Objective By means of High Performance Liquid Chromatography (HPLC), the extraction method and content determination method of $\alpha$ -cyperone were explored to achieve the purpose of accuracy, reliability, convenience and easy operation. Methods High performance liquid chromatography was used. The chromatography was performed on Agilent Eclipse Plus $\left({{150}\times {4.6}\mathrm{\;{mm}},5\mathrm{\;{\mu m}}}\right)$ silica gel column with mobile phase consisted of methanol-water (70:30) and UV-VIS spectrometer. The flow rate was ${1.0}\mathrm{{mL}}/\mathrm{{min}}$ . The detection wavelength was ${240}\mathrm{\;{nm}}$ . The column temperature was ${35}^{\circ }\mathrm{C}$ . Results The results showed a good linear relationship in the linear range of ${0.02314}- {0.46287}\mathrm{{mg}}/\mathrm{{mL}}$ with ${\mathrm{R}}^{2}= 1$ , the average recovery rate of ${99.84}\%$ and relative standard deviation (RSD)=0.09%. Conclusion The method has the advantages of simple operation, high sensitivity, good repeatability, accuracy and reliability, and can be used to determine the content of $\alpha$ -cyperone in order to control the quality of cyperone effectively.

目的 通过高效液相色谱法(High Performance Liquid Chromatography, HPLC)探索香附中 $\alpha$ - 香附酮的提取方式和含量的测定方法, 达到方法准确可靠简便易操作的目的。方法 采用高效液相色谱法。色谱柱为 Agilent Eclipse Plus,十八烷基硅烷键合硅胶色谱柱,150×4.6 mm,5μm,流动相为:甲醇一水(70:30),紫外 - 可见分光检测器;流速为 ${1.0}\mathrm{\;{mL}}/\mathrm{{min}}$ ;检测波长为 ${240}\mathrm{\;{nm}}$ ;柱温为 ${35}^{\circ }\mathrm{C}$结果 香附在 ${0.02314}- {0.46287}\mathrm{{mg}}/\mathrm{{mL}}$ 线性范围内呈良好线性关系, ${\mathrm{R}}^{2}= 1$ ,平均回收率 99.84%, RSD (Relative Standard Deviation) $={0.09}\%$结论 该方法操作简便, 灵敏度高,重复性好,准确、可靠,可用于测定香附 $\alpha$ - 香附酮的含量,并有效控制香附的质量。

Objective By means of High Performance Liquid Chromatography (HPLC), the extraction method and content determination method of $\alpha$ -cyperone were explored to achieve the purpose of accuracy, reliability, convenience and easy operation. Methods High performance liquid chromatography was used. The chromatography was performed on Agilent Eclipse Plus $\left({{150}\times {4.6}\mathrm{\;{mm}},5\mathrm{\;{\mu m}}}\right)$ silica gel column with mobile phase consisted of methanol-water (70:30) and UV-VIS spectrometer. The flow rate was ${1.0}\mathrm{{mL}}/\mathrm{{min}}$ . The detection wavelength was ${240}\mathrm{\;{nm}}$ . The column temperature was ${35}^{\circ }\mathrm{C}$ . Results The results showed a good linear relationship in the linear range of ${0.02314}- {0.46287}\mathrm{{mg}}/\mathrm{{mL}}$ with ${\mathrm{R}}^{2}= 1$ , the average recovery rate of ${99.84}\%$ and relative standard deviation (RSD)=0.09%. Conclusion The method has the advantages of simple operation, high sensitivity, good repeatability, accuracy and reliability, and can be used to determine the content of $\alpha$ -cyperone in order to control the quality of cyperone effectively.

