Objective

To establish a method for detecting four arsenic species—As (III), DMA, MMA, and as (V)—in seafood using liquid chromatography-atomic fluorescence spectrometry (LC-AFS).

Methods

The detection conditions were optimized, including the type and concentration of the carrier liquid, concentration of potassium borohydride, lamp current, negative high voltage, mobile phase concentration, and pH. Parameters of the established detection method were evaluated, including linear range, accuracy, precision, and detection limit.

Results

Under the conditions of 5% hydrochloric acid, 5 g/L sodium hydroxide-20 g/L potassium borohydride, a negative high voltage of 270 V, lamp main current of 60 mA, mobile phase A as ammonium dihydrogen phosphate (1 mmol/L, pH 8.5), and mobile phase B as ammonium dihydrogen phosphate (25 mmol/L, pH 8.0), the linear range for As(III), DMA, MMA, and As(V) was 0-100 μg/L, with correlation coefficients exceeding 0.999. The relative standard deviation (RSD) for retention time ranged from 0.2% to 1.2%, while the RSD for peak area ranged from 2.1% to 4.4%. The recovery rate for spiked samples was between 81.2% and 111.6%.

Conclusion

The method demonstrates high sensitivity, accuracy, and precision, with a short analysis time and high detection efficiency, while requiring minimal reagent usage, yielding satisfactory results.

目的

建立液相色谱-原子荧光联用技术(LC-AFS)检测海产品中As(III)、DMA、MMA、As(V)四种形态砷的方法。

方法

优化检测条件,包括载流的类型及浓度、硼氢化钾浓度、灯电流、负高压、流动相浓度及pH等,考察所建立检测方法的参数,包括线性范围、准确度、精密度、检出限等。

结果

在5 %盐酸、5 g/L氢氧化钠-20 g/L硼氢化钾、负高压270 V、灯主电流60 mA、流动相A为磷酸二氢铵(1 mmol/L,pH 8.5)、流动相B为磷酸二氢铵(25 mmol/L,pH 8.0)的条件下,As(III)、DMA、MMA、As(V)线性范围为0~100 μg/L,相关系数均大于0.999;保留时间RSD为0.2%~1.2%,峰面积RSD为2.1%~4.4%;样品加标回收率为81.2%~111.6%。

结论

方法灵敏度高、准确性好、精密度高,分析时间短,检测效率高,试剂用量少,结果满意。

目的

建立液相色谱-原子荧光联用技术(LC-AFS)检测海产品中As(III)、DMA、MMA、As(V)四种形态砷的方法。

方法

优化检测条件,包括载流的类型及浓度、硼氢化钾浓度、灯电流、负高压、流动相浓度及pH等,考察所建立检测方法的参数,包括线性范围、准确度、精密度、检出限等。

结果

在5 %盐酸、5 g/L氢氧化钠-20 g/L硼氢化钾、负高压270 V、灯主电流60 mA、流动相A为磷酸二氢铵(1 mmol/L,pH 8.5)、流动相B为磷酸二氢铵(25 mmol/L,pH 8.0)的条件下,As(III)、DMA、MMA、As(V)线性范围为0~100 μg/L,相关系数均大于0.999;保留时间RSD为0.2%~1.2%,峰面积RSD为2.1%~4.4%;样品加标回收率为81.2%~111.6%。

结论

方法灵敏度高、准确性好、精密度高,分析时间短,检测效率高,试剂用量少,结果满意。

Objective

To establish a method for detecting four arsenic species—As (III), DMA, MMA, and as (V)—in seafood using liquid chromatography-atomic fluorescence spectrometry (LC-AFS).

Methods

The detection conditions were optimized, including the type and concentration of the carrier liquid, concentration of potassium borohydride, lamp current, negative high voltage, mobile phase concentration, and pH. Parameters of the established detection method were evaluated, including linear range, accuracy, precision, and detection limit.

Results

Under the conditions of 5% hydrochloric acid, 5 g/L sodium hydroxide-20 g/L potassium borohydride, a negative high voltage of 270 V, lamp main current of 60 mA, mobile phase A as ammonium dihydrogen phosphate (1 mmol/L, pH 8.5), and mobile phase B as ammonium dihydrogen phosphate (25 mmol/L, pH 8.0), the linear range for As(III), DMA, MMA, and As(V) was 0-100 μg/L, with correlation coefficients exceeding 0.999. The relative standard deviation (RSD) for retention time ranged from 0.2% to 1.2%, while the RSD for peak area ranged from 2.1% to 4.4%. The recovery rate for spiked samples was between 81.2% and 111.6%.

Conclusion

The method demonstrates high sensitivity, accuracy, and precision, with a short analysis time and high detection efficiency, while requiring minimal reagent usage, yielding satisfactory results.

