Objective

To investigate the effects of walnut-derived peptides (WDP) on miR-124-3p, STAT3, TGF-β1, and Smad3 proteins in aluminum-induced cognitive impairment.

Methods

Forty male SD rats were divided into control (saline),aluminum model (20 μmol/kg aluminum maltolate, Al (mal)3), 45-day WDP treatment (Al (mal)3 + 800 mg/kg WDP from day 45), and 90-day WDP prevention (Al (mal)3 + 800 mg/kg WDP from day 1) groups (n=10 each). Cognitive function was assessed by Morris water maze. Hippocampal protein expressions (p-STAT3 Ser727, p-STAT3 Tyr705, TGF-β1, p-Smad3 Ser423/425) and miR-124-3p levels were measured.

Results

Aluminum exposure prolonged escape latency from day 2 (P<0.05), which was attenuated by WDP (P<0.05). Aluminum increased hippocampal p-STAT3, TGF-β1, and p-Smad3 expressions while decreasing miR-124-3p (P<0.05 vs control). WDP interventions reversed these alterations (P<0.05 vs model).

Conclusion

WDP exerts protective effects against aluminum-induced cognitive dysfunction potentially through modulating miR-124-3p/STAT3-TGF-β1/Smad3 signaling pathway.

目的

观察探究核桃肽对铝暴露认知功能损害大鼠的miR-124-3p、信号转导和转录激活因子3(STAT3)、转化生长因子(TGF-β1)、SMAD同源物3(Smad3)重组蛋白的影响。

方法

以40只健康雄性SD大鼠为对象,分为对照组(腹腔注射生理盐水)、铝染毒模型组(腹腔注射20 μmol/kg麦芽酚铝)、核桃肽45 d治疗组(腹腔注射20 μmol/kg麦芽酚铝,在染毒第45 d开始灌胃800 mg/kg核桃肽,持续45 d)、核桃肽90 d预防组(腹腔注射20 μmol/kg麦芽酚铝,在染毒第1 d开始灌胃800 mg/kg核桃肽,持续90 d),每组10只。周一至五染毒,连续90 d。其后,通过Morris水迷宫评估大鼠学习记忆功能,Western Blot检测大鼠海马组织中p-STAT3 Ser727、p-STAT3 Tyr705、TGF-β1、p-Smad3 Ser423、p-Smad3 Ser425蛋白表达,实时荧光定量聚合酶链反应检测miR-124-3p表达。以上计量资料各组比较时均使用方差分析方法,体重变化比较用重复测量方差分析,分子表达变化用单因素方差分析。

结果

铝暴露各组大鼠从Morris水迷宫第2 d起逃避潜伏期均延长(P<0.05),其中核桃肽干预组大鼠的逃避潜伏期相较于模型组减少(P<0.05)。与对照组比,铝暴露大鼠海马组织的p-STAT3 Ser727、p-STAT3 Tyr705、TGF-β1、p-Smad3 Ser423、p-Smad3 Ser425的相对表达水平升高,miR-124-3p相对表达水平下降(P<0.05);与染毒模型组比,核桃肽干预组大鼠海马组织的p-STAT3 Ser727、p-STAT3 Tyr705、TGF-β1、p-Smad3 Ser423、p-Smad3 Ser425的相对表达水平下降,miR-124-3p相对表达水平上升(P<0.05)。

结论

核桃肽可在miR-124-3p调节STAT3-TGF-β1/Smad3通路,对铝诱导大鼠的认知功能障碍发挥保护作用。

目的

观察探究核桃肽对铝暴露认知功能损害大鼠的miR-124-3p、信号转导和转录激活因子3(STAT3)、转化生长因子(TGF-β1)、SMAD同源物3(Smad3)重组蛋白的影响。

