Objective

To explore the role and mechanism of ferroptosis in exacerbating airway inflammation in asthmatic mice exposed to traffic-related PM2.5.

Methods

Fifty BALB/c mice were randomly divided into five groups: saline (NS), asthma (ovalbumin, OVA), and OVA combined with low, medium, and high exposure to traffic-derived PM2.5 (1.8, 3.6, and 7.2 mg/kg·bw, respectively). The levels of IL-6 and TNF-α in lung tissue were measured using ELISA kits. The concentrations of reduced glutathione (GSH), malondialdehyde (MDA), and Fe2+ in lung homogenate supernatants were assessed by colorimetry. Protein expressions of prostaglandin-endoperoxide synthase 2 (PTGS2), acyl-CoA synthetase long chain family member 4 (ACSL4), glutathione peroxidase 4 (GPX4), tumor protein p53 (p53), phospho-p53 (p-p53), spermidine/spermine N1-acetyltransferase 1 (SAT1), and arachidonate 15-lipoxygenase (ALOX15) were detected using western blot. mRNA expressions of PTGS2, ACSL4, GPX4, p53, SAT1, and ALOX15 were examined by qRT-PCR. One-way ANOVA was used for multi-group data comparison, followed by Tukey’s HSD for further multiple comparisons.

Results

PM2.5 exposure significantly increased IL-6 and TNF-α levels in asthmatic mouse lung tissues, especially in the high PM2.5 exposure group (FIL-6=17.910, P<0.001; FTNF-α=5.414, P<0.05). All PM2.5 exposure groups showed a reduction in GSH, most notably in the high exposure group (FGSH=13.560, P<0.001). MDA and Fe2+ levels were significantly higher in the medium and high PM2.5 groups compared to the OVA group (FMDA=55.230, P<0.001; FFe=17.660, P<0.001). PM2.5 exposure also elevated the protein and mRNA expressions of PTGS2, ACSL4, and ALOX15, while reducing GPX4 expression. The protein and mRNA expression levels of p53, SAT1, and ALOX15 showed a clear dose-response to PM2.5, with p-p53 following a similar trend as p53 protein expression.

Conclusion

Traffic-related PM2.5 may induce ferroptosis in the lungs of asthmatic mice and exacerbate asthma inflammation through the p53/SAT1/ALOX15 pathway.

目的

探讨铁死亡在交通相关PM2.5加重哮喘小鼠气道炎症中的作用及其机制。

方法

50只BALB/c小鼠随机分为5组:生理盐水(NS)组、哮喘(卵清蛋白OVA)组、OVA联合交通源PM2.5低、中、高暴露(分别为1.8、3.6、7.2 mg/kg·bw)组。使用ELISA方法检测肺组织中IL-6和TNF-α的水平;比色法评估肺组织匀浆上清中的还原性谷胱甘肽(GSH)、丙二醛(MDA)以及Fe2+的水平;用western blot法检测前列腺素内过氧化物合酶2(PTGS2)、酰基辅酶A合成酶长链家族成员4(ACSL4)、谷胱甘肽过氧化物酶4(GPX4)和肿瘤蛋白p53(p53)、磷酸化p53(p-p53)、亚精胺/精胺 N1-乙酰转移酶1(SAT1)、花生四烯酸15-脂氧合酶(ALOX15)的蛋白表达,并通过qRT-PCR法检测以上除p-p53外其他指标的mRNA表达。采用单因素方差分析比较多组数据,用Tukey检验进一步多重比较。

结果

PM2.5刺激明显增加了哮喘小鼠肺组织中IL-6和TNF-α水平,尤其在PM2.5高暴露组(FIL-6=17.910,P<0.001;FTNF-α=5.414,P<0.05)。所有PM2.5暴露组的GSH均下降,其中PM2.5高暴露组的GSH降低最显著(FGSH=13.560,P<0.001)。PM2.5中、高暴露组MDA和Fe2+显著高于OVA组(FMDA=55.230,P<0.001;F=17.660,P<0.001)。PM2.5暴露后PTGS2、ACSL4、ALOX15的蛋白和mRNA表达增高,而GPX4的表达降低。p53、SAT1和ALOX15的蛋白及mRNA表达水平随PM2.5剂量增加而呈现明显的剂量效应,p-p53与p53的蛋白表达趋势一致。

