Objective

The differential metabolites of Sangshen(Mori Fructus)with different maturities were studied to provide reference for the determination of Sangshen(Mori Fructus)harvest time.

Methods

Based on the metabolo micsmethod,the changes of chemical components in Sangshen(Mori Fructus)with different maturities were explored.The data of secondary metabolites in mulberry fruits was collected by ultra-high performance liquid chromatography-quadrupole/exactive Orbitrap mass spectrometry(UPLC-Q-Exactive Orbitrap MS).Multivariate statistical analysis methods such as principal component analysis(PCA)and partial least squares discriminant analysis(PLS-DA)were used to explore the differences in metabolic components of Sangshen(Mori Fructus)with different maturities.

Results

There were significant differences in the metabolites of Sangshen(Mori Fructus)with different maturities.A total of176 compounds in 11 categories were screened.Flavonoids and organic acids accounted for 23.30% and 21.02%,respectively.The contents of most screened flavonoids and organic acid differential metabolites increased with the increase of maturity,and the flavonoids and organic acid metabolites of Sangshen(Mori Fructus)of different germplasms were different.The contents of extract,total flavonoids,total polyphenols and rutin in the mature stage were higher than those in the immature stage.The antioxidant activity in the mature stage was stronger than that in the immature stage.The content of chlorogenic acid in the immature stage was higher than that in the mature stage,and the inhibitory effectonα-glucosidase was also stronger than that in the mature stage.

Conclusion

There are obvious differences in the metabolites of Sangshen(Mori Fructus)with different maturities.The contents of chemical components in Sangshen(Mori Fructus)are related to different maturities.Based on the antioxidant effect,it is better to harvest when Sangshen(Mori Fructus)is dark purple(mature).Based on the hypoglycemic effect,it is better to harvest when the Sangshen(Mori Fructus)becomes red(immature),which provides a basis for the quality control of Sangshen(Mori Fructus)and the determination of harvesting time.

目的

研究不同成熟度桑椹差异代谢物,为桑椹采收时间的确定提供参考。

方法

基于代谢组学方法探索不同成熟度桑椹化学成分的变化规律,采用超高效液相色谱-四极杆-静电场轨道阱质谱法(UPLC-Q-Exactive Orbitrap MS)对桑椹次生代谢产物数据进行采集,利用主成分分析(Principal Component Analysis,PCA)和偏最小二乘判别分析(Partial Least Squares Discriminant Analysis,PLS-DA)等多元统计分析方法探讨不同成熟度桑椹的代谢成分差异。

结果

不同成熟度桑椹的代谢物成分显著差异,共筛选到差异代谢物11类共176个化合物,黄酮类和有机酸占比较大,分别为23.30%和21.02%;大部分筛选出的黄酮类和有机酸类差异代谢成分含量随着成熟度增加而增加,且不同种质桑椹的黄酮类和有机酸类代谢化合物存在差异;成熟期样品浸出物、总黄酮、总多酚、芦丁含量均高于未成熟期,成熟期的抗氧化活性均强于未成熟期,未成熟期桑椹样品绿原酸的含量高于成熟期,对α-葡萄糖苷酶抑制作用也强于成熟期。

结论

不同成熟度桑椹代谢产物具有明显的差异,桑椹中化学成分含量与不同成熟度有关,基于抗氧化作用,在桑椹果实暗紫色(成熟)时采收更佳;基于降血糖作用,在桑椹果实变红(未成熟)时采收更佳,为桑椹的质量控制和采收时间的确定提供依据。

目的

研究不同成熟度桑椹差异代谢物,为桑椹采收时间的确定提供参考。

方法

基于代谢组学方法探索不同成熟度桑椹化学成分的变化规律,采用超高效液相色谱-四极杆-静电场轨道阱质谱法(UPLC-Q-Exactive Orbitrap MS)对桑椹次生代谢产物数据进行采集,利用主成分分析(Principal Component Analysis,PCA)和偏最小二乘判别分析(Partial Least Squares Discriminant Analysis,PLS-DA)等多元统计分析方法探讨不同成熟度桑椹的代谢成分差异。

