Objective

To establish and validate a highly sensitive and selective high performance liquid chromatography-tandem mass spectrometry (HPLC-MS/MS) method for the determination of lurasidone, which was used subsequently to clinical lurasidone blood drug concentration monitoring.

Methods

Tadalafil was used as internal standard. Following a deproteinization procedure, lurasidone and the internal standard (tadalafil) were isostatically eluted using a mobile phase composed of methanol and 0.1% aqueous formic acid (50:50, v/v) at a flow rate of 0.70 mL·min-1. The chromatographic separation was achieved within 4.0 min on an Agilent ZORBAX Eclipse plus C8(4.6 mm×100.0 mm,3.5 μm). Quantification was performed using a triple-quadrupole mass spectrometer operating in positive electrospray ionization (ESI) mode with multiple reaction monitoring (MRM). The method was validated for selectivity, linearity (calibration curve), precision and accuracy, matrix effect, extraction recovesise, stability and dilutive integrity. The concentrations of 14 clinical samples were measured after this method was validated.

Results

The calibration curve for lurasidone in human plasma demonstrated linearity over the concentration range of 0.50-500.00 ng·mL-1. The precision data (both intra- and inter-day) for the three QC levels ranged from 2.87% to 10.03%. Accuracy (relative error) was within±15% of the nominal values. The plasma samples maintained stability for 28 h at room temperature, for 85 days at -20 ℃ and through five freeze-thaw cycles. The measured concentrations of clinical samples were within the range of the standard curve, with concentrations ranging from 2.63 to 21.17 ng·mL-1.

Conclusion

The validated method is proved to be convenient, accurate, and sensitive for the quantification of lurasidone in human plasma. The method is proved to be suitable for the monitoring of plasma concentration and pharmacokinetics study of lurasidone.

目的

建立和验证一种具有高灵敏度和高选择性的液相色谱-串联质谱方法(HPLC-MS/MS),用于人血浆中鲁拉西酮的浓度和进行血药浓度监测。

方法

选用他达拉非作为内标,通过蛋白沉淀法对样品进行前处理,以甲醇和0.1%甲酸水溶液流动相,采用50:50 (v/v)等度洗脱,流速为0.70 mL·min-1,Agilent公司色谱柱,型号为ZORBAX Eclipse plus C8 (4.6 mm×100.0 mm,3.5 μm),质谱采集时间为4.0 min。利用电喷雾电离(ESI),在多反应监测(MRM)模式下,使用HPLC-MS/MS分析测定血浆样品浓度。考察该方法的标准曲线和定量下限、精密度、准确度、选择性(干扰)、稳定性、回收率、基质效应和稀释可靠性。该方法经过方法学验证后,对收集的14例服用鲁拉西酮患者血浆标本药物浓度分析测定。

结果

鲁拉西酮的血浆浓度在0.50~500.00 ng·mL-1范围内具有良好线性,批内、批间样品检测的精密度相对标准偏差均在2.87%~10.03%以内,准确度与理论浓度的偏差在±15%以内。鲁拉西酮血浆样品前处理过程室温放置28 h,血浆样品在经历5次冻融循环(-20 ℃)及长期冷冻保存(-20 ℃,85 d)条件下均表现出良好的稳定性。临床样品测定浓度均在标准曲线浓度范围内,浓度为2.63~21.17 ng·mL-1

结论

该研究建立的分析方法简便、灵敏度高、选择性好,可有效支持鲁拉西酮的临床血药浓度监测及药代动力学研究。

目的

建立和验证一种具有高灵敏度和高选择性的液相色谱-串联质谱方法(HPLC-MS/MS),用于人血浆中鲁拉西酮的浓度和进行血药浓度监测。

方法

选用他达拉非作为内标,通过蛋白沉淀法对样品进行前处理,以甲醇和0.1%甲酸水溶液流动相,采用50:50 (v/v)等度洗脱,流速为0.70 mL·min-1,Agilent公司色谱柱,型号为ZORBAX Eclipse plus C8 (4.6 mm×100.0 mm,3.5 μm),质谱采集时间为4.0 min。利用电喷雾电离(ESI),在多反应监测(MRM)模式下,使用HPLC-MS/MS分析测定血浆样品浓度。考察该方法的标准曲线和定量下限、精密度、准确度、选择性(干扰)、稳定性、回收率、基质效应和稀释可靠性。该方法经过方法学验证后,对收集的14例服用鲁拉西酮患者血浆标本药物浓度分析测定。

