Objective To establish a method for the rapid detection of 15 N-nitrosamines in bacon by enhanced matrix removal dispersive solid phase extraction combined with gas chromatography-tandem mass spectrometry (GC-MS/MS). Methods The samples were extracted with acetonitrile oscillation, frozen for 30 minutes, and then centrifuged at low temperature. The extract was further purified using an enhanced lipid removal purification tube (QuEChERS EMR-lipid) and an EMR-lipid stripping tube. The N-nitrosamines were separated on a polar capillary column, detected in the multiple reaction monitoring (MRM) mode and quantified by the matrix-matched external standard method. Results The linear correlation coefficients of the method were greater than 0.990 in the range of 0.2-40.0 μg/L, with the detection and quantification limits of 0.03-0.30 and 0.1-1.0 μg/kg, respectively. The recoveries were 64.1%-106.1% at three spiked levels (2.0, 5.0, 10.0 μg/kg),with the relative standard deviations (RSDs) ranging from 3.48% to 14.55%. Conclusion The method does not require activation, equilibration, or elution steps, consumes less solvent, is simple and rapid, and is suitable for the rapid detection of 15 N-nitrosamines in bacon.

目的 建立腊肉中15种N-亚硝胺类化合物的增强型基质去除分散固相萃取结合气相色谱串联质谱(GC-MS/MS)快速测定方法。方法 样品经乙腈振荡提取,冷冻30 min后低温离心,提取液经增强型脂质去除净化管(QuEChERS EMR-Lipid)和EMR-Lipid反萃管进一步净化,目标物经极性毛细管柱分离后以多反应监测模式(MRM)检测和基质匹配外标法定量。结果 在0.2~40.0 μg/L范围内线性相关系数均大于0.990,方法检出限及定量限分别为0.03~0.30 μg/kg和0. 1~1.0 μg/kg。三个水平(2.0、5.0和10.0 μg/kg)的加标回收率范围为64.1%~106.1%,相对标准偏差为3.48%~14.55%。结论 本法无需活化、平衡、洗脱步骤,消耗溶剂较少,操作简单,适用于腊肉中N-亚硝胺类化合物的快速检测。

Objective To establish a method for the rapid detection of 15 N-nitrosamines in bacon by enhanced matrix removal dispersive solid phase extraction combined with gas chromatography-tandem mass spectrometry (GC-MS/MS). Methods The samples were extracted with acetonitrile oscillation, frozen for 30 minutes, and then centrifuged at low temperature. The extract was further purified using an enhanced lipid removal purification tube (QuEChERS EMR-lipid) and an EMR-lipid stripping tube. The N-nitrosamines were separated on a polar capillary column, detected in the multiple reaction monitoring (MRM) mode and quantified by the matrix-matched external standard method. Results The linear correlation coefficients of the method were greater than 0.990 in the range of 0.2-40.0 μg/L, with the detection and quantification limits of 0.03-0.30 and 0.1-1.0 μg/kg, respectively. The recoveries were 64.1%-106.1% at three spiked levels (2.0, 5.0, 10.0 μg/kg),with the relative standard deviations (RSDs) ranging from 3.48% to 14.55%. Conclusion The method does not require activation, equilibration, or elution steps, consumes less solvent, is simple and rapid, and is suitable for the rapid detection of 15 N-nitrosamines in bacon.

