Objective

To establish the infrared fingerprint spectra of standard decoctions of Xiangfu(Cyperi Rhizoma)and its processed products,and quickly identify the standard decoctions of Xiangfu(Cyperi Rhizoma)and its processed products.

Methods

Infrared fingerprint spectra were established for the decoctions of Xiangfu(Cyperi Rhizoma),wine-processed Xiangfu(Cyperi Rhizoma),vinegar-processed Xiangfu(Cyperi Rhizoma)and four-processed Xiangfu(Cyperi Rhizoma).The differences among the average infrared spectra of the decoctions of Xiangfu(Cyperi Rhizoma),wine-processed Xiangfu(Cyperi Rhizoma),vinegar-processed Xiangfu(Cyperi Rhizoma)and four-processed Xiangfu(Cyperi Rhizoma)were analyzed using OMNIC 9.2 software.The common peaks of Xiangfu(Cyperi Rhizoma)and its processed product decoctions were identified using Spectrum forWindow 3.02 software and the relative peak heights were calculated.The differences among the processed Xiangfu(Cyperi Rhizoma)decoctions were compared using principal component analysis,orthogonal partial least squares-discriminant analysis and clustering analysis.

Results

A total of 12 common peaks were identified and standardized for the decoctions of Xiangfu(Cyperi Rhizoma),vinegar-processed Xiangfu(Cyperi Rhizoma),wine-processed Xiangfu(Cyperi Rhizoma)and four-processed Xiangfu(Cyperi Rhizoma)using baseline correction and normalization.The differences were obvious in the 1700-1000 cm-1 waveband,and fourmain contribution wavebands were selected,including2929 cm-1,1053 cm-1,997 cm-1 and 1639 cm-1.The processed Xiangfu(Cyperi Rhizoma)decoctionswere all clustered into four categories through analysis.

Conclusion

There are obvious differences among the decoctions of Xiangfu(Cyperi Rhizoma),vinegar-processed Xiangfu(Cyperi Rhizoma),wine-processed Xiangfu(Cyperi Rhizoma)and four-processed Xiangfu(Cyperi Rhizoma),and they can be quickly identified using infrared spectroscopy.The method is fast,effective and accurate.

目的

建立香附及其炮制品标准汤剂红外指纹图谱,快速鉴别香附及其炮制品标准汤剂。

方法

建立香附、酒香附、醋香附、四制香附汤剂指纹图谱,利用OMNIC 9.2软件分析香附、酒香附、醋香附、四制香附汤剂的平均红外光谱间的差异,采用Spectrum for Window 3.02软件标定香附及其炮制香附汤剂的共有峰,计算相对峰高;通过主成分分析、正交偏最小二乘法-判别分析、聚类分析比较香附炮制品汤剂之间差异。

结果

通过基线校正、归一化处理对香附汤剂、醋香附汤剂、酒香附汤剂、四制香附汤剂共标定了12个共有峰,在1700~1000 cm-1波段处差异明显,筛选出2929、1053、997、1639 cm-1共4个波段为差异主要贡献波段,通过分析4种炮制品汤剂均聚为四类。

结论

香附汤剂、醋香附汤剂、酒香附汤剂、四制香附汤剂之间有明显差异,通过红外可以快速鉴别香附及其炮制品汤剂,所用方法快速有效,且结果准确。

目的

建立香附及其炮制品标准汤剂红外指纹图谱,快速鉴别香附及其炮制品标准汤剂。

方法

建立香附、酒香附、醋香附、四制香附汤剂指纹图谱,利用OMNIC 9.2软件分析香附、酒香附、醋香附、四制香附汤剂的平均红外光谱间的差异,采用Spectrum for Window 3.02软件标定香附及其炮制香附汤剂的共有峰,计算相对峰高;通过主成分分析、正交偏最小二乘法-判别分析、聚类分析比较香附炮制品汤剂之间差异。

