Objective

To investigate the veterinary drug residue status of commercially available freshwater fish in Henan Province from 2021 to 2024 and evaluate the dietary exposure risk.

Methods

A total of 186 samples of freshwater fish were randomly collected from 2021 to 2024, and six quinolone antibiotics, two anesthetics and two sedative-hypnotics were detected according to the standard procedures in the “National Food Contamination and Hazardous Factors Risk Monitoring Operation Manual”. General descriptive analysis and χ2 tests were performed using Excel and SAS software. The probability distribution of drug exposure was calculated using Monte Carlo combined with the consumption of freshwater fish in Henan Province, and the health risk was evaluated using the hazard quotient (HQ).

Results

A total of 152 out of 186 freshwater fish samples were detected with veterinary drug residues, with a detection rate of 81.72%; 39 samples were unqualified, with an unqualified rate of 20.96%. The veterinary drug detection rates in descending order were: enrofloxacin, eugenol > diazepam > ofloxacin, oxazepam (χ2=309.867, P < 0.001), and the unqualified rate of diazepam was the highest (χ2=61.822, P < 0.001). There were significantly statistical differences in the detection rates and unqualified rates in different years (χ2=24.962; χ2=19.731, all P < 0.001). There were no statistically significant differences in the detection rates and unqualified rates of veterinary drugs in freshwater fish between farmers’ markets and stores (χ2=0.806, P=0.369; χ2=2.788, P=0.095). The P95 exposure amount of enrofloxacin in freshwater fish consumed by adults in Henan Province was 0.11 μg/(kg·BW), accounting for 1.77% of the ADI. The P95 exposure of eugenol was 4.02 μg/(kg·BW), accounting for 0.16% of the ADI.

Conclusion

Veterinary drug residues were present in freshwater fish sold in Henan Province, but the overall exposure risk was low, with all HQ less than 1.

目的

了解2021—2024年河南省市售淡水鱼兽药残留状况,并评估膳食暴露风险。

方法

2021—2024年随机采集淡水鱼样品186份,依据《国家食品污染和有害因素风险监测工作手册》中的标准程序检测6种喹诺酮类抗生素、2种麻醉剂和2种镇静催眠剂,通过Excel和SAS软件进行一般性描述分析和χ2检验;结合河南省居民淡水鱼消费量,运用蒙特卡洛原理模拟计算兽药暴露量的概率分布,采用危害商评估健康风险。

结果

186份淡水鱼样品中152份检出兽药残留,检出率为81.72%;不合格样品39份,不合格率为20.97%。兽药检出率从高到低依次为恩诺沙星、丁香酚>地西泮>氧氟沙星、奥沙西泮(χ2=309.867,P<0.001),地西泮的不合格率最高(χ2=61.822,P<0.001)。不同年度的检出率和不合格率均有统计学差异(χ2=24.962、19.731,均P<0.001)。农贸市场和商店淡水鱼兽药的检出率和不合格率差异均无统计学意义(χ2=0.806,P=0.369;χ2=2.788,P=0.095)。河南省成年人食用淡水鱼恩诺沙星的P95暴露量为0.11 μg/(kg·BW),占每日允许摄入量(ADI)的1.77%。丁香酚P95暴露量为4.02 μg/(kg·BW),占ADI的0.16%。

结论

河南省市售淡水鱼存在一定的兽药残留,但暴露风险总体较低,危害商均小于1。

目的

了解2021—2024年河南省市售淡水鱼兽药残留状况,并评估膳食暴露风险。

方法

2021—2024年随机采集淡水鱼样品186份,依据《国家食品污染和有害因素风险监测工作手册》中的标准程序检测6种喹诺酮类抗生素、2种麻醉剂和2种镇静催眠剂,通过Excel和SAS软件进行一般性描述分析和χ2检验;结合河南省居民淡水鱼消费量,运用蒙特卡洛原理模拟计算兽药暴露量的概率分布,采用危害商评估健康风险。

