Yanhusuo(Corydalis Rhizoma)is a commonly used medicine for promoting blood circulation and relieving pain,and is one of the“eight flavours of Zhejiang”,whose traditional local production areas are Dongyang and Pan’an in Zhejiang.Modern research shows that the main chemical components of Yanhusuo(Corydalis Rhizoma)include alkaloids,organic acids,nucleosides,steroids,etc.Alkaloids are the material basis for the pharmacological effects of Yanhusuo(Corydalis Rhizoma).Yanhusuo(Corydalis Rhizoma)has analgesic,sedative and other effects on regulating the central nervous system,as well as vasodilatory and anti myocardial is chemia effects on regulating the cardiovascular system.It also has various pharmacological activities such as anti-tumor,anti-inflammatory,hypoglycemic and lipid-lowering effects.Based on the theory of quality marker(QMarker)in traditional Chinese medicine,through the Q-marker prediction analysis of different dimensions of Yanhusuo(Corydalis Rhizoma),it is suggested that the quality markers of tetrahydropalmatine,d-corydaline,dehydrocorydaline,protopine,palmatine,berberine,canadine,bulbocapnine,yuanhunine and d-glaucine are suggested as the quality markers of Yanhusuo(Corydalis Rhizoma),which can be used for the provide scientific basis for the rationalevaluation of the quality of Yanhusuo(Corydalis Rhizoma).

延胡索为常用的活血止痛药,是“浙八味”之一,其传统道地产区为浙江东阳、磬安等地。现代研究表明,延胡索的主要化学成分包括生物碱类、有机酸类、核苷类、甾体类等,其中,生物碱类为延胡索药效的物质基础。延胡索具有镇痛、镇静等调节神经中枢系统的作用,有扩张血管、抗心肌缺血等调节心血管系统的作用,还有抗肿瘤、抗炎、降血糖、降血脂等多种药理活性。基于中药质量标志物(Quality marker,Q-Marker)理论,通过对延胡索不同维度的Q-marker预测分析,建议将延胡索乙素、延胡索甲素、去氢延胡索甲素、原阿片碱、巴马汀、小檗碱、四氢小檗碱、紫堇球碱、元胡宁、海罂粟碱等作为延胡索的质量标志物,为合理评价延胡索药材质量提供科学依据。

延胡索为常用的活血止痛药,是“浙八味”之一,其传统道地产区为浙江东阳、磬安等地。现代研究表明,延胡索的主要化学成分包括生物碱类、有机酸类、核苷类、甾体类等,其中,生物碱类为延胡索药效的物质基础。延胡索具有镇痛、镇静等调节神经中枢系统的作用,有扩张血管、抗心肌缺血等调节心血管系统的作用,还有抗肿瘤、抗炎、降血糖、降血脂等多种药理活性。基于中药质量标志物(Quality marker,Q-Marker)理论,通过对延胡索不同维度的Q-marker预测分析,建议将延胡索乙素、延胡索甲素、去氢延胡索甲素、原阿片碱、巴马汀、小檗碱、四氢小檗碱、紫堇球碱、元胡宁、海罂粟碱等作为延胡索的质量标志物,为合理评价延胡索药材质量提供科学依据。

Yanhusuo(Corydalis Rhizoma)is a commonly used medicine for promoting blood circulation and relieving pain,and is one of the“eight flavours of Zhejiang”,whose traditional local production areas are Dongyang and Pan’an in Zhejiang.Modern research shows that the main chemical components of Yanhusuo(Corydalis Rhizoma)include alkaloids,organic acids,nucleosides,steroids,etc.Alkaloids are the material basis for the pharmacological effects of Yanhusuo(Corydalis Rhizoma).Yanhusuo(Corydalis Rhizoma)has analgesic,sedative and other effects on regulating the central nervous system,as well as vasodilatory and anti myocardial is chemia effects on regulating the cardiovascular system.It also has various pharmacological activities such as anti-tumor,anti-inflammatory,hypoglycemic and lipid-lowering effects.Based on the theory of quality marker(QMarker)in traditional Chinese medicine,through the Q-marker prediction analysis of different dimensions of Yanhusuo(Corydalis Rhizoma),it is suggested that the quality markers of tetrahydropalmatine,d-corydaline,dehydrocorydaline,protopine,palmatine,berberine,canadine,bulbocapnine,yuanhunine and d-glaucine are suggested as the quality markers of Yanhusuo(Corydalis Rhizoma),which can be used for the provide scientific basis for the rationalevaluation of the quality of Yanhusuo(Corydalis Rhizoma).