结论

交通相关PM2.5可能通过p53/SAT1/ALOX15通路诱导哮喘小鼠肺部铁死亡并加重哮喘炎症。

目的

探讨铁死亡在交通相关PM2.5加重哮喘小鼠气道炎症中的作用及其机制。

方法

50只BALB/c小鼠随机分为5组:生理盐水(NS)组、哮喘(卵清蛋白OVA)组、OVA联合交通源PM2.5低、中、高暴露(分别为1.8、3.6、7.2 mg/kg·bw)组。使用ELISA方法检测肺组织中IL-6和TNF-α的水平;比色法评估肺组织匀浆上清中的还原性谷胱甘肽(GSH)、丙二醛(MDA)以及Fe2+的水平;用western blot法检测前列腺素内过氧化物合酶2(PTGS2)、酰基辅酶A合成酶长链家族成员4(ACSL4)、谷胱甘肽过氧化物酶4(GPX4)和肿瘤蛋白p53(p53)、磷酸化p53(p-p53)、亚精胺/精胺 N1-乙酰转移酶1(SAT1)、花生四烯酸15-脂氧合酶(ALOX15)的蛋白表达,并通过qRT-PCR法检测以上除p-p53外其他指标的mRNA表达。采用单因素方差分析比较多组数据,用Tukey检验进一步多重比较。

结果

PM2.5刺激明显增加了哮喘小鼠肺组织中IL-6和TNF-α水平,尤其在PM2.5高暴露组(FIL-6=17.910,P<0.001;FTNF-α=5.414,P<0.05)。所有PM2.5暴露组的GSH均下降,其中PM2.5高暴露组的GSH降低最显著(FGSH=13.560,P<0.001)。PM2.5中、高暴露组MDA和Fe2+显著高于OVA组(FMDA=55.230,P<0.001;F=17.660,P<0.001)。PM2.5暴露后PTGS2、ACSL4、ALOX15的蛋白和mRNA表达增高,而GPX4的表达降低。p53、SAT1和ALOX15的蛋白及mRNA表达水平随PM2.5剂量增加而呈现明显的剂量效应,p-p53与p53的蛋白表达趋势一致。

结论

交通相关PM2.5可能通过p53/SAT1/ALOX15通路诱导哮喘小鼠肺部铁死亡并加重哮喘炎症。

Objective

To explore the role and mechanism of ferroptosis in exacerbating airway inflammation in asthmatic mice exposed to traffic-related PM2.5.

Methods

Fifty BALB/c mice were randomly divided into five groups: saline (NS), asthma (ovalbumin, OVA), and OVA combined with low, medium, and high exposure to traffic-derived PM2.5 (1.8, 3.6, and 7.2 mg/kg·bw, respectively). The levels of IL-6 and TNF-α in lung tissue were measured using ELISA kits. The concentrations of reduced glutathione (GSH), malondialdehyde (MDA), and Fe2+ in lung homogenate supernatants were assessed by colorimetry. Protein expressions of prostaglandin-endoperoxide synthase 2 (PTGS2), acyl-CoA synthetase long chain family member 4 (ACSL4), glutathione peroxidase 4 (GPX4), tumor protein p53 (p53), phospho-p53 (p-p53), spermidine/spermine N1-acetyltransferase 1 (SAT1), and arachidonate 15-lipoxygenase (ALOX15) were detected using western blot. mRNA expressions of PTGS2, ACSL4, GPX4, p53, SAT1, and ALOX15 were examined by qRT-PCR. One-way ANOVA was used for multi-group data comparison, followed by Tukey’s HSD for further multiple comparisons.

Results

PM2.5 exposure significantly increased IL-6 and TNF-α levels in asthmatic mouse lung tissues, especially in the high PM2.5 exposure group (FIL-6=17.910, P<0.001; FTNF-α=5.414, P<0.05). All PM2.5 exposure groups showed a reduction in GSH, most notably in the high exposure group (FGSH=13.560, P<0.001). MDA and Fe2+ levels were significantly higher in the medium and high PM2.5 groups compared to the OVA group (FMDA=55.230, P<0.001; FFe=17.660, P<0.001). PM2.5 exposure also elevated the protein and mRNA expressions of PTGS2, ACSL4, and ALOX15, while reducing GPX4 expression. The protein and mRNA expression levels of p53, SAT1, and ALOX15 showed a clear dose-response to PM2.5, with p-p53 following a similar trend as p53 protein expression.