结果

186份淡水鱼样品中152份检出兽药残留,检出率为81.72%;不合格样品39份,不合格率为20.97%。兽药检出率从高到低依次为恩诺沙星、丁香酚>地西泮>氧氟沙星、奥沙西泮(χ2=309.867,P<0.001),地西泮的不合格率最高(χ2=61.822,P<0.001)。不同年度的检出率和不合格率均有统计学差异(χ2=24.962、19.731,均P<0.001)。农贸市场和商店淡水鱼兽药的检出率和不合格率差异均无统计学意义(χ2=0.806,P=0.369;χ2=2.788,P=0.095)。河南省成年人食用淡水鱼恩诺沙星的P95暴露量为0.11 μg/(kg·BW),占每日允许摄入量(ADI)的1.77%。丁香酚P95暴露量为4.02 μg/(kg·BW),占ADI的0.16%。

结论

河南省市售淡水鱼存在一定的兽药残留,但暴露风险总体较低,危害商均小于1。

Objective

To investigate the veterinary drug residue status of commercially available freshwater fish in Henan Province from 2021 to 2024 and evaluate the dietary exposure risk.

Methods

A total of 186 samples of freshwater fish were randomly collected from 2021 to 2024, and six quinolone antibiotics, two anesthetics and two sedative-hypnotics were detected according to the standard procedures in the “National Food Contamination and Hazardous Factors Risk Monitoring Operation Manual”. General descriptive analysis and χ2 tests were performed using Excel and SAS software. The probability distribution of drug exposure was calculated using Monte Carlo combined with the consumption of freshwater fish in Henan Province, and the health risk was evaluated using the hazard quotient (HQ).

Results

A total of 152 out of 186 freshwater fish samples were detected with veterinary drug residues, with a detection rate of 81.72%; 39 samples were unqualified, with an unqualified rate of 20.96%. The veterinary drug detection rates in descending order were: enrofloxacin, eugenol > diazepam > ofloxacin, oxazepam (χ2=309.867, P < 0.001), and the unqualified rate of diazepam was the highest (χ2=61.822, P < 0.001). There were significantly statistical differences in the detection rates and unqualified rates in different years (χ2=24.962; χ2=19.731, all P < 0.001). There were no statistically significant differences in the detection rates and unqualified rates of veterinary drugs in freshwater fish between farmers’ markets and stores (χ2=0.806, P=0.369; χ2=2.788, P=0.095). The P95 exposure amount of enrofloxacin in freshwater fish consumed by adults in Henan Province was 0.11 μg/(kg·BW), accounting for 1.77% of the ADI. The P95 exposure of eugenol was 4.02 μg/(kg·BW), accounting for 0.16% of the ADI.

Conclusion

Veterinary drug residues were present in freshwater fish sold in Henan Province, but the overall exposure risk was low, with all HQ less than 1.