结果

不同成熟度桑椹的代谢物成分显著差异,共筛选到差异代谢物11类共176个化合物,黄酮类和有机酸占比较大,分别为23.30%和21.02%;大部分筛选出的黄酮类和有机酸类差异代谢成分含量随着成熟度增加而增加,且不同种质桑椹的黄酮类和有机酸类代谢化合物存在差异;成熟期样品浸出物、总黄酮、总多酚、芦丁含量均高于未成熟期,成熟期的抗氧化活性均强于未成熟期,未成熟期桑椹样品绿原酸的含量高于成熟期,对α-葡萄糖苷酶抑制作用也强于成熟期。

结论

不同成熟度桑椹代谢产物具有明显的差异,桑椹中化学成分含量与不同成熟度有关,基于抗氧化作用,在桑椹果实暗紫色(成熟)时采收更佳;基于降血糖作用,在桑椹果实变红(未成熟)时采收更佳,为桑椹的质量控制和采收时间的确定提供依据。

Objective

The differential metabolites of Sangshen(Mori Fructus)with different maturities were studied to provide reference for the determination of Sangshen(Mori Fructus)harvest time.

Methods

Based on the metabolo micsmethod,the changes of chemical components in Sangshen(Mori Fructus)with different maturities were explored.The data of secondary metabolites in mulberry fruits was collected by ultra-high performance liquid chromatography-quadrupole/exactive Orbitrap mass spectrometry(UPLC-Q-Exactive Orbitrap MS).Multivariate statistical analysis methods such as principal component analysis(PCA)and partial least squares discriminant analysis(PLS-DA)were used to explore the differences in metabolic components of Sangshen(Mori Fructus)with different maturities.

Results

There were significant differences in the metabolites of Sangshen(Mori Fructus)with different maturities.A total of176 compounds in 11 categories were screened.Flavonoids and organic acids accounted for 23.30% and 21.02%,respectively.The contents of most screened flavonoids and organic acid differential metabolites increased with the increase of maturity,and the flavonoids and organic acid metabolites of Sangshen(Mori Fructus)of different germplasms were different.The contents of extract,total flavonoids,total polyphenols and rutin in the mature stage were higher than those in the immature stage.The antioxidant activity in the mature stage was stronger than that in the immature stage.The content of chlorogenic acid in the immature stage was higher than that in the mature stage,and the inhibitory effectonα-glucosidase was also stronger than that in the mature stage.

Conclusion

There are obvious differences in the metabolites of Sangshen(Mori Fructus)with different maturities.The contents of chemical components in Sangshen(Mori Fructus)are related to different maturities.Based on the antioxidant effect,it is better to harvest when Sangshen(Mori Fructus)is dark purple(mature).Based on the hypoglycemic effect,it is better to harvest when the Sangshen(Mori Fructus)becomes red(immature),which provides a basis for the quality control of Sangshen(Mori Fructus)and the determination of harvesting time.