方法

以40只健康雄性SD大鼠为对象,分为对照组(腹腔注射生理盐水)、铝染毒模型组(腹腔注射20 μmol/kg麦芽酚铝)、核桃肽45 d治疗组(腹腔注射20 μmol/kg麦芽酚铝,在染毒第45 d开始灌胃800 mg/kg核桃肽,持续45 d)、核桃肽90 d预防组(腹腔注射20 μmol/kg麦芽酚铝,在染毒第1 d开始灌胃800 mg/kg核桃肽,持续90 d),每组10只。周一至五染毒,连续90 d。其后,通过Morris水迷宫评估大鼠学习记忆功能,Western Blot检测大鼠海马组织中p-STAT3 Ser727、p-STAT3 Tyr705、TGF-β1、p-Smad3 Ser423、p-Smad3 Ser425蛋白表达,实时荧光定量聚合酶链反应检测miR-124-3p表达。以上计量资料各组比较时均使用方差分析方法,体重变化比较用重复测量方差分析,分子表达变化用单因素方差分析。

结果

铝暴露各组大鼠从Morris水迷宫第2 d起逃避潜伏期均延长(P<0.05),其中核桃肽干预组大鼠的逃避潜伏期相较于模型组减少(P<0.05)。与对照组比,铝暴露大鼠海马组织的p-STAT3 Ser727、p-STAT3 Tyr705、TGF-β1、p-Smad3 Ser423、p-Smad3 Ser425的相对表达水平升高,miR-124-3p相对表达水平下降(P<0.05);与染毒模型组比,核桃肽干预组大鼠海马组织的p-STAT3 Ser727、p-STAT3 Tyr705、TGF-β1、p-Smad3 Ser423、p-Smad3 Ser425的相对表达水平下降,miR-124-3p相对表达水平上升(P<0.05)。

结论

核桃肽可在miR-124-3p调节STAT3-TGF-β1/Smad3通路,对铝诱导大鼠的认知功能障碍发挥保护作用。

Objective

To investigate the effects of walnut-derived peptides (WDP) on miR-124-3p, STAT3, TGF-β1, and Smad3 proteins in aluminum-induced cognitive impairment.

Methods

Forty male SD rats were divided into control (saline),aluminum model (20 μmol/kg aluminum maltolate, Al (mal)3), 45-day WDP treatment (Al (mal)3 + 800 mg/kg WDP from day 45), and 90-day WDP prevention (Al (mal)3 + 800 mg/kg WDP from day 1) groups (n=10 each). Cognitive function was assessed by Morris water maze. Hippocampal protein expressions (p-STAT3 Ser727, p-STAT3 Tyr705, TGF-β1, p-Smad3 Ser423/425) and miR-124-3p levels were measured.

Results

Aluminum exposure prolonged escape latency from day 2 (P<0.05), which was attenuated by WDP (P<0.05). Aluminum increased hippocampal p-STAT3, TGF-β1, and p-Smad3 expressions while decreasing miR-124-3p (P<0.05 vs control). WDP interventions reversed these alterations (P<0.05 vs model).

Conclusion

WDP exerts protective effects against aluminum-induced cognitive dysfunction potentially through modulating miR-124-3p/STAT3-TGF-β1/Smad3 signaling pathway.