结果

通过基线校正、归一化处理对香附汤剂、醋香附汤剂、酒香附汤剂、四制香附汤剂共标定了12个共有峰,在1700~1000 cm-1波段处差异明显,筛选出2929、1053、997、1639 cm-1共4个波段为差异主要贡献波段,通过分析4种炮制品汤剂均聚为四类。

结论

香附汤剂、醋香附汤剂、酒香附汤剂、四制香附汤剂之间有明显差异,通过红外可以快速鉴别香附及其炮制品汤剂,所用方法快速有效,且结果准确。

Objective

To establish the infrared fingerprint spectra of standard decoctions of Xiangfu(Cyperi Rhizoma)and its processed products,and quickly identify the standard decoctions of Xiangfu(Cyperi Rhizoma)and its processed products.

Methods

Infrared fingerprint spectra were established for the decoctions of Xiangfu(Cyperi Rhizoma),wine-processed Xiangfu(Cyperi Rhizoma),vinegar-processed Xiangfu(Cyperi Rhizoma)and four-processed Xiangfu(Cyperi Rhizoma).The differences among the average infrared spectra of the decoctions of Xiangfu(Cyperi Rhizoma),wine-processed Xiangfu(Cyperi Rhizoma),vinegar-processed Xiangfu(Cyperi Rhizoma)and four-processed Xiangfu(Cyperi Rhizoma)were analyzed using OMNIC 9.2 software.The common peaks of Xiangfu(Cyperi Rhizoma)and its processed product decoctions were identified using Spectrum forWindow 3.02 software and the relative peak heights were calculated.The differences among the processed Xiangfu(Cyperi Rhizoma)decoctions were compared using principal component analysis,orthogonal partial least squares-discriminant analysis and clustering analysis.

Results

A total of 12 common peaks were identified and standardized for the decoctions of Xiangfu(Cyperi Rhizoma),vinegar-processed Xiangfu(Cyperi Rhizoma),wine-processed Xiangfu(Cyperi Rhizoma)and four-processed Xiangfu(Cyperi Rhizoma)using baseline correction and normalization.The differences were obvious in the 1700-1000 cm-1 waveband,and fourmain contribution wavebands were selected,including2929 cm-1,1053 cm-1,997 cm-1 and 1639 cm-1.The processed Xiangfu(Cyperi Rhizoma)decoctionswere all clustered into four categories through analysis.

Conclusion

There are obvious differences among the decoctions of Xiangfu(Cyperi Rhizoma),vinegar-processed Xiangfu(Cyperi Rhizoma),wine-processed Xiangfu(Cyperi Rhizoma)and four-processed Xiangfu(Cyperi Rhizoma),and they can be quickly identified using infrared spectroscopy.The method is fast,effective and accurate.