随着全球水产养殖业的快速发展,淡水鱼作为重要的蛋白质来源,在满足人类日益增长的食品需求方面发挥着重要作用。然而,为了提高养殖效率和预防疾病,部分养殖户和商家可能会违规使用药物,其中喹诺酮类、丁香酚类和地西泮类药物的使用尤为引人关注。这些药物虽然在短期内能够有效控制疾病,提高鱼虾存活率,但残留可能对人类健康和生态环境构成潜在威胁。
喹诺酮类药物作为一类高效广谱抗菌剂,在水产养殖中被广泛用于防治细菌性疾病。然而,大量研究表明[1-2],喹诺酮类药物残留可能导致细菌耐药性增加,进而降低人类临床治疗的效果。丁香酚类化合物具有麻醉、抗应激等特性,广泛用于鱼类的运输和暂养过程中。有报道[3],丁香酚具有肝毒性、胚胎毒性、并能导致代谢性酸中毒。地西泮类化合物作为一种苯二氮卓类中枢神经系统抑制剂,被用于水产养殖和运输过程中。然而,地西泮类药物在鱼体内残留是永久性的,可通过食物链传递给人类,可能引起人体嗜睡疲乏、动作失调、精神混乱等症状[4]
近年来,多地[5-7]的淡水鱼检测结果显示,喹诺酮类药物尤其是恩诺沙星(恩诺沙星与环丙沙星之和)的使用频率较高。此外,丁香酚类和地西泮类药物在鱼类运输过程中具有显著的镇静等作用,应用前景广阔,已成为研究热点。值得注意的是,我国目前对丁香酚作为渔用麻醉剂的使用尚无明确规定。然而,河南省关于淡水鱼中上述兽药残留的研究相对较少。为全面了解河南省市售淡水鱼中兽药残留现状,本研究对2021—2024年河南地区淡水鱼中10种兽药(包括喹诺酮类、丁香酚类、地西泮类)的监测结果进行分析,并采用概率评估方法对残留兽药的膳食暴露风险进行科学评估,旨在为有关部门加强监管和制定限量标准提供科学依据和数据支撑。
参考国家食品化学污染物监测计划,结合河南省地域特征和人口分布情况,2021—2024年在河南省郑州市、济源市、开封市、洛阳市、南阳市、平顶山市、信阳市、新乡市、周口市、安阳市、漯河市、三门峡市和商丘市共计13个地市随机采集鲜活淡水鱼样品186份,其中农贸市场102份,商店(超市、便利店、专营店)84份。2021年45份,2022年45份,2023年46份,2024年50份。
参照《国家食品污染和有害因素风险监测工作手册》的标准程序分别监测淡水鱼中6种喹诺酮类抗生素(恩诺沙星、环丙沙星、诺氟沙星、氧氟沙星、培氟沙星、洛美沙星)、2种麻醉剂[丁香酚、3-氨基苯甲酸乙酯甲基磺酸盐(MS-222)]、2种镇静催眠剂(地西泮、奥沙西泮),采用超高效液相色谱-串联质谱法分别进行测定,内标法定量。
依据《食品安全国家标准 食品中兽药最大残留限量》(GB 31650—2019)[8]中恩诺沙星(恩诺沙星与环丙沙星之和)≤100 μg/kg、地西泮不得检出(允许做治疗使用,但不得在动物性食品中检出),《食品安全国家标准 食品中41种兽药最大残留限量》(GB 31650.1—2022)[9]中洛美沙星、诺氟沙星、氧氟沙星、培氟沙星均为≤2 μg/kg,农业农村部第176号公告《禁止在饲料和动物饮用水中使用的药物品种目录》[10]中奥沙西泮禁止使用。对有限量标准的兽药,残留量超过标准限值即判定为不合格;禁止检出的兽药,检出则判定为不合格。丁香酚在水产品的应用我国目前暂无相关标准,因此暂不做合格评价,仅考虑检出情况。
采用联合国粮食及农业组织和世界卫生组织(FAO/WHO)推荐的《食品中化学物风险评估原则和方法》中概率评估方法[11]建立淡水鱼中兽药摄入评估模型:EDIc=(Ci×Fi)/BWEDIc为人群通过淡水鱼暴露于兽药的量[μg/(kg·BW·d)],Ci为食品中兽药含量(μg/kg),Fi为食物消费量(g/d),BW为成年人体重(kg)。利用Crystal ball软件进行变量分布拟合,采用拉丁超立方抽样方法,随机抽取10 000次,利用Monte Carlo模拟得到成人通过淡水鱼兽药暴露量概率分布。
用危害商(hazard quotient,HQ)评估食用淡水鱼中兽药暴露风险,计算公式为:HQ=EDIc/ADIADI(每日允许摄入量)为健康指导值。HQ<1,认为药物暴露风险较低,可以接受;HQ≥1,认为药物暴露风险较高,应予以关注。
