Objective

To investigate the impact of Schisandrol A(SA)on lipopolysaccharide(LPS)-induced and differentiated small intestinal epithelial cell damage in human colorectal cancer Caco-2 cells.

Methods

Caco-2 cells were random ly divided into a normal control group(without drug intervention,Con),a model group(0.7 mg·mL-1,LPS)and SA(2.5,5,10 μmol·L-1)dose groups.The cell viability was assessed by the CCK-8 method.The cell apoptosis,cell cycle,mitochondrial membrane potential and intracellular reactive oxygen species(ROS)were detected by flow cytometry.The levels of inflammatory factors in the cell supernatant were measured using ELISA.Oxidative stress was assessed using an enzyme-labeled method.The expressions of proteins related to the NF-κB signaling pathway and apoptosis-related pathways were determined by Western Blot analysis.

Results

The findings indicated that compared with the LPS group,SA at concentrations between 2.5-10μmol·L-1 exhibited a significant protective effect on cells,exhibiting a certain dose-dependent relationship(P<0.01,P<0.001).It reduced the contents of malondialdehyde(MDA),lactate dehydrogenase(LDH)and ROS(P<0.05,P<0.01,P<0.001),increased the activities of superoxide dismutase(SOD)and GSH(P<0.01,P<0.001),inhibited the secretion of pro-inflammatory factors tumor necrosis factor-α(TNF-α),interleukin-6IL-6 and interleukin-18(IL-18)(P<0.05,P<0.01,P<0.001),promoted the release of anti-inflammatory factor interleukin-10(IL-10)(P<0.001),increased mitochondrial membrane potential(P<0.05,P<0.01),improved cell cycle arrest(P<0.05)and modulated the expression levels of inflammation and apoptosis-related pathway proteins(P<0.05,P<0.01,P<0.001).

Conclusion

This study has confirmed in vitro that SA is capable of improving LPS-induced damage to Caco-2 cells.The mechanism of action appears to be related to the inhibition of the NF-κB signaling pathway.

目的

探究五味子醇甲(Schisandrol A,SA)对脂多糖(lipopolysaccharide,LPS)诱导与分化小肠上皮细胞类似的人结直肠癌Caco-2细胞损伤的影响。

方法

将Caco-2细胞随机分为正常对照(不受药物干预,Con)组,模型(0.7 mg·mL-1,LPS)组、SA(2.5、5、10μmol·L-1)剂量组。CCK-8法检测细胞活力;流式细胞术检测细胞凋亡、细胞周期、线粒体膜电位和细胞内活性氧(reactive oxygen species,ROS)水平;ELISA检测细胞上清中炎症因子的含量;酶标法检测氧化应激水平;Western Blot检测NF-κB信号通路蛋白表达和凋亡相关通路蛋白表达。

结果

与LPS组相比,SA在2.5~10μmol·L-1范围内对细胞有明显的保护作用且呈一定剂量依赖性(P<0.01,P<0.001),其减少了丙二醛(malondialdehyde,MDA)、乳酸脱氢酶(lactate dehydrogenase,LDH)及ROS的含量(P<0.05、P<0.01、P<0.001),增加了超氧化物歧化酶(superoxide dismutase,SOD)和谷胱甘肽(glutathione,GSH)的活性(P<0.01、P<0.001),抑制了促炎因子[肿瘤坏死因子-α(tumor necrosis factor-α,TNF-α)、白细胞介素-6(interleukin-6,IL-6)、白细胞介素-18(interleukin-18,IL-18)]的分泌(P<0.05、P<0.01、P<0.001),促进了抗炎因子白细胞介素-10(interleukin-6,IL-10)的释放(P<0.001),升高了线粒体膜电位(P<0.05、P<0.01),改善了细胞周期的阻滞(P<0.05),同时调节了凋亡通路与NF-κB通路相关蛋白的表达水平(P<0.05、P<0.01、P<0.001)。