桑椹为桑科植物桑(Morus alba L.)的干燥果穗,4~6月果实变红时采收[1],在广西成熟季大多在3~4月。《本草纲目》《中药大辞典》《中药志》等都有桑椹入药的记载。中医认为:桑椹味甘酸,归心、肝、肾经,具有滋阴补血、生津润燥、乌发明目、生津止渴、镇静安神、通气血、润肠燥的功效[1]。桑椹被国家卫生健康委员会认定为“既是食品又是药品”的物质之一,其含有丰富的黄酮、多糖、酚酸、蛋白质、总糖、维生素、矿物质、人体必需氨基酸等成分,具有抗氧化[2-3]、降血糖[4]、延缓衰老、调节免疫力、抗肿瘤[5]、抗肥胖[6]、抗炎保肝[7-8]等广泛的药理活性和较高的营养价值[9]。近年来,研究植物不同生长发育阶段果实中代谢产物的累积规律,逐渐成为研究的热点[10-11]。李勋兰等[12]以35份果桑成熟果的营养和药用品质进行综合评价,并通过因子分析法和灰色关联分析法筛选出4份品质优良果桑资源。张芳等[13]利用主成分分析和聚类分析从15份贵州野生桑果中筛选出4份桑果可作为贵州优良果桑种质资源。课题组前期对广西地区栽培的19个桑椹品种的农艺性状、药用成分和抗氧化活性差异分析和综合评价,筛选出的桂诱2172、桂椹94208、桂椹92L38、桂椹90191和桂椹92L44可作为基于抗氧化作用的桑椹优良种质选育资源[14]。冀冰聪等[15]通过高效液相色谱联合质谱分析,从代谢组水平对果桑品种大10和白玉王桑椹的成熟果实的小分子化合物进行检测分析,结果显示,大10和白玉王桑椹的代谢组存在明显差异。目前研究主要集中在桑椹的营养和药用成分综合评价和种质资源筛选方面,对桑椹成熟过程中代谢成分的差异和变化的研究较少。代谢组学从植物生物体系的整体性和动态性出发,研究植物代谢产物的动态变化,不仅可以观察到化学成分的积累过程,还可以发现各成分之间的相互转化规律,同时能检测到常规分析方法难以检测到的物质,系统完整地阐明影响植物生长以及作物果实品质的因素,可从整体上科学评价药材的质量,逐渐被应用于农业、医药和食品领域[16]。中药质量的优劣取决于活性成分的含量[17],活性成分的积累会随药材生长过程发生不同程度的动态变化,因此适时采收是保证中药材质量的先决因素之一。中药药效物质基础与植物次生代谢物密切相关[18],桑椹果实初熟时呈红色,成熟后逐渐转变为紫黑色,桑椹逐渐成熟的过程中代谢成分会发生不同程度的动态变化,因此进一步研究不同成熟度桑椹代谢物差异,研究不同成熟度桑椹的活性成分变化和药理活性,对桑椹的品质差异、采收时间和产业发展具有重要意义。本研究采用超高效液相色谱-四极杆-静电场轨道阱质谱法(UPLC-Q-Exactive Orbitrap MS)技术,结合多元统计分析方法探讨不同生长成熟阶段桑椹差异代谢物及品质差异,并探索不同成熟度桑椹的化学成分含量差异和对抗氧化、降血糖药理活性的影响,为桑椹采收时间的确定提供基础。
3个不同种质不同成熟期桑椹药材均于2022年3月30日采自广西壮族自治区蚕业技术推广站桑树资源圃,经广西中医药大学朱意麟园艺师、滕建北教授鉴定为桑科植物桑(Morus alba L.)栽培品种的干燥果穗。桑椹样品信息见表1
甲醇、乙腈(色谱纯,赛默飞世尔科技有限公司);甲酸(色谱纯,天津市科密欧化学试剂有限公司);芦丁(批号:SR8250)、福林酚(批号:921O036);2,4,6-三吡啶基三嗪(2,4,6-Tris(2-pyridyl)-s-triazine,TPTZ)(批号:1029G022)、α-葡萄糖苷酶(批号:1202I012,13.2 mg/1 kU)、对硝基苯基-α-D-吡喃葡萄糖苷(p-Nitrophenyl-α-D-glucopyranoside,PNPG)(批号:409J021)(均购于北京索莱宝科技有限公司);没食子酸(批号:190406,上海融禾医药科技有限公司);1,1-二苯基-2-三硝基苯肼(2,2-Diphenyl-1-picrylhydrazyl,DPPH)(批号:Z6B8N-TF,梯希爱(上海)化成工业发展有限公司);2,2-联氮-二(3-乙基-苯并噻唑-6-磺酸)二铵盐(2,2'-Azinobis(3-ethylbenzothiazoline-6-sulfonic acid Ammonium Salt,ABTS)(批号:C13178583,上海麦克林生化科技有限公司)。