铝(aluminum)是地壳中含量最高的金属元素,已广泛应用于工业生产和日产生活,可从多种途径进入人体[1]。过量摄入的铝易在组织器官中积累,尤其是大脑,铝在脑组织的蓄积主要集中在海马体区域,长期铝暴露会引起认知功能损害,与神经退行性疾病如阿尔兹海默症(Alzheimer disease, AD),帕金森病(Parkinsonism disease, PD)等神经系统疾病的发病机制密切相关[2]。核桃肽(walnut peptide)作为植物源性活性物质,有多种生理调节功能[3]。现有研究显示,核桃肽可以有效增强小鼠空间学习记忆能力,并对PC12细胞氧化损伤表现出明显的保护作用[4]
研究表明miRNA在铝诱导神经毒性的过程中发挥关键调控作用[5]。在认知功能障碍的大鼠中,海马区小胶质细胞外泌体miR-124-3p表达显著下调[6]。而且miR-124-3p低表达已经成为海马依赖性记忆障碍的共同特征[7]。STAT3是信号转导和转录激活因子(STAT)蛋白家族的一员,是miR-124-3p的直接作用靶点,在中枢神经系统中扮演着重要角色[7]。STAT3调节神经胶质细胞的活化、增殖和分化过程,进而影响整体神经功能[8]。分子机制研究表明,miR-124-3p与STAT3在结合位点特异性结合,可进一步调控下游TGF-β1/Smad3信号通路[9]。TGF-β1信号通路参与多种神经精神疾病的病理进程,特别是在AD的发病机制中,该通路的异常激活可导致TGF-β1和Smad3蛋白异常表达,影响神经元凋亡过程[10]
基于上述研究背景,本研究提出miR-124-3p调控STAT3-TGF-β1/Smad3信号通路可能是核桃肽改善铝致认知功能障碍的关键分子机制,通过建立亚慢性铝暴露大鼠模型,用核桃肽进行干预,检测海马组织miR-124-3p、STAT3、TGF-β1、Smad3的表达变化,探讨miR-124-3p调控STAT3-TGF-β1/Smad3通路在核桃肽改善铝致认知功能障碍的作用。
40只6周龄健康雄性SD大鼠[SPF级,由济南朋悦实验动物繁育有限公司提供,实验动物生产许可证号:SCXK(鲁)2022 0006],体重(190±20)g,动物房饲养,温度保持22~26 ℃,每天早上8点开灯到晚上20点,用普通饲料喂养。适应性喂养后,按重量随机分组:对照组(腹腔注射生理盐水)、模型组(腹腔注射20 μmol/kg麦芽酚铝)、治疗组(腹腔注射20 μmol/kg麦芽酚铝,在染毒第45 d开始灌胃800 mg/kg核桃肽,持续45 d)、预防组(腹腔注射20 μmol/kg麦芽酚铝,在染毒第1 d开始灌胃800 mg/kg核桃肽,持续90 d)。周一至五按照1 ml/kg腹腔注射麦芽酚铝染毒,连续90 d。每周一对大鼠称重记录,每天观察大鼠精神状态和生长情况以及腹腔注射的针口愈合程度。所有实验程序均得到济宁医学院动物伦理委员会的批准,实验动物伦理委员会批准编号:JNMC-2021-DW-013。
三氯化铝(结晶)(AlCl3·6H2O,天津风船化学试剂有限公司);麦芽酚(分析纯,美国Sigma-Aldrich);核桃肽(斯诺特生物);戊巴比妥钠(山东鲁抗辰欣药业有限公司);氯仿、异丙醇;Total RNA Extractor (Trizol)、M-MuLV第一链cDNA合成试剂盒、2Xsg Fast qPCR Master Mix(上海生工);RIPA裂解液、蛋白酶、磷酸酶抑制剂、BCA定量试剂盒、彩虹180广谱蛋白marker(北京康为世纪生物公司);电泳、转膜液、TBS(武汉赛维尔生物科技有限公司);SDS-PAGE制备试剂盒(上海雅酶);TGF-β1一抗,Anti-GADPH antibody (ERP16891)、Goat Anti-Rabbit IgG HL(HRP)(英国Abcam);p-Smad3 Ser423、p-Smad3 Ser425、p-STAT3 Ser727、p-STAT3 Tyr705一抗(美国Thermo Fisher Scientific);化学发光成像系统(Gelpro32)。