Conclusion

Traffic-related PM2.5 may induce ferroptosis in the lungs of asthmatic mice and exacerbate asthma inflammation through the p53/SAT1/ALOX15 pathway.

哮喘是以气道炎症为主的复杂多表型疾病,与空气污染、过敏原及微生物密切相关。全球约超过3.3亿人受到哮喘的困扰,过敏性哮喘是哮喘最常见的类型,我国过敏性哮喘占成年哮喘的50%以上[1]。因此,从哮喘加重的角度探索针对性的防控措施,对提升患者生活品质至关重要。
PM2.5来源广泛,与人类健康风险密切相关。交通排放是城市PM2.5的主要来源,流行病学研究显示,PM2.5浓度上升与哮喘就诊率增加相关[2]。Schultz等人的研究表明与工业来源相比,道路交通来源的PM2.5与哮喘的关联更加显著[3]。实验研究表明PM2.5暴露可以加剧小鼠肺部炎症,导致促炎因子如TNF-α、IL-6水平显著升高[4]。课题组前期发现,交通来源的PM2.5可引起哮喘大鼠脾脏的炎症反应,导致免疫功能损伤[5]。以上表明交通来源的PM2.5与哮喘的发生发展关系密切。
研究发现PM2.5暴露导致人支气管上皮(BEAS-2B)细胞铁积累和脂质过氧化,引发与铁死亡相关的变化[6],提示PM2.5暴露可诱发铁死亡。然而铁死亡在交通源PM2.5诱导哮喘加重中的角色尚不明确。最新研究发现哮喘小鼠模型中出现铁死亡现象,抑制铁死亡可以缓解小鼠气道炎症反应从而改善哮喘,表明铁死亡在哮喘的发展中可能发挥着重要作用[7]。铁死亡的调控途径包括XC -/GSH/GPX4、p62/Keap1/Nrf2、p53/SAT1/ALOX15等[8]。p53是铁死亡的核心调控基因,在诱导细胞脂质过氧化和铁死亡中发挥重要作用。近期研究发现p53可以通过调控精脒/精胺N1-乙酰基转移酶1(SAT1)的转录,引起脂氧合酶15(ALOX15)依赖性的脂质过氧化进而导致铁死亡[9]。但目前关于p53/SAT1/ALOX15介导的铁死亡通路在PM2.5加重哮喘中的具体作用仍待阐明。
因此,本研究以小鼠为研究对象,通过构建哮喘小鼠模型,探讨铁死亡在PM2.5诱导哮喘加重中的潜在作用,并探究p53/SAT1/ALOX15通路介导的铁死亡在哮喘发展过程中的调控作用,为探究大气污染引起哮喘加重的机制提供实验依据。
本研究选用50只6~8周龄的SPF级BALB/c雄性小鼠,体重在18~22 g之间,由北京维通利华实验动物技术有限公司提供,许可证号:SCXK(京)2021-0006。实验材料包括增强型RIPA裂解液、BCA试剂盒(武汉博士德生物工程有限公司);ECL发光液(大连美仑生物公司);微量还原型谷胱甘肽(GSH)检测试剂盒(南京建成生物工程研究所);丙二醛(MDA)含量检测试剂盒(北京金克隆生物技术有限公司);亚铁离子含量检测试剂盒(北京索莱宝科技有限公司);小鼠白介素6(IL-6)和肿瘤坏死因子α(TNF-α)ELISA试剂盒(上海酶联生物科技公司);谷胱甘肽过氧化酶4(GPX4)鼠单克隆抗体、前列腺素内过氧化物合酶2(PTGS2)兔多克隆抗体、亚精胺/精胺 N1-乙酰转移酶1(SAT1)兔多克隆抗体、p53鼠单克隆抗体 和p-p53兔多克隆抗体(武汉三鹰生物技术有限公司);花生四烯酸 15-脂氧合酶(ALOX15)兔单克隆抗体和长链脂酰辅酶A合成酶4(ACSL4)兔单克隆抗体(英国 Abcam公司);β-actin鼠单克隆抗体(美国SAB公司);羊抗鼠二抗、羊抗兔二抗(武汉博士德生物工程有限公司)。