结果

鲁拉西酮的血浆浓度在0.50~500.00 ng·mL-1范围内具有良好线性,批内、批间样品检测的精密度相对标准偏差均在2.87%~10.03%以内,准确度与理论浓度的偏差在±15%以内。鲁拉西酮血浆样品前处理过程室温放置28 h,血浆样品在经历5次冻融循环(-20 ℃)及长期冷冻保存(-20 ℃,85 d)条件下均表现出良好的稳定性。临床样品测定浓度均在标准曲线浓度范围内,浓度为2.63~21.17 ng·mL-1

结论

该研究建立的分析方法简便、灵敏度高、选择性好,可有效支持鲁拉西酮的临床血药浓度监测及药代动力学研究。

Objective

To establish and validate a highly sensitive and selective high performance liquid chromatography-tandem mass spectrometry (HPLC-MS/MS) method for the determination of lurasidone, which was used subsequently to clinical lurasidone blood drug concentration monitoring.

Methods

Tadalafil was used as internal standard. Following a deproteinization procedure, lurasidone and the internal standard (tadalafil) were isostatically eluted using a mobile phase composed of methanol and 0.1% aqueous formic acid (50:50, v/v) at a flow rate of 0.70 mL·min-1. The chromatographic separation was achieved within 4.0 min on an Agilent ZORBAX Eclipse plus C8(4.6 mm×100.0 mm,3.5 μm). Quantification was performed using a triple-quadrupole mass spectrometer operating in positive electrospray ionization (ESI) mode with multiple reaction monitoring (MRM). The method was validated for selectivity, linearity (calibration curve), precision and accuracy, matrix effect, extraction recovesise, stability and dilutive integrity. The concentrations of 14 clinical samples were measured after this method was validated.

Results

The calibration curve for lurasidone in human plasma demonstrated linearity over the concentration range of 0.50-500.00 ng·mL-1. The precision data (both intra- and inter-day) for the three QC levels ranged from 2.87% to 10.03%. Accuracy (relative error) was within±15% of the nominal values. The plasma samples maintained stability for 28 h at room temperature, for 85 days at -20 ℃ and through five freeze-thaw cycles. The measured concentrations of clinical samples were within the range of the standard curve, with concentrations ranging from 2.63 to 21.17 ng·mL-1.

Conclusion

The validated method is proved to be convenient, accurate, and sensitive for the quantification of lurasidone in human plasma. The method is proved to be suitable for the monitoring of plasma concentration and pharmacokinetics study of lurasidone.

