Objective To evaluate the uncertainty in determination of olive oil total migration by gas chromatography spectrometry, determine the factors affecting the accuracy of the results and provide a basis for accurate control of the olive oil total migration. Methods Gas chromatography spectrometry was used to determine the olive oil total migration, and a mathematical model was established. According to relevant rules and standards for uncertainty evaluation, the uncertainty was evaluated including repeatability, sample quality and adsorbed olive oil quality, instrument calibration, standard curve fitting, soaking area, etc. Results When The total migration of olive oil in the sample is ${4.0}\mathrm{{mg}}/{\mathrm{{dm}}}^{2}$, the expanded uncertainty was ${1.3}\mathrm{{mg}}/{\mathrm{{dm}}}^{2}\left({P={95}\%, k = 2}\right)$ . Conclusion The uncertainty of olive oil total migration mainly comes from repeatability, standard solution preparation and standard curve fitting.

目的 评定气相色谱法测定橄榄油总迁移量的不确定度,确定影响结果准确性的因素, 为橄榄油总迁量检测的准确度控制提供依据。方法 采用气相色谱法测定橄榄油总迁移量,建立数学模型。根据不确定度评定相关规则和标准对实验过程中的不确定度因素包括重复性、试样质量、仪器校准、标准曲线拟合、浸泡面积等进行不确定度评定。结果 试样的橄榄油总迁移量为 ${4.0}\mathrm{{mg}}/{\mathrm{{dm}}}^{2}$ ,其扩展不确定度为 ${1.4}\mathrm{{mg}}/{\mathrm{{dm}}}^{2}$ ($P ={95}\%$ ,k=2)。结论 橄榄油总迁移量的不确定度主要来源有重复性测定和试样提取的橄榄油质量。

Objective To evaluate the uncertainty in determination of olive oil total migration by gas chromatography spectrometry, determine the factors affecting the accuracy of the results and provide a basis for accurate control of the olive oil total migration. Methods Gas chromatography spectrometry was used to determine the olive oil total migration, and a mathematical model was established. According to relevant rules and standards for uncertainty evaluation, the uncertainty was evaluated including repeatability, sample quality and adsorbed olive oil quality, instrument calibration, standard curve fitting, soaking area, etc. Results When The total migration of olive oil in the sample is ${4.0}\mathrm{{mg}}/{\mathrm{{dm}}}^{2}$, the expanded uncertainty was ${1.3}\mathrm{{mg}}/{\mathrm{{dm}}}^{2}\left({P={95}\%, k = 2}\right)$ . Conclusion The uncertainty of olive oil total migration mainly comes from repeatability, standard solution preparation and standard curve fitting.