香附,莎草科植物莎草Cyperus rotundus L.的干燥根茎,始载于《名医别录》[1],归肝、脾、三焦经。传统中医理论认为,香附具有行气解郁、调经止痛的功效[2],香附在中医学中具有生熟异治的作用[3],《本草纲目》[4]记载不同炮制品其功能主治也略有差别,酒制香附有行经络疏肝理气,止痛,消积化滞功效,临床用于饮食内停,心气痛,腹痛[5];四制香附有增强理气、理血、解郁止痛功效;醋制香附则有消积聚、理气止痛功效[6]。现代研究表明,香附有抗癌、抗抑郁、抗氧化、抑菌抗炎等药理作用[7-13]。临床上的香附炮品有醋制、炒制、蜜制、盐制、酒制、姜制、四制和七制等20多种[14]。2020版《中华人民共和国药典》仅收录醋香附,而在各省份的炮制规程中有醋制、酒制、四制、七制等,其中醋制和四制是应用最为广泛。
本实验采用傅里叶变换红外光谱(FTIR)鉴别香附及炮制品标准汤剂,傅里叶变换红外光谱具有快速、样品无损坏、绿色无污染、重复性高等优点[15-18]。通过红外光谱可以快速鉴别出香附及不同炮制品,比较不同炮制品之间的差异,分析香附及其炮制品红外光谱峰型、峰位、吸收强度等差异,利用化学计量学方法对香附及其炮制品进行差异分析,鉴别香附及其炮制品种类,从而可规范临床用药以及为不同炮制品制剂提供参考鉴定标准。
粉末压片机(型号:FW-4A,天津市拓扑仪器有限公司);傅里叶变换红外分光光度计(型号:Spectrum100,美国Pekin Elmer公司);电热恒温鼓风干燥箱(型号:101-3AB,北京中兴伟业仪器有限公司);溴化钾(光谱纯);无水乙醇(分析纯)。
香附药材采于2022年河南省焦作市,将药材火燎后直接晒干,切成厚度相同的片,即香附饮片,备用。
醋香附饮片、酒香附饮片、四制香附饮片制备参考乔璐等[19]、乔俊昊等[24]的炮制方法并稍作修改。
标准汤剂样品制备:取50 g样品,置于砂锅中,煎煮两次,分别加入7、6倍水,第一次煎煮前,浸泡30 min,武火煮沸腾后转文火煎煮25 min,第二次煎煮武火煮沸腾后转文火煎煮15 min,趁热过滤,合并滤液,减压浓缩,冷冻干燥,即得标准汤剂冻干粉末(编号与对应的饮片一致)[20-21]。样品信息见表1
取冻干样品粉末(过九号筛)适量于研钵中,加入光谱纯溴化钾,研磨均匀,取适量混合样品置于专用压片模具中,压成均匀半透明薄片,同法制备空白背景片。中红外光谱扫描范围为400~4000 cm-1,首先扫描空白样品片,扣除背景干扰,分辨率为8 cm-1,扫描6次/s,扫描速度0.2 cm/s[3]
按照“2.1”项采集各样品的红外光谱,由红外图谱可以看到不同样品光谱总体趋势基本相同,包括的特征峰和形状都差不多,但也有一些特征峰的强度比较明显,表现出了明显的差别,这可能是因为不同的炮制加工方式所引起的成分变化,结果见插页ⅩLⅧ图1
将香附标准汤剂、醋香附标准汤剂、酒香附标准汤剂、四制香附标准汤剂的红外光谱图分别进行处理后,得到红外光谱平均值图谱,结果见插页ⅩLⅧ图2
由插页ⅩLⅧ图2红外图谱中可知,羟基O-H所形成的伸缩振动峰在3388 cm-1波段处;由于亚甲基C-H2是不对称分子其所产生的振动伸缩吸收峰在2929 cm-1波段处;1700~1600 cm-1附近存在较强的吸收峰,可能为C=O和C=C的不对称伸缩振动产生;1300~1400 cm-1波段处为C-O-C的对称以及不对称伸缩振动所产生,可能为α,β-不饱和酸酯;糖、糖苷类等物质C-O键的伸缩振动吸收峰主要在1053、997、932 cm-1波段处[322]
由插页ⅩLⅨ图3可知,香附标准汤剂、酒香附标准汤剂、四制香附标准汤剂在1518、1420、1109 cm-1波段处存在新峰,香附标准汤剂、四制香附标准汤剂在1678、1375 cm-1波段处存在新峰,在1200~1300 cm-1波段处醋香附标准汤剂、酒香附标准汤剂为1263 cm-1的单峰,香附标准汤剂、四制香附标准汤剂为1291、1252 cm-1的“双峰”,四制香附标准汤剂在1030 cm-1波段处存在新峰。