Ultimate 3000型超高效液相色谱,Q-Exactive型高分辨质谱,HSST3色谱柱(2.1 mm×100 mm,1.8μm)(美国赛默飞世尔科技(中国)有限公司);Waters e2695高效液相色谱仪(上海沃特世科技有限公司);Agilent 5 TC-C18色谱柱(4.6 mm×250 mm,5μm),安捷伦科技(中国)有限公司);SQP型电子分析天平(德国Sartorius公司);KQ5200B型超声波清洗器(昆山市超声仪器有限公司);UV-2600型紫外分光光度计(苏州岛津仪器有限公司);SYNERGYH1酶标仪(美国伯腾公司)。
精密称取不同成熟度的桑椹粉末各20mg,加入50%乙醇1mL,涡旋混匀30 s;冰水浴超声处理30 min;于4℃,13 000 rpm,离心2 min;吸取上清,过0.45 μm微孔滤膜,备用。质控样本(QC)由样本提取物混合制备而成,每10个检测分析样本中插入一个质控样本,以监测分析过程的重复性。
色谱条件:色谱柱:Waters HSS T3(100 mm×2.1 mm 1.8μm),正离子模式流动相为含0.1%甲酸的水溶液(A液)和含0.1%甲酸的甲醇(B液),负离子模式流动相10μmol/L甲酸氨的水溶液(A液)和含10μmol/L甲酸氨的95%甲醇(B液);洗脱梯度:0 min,90%A;1~13 min,90%~2%A;13~18 min,2%A;18~18.5 min,2% ~90%A;18.5~20 min,90%A;流速:0.3 mL/min;柱温:35℃;进样量2μL。
质谱条件:扫描模式:正、负离子分开扫描;检测方式:Full mass/dd-MS2;分辨率:70 000(Full mass),17 500(dd-MS2);电喷雾电压3.8 kV(Positive),3.2 kV(Negative);毛细管温度:300℃;鞘气流速:40 Arb;雾化器温度:350℃。
浸出物含量测定参照醇溶性浸出物测定法(通则2201)项下的热浸法测定,用85%乙醇作溶剂浸出物采用热浸法测定[1]。总黄酮含量采用Al(NO33分光光度比色法测定,以芦丁为对照品[19]。分别精密称取不同成熟度桑椹药材粉末0.2 g,加入80%乙醇40 mL,平行3份,超声处理30 min,滤过。精密移取1 mL样品溶液,加入1 mL 4% NaNO2 溶液,反应6 min,再加入1 mL 10% Al(NO33 溶液,反应6 min,再加入4 mL 8% NaOH溶液,反应15min。以纯水为空白,于510 nm下测定。总多酚含量采用福林-酚比色法测定,以没食子酸为对照品[19]。分别精密称取不同成熟度桑椹药材粉末0.1 g,加入60%乙醇30 mL,平行3份,超声处理40 min,滤过。精密移取1 mL样品溶液,避光加入福林酚0.5 m L,反应5 min,再加入2 mL 7.5% Na2 CO3溶液,70℃水浴1h(避光),取出放至室温,以60%乙醇为空白,于754 nm下测定。
