Objective To screen lactic acid bacteria from Viili fermented milk and explore the proportion of bacteria in the compound starter cultures. Methods Lactic acid bacteria were isolated using streak plating, and their taxonomic classification was determined through phylogenetic analysis based on 16S rRNA sequences. The composition of strain ratios in mixed cultures was designed with reference to the bacterial abundance observed in Viili fermented milk. Comprehensive analyses were performed to evaluate the acidity, exopolysaccharide production, textural properties and volatile flavor compounds of the mixed fermentation groups. Results Diversity analysis identified 4 bacterial genera, among which Lactococcus, Streptococcus and Leuconostoc were the dominant genera, comprising 49.12%, 40.67% and 10.21% of the total bacterial composition, respectively. Lactococcus lactis H2, Streptococcus thermophilus H17, Leuconostoc mesenteroides H30 and Lactobacillus paracasei H33 were isolated and screened. A total of 20 fermentation trials with different strain compositions were designed to conduct a comprehensive assessment of key parameters, including pH, coagulation time, textural properties, exopolysaccharide content and flavor profile. The results indicated that formulations S13 (H2:H17:H30:H33=11:4:4:1) and S17 (H2:H17:H30:H33=30:35:16:19) demonstrated the highest similarity to Viili fermented milk in terms of physicochemical and sensory properties. Conclusion The blended formulations S13 (H2:H17:H30:H33=11:4:4:1) and S17 (H2:H17:H30:H33=30:35:16:19) exhibit promising potential as compound starter cultures. This study provides a theoretical basis for developing compound starter cultures in Viili fermented milk production.

目的 筛选Viili发酵乳中的乳酸菌, 探讨复配发酵剂中菌种比例。方法 通过平板划线分离筛选其微生物, 根据16S rRNA序列构建系统发育树确定菌株的种属。并结合Viili发酵乳中细菌的丰度对复配比例进行设计, 对复配发酵组的酸度、胞外多糖含量、质构、挥发性风味物质进行综合分析。结果 多样性结果显示检测出4个细菌属, 其中乳球菌属(Lactococcus)、链球菌属(Streptococcus)、明串珠菌属(Leucanostoc)作为优势细菌属, 占比分别为49.12%、40.67%、10.21%。分离筛选得到乳酸乳球菌H2、嗜热链球菌H17、肠膜明串珠菌H30、副干酪乳杆菌H33。通过复配设计20组不同比例的发酵实验, 综合评估pH、凝乳时间、质构特性、胞外多糖含量及风味等指标, 结果表明, 复配组S13(活菌数之比为H2:H17:H30:H33=11:4:4:1)和S17(活菌数之比为H2:H17:H30:H33=30:35:16:19)在各项性能上与Viili发酵乳最为接近。结论 复配组S13 (H2:H17:H30:H33=11:4:4:1)和S17 (H2:H17:H30:H33=30:35:16:19 )具有作为复合发酵剂的应用价值。本研究为Viili发酵乳复配发酵剂提供了理论基础。