以香附、醋香附、酒香附、四制香附标准汤剂的共有峰面积为基础,利用SPSS 22.0软件进行聚类分析,结果如图4,当组间距离为5时,香附样品聚为2类,香附(TY1~TY10)、酒香附(TJ1~TJ10)和四制香附(TS1~TS10)聚为一类,醋香附(TC1~TC10)单独聚为一类。说明通过标准汤剂的12个共有峰,可以区分不同香附及其炮制品。
将12个共有峰数据导入多元统计分析软件SIMCA 14.1进行主成分分析,得分矩阵R2X =0.994,Q2 =0.948,如插页ⅩLⅨ图5所示。从图中可以看出,香附样品可聚为4类,其中醋香附与酒香附汤剂距离较大,四制香附和香附汤剂之间距离较小,提示醋香附和酒香附在加工炮制的过程中成分含量变化较大,导致峰面积变化较大,而四制香附在炮制加工的过程中虽然伴随着成分含量的变化,但是在得分矩阵中四制香附炮制品间距离相较于香附饮片较近。
为更好挖掘出香附炮制品之间的差别,以各标准汤剂的共有峰的相对峰高为变量,通过正交偏最小二乘法-判别分析(OPLS-DA),结果数据矩阵的模型区分参数(R2Y=0.937)、解释率参数(R2X =0.988)、模型预测能力参数(Q2=0.915)均接近1,表明所建立的模型预测能力良好、稳定可靠[23]。40批样品可聚为4类,醋香附(TC1~TC10)、四制香附(TS1~TS10)、香附(TY1~TY10)、酒香附(TJ1~TJ10)分别单独聚为一类。结果 见插页ⅩLⅨ图6
通过200次置换实验,对已构建的OPLS-DA模型进行了验证,发现R2Q2均小于0.3,说明该模型是精确、可信的,能有效地避免过拟合,可为后续的标记筛选提供依据[3]。结果 见插页L图7
为进一步筛选出不同香附汤剂中的差异性成分,以VIP>1为作为筛选指标,筛选出不同香附汤剂间差异的吸收峰依次为2929、1053、997、1639 cm-1,表明这4个吸收峰在40批香附聚类分组中占有显著的重要性,可作为区分不同香附汤剂间差异标志物。结果 见图8
本实验采用傅里叶变换红外光谱快速鉴别香附及其炮制品标准汤剂,通过基线校正、归一化处理后,共标定了12个共有峰。通过图谱差异性比较在1518、1420、1109 cm-1波段处醋香附缺少相对应的峰位,香附标准汤剂、四制香附标准汤剂在1678、1375 cm-1波段处存在新峰,在1200~1300 cm-1波段处醋香附标准汤剂、酒香附标准汤剂为1263 cm-1的单峰,香附标准汤剂、四制香附标准汤剂为1291、1252 cm-1的“双峰”,四制香附标准汤剂在1030 cm-1波段处存在新峰。通过主成分分析(PCA)以及正交偏最小二乘法-判别分析(OPLS-DA)结果均显示40批香附样品可聚为4类,醋香附(TC1~TC10)、四制香附(TS1~TS10)、香附(TY1~TY10)、酒香附(TJ1~TJ10)分别单独聚为一类。表明香附标准汤剂、醋香附标准汤剂、酒香附标准汤剂、四制香附标准汤剂间存在差异。对各标准汤剂进行差异性评价,通过VIP法(VIP>1)筛选引起样品间差异的主要波段,筛选出2929、1053、997、1639 cm-1共4个波段为差异主要贡献波段,其中1053、997 cm-1波段处属于糖类成分的特征吸收峰,1639 cm-1波段处属于酸类成分的特征吸收峰,提示区分香附饮片标准汤剂、醋香附标准汤剂、酒香附标准汤剂、四制香附标准汤剂应重点关注糖类成分、酸类成分的变化[24]。红外光谱能够体现出药材整体化学物质的相关差异[25],红外光谱技术能够很好并快速区分香附以及各炮制品标准汤剂。
综上所述,本实验中香附及各炮制品标准汤剂间的红外光谱图差异明显,能够很好并快速对各标准汤剂进行鉴别分析,可为标准汤剂相关研究提供参考。