结论

通过体外实验证实了五味子醇甲能够改善LPS诱导的Caco-2细胞损伤,其作用机制可能与抑制NF-κB信号通路相关。

目的

探究五味子醇甲(Schisandrol A,SA)对脂多糖(lipopolysaccharide,LPS)诱导与分化小肠上皮细胞类似的人结直肠癌Caco-2细胞损伤的影响。

方法

将Caco-2细胞随机分为正常对照(不受药物干预,Con)组,模型(0.7 mg·mL-1,LPS)组、SA(2.5、5、10μmol·L-1)剂量组。CCK-8法检测细胞活力;流式细胞术检测细胞凋亡、细胞周期、线粒体膜电位和细胞内活性氧(reactive oxygen species,ROS)水平;ELISA检测细胞上清中炎症因子的含量;酶标法检测氧化应激水平;Western Blot检测NF-κB信号通路蛋白表达和凋亡相关通路蛋白表达。

结果

与LPS组相比,SA在2.5~10μmol·L-1范围内对细胞有明显的保护作用且呈一定剂量依赖性(P<0.01,P<0.001),其减少了丙二醛(malondialdehyde,MDA)、乳酸脱氢酶(lactate dehydrogenase,LDH)及ROS的含量(P<0.05、P<0.01、P<0.001),增加了超氧化物歧化酶(superoxide dismutase,SOD)和谷胱甘肽(glutathione,GSH)的活性(P<0.01、P<0.001),抑制了促炎因子[肿瘤坏死因子-α(tumor necrosis factor-α,TNF-α)、白细胞介素-6(interleukin-6,IL-6)、白细胞介素-18(interleukin-18,IL-18)]的分泌(P<0.05、P<0.01、P<0.001),促进了抗炎因子白细胞介素-10(interleukin-6,IL-10)的释放(P<0.001),升高了线粒体膜电位(P<0.05、P<0.01),改善了细胞周期的阻滞(P<0.05),同时调节了凋亡通路与NF-κB通路相关蛋白的表达水平(P<0.05、P<0.01、P<0.001)。

结论

通过体外实验证实了五味子醇甲能够改善LPS诱导的Caco-2细胞损伤,其作用机制可能与抑制NF-κB信号通路相关。

Objective

To investigate the impact of Schisandrol A(SA)on lipopolysaccharide(LPS)-induced and differentiated small intestinal epithelial cell damage in human colorectal cancer Caco-2 cells.

Methods

Caco-2 cells were random ly divided into a normal control group(without drug intervention,Con),a model group(0.7 mg·mL-1,LPS)and SA(2.5,5,10 μmol·L-1)dose groups.The cell viability was assessed by the CCK-8 method.The cell apoptosis,cell cycle,mitochondrial membrane potential and intracellular reactive oxygen species(ROS)were detected by flow cytometry.The levels of inflammatory factors in the cell supernatant were measured using ELISA.Oxidative stress was assessed using an enzyme-labeled method.The expressions of proteins related to the NF-κB signaling pathway and apoptosis-related pathways were determined by Western Blot analysis.

Results

The findings indicated that compared with the LPS group,SA at concentrations between 2.5-10μmol·L-1 exhibited a significant protective effect on cells,exhibiting a certain dose-dependent relationship(P<0.01,P<0.001).It reduced the contents of malondialdehyde(MDA),lactate dehydrogenase(LDH)and ROS(P<0.05,P<0.01,P<0.001),increased the activities of superoxide dismutase(SOD)and GSH(P<0.01,P<0.001),inhibited the secretion of pro-inflammatory factors tumor necrosis factor-α(TNF-α),interleukin-6IL-6 and interleukin-18(IL-18)(P<0.05,P<0.01,P<0.001),promoted the release of anti-inflammatory factor interleukin-10(IL-10)(P<0.001),increased mitochondrial membrane potential(P<0.05,P<0.01),improved cell cycle arrest(P<0.05)and modulated the expression levels of inflammation and apoptosis-related pathway proteins(P<0.05,P<0.01,P<0.001).

Conclusion

This study has confirmed in vitro that SA is capable of improving LPS-induced damage to Caco-2 cells.The mechanism of action appears to be related to the inhibition of the NF-κB signaling pathway.

