目的

针对口服易降解、基质复杂药物(阿司匹林、布洛芬、硝苯地平)的含量检测难题,优化高效液相色谱法(high performance liquid chromatography, HPLC)检测条件并结合固相萃取(solid phase extraction, SPE)前处理技术,建立准确、灵敏的分析方法。

方法

以阿司匹林(水解敏感)、布洛芬(脂溶性干扰)、硝苯地平(光热不稳定)为研究对象,通过优化流动相组成(pH、有机相比例)、色谱柱类型及柱温,结合HLB SPE小柱净化消除基质效应;采用外标法定量,验证线性范围、回收率、精密度等指标。

结果

3种药物检测方法均达到药典要求:阿司匹林线性范围为5~500 μg/mL (r=0.9998),加样回收率为98.2%~101.5%{日内/日间精密度[相对标准偏差(relative standard deviation, RSD)]≤1.5%};布洛芬线性范围为2~200 μg/mL (r=0.9997),回收率为97.8%~102.1%(RSD≤1.8%);硝苯地平线性范围为1~100 μg/mL (r=0.9999),回收率为98.5%~101.8%(RSD≤1.2%)。实际样品检测结果显示,方法抗干扰能力强,适用于常规实验室质量控制。

结论

优化的HPLC-SPE方法可有效解决易降解/复杂基质口服药物的含量检测问题,操作简便、结果可靠,具有显著实验室应用价值。

目的