延胡索(Corydalis rhizoma)为罂粟科植物延胡索Corydalis yanhusuo W.T.Wang的干燥块茎,别名元胡、玄胡、滴金卵等,是“浙八味”之一,其传统道地产区为浙江东阳、磬安等地[1]。延胡索归肝、脾经,具有活血、行气、止痛的功效,用于胸胁、脘腹疼痛,胸痹心痛,经闭痛经,产后瘀阻,跌打肿痛[2]。《景岳全书》记载:“延胡索善行滞气,破滞血,血中气药。故能止腹痛,通经,调月水淋滞,心气疼痛,破癥癖跌扑凝瘀。”
延胡索的主要成分为生物碱类,其中,叔胺碱、季胺碱为主要活性成分,也是其主要的镇痛成分[3]。延胡索乙素为《中华人民共和国药典》唯一收录的质量标志物(Quality marker,Q-Marker),随着延胡索化学成分、药理作用、临床应用的研究逐渐深入,更多的活性部位受到广泛研究,结合植物基原、炮制方法、药理作用、配伍用药等方面预测延胡索的质量标志物,以期为延胡索的质量评估提供参考。
延胡索中的化学成分主要有生物碱类、有机酸类、甾醇类、蒽醌类、核苷类等。
生物碱类是延胡索最主要的化学成分,其含量最高,种类最多,到目前为止,从延胡索中发现的生物碱已达50余种,见表1。延胡索中的生物碱主要为异喹啉生物碱,涉及原小檗碱类、阿朴菲类、原阿片类等[4]
张晓丽等首次于延胡索中分离得到山嵛酸、香草酸、对羟基苯甲酸[7],刘振华首次在延胡索中发现苹果酸、枸橼酸、软脂酸、硬脂酸、丁二酸、2-羟基丙酸、2,3-二羟基丙酸[9],延胡索中含有的有机酸类还有油酸、亚油酸、亚麻酸、延胡索酸[16]、反式-9-十八(碳)烯酸、2-哌啶甲酸等[19]
刘焱等[20]用高效液相色谱(High Performance Liquid Chromatography,HPLC)法在延胡索中分离出11种核苷类化合物,分别为腺嘌呤、胸腺嘧啶、尿嘧啶、肌苷、尿苷、鸟苷、胞苷、腺苷、2′-脱氧肌苷、2′-脱氧腺苷及2′-脱氧胸苷。
延胡索中甾体类化合物较少,文献报道目前仅4种甾体被分离鉴定出来,分别是豆甾醇、β-谷甾醇、胡萝卜苷[6]、麦角甾-4-烯-3-酮[21]
延胡索中含有三萜类化合物,3β-羟基-齐墩果烷-111,3(18)-二烯-28-酸,该化合物由石俊敏等首次分离得到[6]。还有蒽醌类化合物如大黄素、大黄素甲醚[9],以及糖类、甘油、磷酸、内酯、无机微量元素等其他成分[15-21]
延胡索具有行气止痛的作用,可用于治疗神经性疼痛、关节痛、内脏痛、痛经、癌痛等。延胡索发挥镇痛作用的主要活性成分为生物碱,其中,延胡索乙素的镇痛作用最强[22]。程连芝等[23]通过制备糖尿病性神经病理性疼痛大鼠模型,并诱导BV2小胶质细胞激活,建立糖尿病炎症损伤细胞模型,发现延胡索乙素可通过抑制p38丝裂原活化蛋白激酶(p38 Mitogen-activated protein kinase,p38 MAPK)通路显著缓解大鼠的疼痛症状。ZHOU等[24]发现延胡索总生物碱可显著缓解慢性缩窄损伤大鼠神经性疼痛,并抑制体外PC12神经元细胞、C6星形胶质细胞和BV2小胶质细胞的活化。