五味子Schisandra chinensis(Turcz).Baill为木兰科植物五味子的干燥成熟果实。《神农本草经》与《本草备要》中记载,五味子具有补肾宁心、收敛固涩之功效[1-2]。五味子醇甲(Schisandrol A,SA)为五味子的标志性成分[3]。现代药理研究表明,五味子能够改善肠道损伤。其中,五味子提取物通过抗炎、抗氧化、抗凋亡等途径改善肠道损伤的作用近年来尤为受重视[4-7]
脂多糖(lipopolysaccharides,LPS)是革兰氏阴性菌内壁的主要成分,入血后会产生毒性,是一种常见的强效诱导炎症、致凋亡的造模药物。LPS诱导的模型是评价肠道损伤的经典实验模型[8]。人结直肠腺癌细胞(Caco-2),其结构和功能与分化的小肠上皮细胞类似,常用来进行模拟体内肠功能的细胞实验[9]
课题组前期研究表明,五味子醇甲能够保护肠道组织性状,恢复菌群平衡,从而缓解小鼠的腹泻症状[10]。但其对肠上皮损伤的影响和作用机制还不清楚。本研究利用LPS诱导Caco-2细胞建立模拟肠上皮细胞损伤模型,探讨五味子醇甲改善肠上皮损伤的作用及机制,为五味子提取物改善肠道健康产品的研发提供数据基础和指导。