鲁拉西酮是2010年美国批准上市的新型抗精神病药,用于成人/青少年精神分裂症及成人/儿童双相抑郁障碍的治疗[1-3]。鲁拉西酮片(商品名:罗舒达®)于2019年登陆中国,用于治疗成年患者精神分裂症,每日口服1次。《鲁拉西酮治疗成人精神分裂症的实践指南》[4]中确定的鲁拉西酮有效剂量范围为37~148 mg·d-1,美国食品药品监督管理局(Food and drug administration, FDA)批准的剂量为40~160 mg·d-1,中国获批剂量为40~80 mg·d-1,国内使用剂量与欧洲/美国等地的差异,对临床医生用药产生了一定的困扰。随餐给药(大于350焦耳)[5]可促进鲁拉西酮的药物利用,患者需在每日进餐时服用鲁拉西酮片,对于精神疾病患者坚持饮食指导可能存在一定困难。《神经精神药理学治疗药物监测共识指南:2017版》[6-7]建议对鲁拉西酮进行治疗药物监测(therapeutic drug monitoring, TDM),以便对用药方案进行及时调整。鲁拉西酮在生物样品中内源性干扰物质多、基质效应显著,且血药浓度较低,因此需要建立高灵敏度、高选择性的检测分析方法。本研究拟采用高效液相色谱-串联质谱法(high performance liquid chromatography-tandem mass spectrometry, HPLC-MS/MS)测定人血浆中的鲁拉西酮浓度,并应用于鲁拉西酮血药浓度监测。
血浆 空白人血浆、鲁拉西酮血浆样品来自苏州市广济医院就诊门诊和住院患者,本研究经苏州市广济医院伦理委员会批准,伦理批号:苏广伦审2023-036号。
药品 盐酸鲁拉西酮对照标准物质,纯度:99.0%,批号:D1225B,大连美伦生物技术有限公司生产;他达拉非对照标准物质,纯度:98.97%,批号:TDF-0415002,印度SMS Pharmaceuticals Ltd.生产;肝素抗凝剂,批号:2307209,浙江拱东医疗器械股份有限公司生产。
仪器 Agilent 1260型高效液相色谱仪,美国Agilent公司产品;API4000三重四极杆质谱仪,美国AB Sciex公司产品,配备Analyst 1.6.3数据处理软件。
色谱条件 色谱柱:ZORBAX Eclipse plus C8 (4.6 mm×100.0 mm,3.5 μm),保护柱:Phenomenex Security Guard TM C8 (4.0 mm×3.0 mm);流动相:甲醇-0.1%甲酸水溶液=50:50 (v/v);流速0.7 mL·min-1,进样体积10 μL,柱温35 ℃,分析时间4.0 min。
质谱条件 采用电喷雾电离源,以正离子模式进行检测;雾化气压力60 psi,干燥气(加热辅助气)压力60 psi,离子源温度650 ℃,电喷雾电压5500 V;采用多反应监测;鲁拉西酮和内标他达拉非监测离子对分别为m/z 493.5→166.2和m/z 390.2→268.2,去簇电压分别为130和50 V,入口电压均为10 V,碰撞能量分别为55和18 V,出口电压分别为10和15 V,停留时间均为200 ms。
鲁拉西酮溶液配制 称量盐酸鲁拉西酮对照标准物质10.85 mg(含量校正为10.00 mg鲁拉西酮),置于10 mL容量瓶中,用甲醇溶解并定容至刻度,混匀,制得质量浓度为1.00 mg·mL-1鲁拉西酮标准曲线储备液。以甲醇为稀释剂,逐级稀释制备系列浓度的标准曲线工作溶液,最终目标质量浓度分别为0.01、0.02、0.04、0.20、0.40、2.00、4.00和10.00 μg·mL-1。重新称量盐酸鲁拉西酮对照标准物质,以甲醇为稀释剂,配制最低定量下限、低、中、高4个浓度的质控工作液,其质量浓度分别0.01、0.03、0.30和8.00 μg·mL-1
内标他达拉非溶液配制 称量他达拉非对照标准物质10.10 mg(含量校正为10.00 mg他达拉非),置于10 mL容量瓶中,用甲醇溶解并定容至刻度,混匀,内标他达拉非储备液质量浓度为1.00 mg·mL-1。移取内标他达拉非储备液,以甲醇为稀释剂,将样品配制成目标质量浓度为2.00 μg·mL-1的内标他达拉非工作溶液。
