Objective To establish a detection method for soybean allergens in plant-based protein products based on ultra performance liquid chromatography-tandem mass spectrometry, and conduct research on the quantification and authenticity determination of allergy protein in plant-based protein products. Methods After the soybean protein isolate was hydrolyzed by trypsin, the data were collected by high resolution mass spectrometry and matched with the Uniprot database to screen out the soybean specific allergen peptides. The response surface design was adopted to optimize the extraction conditions of the allergen protein, thereby constructing the quantitative method of allergens. Actual sample tests were conducted on commercially available plant-based protein products to preliminarily obtain the content of soybean allergy proteins in different plant-based protein products. Results The 6 kinds of soybean quantitative peptides with good linearity and correlation coefficients above 0.995 were selected. The limits of detection and the limits of quantification of the method were 0.182-2.08 µg/g and 0.607-6.920 µg/g. The spiked recoveries were 97.1%-126.6% and the intra-day and inter-day precisions were 5.17%-10.20% and 5.02%-11.70%. Conclusion This method has strong specificity and high sensitivity, and can well detect plant-based protein products, achieving accurate quantification of soybean allergens without isotope-labeled peptide segments. It has a good application prospect in the detection of allergen in plant-based protein products. Meanwhile, it can achieve the identification of adulteration in plant-based protein products and the discrimination of allergic information on food labels.

目的 基于超高效液相色谱-串联质谱法建立植物替代蛋白产品中大豆过敏蛋白的检测方法, 开展植物替代蛋白产品中过敏蛋白的定量和真实性判定研究。方法 大豆分离蛋白经胰蛋白酶水解后, 通过高分辨质谱采集数据并与Uniprot数据库匹配, 筛选出大豆特异性过敏蛋白多肽, 并采用响应面设计优化过敏蛋白提取条件, 从而构建过敏原定量方法。对市售植物替代蛋白产品进行实际样品检测, 初步得到不同植物替代蛋白产品中大豆过敏蛋白含量。结果 筛选出线性较好的6种大豆定量多肽、相关系数均大于0.995, 方法检出限为0.182~2.080 µg/g、方法定量限为0.607~6.920 µg/g, 回收率为97.1%~126.6%, 日内精密度为5.17%~10.20%, 日间精密度为5.02%~11.70%。结论 本方法特异性强、灵敏度高, 可以较好地针对植物替代蛋白产品进行检测, 实现了在无同位素标记肽段的情况下对大豆过敏蛋白的准确定量。在植物替代蛋白产品的过敏蛋白检测上具有较好的应用前景, 同时可实现植物替代蛋白产品的掺假鉴别及食品标签的过敏信息判别。

目的 基于超高效液相色谱-串联质谱法建立植物替代蛋白产品中大豆过敏蛋白的检测方法, 开展植物替代蛋白产品中过敏蛋白的定量和真实性判定研究。方法 大豆分离蛋白经胰蛋白酶水解后, 通过高分辨质谱采集数据并与Uniprot数据库匹配, 筛选出大豆特异性过敏蛋白多肽, 并采用响应面设计优化过敏蛋白提取条件, 从而构建过敏原定量方法。对市售植物替代蛋白产品进行实际样品检测, 初步得到不同植物替代蛋白产品中大豆过敏蛋白含量。结果 筛选出线性较好的6种大豆定量多肽、相关系数均大于0.995, 方法检出限为0.182~2.080 µg/g、方法定量限为0.607~6.920 µg/g, 回收率为97.1%~126.6%, 日内精密度为5.17%~10.20%, 日间精密度为5.02%~11.70%。结论 本方法特异性强、灵敏度高, 可以较好地针对植物替代蛋白产品进行检测, 实现了在无同位素标记肽段的情况下对大豆过敏蛋白的准确定量。在植物替代蛋白产品的过敏蛋白检测上具有较好的应用前景, 同时可实现植物替代蛋白产品的掺假鉴别及食品标签的过敏信息判别。

Objective To establish a detection method for soybean allergens in plant-based protein products based on ultra performance liquid chromatography-tandem mass spectrometry, and conduct research on the quantification and authenticity determination of allergy protein in plant-based protein products. Methods After the soybean protein isolate was hydrolyzed by trypsin, the data were collected by high resolution mass spectrometry and matched with the Uniprot database to screen out the soybean specific allergen peptides. The response surface design was adopted to optimize the extraction conditions of the allergen protein, thereby constructing the quantitative method of allergens. Actual sample tests were conducted on commercially available plant-based protein products to preliminarily obtain the content of soybean allergy proteins in different plant-based protein products. Results The 6 kinds of soybean quantitative peptides with good linearity and correlation coefficients above 0.995 were selected. The limits of detection and the limits of quantification of the method were 0.182-2.08 µg/g and 0.607-6.920 µg/g. The spiked recoveries were 97.1%-126.6% and the intra-day and inter-day precisions were 5.17%-10.20% and 5.02%-11.70%. Conclusion This method has strong specificity and high sensitivity, and can well detect plant-based protein products, achieving accurate quantification of soybean allergens without isotope-labeled peptide segments. It has a good application prospect in the detection of allergen in plant-based protein products. Meanwhile, it can achieve the identification of adulteration in plant-based protein products and the discrimination of allergic information on food labels.