用EXCEL录入样品信息及数据信息,用SAS 9.2软件进行统计分析,用平均值、中位数、P75P90P95、最大值进行定量描述,用χ2检验或Fisher确切概率法比较率间差异,检验水准α=0.05。
分析实际样品前,对检测方法进行方法确认,确保其适用性和可靠性;参加国家有关机构组织的能力验证,保证实验室达到可接受的准确度和精密度水平。实际分析样品时,每15个样品做一次方法空白试验、一个平行样、一个标准点、一个加标样品、一个质控样品(标准参考物质),以监测实验过程的稳定性和准确性,各试验结果应在预期范围内,否则需要查找原因并重新进行实验。
186份淡水鱼样品共152份检出兽药残留,检出率为81.72%;不合格样品39份,不合格率为20.97%。检出样品中,64份样品检出2种兽药,占检出样品的42.10%;15份样品检出3种兽药,占检出样品的9.87%。不合格样品中,5份样品恩诺沙星超出限量标准,占比为12.82%;29份样品检出地西泮,占比为74.36%;3份样品检出奥沙西泮,占比为7.69%;2份样品检出地西泮及恩诺沙星超出限量标准,占比为5.13%。
共检出5种兽药残留,恩诺沙星、氧氟沙星、丁香酚、地西泮、奥沙西泮的检出率差异有统计学意义(χ2=309.867,P<0.001),检出率从高到低依次为恩诺沙星、丁香酚>地西泮>氧氟沙星、奥沙西泮。3种兽药残留不符合国家限量标准,恩诺沙星、地西泮、奥沙西泮的不合格率差异有统计学差异(χ2=61.822,P<0.001),地西泮的不合格率最高。诺氟沙星、培氟沙星、洛美沙星、MS-222没有检出。见表1
2021—2024年淡水鱼中兽药的检出率分别为60.00%(27/45)、77.78%(35/45)、97.83%(45/46)、90.00%(45/50),不合格率分别为4.44%(2/45)、31.1%(14/45)、37.8%(17/46)、12.0%(6/50)。年度检出率和不合格率差异均有统计学意义(χ2=24.962、19.731,P均<0.001)。其中,2023和2024年的检出率高于2021和2022年,2023年的不合格率最高。见表2
农贸市场淡水鱼样品的兽药检出率为79.41%(81/102),不合格率为25.49%(26/102);商店(超市、便利店、专营店)的检出率为84.52%(71/84),不合格率为15.48%(13/84)。农贸市场和商店淡水鱼兽药的检出率和不合格率差异均无统计学意义(χ2=0.806,P= 0.369;χ2=2.788,P=0.095)。见表3
本研究仅对检出率较高且具有ADI值的恩诺沙星和丁香酚开展暴露评估。暴露风险评估中的变量拟合分布见表4。由表5可见,河南省成年人每天通过淡水鱼恩诺沙星的平均暴露量为0.03 μg/(kg·BW),占ADI(6.2 [μg/(kg·BW·d)])的0.48%,P95暴露量为0.11 μg/(kg·BW),占ADI的1.77%。河南省成年人每天通过淡水鱼丁香酚的平均暴露量为1.25 μg/(kg·BW),占ADI(2.5 [mg/(kg·BW·d)])的0.05%,P95暴露量为4.02 μg/(kg·BW),占ADI的0.16%。河南省成年人食用淡水鱼暴露量均未超过对应的ADI值,HQ均小于1,提示河南省成年人通过淡水鱼暴露恩诺沙星、丁香酚的风险较低。
监测结果显示,186份样品中兽药的检出率为81.72%,提示淡水鱼中兽药的普遍使用。检出样品中,检出2种及2种以上兽药比例为51.97%;不合格样品中,两种兽药均不合格占比为5.13%。提示需警惕联合用药所带来的毒性增强、耐药性增加等多种复杂风险。