王志成等[25]用延胡索总碱贴片于神经病理性疼痛大鼠模型腹部贴敷,结果显示,延胡索总碱能抑制脊髓中p38 MAPK、Toll样受体4(Toll-like receptor 4,TLR4)、核因子κB(Nuclear factor kappa-B,NF-κB)表达,并上调信号传导与转录激活因子3(Signal transducer and activator of transcription 3,STAT3)的表达从而发挥镇痛作用。研究表明,延胡索对痛风性关节炎有明显的抑制作用,且呈浓度依赖性[26]。临床上将血尿酸水平持续高于6.8 mg/dL定义为高尿酸血症,是痛风发病的重要病理基础,张旭等[27]发现延胡索提取物可降低高尿酸血症小鼠血清尿酸水平与氧化应激相关因子水平,降低痛风发生的可能。邵敬宝等[28]研究显示延胡索总生物碱对小鼠外周及中枢均有明显的镇痛作用,提高痛阈值,且效果优于延胡索乙素。另有研究发现,大剂量的延胡索配合癌痛三阶梯疗法,可改善中重度癌症患者的疼痛程度[29],延胡索经产地鲜切加工或传统加工均能对痛经发挥良好的治疗作用[30]
延胡索中的生物碱具有镇静、保护神经元的作用。王艳艳等[31]采用多平台水环境法剥夺大鼠快速眼动睡眠,考察左旋延胡索乙素对睡眠剥夺大鼠学习记忆的影响,结果显示,延胡索乙素可提高大鼠的认知能力,增加睡眠发生次数,延长睡眠时间,其药效维持时间与地西泮相比较短,且无宿醉现象。王婷婷等[32]制备肝郁型失眠小鼠模型,发现延胡索可显著延长小鼠睡眠时间,降低小鼠血清去甲肾上腺素与多巴胺水平,提高5-羟色胺水平,抑制下丘脑-垂体-肾上腺轴激活,从而发挥镇静催眠的作用。YAN等[33]发现延胡索中的原小檗碱型生物碱可改善阿尔茨海默病小鼠的焦虑样行为、空间和非空间识别以及记忆能力,调节神经递质水平。田冰等[34]采用双侧颈总动脉永久性结扎制备脑损伤大鼠模型,结果表明,延胡索总生物碱能改善大鼠的运动能力与认知功能,减轻神经元损伤,验证延胡索对脑神经元有保护作用。甘椿椿等[35]研究表明延胡索乙素对乳鼠的缺糖缺氧性海马神经元细胞损伤显示出良好的保护作用,其作用机制可能与抗氧化、抑制细胞凋亡有关。
阿片类药物成瘾是一种慢性复发性疾病,复发率高,具有强迫性用药症状与持续性药物渴求的特点。YUE等[36]研究显示延胡索乙素与多巴胺受体具有拮抗作用,可降低大鼠对海洛因的依赖性,抑制海洛因诱导的寻求行为恢复。余守洋等[37]建立对吗啡条件性位置偏爱大鼠模型,发现延胡索和延胡索乙素可通过下调大鼠海马与纹状体脑区的多巴胺递质含量,上调多巴胺转运体和多巴胺D2受体的表达,降低大鼠对吗啡的成瘾性,结果显示,1倍量的延胡索饮片相当于约14倍单独应用延胡索乙素的效果。另有研究表明[38],连续递增剂量给予吗啡可显著降低大鼠血浆及下丘脑中β-内啡肽含量,纹状体中亮氨酸脑啡肽含量,而左旋延胡索乙素可通过阻断多巴胺受体促进β-内啡肽的释放,促进大鼠自然戒断。