近年来, 随着动物蛋白需求增加导致环境、供给、农兽药残留等问题的凸显, 作为优质蛋白来源的植物替代蛋白, 逐渐受到消费者们的关注[1-3]。植物替代蛋白产品不仅可以满足人体所需的蛋白营养需求, 还可避免环境污染及动物蛋白供给不充分等问题, 具有安全、稳定、高效、可持续等特点[4]。植物替代蛋白产品主要以大豆蛋白、花生蛋白、小麦蛋白、豌豆蛋白为原材料[5]。其中, 大豆蛋白是最主要的植物替代蛋白产品成分, 具有与动物蛋白相当的营养价值、更易消化、热量更低、产量高、价格低廉等特点, 常被用于生产减脂代餐等各类功能性食品。随着植物替代蛋白市场接受度的提高[3,6], 掺假问题也开始频发, 有不良商家将价格更低廉的其他蛋白掺进植物替代蛋白产品中, 生产大量功能性食品来牟取暴利, 并且在食品标签上不正确标识或隐藏过敏原信息, 严重损害了消费者的合法权益, 甚至威胁到过敏人群的健康[7]。食物过敏作为一个严重的食品健康问题, 影响着全球5%的人口, 且发病率呈现逐年上涨的趋势[8], 其中大豆蛋白中的大豆球蛋白(glycinin G1-G5)和β-伴大豆球蛋白的α亚基及β亚基为主要的过敏蛋白[9]。鸡蛋、牛奶、鱼、花生、甲壳类动物、大豆、小麦和坚果是最易过敏的食物[10], 植物替代蛋白产品中就普遍包含3类。但目前, 针对植物替代蛋白产品中过敏蛋白定量方法的研究较少, 无法对食品中过敏原进行准确定量等问题也制约着植物替代蛋白产业的发展, 因此, 亟待建立出一套针对植物替代蛋白产品中大豆过敏蛋白的定量方法, 实现植物替代蛋白产品中大豆过敏蛋白含量的确定与真实性判定。
目前, 针对蛋白类过敏原的检测技术主要包含聚合酶链式反应(polymerase chain reaction, PCR)[11-13]、酶联免疫吸附法(enzyme-linked immunosorben assay, ELISA)[14]、液相色谱-串联质谱法(liquid chromatography-tandem mass spectrometry, LC-MS/MS)[15-17]。PCR方法并不直接检测过敏蛋白, 而是以植物内源基因为基础, 通过设计特异性引物及探针进行过敏鉴别, 所以无法实现对过敏蛋白的准确定量[18]。ELISA方法利用抗原抗体的特异性免疫反应, 通过酶作用于底物后颜色反应的强弱实现定性和定量分析[9], ZHU等[19]开发了一种特异性夹心ELISA方法用于检测预包装食品中的过敏原成分的方法, 该方法以全大豆蛋白产生的多克隆抗体作为检测和捕获抗体, 定量限为0.0625~6.0 μg/mL。但是该方法在测定火腿肠中大豆过敏原时会出现假阳性现象, 可能与其烘焙加工条件有较大关系。尽管该方法灵敏度高、特异性强, 但由于美拉德反应及聚合反应产物的生成, 抗体无法准确识别过敏蛋白, 进而产生假阴性或定量结果偏低等现象。因此, 这两种方法均无法实现对过敏蛋白的准确定量。现阶段, 由于食品加工复杂程度的提高, 对于食品过敏原的检测提出了更高的要求, 已有研究学者将蛋白质组学与LC-MS/MS相结合, 开发出准确度、精密度、灵敏度都更高的检测方法, 并将其应用于过敏蛋白的检测且取得了较好的效果[20]。LC-MS/MS主要是通过质谱仪测定出经胰蛋白消化后的蛋白肽段混合物, 再与相关蛋白数据库进行匹配, 从而达到筛选特异性定量多肽的目的。
现阶段已有研究测定食品中的大豆过敏蛋白: 李丽芳等[21]开发了一种基于超高效液相色谱-串联质谱法测定大豆主要过敏原的方法, 优化了蛋白提取方法。但该方法仅停留在定性阶段, 缺少必要的方法学要素的考察。MONTOWSKA等[22]运用肽组学, 非靶向筛选商业加工的畜禽肉制品的特征蛋白消化物, 最终确定Glycinin为大豆过敏原热稳定肽标记物, 该方法实现了深加工肉类制品中大豆过敏蛋白的准确测定。YOSHIMITSU等[23]开发的LC-MS/MS使用重组Gly m 4 (rGly m 4)作为校准品定量, 确定方法回收率为128%, 定量限为0.1 μg/mL, 实现了对大豆加工制品中Gly m4的定量。