生物安全柜(美国Thermo公司,型号MSC Advantage 1.8);CO2 恒温培养箱(美国Thermo公司,型号HERAcell 240i);全波长酶标仪(美国Thermo公司,型号1510);流式细胞仪(美国贝克曼库尔特公司,型号A00-1-1102);高速冷冻离心机(北京大龙兴创实验仪器公司,型号D1524R);电泳仪(上海天能科技有限公司,型号EPS 300);近红外荧光分子成像仪(美国Azure公司,c500)。
Caco-2细胞采用MEM with NEAA培养基(含20%FBS)于5%CO2、37℃的培养箱中培养,待细胞贴壁且生长至对数生长期时进行后续实验。
不同质量浓度SA对Caco-2细胞活性的影响:将细胞随机分为12组,除正常对照(Con)组外,分别用浓度为0.5、1、2、2.5、5、7.5、10、20、25μmol·L-1的SA与细胞共培养24 h。
LPS对Caco-2细胞活性的影响:将细胞随机分为10组,除正常对照(Con)组外,分别用浓度为0.1、0.2、0.3、0.4、0.5、0.6、0.7、0.8、0.9、1 mg·mL-1的LPS处理细胞24 h。
SA剂量依赖性保护作用:将细胞随机分为7组,除Con组和LPS组外,分别用2、2.5、5、7.5、10和20μmol·L-1的SA处理24 h后,加入0.7 mg·mL-1的LPS处理24 h。用于进一步确认SA的有效保护剂量以及后续实验。
将Caco-2细胞(2×103 cells/mL)接种于96孔板中,按照上述处理后每孔加入10μL CCK-8溶液,在37℃继续培养2 h,在酶标仪450 nm处记录其吸光度,计算细胞活力。
细胞活力计算公式:细胞存活率(%)=实验组吸光度值/Con组吸光度值
将Caco-2细胞(5×105 cells/mL)接种于6孔板中,孵育48 h后弃去上清,随机分为Con组、LPS组、SA组(分别为2.5、5、10μmol·L-1),SA 3个剂量组加入对应浓度SA的含药培养基,孵育24 h后弃去上清,除Con组外每组加入0.7 mg·mL-1 LPS含药培养基,继续孵育24 h后收集细胞上清液,按照相应ELISA试剂盒要求操作,酶标仪记录其吸光度值,测定TNF-α、IL-6、IL-18、IL-10等细胞因子含量。
将Caco-2细胞(5×105 cells/mL)接种于6孔板中,按照“2.3”项给药处理后收集细胞,按照相应试剂盒说明书操作,用流式细胞仪检测ROS含量水平或用酶标仪测定SOD、MDA、LDH、GSH的吸光度值,分析氧化应激相关因子水平的变化情况。
样品前处理:将Caco-2细胞(5×105 cells/mL)接种于6孔板中,按照“2.3”项给药处理完后收集细胞,以预冷的PBS清洗后离心弃去上清,分别按照相应试剂盒进行操作。
细胞蛋白提取:将Caco-2细胞(5×105 cells/mL)接种于6孔板中,按“2.3”项下分组培养,每孔加入200μL裂解液冰上裂解10~15 min,收集液体后10 000 r·min-1、4℃离心5 min,使用BCA试剂盒测定蛋白浓度。将蛋白裂解液置于金属浴中95℃加热5 min使蛋白变性,分装后冻存于-80℃冰箱保存备用。
检测:按照电泳、电转、封闭、孵一抗、孵二抗等操作,后加入ECL发光液进行显影。Image J进行条带分析处理。
所有数据使用Graphpad Prism 9.5进行统计分析,所有数据均以表示以P<0.05表示差异有统计学意义。
通过CCK-8法测定LPS对Caco-2细胞存活率的影响,结果如图1所示。与Con组相比,当LPS浓度为0.4 mg·mL-1时,细胞存活率明显降低至(92.73±2.27)%(P<0.001),说明LPS能够影响细胞存活率。随着LPS浓度升高,细胞存活率随之降低,当LPS浓度为0.8 mg·mL-1时,细胞存活率低于70%(P<0.01),不适合进行造模。因此,本研究选择0.7 mg·mL-1 LPS处理作为肠损伤模型的造模剂量,此时细胞存活率为(71.33±0.33)%(P<0.001)。
CCK-8法测定SA对Caco-2细胞存活率的影响,结果表明(图2),随着SA浓度的增高并未对细胞造成损伤,即SA浓度在0~25μmol·L-1时对Caco-2细胞无明显毒性,可以在此浓度范围内开展后续实验。