单胺假说作为广为接受的抑郁症发病假说,认为抑郁症是去甲肾上腺素和多巴胺等神经递质失衡所致[39]。研究显示延胡索乙素可降低创伤后应激障碍大鼠的焦虑指数,阻断下丘脑中神经肽Y的减少和促肾上腺皮质激素释放因子表达的增加,有显著的抗焦虑和抗抑郁作用[40]。另有研究发现,延胡索总生物碱处理后,异常水平的13种抑郁症生物标志物如肌醇、甘油、甘氨酸、肌酸等都显示出恢复到正常水平的趋势[41]。癫痫是常见的慢性神经系统疾病,由于脑部神经元间歇性的高同步化异常放电,导致癫痫长期反复的发作。延胡索总生物碱可显著延长癫痫大鼠的阵挛发作潜伏期,减少皮层神经细胞的凋亡,通过调控NOD样受体热蛋白结构域相关蛋白3(NOD-like receptor thermal protein domain associated protein 3,NLRP3)/含半胱氨酸的天冬氨酸蛋白水解酶-1(Cysteinyl aspartate specific proteinase-1,Caspase-1)/焦孔素家族蛋白D(Gasdermin-D,GSDMD)信号通路减少皮层组织炎症因子分泌,从而对癫痫大鼠起到治疗作用[42]
延胡索具有扩张冠脉血管、提高冠脉流量以及对心肌缺血、心肌梗死的保护作用。心脏冠状动脉血液灌注减少,心肌细胞供氧不足以致不能维持心肌正常机能,是心肌缺血的主要诱因,而心肌梗死是冠脉血流持续性供应不足引起的心肌缺血性损伤和心肌细胞坏死[43]。田冰等采用结扎左冠状动脉前降支法制备心肌缺血比格犬模型,结果显示,延胡索总生物碱可通过降低血压、升高心率、降低心肌酶的浓度来降低心肌缺血程度和梗死面积,防止血栓形成[44-45]。张萍等[46]发现延胡索提取物有明显的缩小心肌梗死面积、提高钠钾-ATP酶(Na+-K+-ATPase)、钙-ATP酶(Ca2+-ATPase)活性的作用,通过促进钠离子与钙离子交换,减轻细胞内钙超载,从而改善心肌缺血。延胡索乙素可抑制MAPK/细胞外调节蛋白激酶(Extracellular regulated protein kinases,ERK)通路激活,改善心肌肥厚及心室功能,对心肌梗死后心力衰竭大鼠的心肌损伤有保护作用[47]。临床研究证明[48],延胡索能降低微血管阻力指数、升高冠状动脉血流储备和血流储备分数,降低炎症指标C反应蛋白的水平,提示延胡索能改善心绞痛患者的冠状动脉微循环功能,进一步改善心肌缺血。DING等[49]研究表示延胡索乙素能通过抗氧化应激、抗内质网应激及抗炎多种作用,抑制TLR4-NF-κB信号通路,改善高脂血症引起的脂质代谢紊乱。延胡索还具有抗心律失常[50]、抑制血小板聚集[51]等作用。
现代药理研究表明延胡索可通过多种途径抑制肿瘤细胞生长,诱导细胞凋亡,从而抑制肿瘤细胞的增殖和转移。尹逊哲等[52]研究显示延胡索乙素能上调线粒体活性氧水平,重编程能量代谢表型,抑制线粒体呼吸,从而促进人肝细胞癌细胞凋亡。丁萌等[53]发现异紫堇碱能通过调控人绒毛膜促性腺激素18(Human leukocyte antigen complex group 18,HCG18)/小分子RNA 380-5p(MicroRNA 380-5p,miR-380-5p)通路从而抑制乳腺癌细胞的增殖、克隆、迁移及侵袭。