采用HPLC法测定不同成熟度桑椹中绿原酸、芦丁含量[20]。取桑椹粉末1.0 g,精密称定,于100 mL锥形瓶中,加入50%乙醇20 mL,超声处理30 min,滤过,备用。以乙腈(A)-0.1%磷酸(B)为流动相,色谱柱:Agilent5 TC-C18(4.6 mm×250 mm,5μm),检测波长:330 nm。色谱柱温度:25℃,流速:1.0 m L/min,进样量:10μL。梯度洗脱:0~15min,5%~12%A;15~30 min,12%~15%A;30~70 min,15%~35%A。
精密称取不同成熟度桑椹药材粉末各0.2 g,平行3份,加入50%乙醇20 mL,超声处理30 min,滤过,备用。
采用FRAP法测定样品的总抗氧化能力[21]。FRAP检测试剂配制:10 mmol/L的TPTZ(40 mmol/L HCl水溶液配制),20 mmol/L的FeCl3 水溶液和0.3 mmol/L醋酸钠溶液,体积比为1∶1∶10混匀,37℃水浴中备用,现配现用。取2.4.1项桑椹供试液120μL加入FRAP工作液4 mL,充分混合均匀,避光反应40 min,实验平行3份,在596 nm波长处测定吸光度值。
采用DPPH法测定桑椹样品清除DPPH自由基能力[22]。取2.4.1项桑椹供试品溶液1 mL与3 mL 0.4 mmol/L DPPH溶液混合均匀,避光反应30 min,使用酶标仪在517 nm下测定吸光度值,实验平行3份。清除率% =[1-(A1-A2)/A0]×100%,式中:A1为反应后样品溶液的吸光度值;A2为样品溶液的吸光度值;A0为DPPH溶液的吸光度值。
采用ABTS法测定桑椹样品清除ABTS自由基能力[22]。ABTS工作液:称取过硫酸钾66.23 mg用纯水定容至100 mL容量瓶中,待用;精密称取ABTS 28.2 mg,加入75 mL纯水搅拌溶解,再加入75 mL制备好的过硫酸钾溶液,混合均匀,避光,过夜。取2.4.1项桑椹供试溶液50μL与50μL ABTS工作液混合均匀,室温反应6 min后,在734 nm测定吸光度值,实验平行3份。清除率% =[1-(A1-A2)/A0]×100%,式中:A1为反应后样品溶液的吸光度值;A2 为样品溶液自身的吸光度值;A0为空白对照的吸光度值。
利用桑椹样品对α-葡萄糖苷酶的抑制率评价降血糖活性[23]。取2.4.1项桑椹供试品溶液40 μL加入PBS缓冲液(PH=6.8,40μL),加入α-葡萄糖苷酶(0.2 U/mL,30μL),孵育15 min后,加入PNPG(5 mmol/L,60 μL),孵育15 min后,再加入Na2 CO3(0.1 mol/L,80μL)终止反应,即得样品溶液。在405 nm测定,实验平行3分。α-葡萄糖苷酶活性抑制率% =[A0-(A1-A2)]/A0×100%,式中:A1为加入待测样品的吸光度,A2为背景溶液(不加酶)的吸光度,A0为空白溶液(不加待测样品)的吸光度。
原始数据采用MSDIAL软件进行峰对齐、保留时间校正和提取峰面积。采用精确质量数匹配(质量偏差 mass tolerance<0.01 Da)和二级谱图匹配(质量偏差 mass tolerance<0.02 Da)的方式进行代谢物结构鉴定,使用HMDB公共数据库进行检索。利用Excel、SPSS 20.0等软件对数据进行处理分析。
结合多变量分析PLS-DA模型前两个主成分的VIP值,单变量分析差异倍数(fold change,FC),以VIP≥1.0,fold change≥2.0或≤0.5同时满足两个条件的,即为差异成分。从不同成熟度桑椹共筛选到差异代谢物11类共176个,包括41个黄酮类化合物、37个有机酸类化合物、10个氨基酸及其衍生物化合物、12个生物碱类化合物、8个核苷酸及其衍生物、10个糖和苷类化合物、3个萜类化合物、4个醌类化合物、7个香豆素类化合物、2个吲哚类化合物以及42个其他类化合物,如插页ⅩLⅡ图1所示。可以看出,不同成熟度桑椹中主要的差异成分为黄酮类和有机酸类化合物,分别为23.30%和21.02%。对不同成熟度桑椹的代谢差异产物进行鉴定,从而了解桑椹成熟过程中的代谢物及差异,后续对差异较大的黄酮类成分和酚酸类成分进行聚类分析。