近年来, 我国逐渐加强了对食品中过敏原的重视, 在拓展检测范围和提升检测精度等方面提出了更为严苛的要求。我国于2019年颁布了GB/T 38163—2019《常见过敏蛋白的测定 液相色谱-串联质谱法》, 该标准适用于检测小麦粉、燕麦粉、可可粉等固态基质食品中的常见过敏原, 但目前还鲜少有针对植物替代蛋白产品中过敏原检测的相关标准及文献。
本研究基于质谱技术在过敏原检测中准确、稳定且高效的优点, 通过高分辨质谱技术与Uniprot数据库相结合, 筛选出大豆的特异性过敏蛋白多肽, 通过响应面设计(Box-Behnken)优化过敏蛋白提取条件, 建立基于特征肽段的大豆过敏蛋白多重反应监测(multiplex reaction monitoring, MRM)检测方法, 并应用于市售10种植物替代蛋白产品, 拟建立一种植物替代蛋白产品中大豆过敏蛋白准确定量的检测方法, 进而实现对植物替代蛋白真实性的鉴别, 为植物替代蛋白产品的质量安全监管提供技术支持。
于北京不同区域的市场、超市购入10种植物替代蛋白产品。
大豆分离蛋白(纯度82.32%, 北京美添前景科技有限公司); 胰蛋白酶(测序级IX-S, 来源于猪胰脏, 13000~20000 BAEE units/mg)、碘乙酰胺(iodoacetamide, IAA, 生化级)(美国Sigma公司); 二硫苏糖醇(dithiothreitol, DTT, 生化级, 美国Promega公司); 甲酸、乙酸、乙腈(色谱纯, 德国Merck公司); 三羟甲基氨基甲烷(Tris)、尿素、氯化胆碱、甘油、盐酸[分析纯, 国药集团化学试剂(上海)有限公司]; 超纯水由Milli-Q超纯水净化系统制备获得。
Q Exactive HF-X超高效液相色谱四极杆静电场轨道阱高分辨质谱系统(配有电喷雾离子源)、Sorvall LYNX 4000落地式高速离心机、Hypeisil GOLD C18色谱柱(2.1 mm×100 mm, 1.9 µm)(美国赛默飞世尔科技公司); Agilent1290-6470超效高液相色谱-串联三重四级杆质谱仪、Agilent RRHD C18色谱柱(3.0 mm×100 mm, 2.1 µm)(美国Agilent公司); AUW220D电子分析天平(感量0.001 g, 日本Shimadzu公司); DT 1028 H超声波清洗机(德国Bandelin公司); HLB固相萃取柱(60 mg, 3cc)(美国Waters公司)。
根据前期研究[24-25], 样品前处理方法主要分为蛋白提取、胰蛋白酶水解、固相萃取净化3个步骤, 具体实验操作如下: 称取0.5 g (±0.001 g)均质后的样品, 加入10 mL蛋白提取液(50 mmol/L Tris-HCl, 含6 mmol/L尿素, pH=10), 冷水浴超声30 min, 12000 r/min离心10 min。
取200 µL上清液加入30 µL 100 mmol/L DTT后于56 ℃条件下气浴振荡1 h, 静止至室温后加入30 µL 100 mmol/L IAA置于暗处反应30 min, 取出后加入100 µL胰蛋白酶溶液(2.5 mg/mL)酶解3 h, 加入15 µL乙酸终止反应, 再加入蛋白提取液定容至2 mL, 上述溶液待净化。
采用3 mL乙腈和0.5%乙酸溶液提前活化HLB固相萃取柱, 将反应液全部上样, 使用3 mL 0.5%乙酸淋洗, 抽干后使用2 mL 60%乙腈溶液(含有0.5%乙酸)洗脱, 洗脱液经0.22 µm微孔滤膜过滤测定。