去氢延胡索甲素能下调乳腺癌细胞的增殖、抗凋亡相关蛋白周期蛋白依赖性激酶1(Cyclin-dependent kinase 1,CDK1)、细胞周期蛋白D1(Cyclin D1,CCND1)、B细胞淋巴瘤(B-cell lymphoma-2,BCL2)和转移相关蛋白基质金属蛋白酶2(Matrix metalloproteinase-2,MMP2)、MMP9,上调促凋亡蛋白Caspase-3/8/9的表达来发挥抗癌作用[54]。另有研究证明[55],去氢延胡索甲素能显著阻断黑色素瘤细胞的增殖,导致细胞周期停滞在G0/G1期,下调黑色素瘤细胞中细胞周期调节因子CDK6和CCND1的表达,但对正常黑色素细胞的抑制作用很小,并通过失活MAPK信号抑制黑色素瘤细胞增殖、迁移和侵袭。张国铎等[56]发现延胡索总碱对6种人源胃癌细胞的增殖均有显著的抑制作用,且呈剂量依赖性,其机制可能与诱导细胞凋亡、将细胞阻滞在S期有关。王芳等[57]发现异紫堇定能增加人宫颈癌细胞种凋亡相关蛋白BCL2关联X蛋白(BCL2-associated X protein,Bax)、Caspase-3的表达,减少Bcl-2蛋白的表达,抑制宫颈癌细胞的增殖,且呈剂量-时间依赖关系。
延胡索具有良好的抗炎、抗氧化作用。杨彬等[58]发现延胡索提取物能抑制NLRP3炎症通路,进而抑制神经炎症因子的表达,保护神经根型颈椎病大鼠神经根组织。夏慧明等[59]研究显示延胡索总生物碱能降低溃疡性结肠炎小鼠血清中白细胞介素-6(Interleukin-6,IL-6)、肿瘤坏死因子α(Tumor necrosis factor-α,TNF-α)含量,减少小鼠肠道溃疡灶数量,缩小溃疡面积,调控MAPK通路抑制p38磷酸化发挥抗炎作用。盐酸巴马汀能降低人急性单核白血病细胞炎症模型中磷酸化核因子κB抑制蛋白α(p-NF-κB inhibitorα,p-IκBα)、p-c-Jun氨基末端激酶(c-Jun N-terminal kinase,JNK)、p-p38、Caspase-1 p20和NLRP3蛋白的表达,减少IL-1β和IL-18的释放,提示盐酸巴马汀的抗炎作用机制与NF-κB/p38 MAPK信号通路和NLRP3炎症小体信号通路相关[60]。马宁宁[61]等利用超高效液相色谱-四极杆飞行时间质谱(UPLC-Q-TOF/MS)建立延胡索提取物的指纹图谱,以NF-κB荧光素酶为标志对人体支气管上皮细胞进行抗炎活性实验,分析特征峰与抗炎作用的谱效关系,结果显示,延胡索有效抗炎成分主要为巴马汀、去氢延胡索甲素、小檗碱、二氢血根碱和黄连碱。
宋亚刚等[62]采用链脲佐菌素诱导糖尿病大鼠模型,发现延胡索乙素能降低血糖值,通过提高血清胰岛素、C肽、超氧化歧化物酶(Superoxide dismutase,SOD)、胰腺指数,降低大鼠体内炎症因子水平,抑制胰岛细胞凋亡,从而发挥降糖作用。延胡索乙素能以浓度依赖性方式抑制脂肪细胞前分化,下调脂肪生成相关蛋白和甾醇调节元件结合蛋白的基因表达,通过AMPK途径抑制3T3-L1脂肪细胞的脂肪生成[63]