麦芽酚铝溶液配制方法:将AlCl3·6H2O溶于生理盐水配制40 mmol/L的氯化铝溶液;将麦芽酚溶于磷酸盐缓冲液(PBS)配制120 mmol/L的麦芽酚溶液。随后将两种溶液等体积混合,最终获得20 mmol/L的麦芽酚铝溶液。根据大鼠体质量的变化,每天称取相应的核桃肽粉末,溶解于灭菌的超纯水中,制得800 mg/kg的核桃肽溶液,现配现用。
定位航行实验:将水池均匀划分为四个区域,把隐藏平台固定一个位置。每日随机选择不同区域放入大鼠,将大鼠入水到找到平台时间算作逃避潜伏期。如果大鼠在120 s内没找到,则带领到平台停留10 s,记录为120 s。空间探索实验:撤掉平台,观察120 s内穿越平台和平台象限的次数[11]
称取一定量的海马组织,提取蛋白测浓度。经SDS-PAGE凝胶电泳,结束后转模到PVDF膜,封闭1.5~2 h,封闭液用浓度为5%的伊利脱脂奶粉或5%的牛血清血蛋白(BSA),再Tris缓冲盐溶液(TBST)漂洗3次。接着,分别加入一抗(p-STAT3 Ser727、p-STAT3 Tyr705、p-Smad3 Ser423、p-Smad3 Ser425、TGF-β1的稀释比为1∶1 000;GADPH的稀释比1∶2 000),过夜4 ℃孵育18 h,用TBST漂洗3次,加相应的二抗(稀释比为1∶2 000),洗涤后显色,打开电脑曝光显影,保存图片,根据蛋白条带进行数据分析。
取大鼠海马组织,提取RNA,将RNA逆转录成cDNA,逆转录完成后以cDNA为模板,进行扩增,根据说明书配置相应反应体系,溶解并混匀各组分,短暂离心,运行qPCR,程序:95 ℃15 min;95 ℃34 s,60 ℃34 s,40个循环,以U6为内参基因,用2-ΔΔCt公式法计算所求基因表达量,计算结果进行归一化处理。引物序列见表1
本研究最初始每组10只SD大鼠,去除实验过程中因RNA降解和蛋白提取失败等造成的样本损失及离群值,最终有效样本量为6~8只/组。获得的结果用SPSS 26.0进行统计分析,用GraphPad Prism制图,结果为计量资料且满足正态分布用()表示,重复测量资料则用双因素重复测量方差分析,多组间对比用one-way ANOVA,组间两两比较方差齐用LSD检验,不齐用Dunnett -T3检验,双侧检验水准α=0.05。
实验期间,观察发现各组大鼠的状态存在差异,对照组的大鼠发育良好,精神状态良好,活动自如,毛发柔顺,腹部柔软,摄食和饮水均正常,未见任何异常情况。铝暴露模型组大鼠精神状态最差,活动减少,皮毛粗糙暗淡,腹部硬,核桃肽干预组大鼠精神状态较好,毛发柔顺,腹部略硬,摄食饮水状态较好。大鼠体质量结果显示,染毒前各组大鼠体质量基本一致,染毒后铝暴露的三组大鼠体质量与生理盐水对照组相比,均有不同程度的降低,铝暴露模型组大鼠体重最低(P<0.01),核桃肽预防组大鼠体重与生理盐水对照组大鼠体重相差最小。大鼠体重在染毒时间和核桃肽干预的主效应上,差异均有统计学意义(F=2 435.662、4.224,P<0.05),在染毒时间和核桃肽干预的交互效应上无统计学差异(F=2.822,P= 0.052)。见表2
定位巡航实验中,各处理组大鼠第1 d和第2 d的平均逃避潜伏期之间差异很小;第3~5 d,铝暴露各组大鼠的逃避潜伏期与对照组大鼠的相比均增加(P<0.01);其中,第4、5 d核桃肽治疗组和预防组的大鼠的逃避潜伏期和铝暴露模型组比较均下降(P<0.05),核桃肽治疗组和预防组之间亦有差异(P<0.05)。空间探索实验中,与对照组大鼠相比,铝暴露各组通过平台和平台象限的次数均减少(P<0.05),核桃肽预防组大鼠与铝暴露模型组大鼠和核桃肽治疗组大鼠相比,通过平台和平台象限的次数均增加(P<0.05)。见表34
实验结果显示,与对照组比,铝暴露各组大鼠海马组织的p-STAT3 Ser727、p-STAT3 Tyr705、TGF-β1、p-Smad3 Ser423、p-Smad3 Ser425蛋白表达水平均增加(P<0.01);与模型组比,核桃肽治疗组和预防组大鼠的p-STAT3 Ser727、STAT3 Tyr705、TGF-β1、p-Smad3 Ser423、p-Smad3 Ser425蛋白表达水平均下降(P<0.01);其中预防组较于治疗组蛋白表达量更低(P<0.05)。见表5图1