近年来,海洋生物受到砷等重金属污染日益严重,人体食用受到污染的海产品,可危害健康[1-2]。海产品中的砷包含无机砷和有机砷,具有多种形态。其无机砷主要为剧毒的三价砷(As(III))、五价砷(As(V)),有机砷包含有毒的一甲基砷(MMA)、二甲基砷(DMA)及几乎无毒的砷甜菜碱(AsB)等[3-5]。因此,检测海产品中As(III)、As(V)、MMA、DMA四种形态砷的含量,对评价海产品中砷的毒性大小具有重要价值。目前,海产品中砷形态分析的方法主要有液相色谱­原子荧光光谱法(LC-AFS)、液相色谱­原子吸收光谱法(LC-AAS)、液相色谱­电感耦合等离子体质谱法(LC-ICP-MS)、气相色谱­电感耦合等离子体质谱法(GC-ICP-MS)、毛细管电泳­电感耦合等离子体质谱法(CE-ICP-MS)等[6-8]。其中以LC-AFS法、LC-ICP-MS 法应用较多。王林裴等人[9]应用LC-AFS 法测定水产动物及其制品中的形态砷,检出限低,检测效率较高。LC-AFS 法集成了多组分高效分离优势和原子荧光光度法的高灵敏度,运行成本较低,分析前仪器升温时间短,操作简单。
目前基于LC-AFS分析海产品中As(III)、DMA、MMA、As(V)等形态的研究,有的仅对原子荧光的检测条件作优化[10],有的仅简单对液相色谱的保留时间、分离度作优化[11-12],缺少对色谱条件及原子荧光条件系统的探讨与优化。因此,文章对该方法进行更全面更深入的研究,以探讨液相色谱条件和原子荧光条件对形态砷的响应强度、保留时间、分离度、灵敏度以及检测时间、试剂用量、色谱柱寿命等方面的影响,优化了仪器分析条件,建立了液相色谱-原子荧光联用技术检测海产品中形态砷的方法,并用建立的方法对实际样品进行检测,取得较好效果。
液相色谱-原子荧光联用仪(LC-AFS 9560,北京海光仪器有限公司),pH计(PHSJ-3F,上海仪电科学仪器股份有限公司),超纯水仪(Milli-Q,德国默克集团),电子天平(AL-204,梅特勒-托利多科技有限公司),鼓风干燥箱(DHG-9140A,上海一恒科学仪器有限公司),高速离心机(Eppendorf Centrifuge 5920 R,德国艾本德有限公司),振荡器(DMV-16,广东省科寅实验室设备有限公司),C18小柱,0.45 μm滤膜。
硝酸、盐酸、硼氢化钾均为优级纯;氢氧化钠、氨水、磷酸二氢铵、正己烷均为分析纯;超纯水(电阻率大于18.2 MΩ·cm);亚砷酸根溶液[As(III),GBW08666,75.5 μg/g]、砷酸根溶液[As(V),GBW08667,17.5 μg/g]、一甲基砷溶液(MMA,GBW08668,25.1 μg/g)、二甲基砷溶液(DMA,GBW08669,52.9 μg/g)、砷甜菜碱溶液(AsB,GBW08670,38.8 μg/g)、砷标准物质均购自中国计量科学研究院,均以As计。
采集泉州市本地海产品64份,包含鱼、虾、蟹、壳类等,进行检测分析。
参考《食品中总砷及无机砷的测定》GB 5009.11-2014(简称国标方法)的海产品前处理方法。除以6 000 r/min离心5 min,样品下层清液无需C18柱净化,直接经0.45μm水系滤膜过滤外,其余同国标方法。
分别吸取适量As(III)溶液、As(V)溶液、MMA溶液、DMA溶液,用超纯水配制成1.0 mg/L混合标准中间液。
分别吸取适量四种1.0 mg/L砷混合标准中间液,用超纯水稀释至浓度为0.0、5.0、10.0、20.0、30.0、50.0和100.0 μg/L标准系列。
载流:5%盐酸;还原剂:5 g/L氢氧化钠-20 g/L硼氢化钾;负高压:270 V;砷灯主/辅电流:60/30 mA;原子化器高度:8 mm;载气流速:400 ml/min;屏蔽气流速:900 ml/min;蠕动泵转速:45 r/min。
PRP-X100阴离子交换柱(250×4.1 mm,10 μm);流动相A:1 mmol/L NH4H2PO4,pH 8.5;流动相B:25 mmol/L NH4H2PO4,pH 8.0;流速1.0 ml/min;进样量:100 μl;柱温:40 ℃。洗脱程序见表1