香附(Cyperi Rhizoma)来源于莎草科植物莎草Cyperus rotundus L. 的干燥根茎 [ 1 ] ,具有疏肝解郁,理气宽中,调经止痛的功效 [ 1 ] 。香附,原名 “莎草”,始载于《名医别录》,列为中品 [ 2 ] 。 《唐本草》始称香附子 [ 3 ] 。《本草纲目》列入草部芳草类,名 “莎草香附子” [ 4 ] ,2020 年版《中国药典》收载的香附,【含量测定】 项为挥发油,不得少于 ${1.0}\%\left({\mathrm{{mL}}/\mathrm{g}}\right)$ ,其中主要成分为 $\alpha$ - 香附酮, 本文采用 HPLC 法测定香附中 $\alpha$ - 香附酮的含量。方法简便、快速、 准确, 专属性强, 可用于香附的质量控制。
高效液相色谱仪 (1260, 安捷伦), 包括紫外 - 可见分光检测器; 安捷伦十八烷基硅烷键合硅胶(150×4.6 mm,5μm)色谱柱;XS205DU 十万分之一电子天平 (梅特勒 - 托利多科技 (中国)有限公司);SB-500DTY 扫频超声波清洗机(宁流新芝生物科技有限公司); UPR-11-10T 优谱超纯水器 (四川优谱超纯科技有限公司)。
对照品 α- 香附酮(批号:110748-202117,特性量值:,购于中国食品药品检定研究院;甲醇为分析纯、色谱纯(成都市科隆化学品有限公司);超纯水自制。香附(广西南药药业有限公司,经绵阳市食品药品检验所魏云主任中药师鉴定为莎草科植物莎草 Cyperus rotundus L. 的干燥根茎)。
采用安捷伦十八烷基硅烷键合硅胶 $\left({{150}\times {4.6}\mathrm{\;{mm}},{5\mu }\mathrm{m}}\right)$ 色谱柱,流动相为甲醇: 水(70:30),柱温为 ${35}^{\circ }\mathrm{C}$ ;紫外 - 可见分光检测器; 检测器温度 ${40}^{\circ }\mathrm{C}$ 。理论板数按 $\alpha$ - 香附酮计算应不低于 4000。检测波长 ${240}{\mathrm{\;{nm}}}^{\left\lbrack 5\right\rbrack }$ ,进样量 ${10\mu }\mathrm{L}$
精密称取 $\alpha$ - 香附酮对照品适量,加甲醇溶解,制成每 $1\mathrm{\;{mL}}$${0.3}\mathrm{{mg}}$ 的对照品溶液,即得。
精密称取香附粉末 $1\mathrm{\;g}$ ,置 ${50}\mathrm{\;{mL}}$ 容量瓶中,加甲醇适量, 超声处理 ${30}\mathrm{\;{min}}$ ,定容至刻度,摇匀,过滤,取续滤液,作为供试品溶液 [ 6 ]
按 1.4.2 项下方法 (不加香附样品, 其他处理方法均一致), 制备阴性供试品溶液。
分别取 1.4.1、1.4.2、1.4.3 项下的对照品溶液、供试品溶液、 阴性样品溶液, 按 1.3 项下的色谱条件进行测定, 进样量为 ${10\mu }\mathrm{L}$ ,测定结果阴性样品溶液对测定成分无干扰,专属性良好 (见图 1-3)。
精密吸取 1.4.1 项下对照品溶液 $1\mathrm{\;{mL}}$ 分别置 $2\text{、}5\text{、}{10}\text{、}{20}\mathrm{\;{mL}}$ 的量瓶中, 加甲醇至刻度, 摇匀, 制得不同质量浓度的对照品溶液, 按 1.3 项下的色谱条件依次进 5 个浓度对照品溶液, ${20\mu }\mathrm{L}$ ,记录色谱图峰面积,以峰面积为纵坐标,以对照品浓度为横坐标,绘制标准曲线,结果显示 $\alpha$ - 香附酮的线性回归方程 $\mathrm{Y}= {55826}\mathrm{X}+ {22.341}$ ,线性范围 0.02314-0.46287 mg/mL,相关系数 ${\mathrm{R}}^{2}= 1,\alpha$ - 香附酮在浓度范围内与其峰面积呈现良好的线性关系。标准曲线 (见 图 4 )。