以不同料液比、酶解时间、酶与蛋白配比进行单因素实验, 考察不同因素对大豆过敏蛋白提取效果的影响, 除变量外, 实验固定蛋白提取液配比(50 mmol/L Tris-HCl, 6 mmol/L尿素, pH=10)、DTT 100 mmol/L、IAA 100 mmol/L、振荡时间1 h、超声时间30 min, 分别研究料液比(1:10、1:15、1:20、1:25、1:30 g:mL)、酶解时间(1、3、6、9、12 h)、酶与蛋白配比(1:10、1:20、1:30、1:40、1:50 mg:g)对过敏蛋白提取效果的影响。
根据单因素实验结果设计响应面实验, 以料液比(A)、酶解时间(B)、酶与蛋白配比(C)为实验因素, 以大豆最佳定量过敏多肽响应值作为考察指标, 进行3因素3水平的响应面实验, 其因素水平见表1
色谱条件: Hypeisil GOLD C18色谱柱(2.1 mm×100 mm, 1.9 µm); 柱温40 ℃, 进样量5 µL; 流动相A为0.1%甲酸水溶液, 流动相B为0.1%甲酸-乙腈溶液, 洗脱梯度为0~0.2 min, 3%~10% B; 0.2~20.0 min, 10%~40% B; 20.0~24.0 min, 40%~80% B; 24.0~26.5 min, 80% B; 26.5~27.0 min, 80%~3% B; 27.0~30.0 min, 3% B。
质谱条件: 喷雾电压3.5 kV; 毛细管温度300 ℃; 离子源雾化温度350℃; 鞘气压力45 arb; 辅气压力15 arb; 扫描模式Full MS/dd-MS2; 离子化模式为正离子; 一级全扫描分辨率120000; 自动增益3e6; 扫描范围150~1500 m/z; 最大离子注入时间20 ms; 二级质谱扫描分辨率15000; 自动增益1e5; 最大离子注入时间100 ms; 分离窗口1.6 m/z; 归一化碰撞能量为20 V。
色谱条件: Agilent RRHD C18色谱柱(3.0 mm×100 mm, 2.1 µm); 柱温40℃, 进样量1 µL; 流动相A为0.1%甲酸水溶液, 流动相B为0.1%甲酸-乙腈溶液, 洗脱梯度为0~1.0 min, 10% B; 1.0~10.0 min, 10%~50% B; 10.0~13.0 min, 50%~90% B; 13.0~14.0 min, 90% B; 14.0~14.1 min, 90%~10% B; 后运行2 min。
质谱条件: 离子源: 电喷雾离子源(electrospray ion source, ESI); 检测模式: MRM; 扫描模式: 正离子模式; 毛细管电压: 3500 V; 干燥气温度: 350 ℃; 干燥气流速: 7 L/min; 鞘气温度: 300 ℃; 鞘气流速: 11 L/min; 雾化气压力: 45 psi。
数据分析: 采用Design Expert 13.0软件对蛋白提取流程的3个主要参数进行响应面优化。采用Proteome Discover (PD 2.8)软件对高分辨采集数据进行分析, PD软件工作流程主要分为Processing Step和Consensus Step, 参数设置最多漏切位点为2, 每个多肽最大平均修饰为3, 多肽长度为6~20, 结合Uniprot蛋白数据库进行大豆过敏蛋白的匹配。
称取0.5 g (±0.001 g)大豆分离蛋白按照1.3.1节前处理方法, 分别移取适量的终溶液以60%乙腈溶液(含有0.5%乙酸)稀释, 配制成0.1、1.0、2.0、5.0、10.0 mg大豆分离蛋白的标准溶液曲线溶液, 以质量浓度作横坐标, 筛选的大豆过敏蛋白多肽响应强度作纵坐标, 计算回归方程。