监测结果显示,淡水鱼中恩诺沙星和丁香酚的检出率均大于50%,分别为60.22%和53.22%,恩诺沙星的不合格率为3.77%。表明这两种药物在水产养殖和运输中被广泛使用。对比国内其它省、市研究结果[12-14],表明不同地区的检测结果存在一定的差异。这种差异与多方面的因素有关,一是各地不同的养殖模式和用药习惯是影响兽药残留的关键因素。池塘养殖、网箱养殖等不同的养殖方式、预防用药和规范用药等不同的给药习惯直接影响药物的使用频率和残留水平。二是土壤和水体等环境因素对兽药的残留水平也有重要影响。不同地区水质、土壤成分的差异,可能会影响药物的代谢和残留情况。三是各地监管力度存在差异。严格的监管能有效降低药物滥用和残留风险,而监管相对薄弱的地区可能面临着更高的药物残留。四是样本种类、采集时间不同也会导致检测结果的差异。不同种类的淡水鱼对药物的代谢能力不同,且药物残留水平可能随时间变化。恩诺沙星残留最大值为432 μg/kg,超过限量标准的4倍。丁香酚残留最大值为9 345 μg/kg,中位数为3.52 μg/kg,相差2 654倍,检出残留差异较大。我国于2021年发布了水产品中丁香酚的检测标准,但尚未制定其在淡水鱼中的残留标准。目前,世界各国对丁香酚能否作为鱼类麻醉剂存有争议,日本、澳大利亚、芬兰等国家允许丁香酚作为合法的水产麻醉剂,但通常要求经过一定的休药期后才可上市;欧盟、美国对丁香酚的使用则较为谨慎。有研究表明,丁香酚对啮齿动物有致癌性或潜在的致癌性[15],因此存在一定的安全隐患,亟待有关部门开展淡水鱼中丁香酚的健康风险评估并制定相应的残留限量标准,加强监管,确保人民群众的生命健康。
监测结果显示,地西泮的不合格率最高,为16.67%,残留最大值为11.80 μg/kg。奥沙西泮不合格率为1.61%,检出最大值为3.12 μg/kg。提示地西泮的使用存在违规现象,如不遵守休药期、超剂量使用等。地西泮化学性质稳定,半衰期长,在生物体内难以代谢掉。奥沙西泮作为地西泮的活性代谢产物,性质不如地西泮稳定,其残留量也相对较低。
监测结果显示,2023和2024年淡水鱼中兽药的检出率高于2021和2022年,年度检出率有增高的趋势,这可能是一是因为检测方法的灵敏度和准确性不断提高,能够检测到更低浓度的兽药残留,从而导致检出率上升。二是滥用兽药,尤其是用于活鱼运输的麻醉剂丁香酚因无限量标准,存在监管盲区,其在2023和2024年的检出率是2021和2022年的二倍左右。三是长期使用兽药可能导致环境中药物残留累积,进而通过土壤、水等途径污染养殖环境,最终影响淡水鱼质量。另外,2023年的不合格率最高,提示地西泮的违规使用,需引起高度重视并加以严格监管。
监测结果显示,农贸市场和商店市售淡水鱼中兽药检出率和不合格率差异均无统计学意义,表明这两种销售渠道均存在兽药残留问题,提示监管力度相当,养殖源头可能相同、运输和存养等条件也可能较为相似。
研究结果显示,河南省成年人每天通过淡水鱼恩诺沙星的P95暴露量为0.11 μg/(kg·BW),HQ为0.017 7;丁香酚的P95暴露量为4.02 μg/(kg·BW),HQ为0.001 6。两者的HQ均小于1,提示河南省食用淡水鱼人群恩诺沙星和丁香酚的健康暴露风险较低。本研究采用基于蒙特卡洛模拟的概率评估方法研究检出率较高的恩诺沙星和丁香酚的暴露风险,与传统的点评估相比较,该方法在样本量较少的情况下,通过计算不同结果的概率分布,能提供相对可靠和科学的评估,更好的反映实际情况,并更全面的反应不确定性,从而为风险管理和决策提供更有力的支持。
综上,本监测研究的是2021—2024年河南省13个地市市售淡水鱼的兽药残留,样品覆盖商店和农贸市场,代表性好;检测前接受全国统一质控,数据质量高;概率评估暴露风险更具科学性和实用性。但由于本次评估中使用的数据和资料的原因,存在一定的局限性和不确定性。一是未考虑未成年人的膳食暴露风险;二是评估模型中使用的膳食摄入量和体重数据来源于2015年河南省居民营养和健康监测,而非基于最新建立的完整的摄入量和体重数据库。随着时间的推移,居民的体重分布和膳食结构会发生变化,这可能会影响评估结果的准确性。此外,兽药暴露风险还来源于畜肉、禽肉等多种动物性食品,仅评估淡水鱼中兽药残留风险是不够的,需考虑多种动物性食品的累积暴露风险,以更全面的评估居民的健康风险。