SA对LPS损伤Caco-2细胞存活率的影响结果如图3所示,与Con组相比,LPS组细胞的存活率降低至72.47%(P<0.001),表明模型建立成功。经SA给药处理后,细胞存活率显著升高,且在2~20μmol·L-1范围内呈剂量依赖性。SA浓度为10μmol·L-1时,对细胞存活率的影响最大,此时细胞存活率为95.37%(P<0.001)。因此,SA能够改善LPS诱导的Caco-2细胞损伤。依据本实验结果,后续SA处理组给药浓度分别为2.5、5和10μmol·L-1
为探究SA对LPS诱导的Caco-2肠损伤模型炎症反应的影响,本实验检测了炎症因子TNFα、IL-6、IL-18和IL-10的含量。如插页ⅩⅩⅩⅣ图4所示,与Con组比较,LPS组中促炎因子TNF-α、IL-6、IL-18均显著升高(P<0.001),抗炎因子IL-10显著降低(P<0.001);与LPS组比较,SA给药组抑制了促炎因子TNF-α、IL-6、IL-18的分泌,促进了抗炎因子IL-10的释放,且呈剂量依赖性。因此,SA能够有效减轻LPS诱导的肠损伤模型细胞的炎症反应。
为了进一步确定SA的抗炎作用机制,通过Western blot对LPS诱导的Caco-2细胞在炎症途径NF-κB通路相关蛋白(p65、IKKα、IKKβ、IκBα、p-IκBα、p-p65)表达量的变化进行了分析。结果 如插页ⅩⅩⅩⅤ图5所示,LPS组的炎症相关通路蛋白表达量均明显上升(P<0.05、P<0.01、P<0.001),而经SA给药后的蛋白表达量得到显著下调(P<0.05、P<0.01、P<0.001),且呈浓度依赖性。因此,SA可通过抑制NF-κB信号通路的激活,来达到调节炎症因子水平、降低肠损伤的作用。
为了探究SA是否能够改善LPS诱导的Caco-2氧化应激反应,本实验检测了ROS、SOD、LDH、GSH和MDA等代表性氧化应激因子的含量。如插页ⅩⅩⅩⅥ图6所示,与Con组相比,LPS组SOD水平和GSH含量显著降低,分别为(125.33±5.33)U·mgprot-1和(5.36±0.37)μmol·gprot-1P<0.001),ROS水平、MDA含量和LDH含量显著上升,可达到(75.33±0.33)%、(0.92±0.02)nmol·mgprot-1、(7.16±0.14)U·gprot-1P<0.001)。经不同浓度SA给药后,与LPS组相比,ROS、SOD、LDH、GSH、MDA的含量均有明显变化,当SA给药浓度为10μmol·L-1时,检测指标含量分别为(27.07±1.03)%、(373.33±19.33)U·mgprot-1、(0.52±0.02)U·gprot-1、(14.66±0.62)μmol·gprot-1、(3.41±0.26)nmol·mgprot-1P<0.001),且该作用呈剂量依赖性。因此,SA能改善LPS诱导的肠上皮细胞氧化损伤。
线粒体膜电位的下降是判断细胞凋亡的重要指标,通过插页ⅩⅩⅩⅦ图7所知。与Con组相比,LPS组细胞的线粒体膜电位显著下降率最高,可达到(32.35±0.67)%(P<0.05),给药不同浓度SA后,线粒体膜电位下降率逐渐降低。当SA浓度达到10μmol·L-1时,线粒体膜电位下降率达(17.95±0.95)%(P<0.01),且呈现明显的剂量依赖性。实验结果表明,SA可通过调节线粒体膜电位水平来改善LPS诱导的Caco-2细胞凋亡。
通过PI染色法检测细胞周期不同时相DNA含量的变化,如插页ⅩⅩⅩⅧ图8所示。结果 表明,与Con组相比,LPS组G1期与S期比例分别为(54.04±0.98)%(P<0.05)和(14.06±0.42)%,说明细胞能够在G1/S期阻滞Caco-2细胞周期进程,使细胞不能进行DNA复制。随着SA浓度增加,S期细胞比例逐步上升,当浓度达到10μmol·L-1时,S期细胞比例为(16.82±0.54)%(P<0.05)。实验结果说明SA能够缓解LPS造成的G1/S期阻滞,使细胞周期恢复正常。
通过流式细胞仪对FITC和PI染色的细胞进行检测,如插页ⅩⅩⅩⅨ图9所示,与Con组相比,LPS组凋亡率可达(54.42±0.42)%(P<0.001)。给药SA浓度为10μmol·L-1时凋亡率显著下降至(25.03±0.68)%(P<0.001),且呈现明显的剂量依赖性。表明SA能够改善由LPS引起的肠上皮细胞凋亡。