待测样品的称样量为m (g), 将待测样品各个多肽的峰面积代入对应的回归方程, 得到相应蛋白的质量为mi (mg), k为对应过敏蛋白与大豆分离蛋白的转化系数, 待测样品中大豆过敏蛋白质量X (µg/g)按式(1)计算:

采用Proteome Discover (PD 2.8)软件对高分辨采集数据进行分析, 采用Origin 2024绘图, 使用IBM SPSS Statistics 27.0进行数据分析。使用Design Expert 13进行响应面分析处理, 并利用Box-Behnken设计进行响应面优化。为减少实验误差, 采用3次独立重复实验。
在建立超高效液相色谱-串联质谱法测定过敏蛋白的检测方法时, 筛选出专一性强、稳定性好、灵敏度高的过敏蛋白特征肽段是至关重要的, 这将直接决定方法的准确性。
本研究将大豆分离蛋白按照上述方法进行前处理, 得到高纯度的多肽蛋白酶解液, 通过超高效液相色谱仪将酶解液喷雾到高分辨质谱仪中进行数据采集, 获取的数据导入至Proteome Discovery (PD 2.8)软件中, 结合Uniprot数据库筛选出大豆蛋白过敏蛋白多肽序列, 共获得大豆过敏蛋白多肽19条, 具体结果见表2。但初步获得的过敏多肽需进一步筛选, 需考察: (1)肽段长度, 氨基酸残基长度在6~20较为合适; (2)专一性, 需确定其多肽仅存在于对应物种的过敏蛋白中, 均是专属的特异多肽; (3)稳定性, 在经过复杂的食品加工处理后仍保持稳定的状态, 不易降解或发生化学反应, 不包含漏切的酶切位点(精氨酸、赖氨酸); (4)响应强度, 需确定多肽离子在质谱分析时具有较低的离子抑制效应, 色谱峰形尖锐对称, 离子响应强度至少大于105
通过将高分辨质谱数据导入Skyline软件, 将筛选后的多肽离子转为离子对信息, 对实际样品进行多重反应监测模式扫描, 为保证结果的准确度及灵敏度, 本研究选取了响应强度较高的6种大豆过敏蛋白多肽, 提取离子色谱图见图1~图2, 本研究提及的全部大豆过敏蛋白见表2, 提取离子色谱图见图3
过敏蛋白的提取效果会影响特征多肽的响应强度, 进而影响过敏蛋白定量的准确度及灵敏度, 因此在研究过程中需最大程度地保证蛋白缓冲溶液的提取效率。本研究采用溶液内消化的蛋白质组学方法, 通过Tris-HCl缓冲液促进细胞或组织的裂解, 利用尿素使蛋白质变性, 进而促进蛋白质溶解, 最终达到较好的蛋白提取效果。在文献调研中发现近年来很多学者使用低共熔溶剂进行蛋白提取且取得较好效果[27-28], 但针对过敏蛋白的提取未见报道, 因此本研究也对低共熔溶剂(deep eutectic solvent, DES)的植物过敏蛋白提取效果进行研究。具体配置的蛋白提取缓冲溶液如下: (1)氯化胆碱:尿素:水(1:1:10 mol/L); (2)氯化胆碱:乙酸:水(1:1:10 mol/L); (3)氯化胆碱:甘油:水(1:1:10 mol/L); (4) 50 mmol/L Tris-HCl; (5) 50 mmol/L Tris-HCl(含6 mol/L尿素)。本研究发现, 加入尿素的DES缓冲液仅能提取少量大豆过敏蛋白且特征肽段的离子响应极低, 但Tris-HCl缓冲溶液的提取效果普遍较好, 其中最佳提取缓冲液为50 mmol/L Tris-HCl(含6 mol/L尿素), 响应较高的特征多肽分别来自大豆球蛋白1、2、4和β-伴大豆球蛋白, 具体结果见图4。DES缓冲液提取效果较差可能是因其较差的热稳定性, 在高于54 ℃时会发生相变, 进而影响其化学结构性质, 而本研究在添加DTT减少二硫化物时需维持在56 ℃体系中[29]。另外, DES缓冲液可能更适宜提取植物蛋白中的生物活性物质[30]。因此, 本研究最终采用50 mmol/L Tris-HCl作为最佳过敏蛋白提取缓冲液。