2020年11月至2021年1月,在山西太原市某交通路口利用TH-1000CII型智能空气采样器搭载PM2.5切割器,距离地面1.6 m,流量为1 m3/min,每日12 h(8:00至20:00)采样。采样完毕滤膜干燥24 h后裁为1 cm2块状,用超纯水超声清洗三次,冷冻干燥后存贮于-20℃备用。
将50只BALB/c鼠随机分为5组,每组10只,分别为生理盐水(NS)组、哮喘(卵清蛋白OVA)组、OVA联合交通源PM2.5低、中、高暴露(分别为1.8、3.6、7.2 mg/kg·bw)组。本研究已获得山西省肿瘤医院动物实验伦理审查委员会的批准(2022026)。
小鼠一周适应性饲养后,于1、8、15天对小鼠进行致敏,NS组腹腔注射PBS,其他组腹腔注射等体积的OVA和氢氧化铝的PBS混悬液。从第21天开始对小鼠进行OVA雾化激发。NS组用无菌PBS雾化,其他各组用2%OVA雾化激发,每次持续30 min。从第20天开始,对小鼠进行鼻腔滴注不同浓度的PM2.5染毒液。OVA雾化和PM2.5染毒交替进行,分别进行10次。
实验观察到哮喘小鼠血浆中OVA特异性IgE水平升高,气管及肺组织的HE染色和PAS染色显示明显的炎性细胞浸润,杯状细胞化生和黏液分泌增加,验证了哮喘模型的成功构建。
使用上海酶联生物科技公司提供的小鼠ELISA试剂盒,依照说明书操作,测定肺组织匀浆中IL-6和TNF-α的浓度。
取适量肺组织加入RIPA裂解液、蛋白酶抑制剂和磷酸酶抑制剂超声处理。离心后上清用于蛋白定量,以确定上样量。电泳分离蛋白样品后转移到PVDF膜上,室温下使用5%脱脂奶粉孵育3 h,加入相应的一抗过夜孵育,抗体浓度如下:ACSL4(1:15 000),PTGS2(1:4 000),GPX4(1:2 500),ALOX15(1:1 000),p53(1:2 500),p-p53(1:2 500),SAT1(1:500),β-actin(1:4 000)。次日洗涤三次,用二抗(1:5 000)孵育1 h后,ECL系统显影,用凝胶成像系统捕获图像并用Image J 1.8.0软件进行定量。
提取肺组织的总RNA,通过Prime Script TM RT 试剂盒将RNA反转录为cDNA,使用TB Green Premix Ex TaqTM Ⅱ和引物对目的基因进行体外扩增,CT值确定mRNA含量,并以β-actin为参照基因,计算相对表达倍数变化。实时荧光定量PCR的引物序列见表1
取适量肺组织制备上清液,于4℃、10 000 g离心10 min。根据说明书设置测定孔和标准孔,于593 nm读取吸光度。
称取适量组织样本,制备组织匀浆,4℃,5 000 r/min离心10 min取上清,按说明书步骤测量并分析 GSH(405 nm吸光度)和MDA(532 nm及600 nm吸光度差值)含量。
实验数据采用GraphPad Prism 8.0软件进行分析和图表绘制,结果使用表示,单因素方差分析用于多组间显著性差异的比较,多重比较采用Tukey检验,检验水准α=0.05。
结果如图1,各组间IL-6、TNF-α水平的差异有统计学意义(FIL-6=17.910,P<0.001;FTNF-α=5.414,P<0.05)。与NS组相比,PM2.5暴露后IL-6水平均显著升高。与OVA组比较,PM2.5中、高暴露组的IL-6水平显著增加(q=4.675,P<0.05;q=7.621,P<0.01)。随着PM2.5暴露浓度增加,IL-6水平呈升高趋势。与NS组(q=6.165,P<0.01)和OVA组(q=4.946,P<0.05)相比,PM2.5高暴露组的TNF-α水平显著提高。