腊肉是我国传统且具有独特风味的肉制品之一,贵州腊肉的制作过程也非常独特,一般选用当地的木材和柴火进行长时间的烟熏,是当地的特色美食之一[1-2]。硝酸盐因具有护色、抑菌、改良肉质等作用,在腊肉的生产加工过程中硝酸盐常被作为主要原材料[3],由于高盐腌制,以及烟熏、发酵等过程,如果控制不当,亚硝酸盐在酸性条件下会和胺类物质形成致癌物亚硝胺,食用后存在健康隐患。
目前,腊肉中亚硝胺检测主要参照《食品中N-亚硝胺类化合物的测定》(GB 5009.26—2023),但标准中只针对N-二甲基亚硝胺(NDMA)[4]。同时该标准中规定四种检测方法,其中气相色谱-热能分析法虽具有检测快速、灵敏度高等优点,但其价格昂贵,应用范围窄[5]。水蒸气蒸馏法虽净化效果好,缺点是需要的样品量较大,分析周期较长,溶剂消耗量较大,操作繁琐,关键步骤较多[6]。而QuEChERS-气相色谱-质谱法中虽然应用较广,但需要配备大体积程序升温进样口,一般实验室并未配备,因此国家标准检测方法还有待进一步优化。
样品前处理是腊肉检测中的关键和难点[7],由于其基质复杂,脂肪和蛋白质的含量较高,以及N-亚硝胺化合物都易溶于水、易挥发,导致在检测过程中,经常出现油脂难以净化完全,目标物容易损失,难以准确定量[8-9]。目前,商品化的适用于肉制品的净化材料众多,在肉制品净化方面都有一定的应用[10-11],本研究通过比较肉制品中目前常用的净化方法,确定最优净化方式并结合GC-MS/MS快速检测腊肉中15种N-亚硝胺化合物,同时对贵州省2024年食品安全风险监测地方专项所收集的130份代表性腊肉样品中N-亚硝胺类化合物进行检测,发现我省腊肉中N-亚硝胺化合物种类,为其他肉制品中N-亚硝胺的测定和法规完善提供参考。
Scion-TQ气相色谱质谱联用仪(美国BRUKER公司);FW100型高速万能粉碎机(天津市泰斯特仪器有限公司);DMV-16型涡旋混合器(广东省科寅实验室设备有限公司);Sigma3-18K型台式高速冷冻离心机(德国西格玛公司)。
12种亚硝胺类混合标准溶液(100 mg/L,上海安谱实验科技股份有限公司);N-亚硝基乙基异丙胺(100 mg/L,纯度>99.8%,曼哈格生物科技有限公司);N-亚硝基二异丙基胺(10 mg,纯度>99.5%,北方伟业计量技术研究院);N-亚硝基二异丁胺(100 mg/L,纯度>99.8%);标准品试剂:乙腈、甲醇、二氯甲烷、乙酸乙酯(色谱纯,纯度均>99.8%,美国TEDIA公司)。Cleanert亚硝胺专用检测管(含分散固相萃取管及Nitro亚硝胺专用净化管,博纳艾杰尔科技有限公司),dSPE亚硝胺检测萃取净化管(含dSPE分散固相萃取管,吡咯烷酮-聚苯乙烯二乙烯基苯净化管,dSPE基质分散固相萃取管,上海安谱实验科技股份有限公司);PLS-A QuEChERS净化管(150 mg,迪马科技有限公司);EMR-Lipid dSPE增强型基质去除净化管及反萃管(安捷伦科技有限公司);Clean-up LPAS多功能净化柱(500 mg/5 ml,北京科德诺斯技术有限公司);HLB固相萃取柱(200 mg/6 ml,安捷伦科技有限公司);Captiva EMR-Lipid HF固相萃取柱(600 mg/6 ml,安捷伦科技有限公司)。
将N-亚硝基二异丙基胺标准品用乙腈溶解后通过稀释配制成浓度为100.0 mg/L标准储备液,然后分别吸取3种亚硝胺单个标准溶液及12种亚硝胺类混合标准溶液各1.00 ml,稀释配制成浓度为1.00 μg/ml的15种亚硝胺混合标准储备液,于-18℃保存,用乙腈稀释成0.20 μg/L~40.0 μg/L的标准系列溶液。空白基质溶液经氮气吹干后分别加入1.00 ml上述混合标准工作液复溶后配制成基质匹配混合标准工作溶液系列。
来自于贵州省2024年食品安全风险监测地方专项,由贵阳市、遵义市、毕节市、黔东南州、铜仁市、黔西南州、黔南州、安顺市、六盘水市9个行政区域大型超市采集的130份腊肉样品。选取250 g以上代表性样品切块后置于粉碎机,捣碎后均匀制样置于玻璃样品瓶中,密封,做好标识,于-18 ℃冷冻保存备用。