将单因素实验得到的大豆特征多肽离子响应强度按列归一化后得到0~1的原始矩阵, 由图5显示, 在料液比为1:20 (g:mL, 下同)时, 大豆中绝大多数特征多肽的离子响应呈现最高, 这表明适当增加料液比可以使得样品能充分浸润于蛋白提取缓冲液中, 有利于过敏蛋白的提取。在低于1:20时, 大豆分离蛋白粉会出现难以充分混合的现象, 上层蛋白粉溶解后结块形成蛋白膜, 阻碍下层蛋白粉与提取液接触, 从而使得提取效率变低。随着料液比的增加, 缓冲液提取的过敏蛋白将达到饱和状态, 继续增加料液比的含量对提取效率没有显著影响。因此, 后续响应面选择料液比1:15、1:20、1:25进行优化。
胰蛋白酶的最佳酶解温度为37 ℃, 酶解时间会直接影响酶解效果, 过长或过短均会导致酶解出的过敏蛋白多肽不充分, 体系中多肽浓度偏低将影响过敏蛋白的定量结果[31]。为研究酶解时间与过敏蛋白特征多肽响应强度间的关系, 本研究考察了不同酶解时间(1、3、6、9、12 h)对大豆蛋白提取效果的影响, 结果如图5所示, 大豆分离蛋白在酶解1~12 h的过程中, 特征多肽均可以被检出, 但不同特征肽段之间的响应差异较大。在酶解1 h时, 绝大多数的特征肽段响应较低, 在酶解时间达到3 h时达到最高, 尽管有少部分肽段在酶解6~9 h后响应强度变高, 但提取溶剂对其提取效率偏低。考虑到时间成本及提取效果, 本研究选取酶解时间3、6、9 h进行响应面优化。
胰蛋白酶与大豆分离蛋白粉的配比也是优化酶解条件的重要参数, 随着配比的增大, 胰蛋白酶将无法酶解过多的底物, 导致特征多肽的响应变差[32-33]。如图5所示, 在酶与蛋白配比在1:40 (mg:g, 下同)时, 选取的大豆过敏蛋白特征肽段均达到最佳的响应强度, 热图中显示, 归一化值均达到0.80以上, 在配比为1:10~1:30时, 绝大多数特征肽段响应强度归一化值在0.60左右。综合考虑, 本研究选取酶与蛋白的配比1:30、1:40、1:50进行响应面优化。
根据2.2.2实验结果, 利用Box-Behnken设计进行响应面优化, 本研究共设计17组实验, 其中包含中心组实验5组, 以β-伴大豆球蛋白的最佳定量离子对响应值为结果。依据Design Expert 13软件对所得的17组实验结果进行线性回归拟合分析, 所得到的二次回归方程为Y=1.051×106-4.829×105A-3.446×105B-20639.1C+21854.1D+1.397×105AC+1.334×105BC+1.003×105A2+1.959×105B2+ 1.674×105C2, 显著性分析见表3
表3可知, 模型F为5.62, P为0.0165<0.05, 具有显著差异, 表明该模型具有较好的准确性及精确性。决定系数R2为0.8784, 说明该模型的预测值与实际值拟合良好。失拟值P为0.4273>0.05, 差异不显著, 表明该响应面模型误差较小。因此, 该模型适用于本研究数据的分析与参数的优化。因素ABP<0.01, 具有极显著差异, 表明料液比及酶解时间对过敏蛋白的提取有显著影响。3D响应面图能够直观地反应3个实验因素(料液比、酶解时间、酶与蛋白配比)对大β-伴大豆球蛋白特征多肽响应的影响。曲面的陡峭程度反映了实验因素对特征多肽的影响程度, 如图6所示, 具有极显著差异的料液比与酶解时间单因素, 相互作用较弱。软件优化的最佳参数为: 料液比1:21.415 g:mL、酶解时间3.086 h、酶与蛋白配比1:44.43 mg:g。考虑到实验的可操作性和简便性, 稍微改动后的参数为料液比1:20、酶解时间3 h、酶与蛋白配比为1:44。通过实验验证, 优化后的β-伴大豆球蛋白特征多肽的响应值达到2.78×106