目的 筛选Viili发酵乳中的乳酸菌, 探讨复配发酵剂中菌种比例。方法 通过平板划线分离筛选其微生物, 根据16S rRNA序列构建系统发育树确定菌株的种属。并结合Viili发酵乳中细菌的丰度对复配比例进行设计, 对复配发酵组的酸度、胞外多糖含量、质构、挥发性风味物质进行综合分析。结果 多样性结果显示检测出4个细菌属, 其中乳球菌属(Lactococcus)、链球菌属(Streptococcus)、明串珠菌属(Leucanostoc)作为优势细菌属, 占比分别为49.12%、40.67%、10.21%。分离筛选得到乳酸乳球菌H2、嗜热链球菌H17、肠膜明串珠菌H30、副干酪乳杆菌H33。通过复配设计20组不同比例的发酵实验, 综合评估pH、凝乳时间、质构特性、胞外多糖含量及风味等指标, 结果表明, 复配组S13(活菌数之比为H2:H17:H30:H33=11:4:4:1)和S17(活菌数之比为H2:H17:H30:H33=30:35:16:19)在各项性能上与Viili发酵乳最为接近。结论 复配组S13 (H2:H17:H30:H33=11:4:4:1)和S17 (H2:H17:H30:H33=30:35:16:19 )具有作为复合发酵剂的应用价值。本研究为Viili发酵乳复配发酵剂提供了理论基础。

Objective To screen lactic acid bacteria from Viili fermented milk and explore the proportion of bacteria in the compound starter cultures. Methods Lactic acid bacteria were isolated using streak plating, and their taxonomic classification was determined through phylogenetic analysis based on 16S rRNA sequences. The composition of strain ratios in mixed cultures was designed with reference to the bacterial abundance observed in Viili fermented milk. Comprehensive analyses were performed to evaluate the acidity, exopolysaccharide production, textural properties and volatile flavor compounds of the mixed fermentation groups. Results Diversity analysis identified 4 bacterial genera, among which Lactococcus, Streptococcus and Leuconostoc were the dominant genera, comprising 49.12%, 40.67% and 10.21% of the total bacterial composition, respectively. Lactococcus lactis H2, Streptococcus thermophilus H17, Leuconostoc mesenteroides H30 and Lactobacillus paracasei H33 were isolated and screened. A total of 20 fermentation trials with different strain compositions were designed to conduct a comprehensive assessment of key parameters, including pH, coagulation time, textural properties, exopolysaccharide content and flavor profile. The results indicated that formulations S13 (H2:H17:H30:H33=11:4:4:1) and S17 (H2:H17:H30:H33=30:35:16:19) demonstrated the highest similarity to Viili fermented milk in terms of physicochemical and sensory properties. Conclusion The blended formulations S13 (H2:H17:H30:H33=11:4:4:1) and S17 (H2:H17:H30:H33=30:35:16:19) exhibit promising potential as compound starter cultures. This study provides a theoretical basis for developing compound starter cultures in Viili fermented milk production.