HP-INNO Wax毛细管色谱柱(30 m×0.25 mm,0.25 μm),载气为高纯氦气,碰撞气为高纯氩气;程序升温条件:起始温度为60 ℃,保持1 min,以10 ℃/min速率升高至140 ℃,以5 ℃/min速率升高至150 ℃保持2.0 min,以20 ℃/min升至250 ℃保持7 min,共25.0 min,溶剂延迟时间为4.5 min;进样口温度为250 ℃,流速为1.0 ml/min,不分流进样,进样量为1.0 μl。
质谱离子源温度为200 ℃,传输线温度为250 ℃;电离模式为电子轰击离子化(EI),电离能量为70 eV;多反应监测模式(MRM);方法的质谱参数见表1
称取5.00 g腊肉样品置于50 ml离心管中,加入10 ml水和10 ml乙腈,涡旋1min,加入6颗玻璃珠均质子,再加入提取包(含4.0 g无水硫酸镁和1.0 g氯化钠),高速振荡1 min,以10 000 r/min条件(离心半径9.5 cm)离心5 min,上清液待净化。取不同体积的上述净化液分别加入到对应的净化管中(Agela取1 ml加入Cleanert Nitro净化管,CNW取5ml加入吡咯烷酮-聚苯乙烯二乙烯基苯净化管、净化前需加5 ml水活化,Dima取5ml加入PLS-A净化管),涡旋混匀后8 000 r/min离心5 min,移取全部净化液于基质分散固相萃取管中(Agela可跳过此步骤直接过滤后上机测定,CNW 和Dima萃取管内为1.6 g硫酸镁和0.4 g氯化钠),振荡提取2 min后放入冷冻离心机中,以8 000 r/min(离心半径9.5 cm)条件于4 ℃离心5 min,取上清液约1.0 ml过0.22 μm针式过滤器后上机测定。
称取5.00 g腊肉样品于50 ml离心管中,加入10 ml乙腈及6颗玻璃珠,振荡提取3 min后放入-20 ℃冰箱冷冻30 min,振荡提取2 min,以8 000 r/min于0 ℃离心5 min,向增强型基质去除净化管中加入5 ml水,高速振荡1 min,取5 ml提取液加入净化管中,振荡提取2 min,以8 000 r/min(离心半径9.5 cm)条件于0 ℃离心5 min,将离心所得全部上清液倒入15 ml反萃管中,高速振荡2 min,以8 000 r/min(离心半径9.5 cm)条件于0 ℃离心5 min,取上清液约1.0 ml过0.22 μm针式过滤器后上机测定。
称取5.00 g腊肉样品于50 ml离心管中,加入10 ml水后高速振荡2 min,加入10 ml乙腈,振荡提取2 min,加入4.0 g无水硫酸镁和1.0g氯化钠,涡旋1 min,以10 000 r/min(离心半径9.5 cm)条件离心5 min,取不同体积的上清液于对应的固相萃取柱上(HLB固相萃取柱上样2 ml,LPAS多功能净化柱上样5 ml后推动柱塞杆以1滴/s的速度收集净化液,Captiva EMR-Lipid HF上样5 ml后依靠重力自流),收集净化液过0.22 μm针式过滤器后上机测定。
提取溶剂是影响回收率的关键因素,15种N-亚硝胺类化合物的极性较强,多易溶于水、醇、醚等[12],本研究利用空白腊肉样品加标方式分别比较二氯甲烷、乙腈、乙酸乙酯、甲醇四种提取溶剂的提取效率,结果见图1
结果表明,当甲醇为提取剂时,盐析后甲醇与水无法分层,无法进行后续操作;当二氯甲烷、乙酸乙酯为提取剂时,N-亚硝基二甲胺、N-亚硝胺吗啉和N-亚硝胺哌啶三种化合物提取回收率较低,其次由于这两种溶剂的极性较弱,易提取出更多油脂,出现严重乳化现象,不利于后续净化过程;当乙腈为提取剂时,目标物回收率较好,且乙腈可沉淀蛋白。因此,选择乙腈为提取溶剂。
腊肉样品中存在脂肪、色素、甾醇等杂质,N-亚硝胺化合物易溶于水、易挥发,导致在检测过程中,经常出现油脂难以净化完全[13]。目前,腊肉基质常用的净化方式有QuEChERS、dSPE和SPE法,本研究分别选取7份腊肉空白样品,分别根据“1.5”所述进行样品前处理,在全扫描模式(扫描范围:50~500 m/z)下分别测定,不同净化方式的总离子流色谱图见图2