总迁移量作为食品接触材料的关键的卫生性能指标之一, 是评估食品接触材料及制品安全的重要参数 [ 1 - 2 ] ,也是企业质量控制和政府监督管理中的重要参数 [ 3 ] 。模拟物的选择是根据食品接触材料及制品预期接触的食品种类确定的, 测试结果能比较真实地反映出食品接触材料的迁移状况 [ 4 - 6 ]
橄榄油总迁移量反映了预期接触油脂类食品接触材料及制品的质量安全。因其测试流程繁多, 过程复杂 [ 7 ] ,影响测试结果准确性的因素较多,所以试验过程中需要把控影响量值的关键控制点, 将影响降至最低, 从而得到较为准确的测试结果。不确定度是衡量实验数据准确程度的重要参数 [ 8 ] 。本研究依据 JJF 1059.1-2012《测量不确定度评定与表示》 [ 9 ] 分析实验过程各因素产生的不确定度 [ 10 - 12 ] , 确定影响检验结果不确定度的主要因素, 以期提高橄榄油总迁移量检测准确度, 对检验过程的质量控制产生指导意义。
XS204 分析天平(梅特勒-托利多公司);HH-4 恒温水浴锅 (常州市凯航仪器有限公司); DLSB-5/20 低温循环冷凝泵(上海越众仪器设备有限公司);R-210 旋转蒸发仪(瑞士 BUCHI 公司); 7890B-7693A 气相色谱 (配有氢火焰离子检测器, 美国 Agilent 公司);B-811 自动索氏提取器 (瑞士 BUCHI 公司);FD115 电加热鼓风干燥箱(德国 BINDER 公司 )。
橄榄油 (化学纯,上海国药集团),符合 $\mathrm{{GB}}$ 5009.156-2016《食品安全国家标准 食品接触材料及制品迁移试验预处理方法通则》附录 $\mathrm{A}$ 的要求 [ 13 ] ; 正戊烷 (分析纯,德国 CNW 科技公司),正庚烷 (分析纯,德国 CNW 科技公司),无水乙醇(分析纯, 德国 CNW 科技公司), 氢氧化钾 (化学纯, 江苏强盛功能化学股份有限公司),甲醇(分析纯, 德国 CNW 科技公司),三氟化硼甲醇混合液(分析纯, 德国 CNW 科技公司), 无水硫酸钠 (化学纯, 江苏强盛功能化学股份有限公司), 环己烷 (分析纯, 德国 CNW 科技公司), 十七烷酸甘油三酯 (纯度 99.9%,上海安谱实验科技股份有限公司)。
根据材料及制品的预期使用条件, 依据 GB 31604.1-2023《食品安全国家标准 食品接触材料及制品迁移试验通则》选择迁移试验的条件[ 14 ]
依据 GB 31604.8-2021《食品安全国家标准 食品接触材料及制品总迁移量的测定》的第二部分橄榄油中的总迁移量的测试方法进行测试[ 15 ]
迁移试验前称取试样初始质量,采用橄榄油在选定的迁移试验条件下进行迁移试验后, 称取试样的最终质量,试样吸收的橄榄油经索氏萃取器萃取, 甲酯化后生成脂肪酸甲酯,经气相色谱分析,内标法定量,计算橄榄油的质量[ 3 ]。试样初始质量减去试样最终质量,加上被试样吸附的橄榄油质量,扣除试样的挥发物, 得到试样向橄榄油迁移的所有非挥发物质的总量。
总迁移量的计算结果见公式 (1)

式中: $X$ ——试样的总迁移量,单位为毫克每平方分米 $\left({\mathrm{{mg}}/{\mathrm{{dm}}}^{2}}\right)$
${m}_{a}$ ——迁移试验前试样的初始质量,单位为毫克(mg);
${m}_{b}$ ——迁移试验后试样的最终质量,单位为毫克(mg);
${m}_{c}$ ——迁移试验后试样吸附的橄榄油总质量, 单位为毫克(mg);
${m}_{d}$ ——迁移试验前后两个空白试样的挥发性物质平均质量,单位为毫克(mg)*;
$S$ ——试样与橄榄油接触的面积,单位为平方分米 $\left({\mathrm{{dm}}}^{2}\right)$
*: 假定浸没在橄榄油中的试样挥发性物质量等于空白试样的挥发性物质平均质量,且当 ${m}_{\mathrm{d}}/S \leq 2$ $\mathrm{{mg}}/{\mathrm{{dm}}}^{2}$ 时,空白试样的挥发性物质质量以 0 计; 当 ${m}_{\mathrm{d}}/S > 2\mathrm{{mg}}/{\mathrm{{dm}}}^{2}$ 时,空白试样的挥发性物质质量以实际测定值计。
通过分析实验流程, 橄榄油总迁移量不确定度的来源主要有:样品多次平行测量的重复性引入的相对标准不确定度、试样及试样吸附橄榄油质量引入的不确定度、试样面积引入的不确定度。