Viili是一种北欧传统的常温发酵乳, 因其黏稠的质地和适宜的口感受到消费者的关注[1]。研究表明, Viili独特的黏稠质地主要归因于乳酸菌在发酵过程中大量合成胞外多糖(exopoly saccharides, EPS), 这些多糖有效提升了发酵乳的黏稠性, 并且可以有效防止乳清析出[2]。Viili中乳酸菌产生的胞外多糖被证明具有提高免疫力、抗疲劳等功效, 并且有研究筛选自Viili的副干酪乳杆菌可以与益生元协同干预结肠炎, 证实了Viili源乳酸菌具有良好的益生潜质[3-5]
传统天然发酵食品中蕴藏着丰富的菌种资源, 通过分离筛选优良的微生物, 可用于提升食品的风味及功能性。目前, 已有多项研究表明传统乳制品、泡菜、食醋等天然发酵食品中菌种具有良好的发酵性能以及益生特性。刘春芳[6]从开菲尔发酵乳中分离筛选出两株发酵性能良好、具有潜在益生特性菌株, 并通过复配开发出风味良好发酵特性稳定的发酵剂。孙悦欣等[7]将分离自不同地区的天然发酵乳的嗜热链球菌和保加利亚乳杆菌进行复配, 得出发酵特性和贮藏特性良好的组合。这些研究表明, 从传统发酵食品中筛选优良微生物用于发酵剂的开发可以提升发酵产品的品质。
Viili作为一种天然多菌种协同发酵的传统乳制品, 不仅富含多样的微生物资源, 还具有独特的风味和质地[8]。在发酵过程中, 乳酸菌与真菌通过代谢产物的积累和互作, 动态调控了Viili的风味形成和质地特性。白地霉作为Viili中的特征真菌, 可以分解乳中的蛋白质和脂肪, 赋予发酵乳独特的风味, 并在发酵乳表面生长, 促进乳酸菌等微生物的共生生长[9]。因此, 筛选优良的Viili源发酵菌种可以为国内发酵乳提供新的可用菌种, 推动国内发酵乳产品的创新。通过研究Viili复合发酵剂, 可以丰富国内发酵剂的种类, 并为开发具有更高附加值和功能性的乳制品提供新的思路。本研究通过分离筛选Viili源微生物, 并优化乳酸菌添加比例, 以期为Viili复配发酵剂开发和产业化提供基础。
Viili发酵乳(购于芬兰); 脱脂奶粉(内蒙古伊利有限公司); 白地霉(Geotrichum candidum, 实验室保藏)。
MRS培养基(北京奥博星生物技术有限公司); 三氯乙酸、苯酚、浓硫酸、无水乙醇(分析纯)、琼脂粉、透析袋(北京索莱宝科技有限公司); 细菌基因组提取试剂盒(北京天根生化科技有限公司); E.Z.N.A. Soil试剂盒(美国Omega公司); FastPfu Polymerase(北京全式金生物技术有限公司)。
SU8010光学显微镜(日本日立公司); DH3600生化培养箱(天津泰斯特仪器有限公司); SW-CJ-1FD超净工作台(苏州净化有限公司); Centrifuge 5424 R低温离心机(德国海蒂诗有限公司); T100 Thermal Cycler聚合酶链式反应(polymerase chain reaction, PCR)仪(美国BIO-RAD公司); JY600C电泳仪(北京君意东方电泳设备有限公司); FE28 pH计(美国梅特勒-托利多仪器有限公司); TMS-Rro质构仪(美国FTC公司); FlavourSpec气相色谱-离子迁移谱联用仪(gas chromatography-ion obility spectrometry, GC-IMS)(山东海能科学仪器有限公司); MXT-WAX毛细管柱(15 m× 0.53 mm, 1 µm)(日本岛津公司)。
将从-80 ℃冰箱中取出的Viili发酵乳以5% (V/V)的接种量接种至经灭菌的12% (m/V)脱脂乳培养基中, 连续传代活化3代后备用。
移取Viili发酵乳1 mL于9 mL生理盐水中进行梯度稀释, 选取10-1~10-7梯度的稀释液100 µL涂布与MRS琼脂培养基平板上。倒置平板于37 ℃培养箱中培养24~48 h。取梯度适宜的平板, 使用接菌环挑取菌落形态不同的菌落, 划线于MRS平板上, 倒置平板于37 ℃培养箱中培养24~48 h后, 挑取单菌落重复划线步骤直至菌落形态一致。将单菌落接种至MRS液体培养基中扩大培养。
接菌环挑菌涂布于载玻片上, 使用革兰氏染色试剂盒染色后, 油镜观察菌种形态。取1 mL菌液, 使用细菌基因组提取试剂盒提取DNA。使用正向引物27F (AGAGTTTGATCMTGGCTCAG)、反向引物1492R (TACGGYTACCTTGTTACGACTT)对提取的DNA进行扩增, 扩增产物经琼脂糖凝胶电泳验证后, 送至苏州金唯智生物科技有限公司进行测序分析。将测序序列上传至美国国家生物技术信息中心(National Center of Biotechnology Information, NCBI)进行同源比对, 下载对比同源性大于99%标准菌株序列, 使用MEGA 11软件进行系统发育树的构建。