鲁拉西酮血浆样品配制(标准曲线和质控) 移取空白生物基质(血浆)200 μL,转移至1.5 mL离心管中,依次添加鲁拉西酮标准曲线工作溶液10 μL(或质控工作溶液)和他达拉非内标工作溶液50 μL,加入乙腈蛋白沉淀600 μL,涡旋振荡1 min,在4 ℃、13 000 r·min-1条件下离心10 min。取200 μL上清液转移至含超纯水200 μL的洁净进样瓶中,涡旋混匀后,经HPLC-MS/MS系统进行鲁拉西酮定量分析。
血浆样品处理 移取受试者待测血浆样品200 μL,转移至1.5 mL离心管中,加入甲醇10 μL(补偿体积)和50 μL他达拉非工作溶液,加入乙腈600 μL蛋白沉淀,涡旋振荡1 min,在4 ℃、13 000 r·min-1离心10 min。取上清液200 μL转移至含200 μL超纯水的洁净进样瓶中,涡旋混匀后,经HPLC-MS/MS系统进行鲁拉西酮定量分析。
专属性 分别取鲁拉西酮对照溶液、他达拉非对照溶液、空白血浆、鲁拉西酮标准含药血浆样品、患者口服鲁拉西酮胶囊80 mg后谷浓度血浆样品、鲁拉西酮定量下限标准含药血浆样品,按“溶液配制与血浆样本处理”项处理后检测。
标准曲线线性范围与定量下限 在空白血浆200 μL中用移液器加入鲁拉西酮标准曲线工作溶液10 μL,配制鲁拉西酮血浆标准曲线样品,各血浆样品中鲁拉西酮质量浓度分别为0.50、1.00、2.00、10.00、20.00、100.00、200.00和500.00 ng·mL-1,处理步骤按照“溶液配制与血浆样本处理”,通过液质联用仪分析,基于标准曲线8个浓度样品中鲁拉西酮与内标的峰面积比定量[As/Ai, 鲁拉西酮峰面积与他达拉非峰面积比值],对理论浓度与响应值(鲁拉西酮与内标峰面积比值)进行线性回归,权重因子为1/x2,定量下限为0.50 ng·mL-1
精密度与回收率 定量下限和质控系列血浆样品(鲁拉西酮质量浓度为0.50、1.50、15.00和400.00 ng·mL-1)。用质控样品(分析批数量为3,每批包含4个浓度水平质控,每个浓度平行6份)进行测定,通过标准曲线计算测定浓度,评估批内和批间准确度和精密度。回收率测试血浆样品:处理步骤按照按照“3溶液配制与血浆样本处理”,鲁拉西酮质量浓度分别为1.50、15.00和400.00 ng·mL-1,平行6份样品,用液质联用仪系统进行分析,经色谱图积分处理后,记录鲁拉西酮(A1)和内标他达拉非(A2)峰面积。回收率参比血浆样品:每个浓度水平(低、中、高)平行制备6份质控样品。移取空白血浆样品200 μL,加入乙腈沉淀蛋白后,完全转移上清液,依次加入鲁拉西酮质控工作溶液10 μL和内标他达拉非工作溶液50 μL混匀,取200 μL转移至含200 μL超纯水的洁净进样瓶中,涡旋混匀后进样测定,计算鲁拉西酮(A3)和内标他达拉非(A4)各自的峰面积。按以下公式计算回收率:鲁拉西酮回收率(%) = (A1/A3) × 100%,内标回收率(%) = (A2/A4) × 100%。
基质效应 取个体空白血浆200 μL,平行制备6份,来源不同个体,转移至1.5 mL离心管中,各加入乙腈蛋白沉淀600 μL,涡旋1 min,在2~8 ℃,13 000 r·min-1条件下,离心10 min,收集上清液作为血浆基质备用。取超纯水200 μL置于1.5 mL离心管中,按上述相同步骤处理,得混合溶液作为非基质对照。取混合溶液(血浆基质上清液或非基质对照溶液)700 μL,依次添加鲁拉西酮质控工作溶液10 μL(低、中、高三个浓度水平)和他达拉非内标工作溶液50 μL,混匀。吸上述混合液200 μL,加入200 μL超纯水,混匀后用液质联用仪系统进行检测分析,获鲁拉西酮和内标他达拉非峰面积。分别记录鲁拉西酮在血浆基质样品中的峰面积B基质和纯溶液中的峰面积B非基质,内标他达拉非在血浆基质样品中的峰面积C基质和纯溶液中的峰面积C非基质,鲁拉西酮基质效应因子(%)=(B基质/B非基质)×100%,内标他达拉非的基质效应因子(%)=(C基质/C非基质)×100%。鲁拉西酮内标校正归一化基质效应因子=鲁拉西酮基质效应因子/内标他达拉非基质效应因子×100%,最终报告鲁拉西酮各浓度水平的内标校正归一化基质效应因子。溶血基质效应通过用溶血的血浆配制低、中、高质控浓度样本进行考察,所有质量浓度水平下,样品测定均值与理论值的相对误差均需控制在±15.0%以内,每个浓度的样品测定值的CV应小于15.0%。高脂血基质效应通过用空白高脂血浆进行考察,高脂血浆配制方法:脂肪乳与空白血浆按照1:19比例混合,脂肪乳占5%。