精密吸取 α- 香附酮对照品溶液 (浓度为 0.4638 mg/mL×99.8%) ${20\mu }\mathrm{L}$ ,按 1.3 项下的色谱条件连续进样 6 次,记录峰面积,其 RSD=0.2%, 表明仪器的精密度良好。
取同一香附样品, 按 1.4.2 项下方法平行制备 10 份供试品溶液,按 1.3 项下的色谱条件依次检测,分别测定并计算含量, $\alpha$ - 香附酮平均含量为 ${0.702}\mathrm{{mg}}/\mathrm{g}$ ,其 RSD 为 ${0.2}\%$ ,表明该方法重复性良好。
取同一供试品溶液,分别在 $0\text{、}1\text{、}2\text{、}4\text{、}6\text{、}8\text{、}{12}$ 小时进行测定,记录峰面积并计算 $\mathrm{{RSD}}$ ,结果 $\alpha$ - 香附酮峰面积的 RSD 为 0.3%,表明供试品溶液在制备后 12 小时内稳定。
精密称取已知含量的供试品 9 份,分别精密加入一定量的 $\alpha$ - 香附酮对照品, 按 1.4.2 中制备供试品溶液的方法进行处理, 测定并计算,结果见 表 1
取 10 批待测香附样品, 按 1.4.2 中制备溶液的方法进行处理并测定, 分别计算含量, 验证结果如下。结果见 表 2
香附中 $\alpha$ - 香附酮属于倍半萜类 [ 7 ] ,易溶于醇及有机溶剂, 难溶于水。 $\alpha$ - 香附酮化学结构式中含有双键的烃基、酯基两大官能团 [ 8 ] ,两者皆属于疏水基团,是非极性官能团,而水是极性 [ 9 ] ,根据相似相溶原理, $\alpha$ - 香附酮难溶于水,易溶于有机溶剂。
实验分别采用了《中国药典》方法加乙醇超声提取 [ 10 ] 、加乙醚回流 [ 11 ] 、加水超声提取 [ 12 ] 三种方法,结果表明加乙醇超声提取样品,色谱峰无杂峰,含 $\alpha$ - 香附酮的量低,方法较简单, 理论塔板数达到要求, 分离效果好; 加乙醚回流, 色谱峰周围有杂峰干扰,含 $\alpha$ - 香附酮的量较高,方法复杂,理论塔板数达到要求,分离效果好; 加水超声提取样品,峰周围无杂峰,含 $\alpha$ - 香附酮的量最高,方法简单,理论塔板数达到要求,分离效果最好,故选择加水超声提取的方法。
通过查阅参考文献 [ 5 - 15 ] $\alpha$ - 香附酮含量测定项,对色谱柱、 流动相、柱温等和检测器温条件进行以下实验:实验中选取氨基键合硅胶柱 $\left({{250}\times {4.6}\mathrm{\;{mm}}}\right)$ 色谱柱和十八烷基硅烷键合硅胶色谱柱, ${250}\times {4.6}\mathrm{\;{mm}},{5\mu }\mathrm{m}$ 进行对比分析,后者分析出的色谱图理论塔板数、分离度、拖尾因子等均优于前者, 故选用十八烷基硅烷键合硅胶色谱柱,250×4.6 mm,5μm。
实验用分别采用了甲醇 : 水( 65:35 ) [ 13 ] 、甲醇 : 水( 45:55 )、 甲醇:水(70:30) [ 14 ] 三种流动相,采用甲醇:水(65:35)流动相,色谱图峰宽较大影响拖尾因子,使其达不到药典标准;甲醇: 水 (45:55) 流动相,色谱图未出峰;采用甲醇:水(70:30)流动相,操作简单,检验时间短,分离度满足需求,杂峰及溶剂峰少,故选择甲醇:水(70:30)流动相。
实验中对柱温和检测器温度条件进行了考察, 分别设置柱温 40、35、30°C [ 15 ] ,检测器温度40、35、 ${30}^{\circ }\mathrm{C}$ ,结果表明当柱温为 ${40}^{\circ }\mathrm{C}$ 、检测器温度 ${40}^{\circ }\mathrm{C}$ 时色谱峰的分离度最大。
通过 HPLC 方法成功摸索出 $\alpha$ - 香附酮的提取方式和含量的测定方法, 具有操作简单快捷、灵敏度高、重复性好、准确可靠、 所用试剂种类少、分离效果好等特点,可用于香附 $\alpha$ - 香附酮的测定, 并有效控制质量。