所用菌种活化3代后, 乳酸菌按照1×107 CFU/mL, 白地霉按照1×105 CFU/mL, 接入灭菌后的脱脂乳中, 30 ℃发酵。
使用采用E.Z.N.A. Soil试剂盒提取发酵乳中微生物总DNA, PCR扩增细菌与真菌的V3-V4可变区和ITS1区并使用2%琼脂糖凝胶回收PCR产物。根据荧光定量检测结果建库后, 利用Illumina Nextseq 2000平台进行测序(上海美吉生物医药科技有限公司)。
参照GB 5009.239—2016《食品安全国家标准 食品酸度的测定》对发酵乳的酸度测定。pH使用pH计进行测定。
参照TIAN等[10]的方法, 取10 mL发酵乳, 95 ℃除酶活5 min, 8000 r/min离心10 min, 取上清。在上清中加入5 mL三氯乙酸(12%), 搅拌处理后静止1 h, 8000 r/min离心15 min, 去除溶液中的蛋白质。加入上清4倍体积的95%乙醇溶液, 静止12 h。8000 r/min离心15 min, 取沉淀, 将沉淀置于8000~14000 kDa透析袋中, 每隔8 h换水一次, 透析处理2 d。
参照王萍[11]的方法, 使用苯酚-硫酸法对所得胞外多糖溶液含量进行测定。
质构仪设置回升参数20 mm, 形变百分量50%, 检测速度60 mm/min, 起始力1 N。对发酵乳的硬度、内聚性、黏度、弹性进行测定。
参照李晏蝶[12]的方法略有修改, 称取2 g发酵乳样本放入20 mL顶空瓶中, 40 ℃孵育20 min, 孵育转速500 r/min, 设置进样量为500 µL。
GC-IMS设置: IMS温度: 45 ℃; 分析时间: 30 min; 进样针温度: 85 ℃; 柱温: 40 ℃, 载气设置见表1
实验均设计3组平行, 使用SPSS 26、Origin 2021、MEGA 11、Grand Pad Prism 10以及美吉生物云平台对数据进行处理分析。
在本研究中, 采用高通量16S rRNA对Viili样品的微生物多样性进行分析。Viili样品中含有较为丰富的细菌, 共检测出4个细菌属, 其中乳球菌属(Lactococcus)、链球菌属(Streptococcus)、明串珠菌属(Leucanostoc)作为优势细菌属, 占比分别为49.12%、40.67%、10.21%。在发酵过程中, 乳酸菌作为关键菌种, 利用发酵环境的糖类等营养物质, 通过糖酵解等生化过程转化为乳酸、乙醇、维生素以及风味物质, 赋予乳制品特殊的质地和风味[13]
对Viili发酵乳中的真菌多样性进行分析, 检测出的均为真菌Saccharomycetes(酵母纲)。在属水平上, 共检测到2个真菌属, 分别为地霉属(Geotrichum)、双足囊菌属(Dipodascaceae), 其中地霉属的相对丰度高, 为优势真菌属。地霉属中的代表菌种白地霉是奶酪以及多种天然发酵食品中存在的真菌, 并且白地霉提供了Viili发酵乳表面特殊的质地, 是Viili发酵乳中的重要真菌[14-15]
革兰氏染液染色后使用光学显微镜观察菌体形态, 对筛选得到的菌种进行初步鉴定。部分微生物镜检结果见图1, 筛选得到的微生物有球状、杆状、链球状, 存在形态差异。
将初步分离纯化筛选所得菌种16S rRNA基因序列上传至NCBI数据库进行比对分析, 选用序列同源性大于99%标准菌株序列为参照。根据细菌多样性分析, 优选各属中生长性能最佳的菌株进行复合发酵培养。通过MEGA 11软件构建系统发育树构建菌种同源进化树[16], 结果如图2所示, 最终确定分离筛选菌株乳酸乳球菌H2、嗜热链球菌H17、肠膜明串珠菌H30、副干酪乳杆菌H33作为发酵菌种。
使用Design Expert 13软件中的混料设计, 模拟Viili发酵乳中细菌添加的最佳配比。根据微生物多样性测定结果中的相对丰度确定不同种细菌在发酵剂体系中添加范围[17]。设定不同菌比例总和100%, 确定乳酸乳球菌H1、嗜热链球菌H2、肠膜明串珠菌H3、副干酪乳杆菌H4添加比例分别为30%~60%、20%~50%、10%~30%、5%~20%。混料设计结果如表2所示。将4株细菌在适宜培养基和培养条件下培养至对数期时, 调整为1×109 CFU/mL的菌悬液。将各复配发酵菌种按比例混合后, 接种至脱脂乳中进行发酵, 初始接种量为1×107 CFU/mL。
同时发酵, 测定发酵终点Viili发酵乳与复配发酵乳的pH。如图3所示, Viili发酵乳在18 h达到发酵终点时pH为4.78, 滴定酸度为136 °T, 达到酪蛋白的等电点范围。如图4所示, 所有复配组合在24 h的pH均能达到4.7左右。复配组与Viili发酵乳相比, 发酵终点时pH均无显著差异。说明复配组中的微生物有良好的代谢活力, 可以达到与Viili发酵乳相当的酸度水平。
Viili发酵乳在18 h时出现凝乳现象, 复配发酵组别均在24 h左右凝乳, 复配组凝乳时间无显著差异, 与Viili发酵乳相比, 达到凝乳所需的时间更长, 说明复配组中菌群具有较好的发酵能力但产酸能力与Viili发酵乳中菌群相比较差, 可能由于复配组微生物菌株数量没有Viili发酵乳中丰富。