稳定性试验 本研究系统评估了鲁拉西酮在不同储存条件下的稳定性;全血稳定性:室温(白光)放置6 h;短期血浆稳定性:室温(白光)放置48 h;长期血浆稳定性:-20 ℃条件下储存14 d;冻融稳定性:血浆样品在-20 ℃条件下反复冻融循环5次。此外,本研究还考察了以下稳定性:鲁拉西酮和内标储备液在室温白光条件下放置48 h,鲁拉西酮和内标储备液在-20 ℃冷冻保存184 d;待测样品在液相自动进样器中放置24 h。
残留效应 用标准曲线最高浓度样品进行残留评估。具体步骤为:首先进样标准曲线最高浓度样品,随后连续进样3针不同来源的空白血浆样品,计算空白样品中鲁拉西酮及内标的响应值,鲁拉西酮要求不超过定量下限(lower limit of quantification, LLOQ)响应值的20%,内标要求不超过平均响应值的5%。
该方法经过方法学验证后,选取2023年9月至2024年12月我院精神科采用鲁拉西酮治疗的14例患者的14个血浆样本进行药物浓度测定。使用肝素采血管采集服用鲁拉西酮患者血液样品,使用离心机将血液样品离心10 min(温度设置4 ℃,离心力3 200 r·min-1),分离上层血浆,保存于-20 ℃或更低温度的冰箱中,待测。患者在常规服药≥8周后评估阳性与阴性症状量表(positive and negative syndrome scalem, PANSS)判断疗效,量表由阴性量表7项(7~49分)、阳性量表7项(7~49分)及一般精神病理量表16项(16-112 分)组成,共30项,评分降低则患者症状得到改善。
专属性 鲁拉西酮和内标他达拉非的保留时间分别为2.10和3.00 min左右,血浆中内源性物质不干扰鲁拉西酮和内标他达拉非的测定,见图1
标准曲线与定量下限 标准曲线方程y=7.07x×10-3+1.27×10-3r=0.9964),血浆基质中鲁拉西酮在0.50~500.00 ng·mL-1内线性关系良好,定量下限为0.50 ng·mL-1
精密度与回收率 鲁拉西酮质控样品(定量下限及低、中、高浓度)的精密度与准确度,以及血浆中鲁拉西酮(低、中、高浓度)和内标的提取回收率见表1
基质效应 血浆中鲁拉西酮低、中、高3个浓度水平的基质效应均值分别为99.15%、99.03%和103.86%;他达拉非内标的基质效应平均为100.27%,低、中、高3个浓度水平的内标归一化基质因子分别为98.89%、98.77%和103.59%。溶血基质、高脂血基质低、中、高浓度样本,与理论值相比,测定结果均值相对误差均在±15.0%范围内。
稳定性试验 鲁拉西酮生物基质样品稳定性:全血样品在室温白光条件下保存6 h后,鲁拉西酮保持稳定;血浆样品在以下条件下均显示良好的稳定性:室温白光暴露48 h,-20 ℃长期储存14 d,-20 ℃经历5次冻融循环(每次循环包括完全融化后重新冷冻);待测样品在4 ℃自动进样器中放置24 h保持稳定;各浓度测定结果均值相对误差均在±15.0%范围内。
溶液稳定性 鲁拉西酮及他达拉非(内标)储备液在室温白光条件下存放48 h和置于冷冻(-20 ℃)保存184 d情况下,储备液均稳定。
残留效应 3个空白血浆样品分析结果显示:鲁拉西酮和内标(他达拉非)均未检出(峰面积为0),残留水平符合方法验证要求,对后续样品测定无干扰。
使用本分析方法测定14例服用鲁拉西酮患者谷浓度血浆标本,样本测定浓度均在标准曲线浓度范围内,浓度为2.63~21.17 ng·mL-1,平均质量浓度为8.12 ng·mL-1。《神经精神药理学治疗药物监测共识指南:2017版》[6]推荐治疗浓度15~40 ng·mL-1 ,14例患者中仅有2例在指南推荐的有效浓度范围,其他患者低于指南推荐的最低有效浓度范围,临床评估14例患者服用的鲁拉西酮(40或80 mg)后,PANSS评分降低,达到预期疗效。