铝暴露组大鼠的miR-124-3p表达水平相较于对照组均下降(P<0.05);与模型组大鼠比,核桃肽治疗组和预防组大鼠的miR-124-3p相对表达水平均增加(P<0.05)。见表6
铝暴露可损害神经系统,引起神经发育障碍和神经退行性病变[2]。麦芽酚作为蔗糖水解或淀粉热解后的副产物,无神经毒性[12],其与三氯化铝结合形成的麦芽酚铝,结构稳定,不易降解,也不易水解。在生理pH条件下,生物利用度较高,能释放大量铝离子,这些铝离子可通过血脑屏障并在脑中蓄积[13-14]。因此,在铝神经毒性的研究中,尤其是在探索认知和记忆功能障碍的机制时,麦芽酚铝是常用的染铝模型的实验材料[15]。而且前期的研究发现,腹腔注射可使麦芽酚铝绕过胃肠道的吸收屏障直接通过腹膜吸收进入血液循环,生物利用度高,从而达到模拟铝在体内的蓄积效应[16]
核桃除了在制成核桃油外,分离纯化制成的核桃肽具有高吸收、高活性、高安全性的特点[17]。核桃肽不经消化即可在机体内被吸收,生物效价极高,能够增强小鼠的学习记忆功能,延缓小鼠的空间记忆衰退[18]。我们通过行为学水迷宫发现,随着实验进行铝暴露大鼠的逃避潜伏期显著增加,穿越平台次数降低,核桃肽干预大鼠逃避潜伏期显著减少,穿越平台次数增加。提示动物造模成功,亚慢性铝暴露破坏大鼠的学习能力,而核桃肽的干预可以显著改善铝暴露所致的学习能力缺陷,证实了核桃肽可以减小铝暴露造成的行为学损害。
miR-124-3p参与中枢神经系统的发育,并在神经元中富集[7]。Esteves M等人[19]发现以细胞外囊泡作为生物载体递送miR-124-3p到侧脑室可以显著改善PD小鼠模型的运动行为症状。Xu Y等人[20]还发现在创伤性脑损伤(traumatic brain injury ,TBI)后小胶质细胞外泌体中的miR-124-3p表达降低,无法抑制神经元自噬,造成神经受损。在本实验中,铝暴露大鼠的海马miR-124-3p表达水平低于对照组,核桃肽干预组的大鼠海马miR-124-3p水平高于铝暴露模型组,表明核桃肽可以缓解铝暴露引起的miR-124-3p降低。
TargetScan预测及双荧光素酶报告基因实验均证实,STAT3是miR-124-3p的直接作用靶点[21]。STAT3在神经退行性疾病中发挥重要的神经保护作用[22]。Yang W等[16]的进一步研究表明,miR-124-3p可通过靶向抑制STAT3的表达,削弱其在PD细胞模型中的神经保护效应。TGF-β1在突触可塑性、神经血管稳态及认知功能调节中具有重要作用,其信号通路的异常激活与AD等神经系统疾病的进展密切相关[23]。研究表明,TGF-β1可通过Smad非依赖性途径迅速诱导STAT3的磷酸化,其中磷酸化位点Tyr705和Ser727是STAT3激活的主要位置[9]。Smad3是TGF-β1信号通路的核心下游信号分子,其结构包括N端(MH1区域,负责DNA结合)、C端(MH2区域,含Ser423和Ser425磷酸化位点)及连接区(含多个丝氨酸/苏氨酸磷酸化位点)[24]。TGF-β1刺激可诱导Smad3在Ser423/Ser425位点的磷酸化,进而调控其核转位及转录活性[24]。本研究发现,亚慢性铝暴露显著上调大鼠海马中p-STAT3 (Ser727/Tyr705)、p-Smad3(Ser423/Ser425)及TGF-β1蛋白的表达水平,提示铝暴露可激活STAT3-TGF-β1/Smad3信号通路,可能通过促进神经炎症或突触功能障碍加剧神经损伤。值得注意的是,核桃肽干预可显著降低上述蛋白的异常表达,表明其可能通过抑制STAT3-TGF-β1/Smad3通路的过度激活,缓解铝诱导的神经毒性。
综上,本研究成功建立了亚慢性铝暴露大鼠模型,揭示了铝暴露通过miR-124-3p调控STAT3-TGF-β1/Smad3信号通路影响神经功能的潜在机制,并证实核桃肽的神经保护作用可能与该通路的调控相关。未来将进一步结合体内外干预实验探索核桃肽的具体活性成分及其分子靶点。