发酵乳的质地是决定产品质量和口感的重要因素, 适宜的口感不仅会提升消费者对产品的喜爱度, 更能保证产品在储存和运输阶段具有稳定性[18]。对Viili发酵乳和复配发酵组的硬度、黏度、内聚性、弹性进行测定。将质构的数据上传至美吉生物云平台, 采用Bray-curtis聚类方法进行层级聚类分析, 结果如图5所示。根据分支间的距离与分支的长度, 将21个实验组划分为3个簇。层级聚类树状图可以清晰的反映复配发酵组与Viili发酵组之间的质构的相似度, 结果表明复配组中编号为S2、S4、S6、S8、S13、S17、S18的组合与Viili发酵乳的质构更相似。将Viili发酵乳所在的分支划分为最小的簇, 可以进一步筛选出复配组S4、S13、S17, 这3组在质构上与Viili发酵乳相似度更高。
为寻找不同簇之间质构的差异, 对上述3簇中个组合的质构数据进行差异分析。由图6可知, 其他两簇与Viili发酵乳所在簇(第3簇)的硬度有明显差异, 簇2硬度与胶黏值显著高于簇3。内聚性与弹性是衡量酸奶结构稳定性的重要指标, 其数值越接近1, 说明发酵乳的质地越均匀, 凝胶结构越稳定[19-20]。结果说明复配组与Viili发酵乳的内聚性与弹性相似。
黏稠的质地是Viili发酵乳的特征, 并与其他发酵乳相比, Viili发酵乳具有独特的丝滑感和延展性。胞外多糖分子通常具有长链结构, 可以在水中形成高度交联的网络结构, 从而限制水分子的流动, 防止乳中的蛋白质沉降, 为发酵乳提供黏稠的质地和顺滑的口感[21-22]。有研究表明, 发酵乳中微生物产生的胞外多糖具有抗氧化、提高免疫力、抑制癌细胞增殖的作用[23]
对实验组的胞外多糖含量进行提取测定, 结果如图7所示, 复配组中胞外多糖的含量差异较大, 其中S11、S17、S18的胞外多糖含量显著高于Viili发酵乳组(P<0.05)。这也解释了质构的结果, 胞外多糖含量增加, 会提升发酵乳的黏度, 可以更好地维持发酵乳的稳定性[24]
风味是决定发酵产品的关键性因素, 发酵食品中多种挥发性风味物质协同作用塑造了特有的整体风味, 发酵乳中的风味物质的形成与原料乳中蛋白质、糖类、脂类被微生物分解转化有关, 乳酸菌和真菌通过代谢途径生成了特定的风味化合物[25-27]。本研究中采用GC-IMS对比复配组发酵乳和Viili发酵乳中的挥发性风味物质, 并对两组所检测出的风味物质进行分析。21组样品中共鉴定出44种挥发性化合物如表3所示, 其中包含14种酯类、3种酸类、5种醇类、8种醛类、5种酮类、4种酚类及5种其他类型的风味物质。酯类物质在所检测出的挥发性风味物质中所占比重高, 这可能与白地霉能够产生脂肪酶与酯化酶有关[28]。此外, 酯类物质种类的多样性也是Viili发酵乳呈现复杂风味的重要原因之一。除了酯类物质外, 还检测到了多种挥发性, 包括醇类、醛类、酸类、酮类和酚类等。这些挥发性风味物质共同作用, 赋予了Viili发酵乳独特的风味和香气[29-30]
GC-IMS指纹图谱可以展示样本组别中挥发性成分的分布以及相对含量[12]。通过将所有实验组的挥发性风味物质导出并生成指纹图谱, 可以直观地揭示复配组与Viili发酵乳之间的风味差异。图8中每点化合物的相对浓度用不同的颜色代表, 白色代表相对浓度低, 红色代表相对浓度高。分析可得, 多数复配组检测出与Viili发酵乳相近的挥发性风味物质, 其中S2、S3等检测出挥发性风味物质较Viili组数量多, 说明复配组可以达到Viili的特征风味。
为比较复配组与Viili发酵乳风味的相似性, 基于风味物质的相对定量数据上传至美吉生物云平台进行进行主成分分析(principal component analysis, PCA), 结果如图9所示。主成分分析结果中, 样本之间距离远近代表风味的整体相似度, 组别之间分离程度越大, 差异越大[31]。分析结果表明, 复配组的整体风味差异明显, 其中S1、S2、S3、S13和S14的风味与Viili发酵乳相似。其余复配组虽然风味较为集中, 但组间仍存在一定差异。
将所有组检测出的挥发性风味物质的相对含量标准化后上传至美吉生物云平台, 采用Bray-curtis的行列距离算法绘制成聚类热图。每一行代表不同的挥发性成分, 每一列代表不同组别。热图中的颜色代表相对含量, 红色代表含量高的物质, 蓝色表示含量较低。由图10可知, 复配组S1、S2、S3、S4、S13、S14、S15中多数挥发性风味物质的相对含量高于其他复配组, 并与Viili发酵乳聚在同一簇, 风味相近。
本研究从Viili中筛选出具有良好发酵能力的, 乳酸乳球菌、肠膜明串珠菌、副干酪乳杆菌、嗜热链球菌, 通过不同组合探究复合发酵剂中4中乳酸菌添加的比例。综合分析表明, 复配组S13 (活菌数之比为H2:H17:H30:H33= 11:4:4:1)在质地和风味方面均接近Viili发酵乳, 复配组S17 (活菌数之比为H2:H17:H30:H33=30:35:16:19)胞外多糖含量高并与Viili发酵乳相似质地, 具有作为发酵剂开发的潜力。后续研究中可以进一步分析Viili发酵乳中微生物复杂的相互作用, 对发酵剂的配比进一步优化, 为国内发酵乳发酵剂提供新选择。