为了进一步确定SA对LPS诱导细胞凋亡的保护作用,我们通过Western Blot对LPS致Caco-2细胞凋亡的相关通路蛋白Caspase-3、Bcl-2、Bax、Cytochrome C 和 cleaved-Caspase-3进行了分析。结果 如插页ⅩL图10所示,与Con组相比,LPS组Caspase-3、Cytochrome C、Bax的表达显著增加(P<0.01、0.05、0.05),Bcl-2蛋白表达下降(P<0.01)。与LPS组比较,SA给药处理后,上调了Bcl-2/Bax的比值(P<0.05、P<0.01、P<0.01),且SA给药组Caspase-3、Cytochrome C蛋白表达显著降低(P<0.01、0.05)。因此,SA能够通过调节线粒体凋亡途径,进而抑制LPS诱导的Caco-2细胞凋亡。
肠道作为人体独一无二的器官,不仅是重要的消化器官,也是最大的免疫器官。肠道屏障是阻止肠道疾病乃至全身疾病的重要防线[11],由于肠道结构的特殊及大量细菌的存在,其具有防止肠道内病原微生物及毒素由肠腔侵入肠外器官的作用[12-13];肠上皮细胞是肠道屏障的基础,其凋亡增加会直接导致腹泻以及炎症性疾病的发生,改善肠上皮屏障破损是治疗肠损伤的关键[14-15]。故本研究采用LPS诱导Caco-2细胞建立肠道细胞屏障损伤模型,模拟肠上皮细胞的受损情况。
ROS保持着氧化还原平衡,其大量增加会触发细胞内脂质、蛋白质以及核酸的氧化,进而造成细胞损伤导致细胞凋亡;MDA是脂质氧化的主要次生产物之一,用来反映机体抗氧化能力和ROS的生成程度;SOD可以清除体内产生的自由基,为主要抗氧剂;GSH可以保护细胞膜结构和功能免受氧化的损害[16];LDH是一种存在于细胞内的酶,当细胞受损或死亡时会释放LDH,使细胞合成SOD能力变弱,细胞内GSH含量降低,ROS,MDA和LDH含量大幅度升高[17],故通过检测LDH的释放量也可以了解细胞受损程度。在LPS诱导后,Caco-2细胞中SOD和GSH水平显著降低导致自由基大量堆积,具有较强的氧化能力,诱发细胞损伤。本研究中经SA给药后细胞中MDA、ROS和LDH水平明显降低,SOD和GSH活性上升,从而平衡Caco-2细胞的氧化应激反应,减少细胞损伤。
氧化应激加速了细胞凋亡[18],氧化应激反应产生大量ROS生成可导致线粒体膜电位分解,使线粒体膜电位下降,进而导致能量缺乏和进一步加重线粒体损伤[19-23],故SA通过上调线粒体膜电位可以抑制细胞凋亡来减少细胞损伤。许多促炎途径都发生在线粒体外膜通透化过程中,在细胞凋亡过程中,调节线粒体膜通道开关的Bcl-2家族,通过激活Bax以及Bcl-2来促进线粒体通透性[24],减少Bcl-2和Bax的比值,改变线粒体膜的通透性,促进Cytochrome C从线粒体大量释放激活Caspase-3,表明凋亡进入不可逆阶段,是细胞发生凋亡的重要标志[25]。本研究表明,在LPS诱导下,Caco-2细胞中Bax、Caspase-3、Cytochrome C的表达水平上升,Bcl-2的表达水平下降,Bcl-2与Bax的比值减小,细胞凋亡趋势明显提高,而SA调节了这一变化,减少了细胞凋亡。
细胞凋亡是细胞周期中自带的检查与制衡机制,细胞周期的正常运行是保证细胞正常增殖的必要条件[26],作为有丝分裂的起始,G1/S期是多数细胞的细胞周期关键点[27-28],本研究中,经LPS诱导后G1期占比明显上升且S期占比明显下降,说明细胞在G1/S期被阻滞,无法进行DNA的合成,抑制了细胞增殖,而经SA保护后缓解了G1/S期阻滞,使细胞周期顺利进行,减轻肠上皮细胞的损伤,从而改善肠损伤。
炎症反应和氧化应激之间存在密切联系,炎症会引起细胞的氧化损伤,导致损伤部位ROS大量释放,从而加重局部损伤导致慢性炎症[29-30]。炎症是机体由内部或外部刺激诱发的复杂生物过程,参与多种疾病的病理进程[31]。在肠道炎症中所产生的促炎因子是肠道屏障损伤的重要促成因素之一[32-33],NF-κB信号通路已被证实是重要的促炎信号通路之一[34-38]。本研究表明了SA可能通过抑制NF-κB途径的激活,增加抗炎因子IL-10的释放,降低促炎因子TNF-α、IL-6和IL-18的分泌,从而有效改善了Caco-2细胞的炎症损伤。
SA能够显著改善LPS诱导的Caco-2肠上皮细胞损伤,修复氧化应激水平,通过改善细胞周期和线粒体膜电位来抑制细胞凋亡,并通过抑制NF-κB信号通路的激活来调节炎症因子水平,进而达到改善肠上皮损伤的作用。研究结果可为五味子功效成分改善胃肠健康产品研制提供基础与指导。