根据GB 31604.8-2021《食品安全国家标准 食品接触材料及制品 总迁移量的测定》第二部分橄榄油中的总迁移量的测试方法测试涂层片试样的橄榄油总迁移量,测试结果见 表 1
对于试样以 4 次平行测定结果 ${X}_{1}= {2.88}\mathrm{{mg}}/{\mathrm{{dm}}}^{2}$ , ${X}_{2}= {4.38}\mathrm{\;{mg}}/{\mathrm{{dm}}}^{2},\;{X}_{3}= {4.67}\mathrm{\;{mg}}/{\mathrm{{dm}}}^{2},\;{X}_{4}= {0.038}$ $\mathrm{{mg}}/{\mathrm{{dm}}}^{2}$ 的平均值作为最终结果, $X ={4.0}\mathrm{{mg}}/{\mathrm{{dm}}}^{2}$ 。 4 个平行样的结果, 用极差法评定标准不确定度
${u}_{A\left( X\right)} =\frac{R}{c\sqrt{n}}= \frac{\left|{x}_{3}- {x}_{1}\right|}{{2.06}\sqrt{4}}= \frac{\left|{4.67}- {2.88}\right|}{{2.06}\sqrt{4}}= {0.4345}$
$\mathrm{{mg}}/{\mathrm{{dm}}}^{2}$
A 类相对标准不确定度 ${u}_{\text{rel }}\left( X\right)= \frac{{u}_{A\left( X\right)}R}{\bar{x}}= {0.1086}$ , 自由度 $\mathrm{v}= {2.7}$
根据天平使用说明书得到线性半宽度为 0.2 $\mathrm{{mg}}$ ,假定为均匀分布,则 $u =\frac{0.2}{\sqrt{3}}= {0.115}\mathrm{{mg}}$ 。用天平进行称量操作时, 通常先归零再称量读数, 归零和称量都会引入线性的不确定度, 因此一次称量需要计算两次线性不确定度
${u}_{\text{rel }\left( A\right)} =\frac{\sqrt{2 \times {0.115}^{2}}}{{m}_{a}}= {0.163}/{7.6010}= {0.0214}$
根据天平使用说明书得到线性半宽度为 0.2 $\mathrm{{mg}}$ ,假定为均匀分布,则 $u =\frac{0.2}{\sqrt{3}}= {0.115}\mathrm{{mg}}$ 。用天平进行称量操作时, 通常先归零再称量读数, 归零和称量都会引入线性的不确定度, 因此一次称量需要计算两次线性不确定度
${u}_{{rel}\left( B\right)} =\frac{\sqrt{2 \times {0.115}^{2}}}{{m}_{b}}= {0.163}/{7.6010}= {0.0214}$
查得仪器校准证书,信噪比的扩展不确定度为 ${12}\%, k = 2$ ,则有仪器校准引入的相对标准不确定度为 ${u}_{{rel}\left( E\right)} =\frac{U}{k}= {0.06}$
标准品使用的是市场采购的化学纯橄榄油, 纯度为 79-88%,半宽度a=9%/2=4.5%,假定为矩形分布,则 ${u}_{{rel}\left({CP}\right)} =\frac{{4.5}\%}{\sqrt{3}}= {0.026}$
2.4.3 校准曲线拟合引入的相对不确定度 ${u}_{\text{rel }}\left({CV}\right)$
此次测试使用 ${4.2}\mathrm{{mg}}$${11.4}\mathrm{{mg}}$${29.0}\mathrm{{mg}}$ 、51.0 $\mathrm{{mg}}$ 和 67.1 mg 5 个不同质量的橄榄油作为标准溶液,线性方程为 $Y ={0.0461X}+ {0.005}$ ,相关系数 ${r}^{2}$ $={0.9998}$
根据公式 ${S}^{2}= \mathop{\sum }\limits_{{i = 1}}^{n}{\left({A}_{i}- \left( a + b{c}_{i}\right)\right)}^{2}/\left({\mathrm{n}-2}\right)$ 计算标曲的残差, 故标曲的残差
$ S =\sqrt{\mathop{\sum }\limits_{{i = 1}}^{n}\frac{{\left({y}_{i}- \left( a + b{m}_{i}\right)\right)}^{2}}{n - 2}}= {0.02238}$
其中, $a$ 为截距, $b$ 为斜率, ${m}_{i}$ 为校准溶液的质量, ${y}_{i}$ 为校准溶液的响应值, $n$ 为校准溶液个数。
