Splice-switching antisense oligonucleotides (ASOs) and engineered U7 small nuclear ribonucleoprotein (U7 Sm OPT) are the most commonly used methods for exon skipping. However, challenges remain, such as limited organ delivery and repeated dosing for ASOs and unknown risks of by-products produced by U7 Sm OPT. Here, we showed that antisense circular RNAs (AS-circRNAs) can effectively mediate exon skipping in both minigene and endogenous transcripts. We also showed a relatively higher exon skipping efficiency at the tested Dmd minigene than U7 Sm OPT. AS-circRNA specifically targets the precursor mRNA splicing without off-target effects. Moreover, AS-circRNAs with adeno-associated virus (AAV) delivery corrected the open reading frame and restored the dystrophin expression in a mouse model of Duchenne muscular dystrophy. In conclusion, we develop an alternative method for regulating RNA splicing, which might be served as a novel tool for genetic disease treatment.

Myofibroblasts, the primary effector cells for implant-induced fibrosis, contribute to this process by secreting excessive collagen-rich matrix and contracting. Thus, approaches that suppress myofibroblasts may achieve desirable suppression effects in the fibrotic process. As one of the important physical properties of materials, material topographical structures have been proven to affect various aspects of cell behaviors, so is it possible to manipulate the formation of myofibroblasts by tailoring the topographical properties of medical devices? In this study, polycaprolactone (PCL) surfaces with typical micropatterns (micro column and micro pit) were fabricated. The regulatory effects of surface micropatterns on the myofibroblastic differentiation of fibroblasts were investigated. Compared to the flat surfaces and surfaces with micro pit, surfaces with micro columns triggered the F- to G-actin transition, inhibiting the nuclear transfer of myocardin-related transcription factor-A. Subsequently, the downstream gene α-smooth muscle actin, which is a marker of myofibroblasts, was suppressed. Further in vivo investigation showed that PCL implants with micro-column-patterned surfaces inhibited the formation of peri-implant fibrotic capsules. Our results demonstrate that surface topographical properties are a potent regulator of fibroblast differentiation into myofibroblasts and highlight the antifibrotic potential of modifying surfaces with micro-column patterns.

Cellular behaviors and functions can be regulated by mechanical cues from microenvironments, which are transmitted to nucleus through the physical connections of cytoskeletons in the cells. How these physical connections determine transcriptional activity were not clearly known. The actomyosin, which generates intracellular traction force, has been recognized to control the nuclear morphology. Here, we have revealed that microtubule, the stiffest cytoskeleton, is also involved in the process of nuclear morphology alteration. The microtubule negatively regulates the actomyosin-induced nuclear invaginations but not the nuclear wrinkles. Moreover, these nuclear shape changes are proven to mediate the chromatin remodeling, which essentially mediates cell gene expression and phenotype determination. The actomyosin disruption leads to the loss of chromatin accessibility, which can be partly recovered by microtubule interference through nuclear shape control. This finding answers the question of how mechanical cues regulate chromatin accessibility and cell behaviors. It also provides new insights into cell mechanotransduction and nuclear mechanics.

As 2 indispensable counterparts in one catalysis system, the independent reduction and oxidation reactions require synergetic regulation for cooperatively promoting redox efficiency. Despite the current success in promoting the catalytic efficiency of half reduction or oxidation reactions, the lack of redox integration leads to low energy efficiency and unsatisfied catalytic performance. Here, we exploit an emerging photoredox catalysis system by combining the reactions of nitrate reduction for ammonia synthesis and formaldehyde oxidation for formic acid production, in which superior photoredox efficiency is achieved on the spatially separated dual active sites of Ba single atoms and Ti3+. High catalytic redox rates are accomplished for respective ammonia synthesis (31.99 ± 0.79 mmol gcat−1 h−1) and formic acid production (54.11 ± 1.12 mmol gcat−1 h−1), reaching a photoredox apparent quantum efficiency of 10.3%. Then, the critical roles of the spatially separated dual active sites are revealed, where Ba single atoms as the oxidation site using h+ and Ti3+ as the reduction site using e− are identified, respectively. The efficient photoredox conversion of contaminants is accomplished with environmental importance and competitive economic value. This study also represents a new opportunity to upgrade the conventional half photocatalysis into the complete paradigm for sustainable solar energy utilization.