将预处理的LC-MS数据进行非监督PCA分析,获得不同成熟度桑椹在空间上的分布情况。结果 如插页ⅩLⅢ图2所示,桂诱2172、桂椹92L38、桂椹90191 3个品种PC1分别解释了总变量的70.3%、74.9%、71.1%,PC2解释了总变量的10.7%、6.4%、9.4%,累计贡献率分别达到81.0%、81.3%、80.5%。3个品种不同成熟桑椹的PCA分析均显示不同成熟度桑椹之间存在明显分离,分别形成两组不同的类群。表明不同成熟度桑椹之间代谢物存在明显差异。
对不同成熟度桑椹数据进行PLS-DA分析。结果 如插页ⅩLⅢ图3所示,不同成熟度桑椹数据分别沿PC1轴明显分开,其中模型参数R2分别为0.998 7、0.999 3、0.999 5;Q2分别为0.973 0、0.983 3、0.976 1。与PCA分析结果一致,不同成熟度桑椹间的代谢物存在明显差异。
将不同成熟度桑椹的黄酮类代谢化合物进行聚类分析,结果如下插页ⅩLⅢ图4,显示黄酮类代谢物在同一成熟度3份样品间含量差异较小,成熟与未成熟桑椹样品间的含量差异较大,且3个不同桑椹种质间的黄酮类代谢物也存在一定差异,3个不同桑椹种质的共有黄酮类代谢物如5-去甲基川陈皮素、圣草酚、麦冬高异黄酮A、白杨素、5,7-二羟基黄烷酮等在成熟期具有较高表达,含量也维持在较高水平。以未成熟桑椹样品为对照,筛选出当fold change≥2表示差异上调,fold change≤0.5表示差异下调的黄酮类差异代谢化合物。结果 见表2,显示桂诱2172、桂椹92L38、桂椹90191 3个不同种质桑椹成熟样品中大部分黄酮类差异代谢成分的log FC≥2.0,部分成分FC值高达10倍以上,表明桑椹中大部分黄酮类差异代谢成分含量随成熟度增加而积累增加。3个种质不同成熟度桑椹样品黄酮类差异代谢产物共有成分有13种,以未成熟桑椹样品为对照,其中有11种成分在成熟桑椹中含量上调,2种成分下调,且同种黄酮类成分在不同种质中含量的差异倍数(FC)存在一定差异。
将不同成熟度桑椹的有机酸类代谢化合物进行聚类分析,结果见插页ⅩLⅢ图5,有机酸类代谢物在相同成熟度3份样品间含量差异较小,不同成熟度的桑椹样品间含量差异较大,表明随着桑椹成熟度的增加有机酸类代谢物含量也随之发生变化,3个不同种质桑椹共有有机酸类代谢物如庚二酸、酮异戊酸、马来酸、甲基琥珀酸、16-羟基十六烷酸等在成熟期表达较高,含量也较高。以未成熟桑椹样品为对照,筛选当fold change≥2表示差异上调,fold change≤0.5表示差异下调的有机酸类差异代谢物。结果 见表2,显示3个种质的成熟样品中大部分有机酸类差异代谢成分log FC≥2.0,部分成分FC值高达10倍以上。桂诱2172、桂椹92L38、桂椹90191 3个种质不同成熟度桑椹样品有机酸类差异代谢化合物共有成分为21种,以未成熟桑椹样品为对照,其中有15种成分在成熟桑椹中含量上调,6种成分下调。可见大部分筛选出的有机酸类差异代谢成分含量随成熟度增加而累积增加,且同种有机酸类成分在不同种质中含量的差异倍数(FC)存在一定差异。
对不同成熟度桑椹的浸出物、总黄酮、总多酚酸、绿原酸、芦丁含量进行测定。结果 见表4,显示桂诱2172、桂椹92L38、桂椹90191 3个种质成熟桑椹样品的浸出物、总黄酮、总多酚、芦丁含量均高于未成熟样品,且差异具有显著性统计学意义(P<0.05)。结果 表明,桑椹浸出物、总黄酮、总多酸、芦丁含量随着成熟度增加而积累增加。但成熟期桑椹样品的绿原酸含量均低于未成熟样品,且差异具有显著性统计学意义(P<0.05),表明绿原酸含量随着成熟度增加而降低。