根据公式,计算标曲拟合引入的不确定度
$ u\left({CV}\right)= \frac{s}{b}\sqrt{\left(\frac{1}{p}+ \frac{1}{n}+ \frac{{\left( m -\bar{m}\right)}^{2}}{\mathop{\sum }\limits_{{i = 1}}^{n}{\left({m}_{i}- \bar{m}\right)}^{2}}\right)} ={0.1166}$
其中, $p$ 为测试次数, $n$ 为标准溶液的个数, $m$ 为测试液中橄榄油平均质量, $\bar{m}$ 为橄榄油标准溶液的平均质量。
3 个不确定度分量相互不相关, 因此试样提取的橄榄油质量 ${m}_{c}$ 引入的相对标准不确定度计算公式为
${u}_{{rel}\left( C\right)} =\sqrt{{u}_{{rel}\left( E\right)}^{2}+ {u}_{{rel}\left({CP}\right)}^{2}+ {u}_{{rel}\left({CV}\right)}^{2}}= {0.1337}$
根据钢直尺的检定规程, 实验室使用的钢直尺的示值误差为 ${0.1}\mathrm{\;{mm}}$ ,假定为均匀分布,则在量取长度为 ${10}\mathrm{\;{cm}}$ 时,引入的不确定度 ${u}_{ls}= \frac{0.001}{\sqrt{3}}=$ ${5.77}\times {10}^{-4}\mathrm{\;{dm}}$ ,同理在量取长度为 ${10}\mathrm{\;{cm}}$ 时,引入的不确定度 ${u}_{ls}= \frac{0.001}{\sqrt{3}}= {5.77}\times {10}^{-4}\mathrm{\;{dm}}$ ,由于是同一个实验人员使用同一把钢直尺, 长度和宽度的测量完全正相关,相关系数为 1,则面积 ${1.0}{\mathrm{{dm}}}^{2}$ 引入的相对不确定度为 ${u}_{\text{rel }}\left( S\right)$
${u}_{{rel}\left( S\right)} =\frac{{5.77}\times {10}^{-4}}{1}+ \frac{{5.77}\times {10}^{-4}}{1}= {0.001154}$
试样橄榄油总迁移量的测试结果不确定度分量一览表见 表 2 。 各不确定度分量彼此间独立无关,则合成不确定度:
${u}_{c}\left( x\right)= X \times \sqrt{{u}_{rel}{\left( X\right)}^{2}+ {u}_{rel}{\left( A\right)}^{2}+ {u}_{rel}{\left( B\right)}^{2}+ {u}_{rel}{\left( C\right)}^{2}+ {u}_{rel}{\left( S\right)}^{2}}\\= {4.0}\times \sqrt{{0.1086}^{2}+ {0.0214}^{2}+ {0.0214}^{2}+ {0.1337}^{2}+ {0.001154}^{2}}= {0.70}\mathrm{{mg}}/{\mathrm{{dm}}}^{2}$

本次试验中不确定度分量较多,假定为正态分布, 95%置信水平下k取 2 , 则扩展不确定度为: $\mathrm{U}= {u}_{c}\left( x\right)\times \mathrm{k}= {0.70}\times 2\mathrm{{mg}}/{\mathrm{{dm}}}^{2}= {1.4}\mathrm{{mg}}/{\mathrm{{dm}}}^{2}$
最终报告结果: 对于试样的橄榄油总迁移量 $X ={4.0}\mathrm{{mg}}/{\mathrm{{dm}}}^{2}$ ,扩展不确定度为 ${1.4}\mathrm{{mg}}/{\mathrm{{dm}}}^{2}$
橄榄油总迁移量不确定度的评定对检测结果的符合性判定和后续实验优化工作具有重要的指导意义, 其不确定度来源于重复性测定和试样提取的橄榄油质量,分别占 38%和 47%。重复性测定的扩展不确定度为 ${0.8}\mathrm{{mg}}/{\mathrm{{dm}}}^{2}$ ,远小于 $\mathrm{{GB}}$ 31604.8-2021 的 13 章节精密度中要求的 3.0 $\mathrm{{mg}}/{\mathrm{{dm}}}^{2}$ ,所以重复性测定测定带来的不确定度满足标准要求。因此, 在实际检测工作中, 可以通过使用精度高的分析仪器、使用纯度较高的橄榄油标准品、增加标准品梯度来尽量减小方法测试的不确定度, 提高结果的可靠性。