本试验参照国标方法的仪器工作条件(以下各优化试验同样依此进行),分别以20% HCl和20% HNO3作载流测定砷混合标准溶液系列。结果表明,当载流为20% HCl溶液时,As(III)、DMA、MMA、As(V)四种形态砷的响应强度均更大;标准曲线的相关系数r、斜率均较大,说明线性更好、灵敏度更高;二者谱图的基线噪声差别不大。经过分析,HNO3具有氧化性,可能会与还原剂硼氢化钾反应,消耗硼氢化钾的量,导致硼氢化钾不足以将形态砷还原成砷化氢,造成响应值偏低、线性差的结果。因此,选择载流类型为HCl溶液。
分别选择3%~30%的HCl溶液作为载流,测定30 μg/L砷标准溶液,探讨形态砷的峰面积、基线噪声随HCl浓度的变化情况。如图1所示,随着HCl浓度增大,As(V)的峰面积急剧增大;As(III)峰面积变化不明显;HCl浓度为5%时,DMA和MMA峰面积达到最大。HCl浓度增大,基线噪声差别不大。因此,选择盐酸浓度为5%。
KBH4作为氢化物生成的还原剂,对氢化物的生成具有重要作用。浓度过小则砷不能彻底还原,浓度过高则使会气流过大导致荧光强度下降[9]。以5 g/L氢氧化钠为溶剂,配制10~50 g/L的KBH4溶液,测定30 μg/L的形态砷混合标准溶液,考察KBH4浓度对As(III)、DMA、MMA、As(V)的峰面积、基线噪声的影响,如图2所示,随着KBH4浓度增加,砷的峰面积增大,但噪声也增大。综合考虑响应强度、噪声、信噪比和KBH4消耗量,选择KBH4浓度为20 g/L,这与王林裴等人[9]的研究一致。
设定230V、250V、270V、300V、330V五种负高压,测定30μg/L砷标准溶液,考察光电倍增管负高压对形态砷峰面积、噪声、信噪比的影响。结果显示,负高压增加,峰面积增大,但噪声也增大;当负高压由270V增至300V时,噪声急剧增加,信噪比逐渐降低。由于负高压过高会减少光电倍增管的使用寿命,综合考虑,选择负高压为270V。
原子荧光光谱分析中,灯电流越大,砷空心阴极灯的激发光强度越强,砷的响应值越高,噪声也越大,但灵敏度不一定增加;若灯电流过大,空心阴极灯会发生自吸现象,且会影响灯的寿命。实验设定30/15 mA、40/20 mA、55/27 mA、60/30 mA、75/37 mA五种主/辅电流,测定30 μg/L砷标准溶液,考察灯电流对形态砷峰面积、信噪比的影响。结果显示,主电流增大,峰面积、噪声不断增加,这与夏拥军[10]的研究结果一致。为了在增加峰面积的同时,提高灵敏度,减小灯自吸现象,延长灯寿命,选择灯电流为60/30 mA。
目前文献对液相色谱工作条件的优化大多只关注流动相浓度或pH对砷形态组分的保留时间和分离情况的影响,较少考虑对峰面积、基线噪声、灵敏度、色谱柱寿命等的影响。李鑫[11]在研究海产品形态砷测定方法时,只探讨流动相浓度、pH对形态砷分离效果的影响。林建杰[12]在测定海藻中形态砷含量时,只研究流动相pH对分离效果的影响。
文章该部分较充分地研究了流动相的影响情况,包括A相、B相磷酸二氢铵溶液的浓度和pH,对形态砷的保留时间、分离效果、峰面积、灵敏度、色谱柱寿命等的影响。
配制20~40 mmol/L磷酸二氢铵溶液,测定50 μg/L砷混合溶液,探究流动相B浓度对形态砷峰面积及保留时间的影响。结果如图34所示,流动相B浓度增大,四种形态砷峰面积变化不大,且分离度始终良好;As(III)和DMA、MMA保留时间几乎稳定;由20 mmol/L升至25 mmol/L时,As(V)保留时间有较明显缩短。流动相浓度越大,则离子强度越高,而离子强度过高会影响阴离子色谱柱的寿命[13]。综合峰面积、分离效果、色谱柱寿命,选择流动相B浓度为25 mmol/L。
配制浓度为25 mmol/L,pH为6.0~10.0的五种磷酸二氢铵溶液,测定50 μg/L砷标准溶液,考察流动相B的pH值对形态砷峰面积、分离效果、基线噪声的影响。结果如图56所示,pH增大,As(III)峰面积减小,保留时间增大;As(V)峰面积先增大后减小,保留时间减少;DMA、MMA峰面积始终较大,保留时间的变化不影响整体的分析时间和分离度;pH为10时,基线噪声过高。pH过大,流动相偏碱性会使部分砷化合物保留在阴离子色谱柱中,可能会缩短色谱柱的寿命[13]。综合考虑峰面积、分离效果,分析时间、噪声、色谱柱寿命等,选择流动相B的pH为8.0。
