期建立的HPLC同时测定上述9种药效成分的方法[13],测定了53批天麻样品中各成分的含量,其色谱条件:色谱柱为Phenomenex Kinetex C18 100Å(250 mm×4.6 mm,5μm);流动相为乙腈(A)-0.1%甲酸水(B),梯度洗脱(0~5 min,0% ~3%A;5~10 min,3% ~3%A;10~20 min,3%~9%A;20~28 min,9% ~10%A;28~40 min,10% ~25%A;40~50 min,25% ~90%A;50~65 min,90% ~90%A;65~75 min,90%~100%A);流速1mL/min;进样量20μL;柱温25℃;检测波长270 nm[14],方法学考察结果表明该方法稳定可靠。结果 见表3
取10 mg的没食子酸配制为1.0 mg/mL的对照品溶液。
称取1.0 g天麻粉末,加入50%乙醇25 mL,超声提取30 min,取5 mL续滤液做为供试品溶液。
取0.1 mL供试品溶液于10 mL的具塞试管中,加水5 mL,混合均匀,加福林酚0.5 mL,充分混合;加1.5 mL 20%碳酸钠溶液,用水定容至10 mL,避光静置2 h,在752 nm处测定吸光度。
取样品溶液和空白对照溶液,按“2.3.3”方法,在400~900 nm范围内进行全波长扫描,由图2可知天麻供试品溶液的最大吸收波的波长是752 nm,且空白对照溶液无吸收,由此说明阴性溶液不会对供试品溶液造成干扰。
分别将没食子酸对照品溶液稀释至0.05、0.10、0.15、0.25、0.50 mg/mL,按“2.3.3”测定吸光度,以浓度为横坐标,752 nm处的吸光度值作为纵坐标来绘制标准曲线,得到线性回归方程Y=1.741 8X+0.017 9和R2 =0.998。说明在0.05~0.50 mg/mL的浓度范围内,没食子酸具有良好的线性关系。
精密移取天麻样品(S22)供试品溶液0.1 mL,按“2.3.3”测定吸光度,平行测定6次,含量RSD为0.27%,表明仪器精密度良好。
吸取0.1 mL的天麻样品(S22)供试品溶液,按“2.3.3”方法,在溶液配制后的0、0.5、1.0、1.5、2.0、2.5 h,测定752 nm吸光度,RSD值为2.12%,表明在2.5 h内,供试品溶液较稳定。
称取6份天麻样品(S22),每份1 g,配制为供试品溶液,分别测定吸光度,其含量的RSD值为1.75%,这证明该方法具有很好的重复性。
分别称取6份天麻样品(S22),每份0.5 g,再取6份没食子酸对照品,每份1.4 mg,混合后分别制成供试品溶液,用“2.3.3”的方法测定吸光度,平均回收率为99.22%,RSD为1.09%,说明此方法较为准确。
分别称取不同批次的天麻样品,制为供试品溶液,按“2.3.3”方法于752 nm处测定吸光度并记录对应的值,带入线性回归方程中计算出这53批样品中多酚的含量。结果 见表3
运用SPSS 24.0统计分析软件对9种差异性气味成分提取主成分,共得到4个主成分,总贡献率为84.953%,其中FAC1、FAC2、FAC3、FAC4 的贡献率分别为 29.143%、26.307%、17.009%、12.493%。采用主成分FAC1、FAC2、FAC3、FAC4与所测得的化学成分的含量进行双变量相关处理。相关分析结果见表4,主成分FAC1与多酚含量呈正相关;FAC2与巴利森苷E呈正相关,与巴利森苷C呈负相关;FAC3与天麻素、原儿茶酸、对羟基苯甲醛、香兰素、巴利森苷B、巴利森苷C、多酚的含量呈正相关,与巴利森苷E呈负相关;FAC4与巴利森苷E的含量呈负相关。而对羟基苯甲醇、巴利森苷A与4个主成分的双侧检验P值均大于0.05,与药材的气无相关性。
对天麻药材气的量化值进行主成分分析,以特征气味差异成分峰面积为变量,采用Alpha soft V14.2统计分析软件,对4种不同等级天麻药材进行分析,结果见图2(左)。不同等级天麻药材分别位于二维图的不同区域,且在PCA二维图上的区分指数为61,说明基于气味上的PCA分析能有效区分不同等级天麻样品。
对天麻药材气的量化值进行判别因子(DFA)分析,以特征气味差异成分峰面积为变量,采用Alpha soft V14.2统计分析软件,将4种等级天麻样品进行判别因子分析,结果见插页ⅩⅩⅥ图3(右),4种等级天麻药材分别位于DFA二维图的不同区域,且判别因子1和2之和为98.442%,说明建立的DFA模型能够用于天麻等级的鉴别。
53批不同产地和不同等级的天麻中,9种药效成分的含量具有一定差异,其中产地为陕西、贵州的天麻对羟基苯甲醇和天麻素的含量与其它产地相比较高,而四川产地的天麻巴利森苷类含量较高,表明四川、陕西、贵州的天麻质量较好,这与文献报道的相一致[15-18]
传统的辨状论质方法包括口尝、鼻闻等,但仅依靠人的感官鉴别药材品质,结果不够准确客观。近年来随着仿生技术的发展,电子鼻技术逐渐被应用到中药品质评价领域。通过电子智能感官技术,模拟人的嗅觉器官[19-23],将药材的气信号转化为电信号,其结果以客观定量的方式呈现出来,进而使传统的气味鉴别更具科学性和说服力。
近年来,有关天麻辨状论质相关研究,文献可见秦梦圆[24]以陕西汉中不同地区的天麻为研究对象,对其外观性状进行研究,分析其品质与产区的相关性;李平[25]通过将天麻“角质样”这一外观性状与多糖等化学成分的含量进行相关性分析,为天麻炮制工艺的研究提供一定依据。但有关不同等级的天麻辨状论质研究尚未见报道,因此本文基于气味对不同等级天麻的辨状论质机制进行研究并构建定性识别模型,为天麻的质量控制及不同等级的鉴别提供参考。
