目的

观察Cyt-c/Apaf-1/Caspase-9/3凋亡信号通路在狼疮性肾炎（LN）中的作用，探讨三七注射液治疗LN的机制。

方法

将15只雌性MRL-faslpr小鼠随机分为模型组、Apaf-1抑制剂组和三七注射液组，每组5只。另设相同周龄C57BL/6小鼠5只，作为正常组。采用免疫荧光、Western Blot检测肾组织凋亡体及相关蛋白表达。

结果

模型组小鼠肾结构损伤严重，炎细胞浸润明显；Apaf-1抑制剂组和三七注射液组肾损伤减轻，炎细胞浸润减轻。免疫荧光分析肾组织凋亡体，模型组小鼠肾组织凋亡体Cyt-c、Apaf-1、Caspase-9表达水平较正常组升高（P<0.01）；Apaf-1抑制剂组和三七注射液组凋亡体Cyt-c、Apaf-1、Caspase-9表达水平较模型组下降（P<0.01）。模型组小鼠肾组织凋亡体Cyt-c、Apaf-1、Cleaved-Caspase-9蛋白和凋亡蛋白Cleaved-Caspase-3、Bax表达水平较正常组升高（P<0.01），Bcl-2蛋白表达较正常组下降（P<0.01）；Apaf-1抑制剂组和三七注射液组凋亡体Cyt-c、Apaf-1、Cleaved-Caspase-9蛋白和凋亡蛋白Cleaved-Caspase-3、Bax表达水平较模型组下降（P<0.01），Bcl-2蛋白表达较模型组升高（P<0.01）。

结论

Cyt-c/Apaf-1/Caspase-9/3凋亡信号通路激活后，促进细胞凋亡，加剧LN的疾病程度。三七注射液可能通过抑制Cyt-c/Apaf-1/Caspase-9/3凋亡信号通路，减轻肾组织细胞凋亡，改善LN。

Objective

To observe the role of Cyt-c/Apaf-1/Caspase-9/3 apoptosis signaling pathway in lupus nephritis (LN) and explore the mechanism of Sanqi Injection (三七注射液) in the treatment of LN.

Methods

Fifteen female MRL-faslpr mice were randomly divided into model group, Apaf-1 inhibitor group and Sanqi Injection group, with 5 mice in each group. Five C57BL/6 mice of the same age were set as the normal control group. The expressions apoptosome and related proteins in renal tissue were detected by immunofluorescence and western blot.

Results

In the model group, the renal structure of the mice was severely damaged, and the infiltration of inflammatory cells was significant. In the Apaf-1 inhibitor group and the Sanqi Injection group, the renal injury was mitigated and the infiltration of inflammatory cells was reduced. Immunofluorescence analysis of kidney tissue apoptosome revealed that the expression levels of Cyt-c, Apaf-1 and Caspase-9 in the model group were higher than those in the normal group (P<0.01). The expression levels of Cyt-c, Apaf-1 and Cleaved-Caspase-9 in the Apaf-1 inhibitor group and Sanqi Injection group were lower than those in the model group (P<0.01). The protein expression levels of Cyt-c, Apaf-1, Cleaved-Caspase-9 and the expression levels of the apoptotic proteins Cleaved-Caspase-3 and Bax in the model group were higher than those in the normal group (P<0.01), while the expression level of Bcl-2 protein was lower than that in the normal group (P<0.01). The protein expression levels of Cyt-c, Apaf-1, Cleaved-Caspase-9, and the expression levels of apoptotic proteins Cleaved-Caspase-3 and Bax in the Apaf-1 inhibitor group and Sanqi Injection group were lower than those in the model group (P<0.01), while the expression level of Bcl-2 protein was higher than that in the model group (P<0.01).

Conclusion

Once the Cyt-c/Apaf-1/Caspase-9/3 apoptotic signaling pathway is activated, it promotes cell apoptosis and exacerbates the disease severity of LN. Sanqi Injection may alleviate LN by inhibiting the Cyt-c/Apaf-1/Caspase-9/3 apoptotic signaling pathway and reducing apoptosis of renal tissue cells.