由于海产品中无毒的AsB含量高,剧毒的As(III)含量较低,两者的保留时间接近,若没有完全分离,As(III)的测定会受AsB干扰,其检测结果将偏高。因此,需要考察流动相A浓度与pH对AsB和As(III)分离度的影响,同时考虑对响应强度的影响。
考察流动相A的pH 9.0,浓度为1、3、5、7和10 mmol/L时的影响情况。结果,随着浓度增加,As(III)峰面积增大,但As(III) 与AsB分离度下降。综合考虑峰面积和分离效果,PH 9.0时,7 mmol/L的效果较好。
考察流动相A的pH 8.5,浓度为1、3、5、7和10 mmol/L时的影响情况。结果,仅当流动相A为1 mmol/L时,As(III) 和AsB能良好分离,其他浓度均未能完全分离。
比较7 mmol/L、pH 9.0和1 mmol/L、pH 8.5,发现7 mmol/L、pH 9.0时基线噪声较大,形态砷的峰面积较低,因此选择流动相A浓度为1 mmol/L,pH 8.5。
国标方法中,原子荧光蠕动泵转速为60 r/min,液相色谱洗脱程序为:0~8 min,100% 流动相A;10~20 min,100% 流动相B;22~32 min,100% 流动相A。此条件下,分析时间长,试剂消耗量大。本试验转速降至45 r/min,洗脱程序改为表1所示的梯度程序时,能大大缩短分析时间,减少HCl溶液和KBH4溶液的消耗量,节约试剂,提高工作效率。且四个形态砷的响应值、标准曲线都良好。因此,选择转速为45 r/min,洗脱程序为表1的梯度程序。
用优化后条件测定0~100 μg/L的 As(III)、DMA、MMA、As(V) 混合标准系列,以峰面积为纵坐标,浓度为横坐标,绘制标准曲线。以3倍、10倍噪音水平所相当的浓度计算检出限、定量限。As(III)、DMA、MMA、As(V)的相关系数均大于0.999;检出限为0.02 mg/kg,定量限为0.05 mg/kg。见表2
以海蛏、鱼为本底,分别加入20、40和80 μg/L三种浓度的砷标准溶液,按方法进行前处理后检测,回收率见表3。As(III)、DMA、MMA、As(V)的加标回收率为81.2%~111.6%。对海蛏为本底,加砷标准溶液40 μg/L的样品提取液,重复测定6次,计算相对标准偏差RSD。As(III)、DMA、MMA、As(V)出峰时间的RSD各为0.6%、0.3%、0.3%、0.4%;峰面积的RSD各为4.1%、4.2%、1.7%、4.5%。可见,峰面积及保留时间的RSD均小于5%,变异程度较小。
以优化条件测定100 μg/L砷标准溶液,色谱图如图7。As(III)、DMA、MMA、As(V)的出峰时间分别为2.767、6.576、8.108和12.474 min。
海产品中的砷包含多种形态,AsB是海产品中砷的主要存在形式,通常占总砷的90%以上,不同形态的砷的毒性不一[3-5]。因此,在评估海产品中砷的污染及毒性时,不应以总砷含量进行判定,而应测定形态砷尤其是无机砷含量。
实际样品测定也证实了这一点,采用ICP-MS方法对64份泉州市本地海产品的总砷含量进行检测,结果为0.42~65.5mg/kg,有62份总砷含量超过0.5 mg/kg(总无机砷限量标准)。进一步采用本文的方法对这62份海产品进行形态分析,结果发现:As(III)、DMA、MMA、As(V)检出份数分别为2、4、6和19份,检测值为0.09~0.13mg/kg、0.06~0.16mg/kg、0.05~0.15 mg/kg、0.05~0.27 mg/kg。总无机砷超标的有6份,含量为0.05~0.27 mg/kg。由此可见,海产品总砷含量虽高,但大部份是AsB,而As(III)、DMA、MMA、As(V)不高。实验结果证实了海产品中总砷含量高时,需要进一步检测形态砷,才能更好地评估海产品中砷的污染及毒性。
本文优化了LC-AFS联用仪的实验条件,建立了用LC-AFS联用技术对海产品中常见的四种砷形态As(III)、DMA、MMA、As(V)进行有效分离和检测的方法。用建立的方法进行测定,四种形态砷在14 min内分离。在0~100 μg/L范围,线性良好,检出限低,精密度高,结果准确可靠。