The spread of the latest SARS-CoV-2 variant Omicron is particularly concerning because of the large number of mutations present in its genome and lack of knowledge about how these mutations would affect the current SARS-CoV-2 vaccines and treatments. Here, by performing phylogenetic analysis using the Omicron spike (S) protein sequence, we found that the Omicron S protein presented the longest evolutionary distance in relation to the other SARS-CoV-2 variants. We predicted the structures of S, M, and N proteins of the Omicron variant using AlphaFold2 and investigated how the mutations have affected the S protein and its parts, S1 NTD and RBD, in detail. We found many amino acids on RBD were mutated, which may influence the interactions between the RBD and ACE2, while also showing the S309 antibody could still be capable of neutralizing Omicron RBD. The Omicron S1 NTD structures display significant differences from the original strain, which could lead to reduced recognition by antibodies resulting in potential immune escape and decreased effectiveness of the existing vaccines. However, this study of the Omicron variant was mainly limited to structural predictions, and these findings should be explored and verified by subsequent experiments. This study provided basic data of the Omicron protein structures that lay the groundwork for future studies related to the SARS-CoV-2 Omicron variant.

Sodium alanate (NaAlH4) with 5.6 wt% of hydrogen capacity suffers seriously from the sluggish kinetics for reversible hydrogen storage. Ti-based dopants such as TiCl4, TiCl3, TiF3, and TiO2 are prominent in enhancing the dehydrogenation kinetics and hence reducing the operation temperature. The tradeoff, however, is a considerable decrease of the reversible hydrogen capacity, which largely lowers the practical value of NaAlH4. Here, we successfully synthesized a new Ti-dopant, i.e., TiH2 as nanoplates with ~50 nm in lateral size and ~15 nm in thickness by an ultrasound-driven metathesis reaction between TiCl4 and LiH in THF with graphene as supports (denoted as NP-TiH2@G). Doping of 7 wt% NP-TiH2@G enables a full dehydrogenation of NaAlH4 at 80°C and rehydrogenation at 30°C under 100 atm H2 with a reversible hydrogen capacity of 5 wt%, superior to all literature results reported so far. This indicates that nanostructured TiH2 is much more effective than Ti-dopants in improving the hydrogen storage performance of NaAlH4. Our finding not only pushes the practical application of NaAlH4 forward greatly but also opens up new opportunities to tailor the kinetics with the minimal capacity loss.

Associative learning is a critical learning principle uniting discrete ideas and percepts to improve individuals’ adaptability. However, enabling high tunability of the association processes as in biological counterparts and thus integration of multiple signals from the environment, ideally in a single device, is challenging. Here, we fabricate an organic ferroelectric neuromem capable of monadically implementing optically modulated associative learning. This approach couples the photogating effect at the interface with ferroelectric polarization switching, enabling highly tunable optical modulation of charge carriers. Our device acts as a smarter Pavlovian dog exhibiting adjustable associative learning with the training cycles tuned from thirteen to two. In particular, we obtain a large output difference (>103), which is very similar to the all-or-nothing biological sensory/motor neuron spiking with decrementless conduction. As proof-of-concept demonstrations, photoferroelectric coupling-based applications in cryptography and logic gates are achieved in a single device, indicating compatibility with biological and digital data processing.

Multifunctionality, interference-free signal readout, and quantum effect are important considerations for flexible sensors equipped within a single unit towards further miniaturization. To address these criteria, we present the slotted carbon nanotube (CNT) junction features tunable Fano resonance driven by flexoelectricity, which could serve as an ideal multimodal sensory receptor. Based on extensive ab initio calculations, we find that the effective Fano factor can be used as a temperature-insensitive extrinsic variable for sensing the bending strain, and the Seebeck coefficient can be used as a strain-insensitive intrinsic variable for detecting temperature. Thus, this dual-parameter permits simultaneous sensing of temperature and strain without signal interference. We further demonstrate the applicability of this slotted junction to ultrasensitive chemical sensing which enables precise determination of donor-type, acceptor-type, and inert molecules. This is due to the enhancement or counterbalance between flexoelectric and chemical gating. Flexoelectric gating would preserve the electron–hole symmetry of the slotted junction whereas chemical gating would break it. As a proof-of-concept demonstration, the slotted CNT junction provides an excellent quantum platform for the development of multistimuli sensation in artificial intelligence at the molecular scale.