结果如图2,各组间GSH和MDA含量的差异均有统计学意义(FGSH=13.560,P<0.001;FMDA=55.230,P<0.001)。与对照组相比,OVA组的GSH(q=3.277,P>0.05)表达降低,MDA(q=4.285,P>0.05)升高,但差异均不显著,PM2.5暴露组的GSH水平均显著降低,MDA水平均显著升高。与OVA组相比,PM2.5高暴露组的GSH水平明显下降(q=6.277,P<0.01),PM2.5中、高暴露组的MDA水平明显上升(q=6.830,P<0.01;q=15.320,P<0.01)。另外,MDA水平随着PM2.5浓度增高而逐渐升高。
结果如图3,各组间Fe2+水平的差异显著(F=17.660,P<0.001)。NS组的Fe2+水平最低,PM2.5暴露组的Fe2+水平均显著升高。相较于哮喘组,PM2.5中、高暴露组的Fe2+水平显著增长(q=5.022,P<0.05;q=8.759,P<0.01)。随着PM2.5暴露浓度的增加,小鼠肺部的Fe2+水平逐步上升,PM2.5高暴露组的Fe2+水平显著高于低暴露组(q=6.050,P<0.01)。
结果如图4,各组间PTGS2、ACSL4和GPX4蛋白表达水平的差异均有统计学意义(FPTGS2=7.456,P<0.01;FACSL4=5.715,P<0.05;FGPX4=16.900,P<0.001)。NS组的PTGS2和ACSL4蛋白表达最低,PM2.5中、高暴露组的PTGS2和ACSL4蛋白表达显著增加。与OVA组相比,PM2.5干预后,PTGS2和ACSL4蛋白表达进一步升高,其中PM2.5高暴露组的PTGS2蛋白表达最高(q=4.729,P<0.05)。与NS组相比,PM2.5高暴露组的GPX4蛋白表达明显降低(q=9.421,P<0.01)。与OVA组相比,PM2.5中、高浓度暴露组的GPX4蛋白表达显著降低(q=6.404,P<0.05;q=6.910,P<0.01)。
结果如图5,各组间PTGS2、ACSL4和GPX4基因表达的差异均有统计学意义(FPTGS2=5.106,P<0.05;FACSL4=66.810,P<0.001;FGPX4=90.260,P<0.001)。NS组的PTGS2和ACSL4的mRNA表达最低。与NS组相比,OVA组和各PM2.5暴露组的PTGS2和ACSL4的mRNA表达均有上升趋势,其中PM2.5高暴露组的增长最为显著(qPTGS2=5.766,P<0.05;qACSL4=21.900,P<0.01)。ACSL4的mRNA表达随PM2.5浓度增长而提升。反之,GPX4的mRNA表达在所有PM2.5暴露组均呈现降低趋势,并随PM2.5浓度升高进一步下调。
结果如图6所示,各组间p53、p-p53、SAT1和ALOX15蛋白表达水平的差异均有统计学意义(Fp53=6.793,P<0.01; Fp-p53=9.457,P<0.001;FSAT1=6.108,P<0.01;FALOX15=20.940,P<0.001)。与NS组相比,所有PM2.5染毒组中p-p53和ALOX15的蛋白表达明显增加,p53(q=6.816,P<0.01)和SAT1(q=5.904,P<0.01)在PM2.5高暴露组中的蛋白表达升高最显著。与OVA组比较,ALOX15蛋白表达在PM2.5染毒后均明显增加,p53(q=5.178,P<0.05)、p-p53(q=4.392,P<0.05)和SAT1(q=4.284,P<0.05)的蛋白表达在PM2.5高暴露组中显著增加。