延胡索含有多种有效成分,其种类与含量受基原、产地、采收加工方式的影响呈现一定的差异,若仅以延胡索乙素的含量作为评判延胡索质量的唯一指标,对建立延胡索质量控制体系的帮助有限。刘昌孝[64]院士首次提出中药质量标志物(Qmarker)的概念,是与中药的功能属性密切相关、能反映中药安全性和有效性的标志性物质。因此,通过对延胡索Q-marker进行预测,以期提出更科学的延胡索药材质量控制方法。
延胡索具有活血、行气、止痛的功效,在调节中枢神经系统与心血管系统方面尤为突出是临床常用的活血止痛药,用于血瘀气滞所致的身体各部位疼痛,现代研究表明,延胡索中以延胡索乙素为代表的生物碱类成分,是发挥活血止痛作用的关键性成分[2245]。其中,延胡索乙素镇痛作用最强,延胡索甲素、左旋四氢非洲防己胺次之,延胡索乙素、延胡索甲素、原阿片碱还具有显著的镇静催眠的功效,是调节中枢神经系统的主要成分[65]。延胡索总生物碱还能调节心血管系统,去氢延胡索甲素能抗动脉粥样硬化[66]、抑制血小板聚集[67],四氢小檗碱能改善血管内皮细胞的损伤[68],原阿片碱能扩张血管[69],药根碱能降糖、降血脂[70],小檗碱对神经系统、心血管系统均有较好的保护作用[71],高风等总结延胡索乙素、去氢延胡索甲素、延胡索甲素、四氢小檗碱、海罂粟碱等成分是舒张血管、抗血栓、抑制血小板聚集的主要成分[72]。以上成分是延胡索发挥“活血止痛”药效的物质基础,均可作为延胡索Q-marker预测的重要依据。
2020年版《中华人民共和国药典》收载的延胡索为罂粟科紫堇属植物延胡索的干燥块茎,全球紫堇属植物约428种,为北温带植物,以中亚和东亚最为集中,我国有298种,主要分布在我国西南部[73]。紫堇属植物在传统中医药中应用广泛,以延胡索、夏天无、紫堇、苦地丁等最为常用。目前从紫堇属中分离出的有效成分主要为异喹啉类生物碱,查阅文献对比延胡索与夏天无、紫堇、苦地丁的化学成分[74-76],发现紫堇球碱、异紫堇鳞茎碱、元胡宁等为延胡索所特有的成分,延胡索乙素、去氢延胡索甲素、延胡索甲素、小檗碱、巴马汀、原阿片碱等成分在不同植物中含量差异明显,可作为延胡索Q-marker的候选成分。
杨敏等[10]使用反相高效液相色谱(RP-HPLC)法同步测定了延胡索中11种生物碱成分的含量,陈俊等[13]采用一测多评法测定了延胡索中7种生物碱成分的含量,曹瑶等[77]对延胡索不同部位和不同生长时期的8种生物碱成分含量进行了动态测定与比较,王欢等[78]建立延胡索药材的HPLC指纹图谱测定了其中9种生物碱成分的含量,基于以上研究,原阿片碱、盐酸小檗碱、盐酸巴马汀、延胡索乙素、延胡索甲素、去氢延胡索甲素、巴马汀、海罂粟碱等成分均可被定量检测,可作为Q-marker预测的重要依据。
中药材加工炮制前后,其化学成分含量与药效会发生明显变化,延胡索常用的炮制方式为醋制,增强其理气止痛之功。徐容[79]等建立延胡索和醋延胡索的HPLC指纹图谱,对其进行含量测定,结果显示,原阿片碱和盐酸巴马汀含量降低,而盐酸小檗碱、去氢延胡索甲素、延胡索乙素和延胡索甲素含量均不同程度升高,可有效的将延胡索与其醋制品进行区分。杨勤等[80]使用HPLC法测定了8个产地延胡索鲜品与干品中6种生物碱的含量,结果显示,延胡索干品中的原阿片碱、盐酸巴马汀、盐酸黄连碱、盐酸小檗碱、去氢延胡索甲素含量均大于鲜品,而延胡索乙素含量小于鲜品。以上成分因延胡索炮制或干燥后含量发生明显变化,可作为延胡索Q-marker的候选成分。