分别采用FRAP、DPPH、ABTS法对不同成熟度桑椹进行抗氧化活性测定,同时考察不同成熟度桑椹对α-葡萄糖苷酶活性抑制作用。结果 见表4,显示不同成熟度桑椹的抗氧化活性与α-葡萄糖苷酶活性抑制作用均呈现差异性,3个不同种质的成熟桑椹样品抗氧化作用均强于未成熟桑椹样品,但未成熟桑椹样品对α-葡萄糖苷酶活性抑制作用均强于成熟桑椹样品,表明随着桑椹不断成熟,桑椹的抗氧化活性越强,降血糖活性越弱。
现代研究表明,桑椹中具有多种生理活性物质和广泛的药理活性桑椹代谢物繁杂,包含黄酮、有机酸、生物碱等代谢成分,并表现出抗氧化[24]、抗衰老、降糖降脂[25]、预防阿尔茨海默病[26]、抑菌等广泛的药理作用[27]。本研究采用非靶向代谢组学分析方法对桂诱2172、桂椹92L38、桂椹90191 3个种质不同成熟度桑椹差异代谢化合物进行分析并对差异较大的黄酮类和有机酸类差异代谢物进行分析,同时对不同成熟度桑椹的浸出物、总黄酮、总多酚、绿原酸、芦丁含量进行测定,并研究了不同成熟度桑椹体外抗氧化和降血糖药理活性。研究结果显示,3个种质不同成熟度桑椹共筛选到差异代谢物11类共176个化合物,以黄酮类和有机酸类成分占比较大,分别为23.30%和21.02%。通过PCA分析和PLS-DA分析表明不同成熟度桑椹间代谢物存在明显差异,同时分别对3个不同种质成熟和未成熟桑椹差异较大的黄酮类和有机酸类成分差异代谢物进行聚类分析发现,以未成熟桑椹为对照,3个种质不同成熟度桑椹药材黄酮类差异代谢物共有成分为13种,其中麦冬高异黄酮A、5,7-二羟基黄烷酮、3-(2-羟基-4-甲氧基苯基)-3,4-二氢-2H-1-苯并吡喃-7-醇、5-去甲基川陈皮素、白杨素等11种成分在成熟桑椹中含量为上调,鸢尾素和表儿茶素2种成分为下调。3个种质不同成熟度桑椹药材有机酸类差异代谢物共有成分为21种,其中马来酸、酮异戊酸、16-羟基十六烷酸、庚二酸、甲基琥珀酸等15种成分上调,组氨酸、DL-硫辛酸、N-甲基丝氨酸、L-谷氨酸、丙氨酸等6种成分下调,表明大部分筛选出的黄酮类和有机酸类差异代谢成分含量随着成熟度增加而增加,且不同种质桑椹药材黄酮类和有机酸类代谢成分含量上存在一定的差异。代谢化合物含量上的差异可能会影响桑椹的品质和药理活性。
本研究发现桑椹的浸出物、总黄酮、总酚酸、芦丁含量随成熟度增加而增加呈现正相关,且成熟桑椹药材抗氧化活性均强于未成熟样品。研究表明,槲皮素[28]、白杨素[29]、芦丁及桑椹总黄酮、总多酚含量与抗氧化能力正相关,提示桑椹总黄酮、总多酚含量越高,羟自由基清除能力越强[30]。成熟桑椹差异代谢物槲皮素、白杨素及主要成分芦丁、总黄酮的含量均较未成熟桑椹含量高,成熟桑椹抗氧化活性也比未成熟桑椹更强。鸢尾素[31]、表儿茶素[32]、绿原酸[33]均具有较好的降血糖活性。成熟桑椹差异代谢物鸢尾素、表儿茶素及主要成分绿原酸的含量均较未成熟桑椹有所下调,即鸢尾素、表儿茶素、绿原酸含量在未成熟桑椹中含量更高,未成熟桑椹降血糖活性比成熟桑椹更强。不同成熟度桑椹药材中差异代谢物及主要活性成分含量的差异对其抗氧化、降血糖活性有较大影响。
《中华人民共和国药典》2023年版收录桑椹在4~6月果实变红时采收。本研究结果表明桑椹成熟期的代谢化合物及其含量与未成熟的样品均存在明显差异,且成熟期样品浸出物、总黄酮、总酚酸、芦丁含量均高于未成熟期,成熟期的抗氧化活性均强于未成熟期;未成熟期桑椹样品绿原酸的含量高于成熟期,对α-葡萄糖苷酶抑制作用也强于成熟期。
综上所述,随着桑椹成熟程度的变化,果穗逐渐由红色转变为黑紫色,桑椹药材的代谢成分及药理活性也随之发生变化。建议根据不同的药效目的选择适宜的采收时间,基于抗氧化作用,在桑椹果实变为暗紫色(成熟)时采收更佳;基于降血糖作用,在桑椹果实变红(未成熟)时采收更佳。