结果如图7,各组间p53、SAT1和ALOX15基因表达的差异均有统计学意义(Fp53=22.770,P<0.001;FSAT1=4.167,P<0.05;FALOX15=32.520,P<0.001)与NS组相比,OVA组和PM2.5染毒组中各项指标均有不同程度的升高,p53的mRNA表达在各组均显著升高,SAT1的mRNA表达在PM2.5高暴露组中显著增加(q=5.219,P<0.05),ALOX15的mRNA表达在PM2.5中、高暴露组中显著增加(q=6.847,P<0.05;q=14.230,P<0.01)。与OVA组相比,p53(q=7.669,P<0.01)和ALOX15(q=12.180,P<0.01)的mRNA表达在PM2.5高暴露组中显著增加。随着PM2.5染毒浓度的提高,p53、SAT1和ALOX15的mRNA表达也呈增高趋势,这种趋势在ALOX15的mRNA表达上最为显著。
PM2.5引发的炎症是肺部疾病的主要致病机制。课题组前期研究发现,交通相关PM2.5可以诱导哮喘大鼠的Th17/Treg失衡,IL-17、IL-23、TGF-β1分泌增加进而加重哮喘[10-11]。IL-17可促进TNF-α、IL-1β、IL-6等促炎因子的分泌,增强组织内中性粒细胞的募集和炎症。另外,Wang Y等人的研究发现PM2.5暴露后健康小鼠肺组织和人支气管上皮BEAS-2B细胞中IL-6、IL-8和TNF-α水平显著增高[12]
铁死亡是以脂质过氧化为核心的新型细胞死亡调控形式,其特点是铁积累、致命性的脂质过氧化及核心还原酶GPX4不能及时清除脂质过氧化产物[13]。脂质过氧化是所有铁死亡调控途径的最终步骤,酰基辅酶A合成酶长链家族成员4(ASCL4)和溶血磷脂酰胆碱酰基转移酶3(LPCAT3)参与脂质合成与重塑,在脂氧合酶的作用下形成大量活性氧(ROS)、自由基和氧化终产物丙二醛(MDA)等[14]。Park等人发现PM2.5可以通过促进铁蓄积和脂质过氧化导致细胞死亡[15]。本研究观察到哮喘小鼠在PM2.5高浓度暴露下显示出Fe2+含量增加、脂质过氧化指标上升,以及GPX4抗氧化作用下降等铁死亡特征。随着PM2.5暴露浓度的增加,铁死亡标志性指标进一步改变,提示PM2.5暴露与铁死亡之间存在剂量-反应关系。
p53是一个关键的细胞代谢调节基因,与铁死亡所涉及的关键代谢途径(包括铁代谢、脂质代谢和氨基酸代谢)都密切相关[16]。p53可以通过转录抑制SLC7A11(胱氨酸-谷氨酸逆转运蛋白的关键组成部分)直接诱导铁死亡或转录促进SAT1调控ALOX15依赖的铁死亡[13]。研究表明氧化锌纳米颗粒诱导的人脐静脉内皮细胞(HUVEC)铁死亡模型中p53、SAT1和ALOX15均升高,敲除p53后SAT1显著降低,ALOX15与SAT1的表达趋势一致[17]。本研究显示,PM2.5暴露后p53、SAT1和ALOX15的表达明显升高,p-p53的表达明显升高,提示p53可能通过磷酸化修饰增加其对下游的转录激活作用。值得注意的是,SAT1是一种调节多胺代谢分解途径的关键酶,本研究首次发现PM2.5可以调控肺中SAT1的表达,提示了PM2.5可能对多胺代谢过程产生影响。
研究发现,铁死亡导致细胞膜破裂,将损伤相关分子模式(DAMPs)释放到细胞外,激活AGER(晚期糖基化终产物特异性受体)介导的炎症信号通路并引起小鼠和细胞促炎因子TNF-α、IL-1β、IL-6 显著增加以及抗炎因子IL-10的下降[18]。Shou等人发现铁死亡可以使Th17/Treg免疫反应失衡,TNF-α、IL-6、IL-1α、IL-1β、IL-17、IL-22和IL-23水平显著增加,而在Fer-1处理后降低[19]。以上证据提示铁死亡可参与并放大炎症反应。
本研究发现,交通源PM2.5加重了哮喘的气道炎症和脂质过氧化,同时导致肺部铁沉积明显。综上所述,我们认为交通相关PM2.5可诱导铁死亡并加重哮喘炎症,p53/SAT1/ALOX15通路可能参与这种调控作用,提示铁死亡可能是过敏性哮喘防治策略的新靶标。