目前各国均未统一过敏原的标准品, 因此本研究采用大豆分离蛋白粉作为标准物质, 通过凯氏定氮法测定其纯度为82.32%。称取0.5 g大豆分离蛋白粉按1.3.1节方法进行前处理, 以大豆分离蛋白质量为横坐标, 筛选出的特征多肽响应为纵坐标, 分别进行线性拟合, 排除响应值较低及相关系数低于0.995的特征多肽, 所得的线性方程见表4。其中最佳定量特征肽段来自β-伴大豆球蛋白, 多肽序列为LITLAIPVNKPGR, 其响应强度比其他特征多肽大一个数量级。
向不含大豆蛋白的植物替代蛋白产品中加入质量浓度为50、100、200 mg/g的大豆分离蛋白粉后按1.3.1节方法进行前处理, 对选取的6条肽段进行检测, 根据线性方程得到的实际浓度与理论浓度计算回收率, 得到的加标回收实验结果见表5, 实际样品不同加标水平的总离子流图见图7。筛选得到的6条特征肽段回收均小于130%, 相对标准偏差小于15%, 作为定量肽段来说具有较好的准确性。在加标浓度为50 mg/g时, 回收率均偏高。对实际样品进行日内及日间精密度测定, 同一试样连续检测3 d, 平行测定3次, 日内精密度为5.17%~10.20%, 日间精密度为5.02%~11.70%。结果表明该方法具有较好的回收率与精密度。方法检出限及定量限通过对标准曲线中最低质量浓度的大豆分离蛋白进行前处理, 以定量离子信噪比的3倍及10倍进行计算, 得出大豆球蛋白G1-1亚基与β-伴大豆球蛋白的检出限均是0.182 µg/g, 定量限均是0.607 µg/g; 大豆球蛋白G1-2亚基的检出限是1.640 µg/g, 定量限是5.470 µg/g; 大豆球蛋白G2-3亚基的检出限是2.080 µg/g, 定量限是6.920 µg/g; 大豆球蛋白G4亚基的检出限是0.583 µg/g, 定量限是1.940 µg/g; 大豆球蛋白前体的检出限是0.255 µg/g, 定量限是0.850 µg/g。结果表明该方法具有较好的灵敏性, 可满足植物替代蛋白产品中大豆过敏蛋白的检测要求。
为了验证方法的适用性, 从北京超市随机抽取10种植物替代蛋白产品, 样品外包装未全部标识大豆过敏原信息, 结果如表6所示, 10种样品均有大豆过敏蛋白检出, 其中大豆蛋白作为首要原材料的实际样品中过敏蛋白含量也相对较高, 大豆球蛋白G2-3亚基含量相对其他过敏蛋白丰富, 其次是大豆球蛋白G1-2亚基, 其中过敏蛋白含量最低的是β-伴大豆球蛋白。
本研究采用Proteome Discovery与Skyline等生物信息学软件与Uniprot数据库相结合, 完成了大豆过敏蛋白肽段的匹配, 通过高分辨质谱技术筛选出用于表征大豆的特异性过敏蛋白肽段, 针对不同肽段分别优化其三重四级杆串联质谱检测参数, 全面、有效、准确的定量大豆蛋白中两种主要过敏原: 大豆球蛋白以及β-伴大豆球蛋白, 并通过响应面设计优化过敏蛋白的提取条件。该方法已成功应用于市售常见植物替代蛋白产品的过敏原检测及真实性鉴别中, 有效弥补了在无同位素标记肽段的情况下对大豆过敏蛋白的准确定量, 为植物替代蛋白产品中大豆过敏蛋白的检测、食物标签中过敏信息的判别提供技术支持和保障, 同时可区分未添加大豆蛋白的植物替代蛋白, 实现了植物替代蛋白的掺假鉴别。
本研究下一步拟将补充其余两种主要的过敏蛋白: Gly m Bd 30K、Gly m Bd 28K, 筛选其离子对信息, 最终建立植物替代蛋白产品中大豆主要过敏原的定性定量检测方法, 并将该方法的研究思路应用于常见植物蛋白过敏原(花生、小麦、豌豆)中, 构建普适性的过敏原检测方法, 弥补植物替代蛋白产品中常见过敏蛋白检测技术的空缺, 为植物替代蛋白产品的质量安全监管提供技术保障。