图2可见,CNW QuEChERS、EMR-Lipid dSPE净化法都可有效降低杂质干扰,净化效果较好,最终确定以EMR-Lipid dSPE为净化柱。腊肉中脂肪含量较高,通常可利用冷冻和低温离心的方法使脂肪从提取液中析出并分离,一般冷冻时间30 min后除脂效果会更加彻底[14],本研究利用腊肉空白样品,按照“1.5.2”节进行操作,在全扫描模式(扫描范围:50~500 m/z)测定,分别对冷冻/低温离心(-20 ℃冰箱冷冻30 min,0 ℃下离心)与常温下操作的除脂效果进行比较,结果表明,腊肉样品经冷冻过后的除脂效果较好,样品全扫谱图基线较平,杂质干扰较小,所以后续实验中样品提取后在放入-20 ℃冰箱冷冻30 min,并采用低温离心。
15种N-亚硝胺化合物分子量和极性差异较大[15],分别比较了DB-5MS、DB-1701MS和HP-INNOWAX色谱柱的分离效果。结果表明,DB-5MS柱和DB-1701MS柱的分离效果较差,特别是NDMA的保留时间短,与溶剂峰近,有拖尾现象且易受杂质峰干扰;15种目标物在HP-INNOWAX柱峰形较好且在25 min内都能有很好的分离,腊肉加标样品的总离子流色谱图见图3
N-亚硝胺类化合物检测通常是按参考文献4中以N-二甲基亚硝胺-D6为内标,其N-二甲基亚硝胺-D6的定性及定量离子对参数(74.0>44.0,碰撞能量3 V;74.0>42.1,碰撞能量19 V),但实验中发现,如按该参数设置,N-二甲基亚硝胺和N-二甲基亚硝胺-D6中存在离子串扰现象,严重影响定量的准确性,且N-二甲基亚硝胺分子量较小,特异性的离子对参数较少,所以采用外标法定量。
基质效应一直影响质谱分析定量的准确性[16-17],通常基质效应以相对响应值法来评价,即基质标准溶液的峰面积与溶剂标准溶液的峰面积的比值乘以100%。当基质效应<80%时,表现为基质抑制效应;当基质效应在 80%~120%范围时,基质效应可忽略不计;当基质效应>120%时,表现为基质增强效应[18-19]。本研究分别取5个批次不同厂家腊肉样品进行混匀制备成混合样品基质,在其中添加混合标准(5 μg/kg)后分别按照“1.5”中的7种方式进行操作,每种方式平行测定5次后计算基质效应。结果表明,腊肉基质效应干扰比较严重,未经净化操作很难进行定量分析。当采用QuEChERS(Agela、Dima)、SPE(LPAS、HLB)净化时15种目标物中基质效应在0~80.0%分别有2、2、2和1个,只有经EMR-Lipid dSPE净化后的15种目标物的基质效应均在80.1%~120.0%,能更有效地减小基质效应。
取空白样品分别加入6个不同浓度的混合标准溶液,按本法测定。以3倍和10倍信噪比(S/N)对应的含量确定方法检出限和定量限。为验证方法的准确度与精密度,取数份腊肉空白样品进行添加回收实验,添加水平为2.00、5.00和10.0 μg/kg,放置隔夜后平行测定6次。结果表明,15种目标物线性关系良好(相关系数r2>0. 99)。方法检出限及定量限分别为0.03~0.3 μg/kg和0.1~1.0 μg/kg;加标回收率为64.1%~106.1%,相对标准偏差(relative standard deviation,RSD)为3.48%~14.55%。见表2
采用本法测定了130份腊肉样品,结果表明,在6份腊肉样品中各检出至少1种亚硝胺化合物残留,检出率最高的分别是NDMA、NDIBA、NMPHA及NDPHA,其中检出NDMA的残留量在0~10.0 μg/kg,超过GB 2762—2022《食品安全国家标准 食品中污染物限量》中的限量值(NDMA<3.0 μg/kg),这可能与腊肉独特的加工工艺有关[20]
本研究建立了增强型基质去除技术结合GC-MS/MS快速筛查贵州产腊肉中15种挥发性N-亚硝胺化合物残留的方法。该方法前处理操作便捷,基质净化效果较好,易于推广,适合腊肉中挥发性N-亚硝胺化合物的批量检测。