中国植物志记载以“延胡索”命名的植物有20余种,全国有东北延胡索、全叶延胡索、齿瓣延胡索等超过15种延胡索类植物的干燥块茎在不同地区作为中药材延胡索使用[81]。目前延胡索的主要产地为浙江、江西、江苏、安徽等地,其中以浙江为道地产地。各产地延胡索中延胡索甲素、延胡索乙素含量差别较大,浙江地区产的延胡索中甲素、乙素含量显著高于其他省份,安徽其次,江苏、河南含量较低[82]。程井等[83]通过UPLC-MS/MS法同时测定不同产地的延胡索化学成分,发现延胡索中5种生物碱的含量在不同产地呈现一定的差异,原阿片碱、盐酸巴马汀、脱氢延胡索甲素、延胡索乙素和延胡索甲素在浙江产地的含量相对其它产地较高。以上成分含量可作为延胡索药材是否道地的重要标准,是Q-marker预测的重要依据。
延胡索常以组方的形式在临床上出现,最常用的为元胡止痛方,由延胡索、白芷组成,于定荣等[84]将延胡索、白芷按不同比例炮制配伍,比较不同配比下延胡索乙素的含量,研究发现,延胡索在醋炙以及与白芷(1∶1、2∶1)配伍过程中,延胡索乙素质量分数显著增加,但醋炙延胡索-白芷(3∶1)配伍反而引起延胡索乙素质量分数显著降低,延胡索与白芷经炮制配伍后,其镇痛作用优于延胡索或醋炙延胡索单用,表明延胡索与白芷在炮制配伍中产生了协同增效的作用。另有研究发现,延胡索与白芷在一定配比范围内,延胡索总生物碱的溶出量、延胡索乙素的溶出量、延胡索乙素的小肠吸收均呈现显著的增加趋势[85]。金铃子散中延胡索与金铃子1∶1配伍,其有效成分延胡索乙素、黄连碱、紫堇碱、巴马汀和去氢延胡索甲素的含量可被稳定地测量[86],蒲元和胃胶囊中延胡索与香附、醋乳香、蒲公英、枯矾、甘草配伍,延胡索乙素、盐酸巴马汀可作为质量控制标志性成分[87],元胡止痛胶囊中延胡索乙素、巴马汀、去氢延胡索甲素、延胡索甲素、欧前胡素及异欧前胡素的含量,可用于元胡止痛胶囊中质量控制标志性成分[88]。以上成分含量在不同的配比条件下或与不同药物配伍时能稳定测定,是Qmarker预测的重要依据。
延胡索是常用的活血止痛类药材,有显著的止痛作用,同时还有镇静、抗抑郁、抗癫痫、抗阿片类药物成瘾等调节神经中枢系统的作用,有突出的扩张血管、抗心肌缺血、抗动脉粥样硬化、抑制血小板聚集等调节心血管系统的作用,还有抗肿瘤、抗炎、降血糖、降血脂等作用,具有广阔的药用前景。延胡索在我国有丰富的药植资源,受产地、采收、加工、炮制等因素的影响,导致质量良莠不齐的现象,因此,建立更加完善、科学、体系的质量评价方法,对延胡索质量控制和资源合理应用有着重要的现实意义。
延胡索的主要成分为生物碱类,基于对其化学成分和药理作用的综述,以及从传统药效、植物亲缘、化学成分特有性与可测性、产地加工和配伍环境等不同维度的Q-marker预测分析,建议将延胡索乙素、延胡索甲素、去氢延胡索甲素、原阿片碱、巴马汀、小檗碱、四氢小檗碱、紫堇球碱、元胡宁、海罂粟碱等作为延胡索的质量标志物,为合理评价延胡索药材质量提供科学依据。