本研究用液质联用法,具有特异性好和灵敏度高等诸多优点,优于文献[8]高效液相色谱-紫外光谱法。质谱的核心是根据离子的质荷比进行分离和检测,它能区分质量差异很小的化合物。LC-MS/MS用多反应监测模式,它先选择目标化合物的母离子,再选择其特定的子离子进行检测。这种“双选择性”(母离子+子离子)大大提高了对目标化合物的识别能力,有效降低了背景噪音和非目标化合物的干扰[9]。本研究乙腈直接沉淀蛋白,简便、快捷,定量下限为0.50 ng·mL-1,优于文献[8,10,11]报道(文献[8]18.00 ng·mL-1,文献[10]2.00 ng·mL-1和文献[11]1.00 ng·mL-1)。鲁拉西酮人体药代动力学研究中,鲁拉西酮在人体内表现出约20 h的消除半衰期特性。基于此药代动力学特征,研究设计需确保血样采集时间至少覆盖3个半衰期。这一较长的采样周期对分析方法的灵敏度提出了较高要求,特别是LLOQ需要满足低浓度样本的准确检测。本研究血浆样品预处理方法与分析优势:本研究采用血浆上清液200 μL与等体积超纯水混合的前处理方法,该设计具有多项技术优势:色谱行为改善:有效抑制二次化学平衡现象,使鲁拉西酮色谱峰形显著改善;基质效应控制:生物基质稀释50%,基质效应值控制在85%~115%范围内,显著降低LC-MS系统污染风险。本研究测定方法每个样本为4 min,可对临床样品进行快速高通量测定,为患者给药方案的调整提供合理依据,满足临床血药浓度监测的要求。本研究采用他达拉非为内标,专属性和基质效应结果均在可接受范围内,内标的基质效应平均为100.27%,鲁拉西酮低、中、高3个浓度水平的内标归一化基质因子分别为98.89%、98.77%和103.59%,验证数据说明测定方法其在实际应用条件下仍然能够提供足够准确和精密的定量结果。《神经精神药理学治疗药物监测共识指南:2017版》[6]推荐鲁拉西酮治疗参考浓度范围为15~40 ng·mL-1,但其参考的文献[12-15]均未列出治疗参考浓度范围,如:PATTEET[14]等描述治疗参考浓度范围时用“—”表示,未列出治疗参考浓度范围。目前中国获批剂量与欧洲/美国等地也存在差异,国外共识指南推荐的治疗参考浓度范围为15~40 ng·mL-1,不一定适合中国患者人群。本研究临床评估12例患者服用的鲁拉西酮(40或80 mg)后均达到预期疗效,但血药浓度低于指南推荐的最低有效浓度范围。精神分裂症患者口服单独或与食物同时服用120 mg鲁拉西酮[15],谷浓度Cmin为7.60~12.30 ng·mL-1,亦不在指南推荐的治疗参考浓度范围内(15~40 ng·mL-1)。对于TDM结果的解释,除了治疗参考浓度范围外还有另一个浓度范围,即剂量相关参考浓度范围。剂量确定后,预期Cmin可以用剂量相关浓度(dose-related concentration, DRC)因子乘以日剂量计算。当患者的血药浓度测得值在剂量相关参考浓度范围内时,就认为患者的血药浓度正常,可以认为高于或低于这个范围的血药浓度就提示可能存在潜在的异常情况,诸如依从性差、药物相互作用、药物代谢酶的基因多态性或者与药物排泄相关的器官存在病变。鲁拉西酮DRC低、高因子分别为0.09和0.13[6],中国日剂量为40和80 mg·d-1,计算鲁拉西酮剂量相关参考浓度范围为3.60~10.40 ng·mL-1。本研究平均质量浓度为8.12 ng·mL-1,在剂量相关参考浓度范围内。若按照美国批准的最大剂量160 mg·d-1,剂量相关参考浓度范围为3.60~20.80 ng·mL-1。鲁拉西酮主要通过CYP3A4酶代谢,血药浓度可能受到同时使用抑制或诱导CYP3A4的药物的影响[12]。酮康唑的存在增加了鲁拉西酮的血清浓度,这一效应与酮康唑对CYP3A4的抑制作用一致。相反,利福平由于诱导CYP3A4,使鲁拉西酮的Cmax和AUC降低了80%以上。因此,不应将CYP3A4的强效抑制剂(如酮康唑、克拉霉素、利托那韦、伏立康唑和米贝拉地尔)或诱导剂(如利福平、圣约翰草、苯妥英和卡马西平)与鲁拉西酮同时使用,并应在开始鲁拉西酮治疗前停用这些药物。精神分裂症患者口服单独或与食物同时服用120 mg 鲁拉西酮[15],Cmax为61.90~173.00 ng·mL-1,本研究建立的检测方法定量上限为500.00 ng·mL-1,可满足未来在中国人群中进行更高剂量或更精细化剂量调整的研究。后续本研究团队将继续开展相关研究,期待制定符合中国人群的治疗参考浓度范围,为临床个体化使用鲁拉西酮提供理论依据,进一步提高临床疗效,降低药物不良反应的发生。
