Objective To establish a method for determining thecontents of paeoniflorin, cinnamaldehyde and paeonol in Guizhi JiliuPills. Methods The RP-HPLC wavelength switching method was used, theanalysis was achieved on an ES Epic ${\mathrm{C}}_{18}\left({{4.6}\mathrm{\;{mm}}\times {250}\mathrm{\;{mm}},{5\mu}\mathrm{m}}\right)$ column, the mobile phase was acetonitrile-0.1% aqueous solution of phosphoric acid gradient elution, the flowrate was ${1.0}\mathrm{\;{mL}}/\mathrm{{min}}$ , thecolumn temperature was ${25}^{\circ }\mathrm{C}$ and thedetection wavelength were ${230}\mathrm{\;{nm}}$ $\left({0\sim {25}\mathrm{\;{min}}}\right),{290}\mathrm{\;{nm}}\left({{25}\sim {41}\mathrm{\;{min}}}\right)$ and ${274}\mathrm{\;{nm}}\left({{41}\sim {51}\mathrm{\;{min}}}\right)$ . Results The linear ranges of paeoniflorin, cinnamaldehyde and paeonolwere 3.6169~90.4232 μg/mL ( $r={0.9993}$ ),1.8388~45.9695 μg/mL ( $r={0.9999}$ ),2.0240~50.5994 μg/mL ( $r={0.9999}$ ). The average sample recovery rates were95.4%(RSD=1.2%), 93.0%(RSD=0.7%) and 94.2%(RSD=1.2%) respectively. Conclusion This method is accurate, sensitive, and has strong specificity. It can be used for the quality control of Guizhi Jiliu Pills.

目的 建立同时测定桂枝肌瘤丸中芍药苷、桂皮醛及丹皮酚 3种成分含量的方法。方法 采用反相高效液相色谱(Reversed Phase HighPerformance Liquid Chromatography, RP-HPLC)波长切换法,色谱柱ES Epic ${\mathrm{C}}_{18}$ ( ${4.6}\mathrm{\;{mm}}\times {250}\mathrm{\;{mm}},{5\mu}\mathrm{m}$ ),流动相为乙腈 $-{0.1}\%$ 磷酸水溶液梯度洗脱,流速 ${1.0}\mathrm{\;{mL}}/\mathrm{{min}}$ ,柱温 ${25}^{\circ }\mathrm{C}$ ,检测波长为 ${230}\mathrm{\;{nm}}\left({0\sim {25}\mathrm{\;{min}}}\right)$${290}\mathrm{\;{nm}}\left({{25}\sim {41}\mathrm{\;{min}}}\right)$${274}\mathrm{\;{nm}}\left({{41}\sim {51}\mathrm{\;{min}}}\right)$结果 芍药苷、桂皮醛、丹皮酚线性范围分别为 ${3.6169}\sim {90.4232\mu}\mathrm{g}/\mathrm{{mL}}\left({r ={0.9993}}\right)$${1.8388}\sim {45.9695\mu}\mathrm{g}/\mathrm{{mL}}\left({r ={0.9999}}\right)$${2.0240}\sim {50.5994\mu}\mathrm{g}/\mathrm{{mL}}\left({r={0.9999}}\right)$ 。平均加样回收率分别为95.4%(RSD=1.2%)、93.0%(RSD=0.7%)和 94.2%(RSD=1.2%)。结论 本方法准确、灵敏, 专属性强,适用于桂枝肌瘤丸的质量控制。

Objective To establish a method for determining thecontents of paeoniflorin, cinnamaldehyde and paeonol in Guizhi JiliuPills. Methods The RP-HPLC wavelength switching method was used, theanalysis was achieved on an ES Epic ${\mathrm{C}}_{18}\left({{4.6}\mathrm{\;{mm}}\times {250}\mathrm{\;{mm}},{5\mu}\mathrm{m}}\right)$ column, the mobile phase was acetonitrile-0.1% aqueous solution of phosphoric acid gradient elution, the flowrate was ${1.0}\mathrm{\;{mL}}/\mathrm{{min}}$ , thecolumn temperature was ${25}^{\circ }\mathrm{C}$ and thedetection wavelength were ${230}\mathrm{\;{nm}}$ $\left({0\sim {25}\mathrm{\;{min}}}\right),{290}\mathrm{\;{nm}}\left({{25}\sim {41}\mathrm{\;{min}}}\right)$ and ${274}\mathrm{\;{nm}}\left({{41}\sim {51}\mathrm{\;{min}}}\right)$ . Results The linear ranges of paeoniflorin, cinnamaldehyde and paeonolwere 3.6169~90.4232 μg/mL ( $r={0.9993}$ ),1.8388~45.9695 μg/mL ( $r={0.9999}$ ),2.0240~50.5994 μg/mL ( $r={0.9999}$ ). The average sample recovery rates were95.4%(RSD=1.2%), 93.0%(RSD=0.7%) and 94.2%(RSD=1.2%) respectively. Conclusion This method is accurate, sensitive, and has strong specificity. It can be used for the quality control of Guizhi Jiliu Pills.

桂枝肌瘤丸是河北省玉田县中医医院制剂, 由桂枝、桃仁、 赤芍、牡丹皮、丹参、茯苓 6 味药材组成,具有活血化瘀,消散癥块的作用。临床主要用于瘀阻胞宫所致漏下不止、血色紫黑灰暗、腹痛拒按;子宫肌瘤、痛经、盆腔炎等症。现行医疗机构制剂质量标准 [ 1 ] 仅有鉴别及检查项,缺少含量测定项,且目前无桂枝肌瘤丸质量标准提升相关文献报道。高效液相色谱仪是目前药品分析实验室常用检测仪器, 利用高压输液泵将流动相泵入色谱柱, 对复杂供试品进行分离分析。目前在中药制剂质量标准提升相关文献中[2-6] ,建立的含量测定方法多为一种高效液相色谱法同时测定多种成分。本文对桂枝肌瘤丸中主要药效成分芍药苷、桂皮醛、丹皮酚展开研究,建立了同时测定三种成分含量的高效液相色谱法, 此方法操作快速、准确, 专属性强, 可用于桂枝肌瘤丸的定量检查, 为医院制剂室及药品监管部门全面控制制剂质量, 保证医院制剂的质量达到临床用药安全、有效提供有利的科学依据。
LC-20A 型高效液相色谱仪 (日本岛津); 8453 紫外 - 可见分光光度计(美国安捷伦);BSA124S-CW 电子天平、BP211D 电子天平(德国赛多利斯);KQ3200E 超声波清洗仪(昆山市超声仪器有限公司)。
芍药苷对照品 (含量: 94.6%,批号: 110736-202145) 桂皮醛对照品(含量:99.5%,批号:110710- 2020 22)、丹皮酚对照品 (含量:,均购自中国食品药品检定研究院;桂枝肌瘤丸样品(规格:每丸重; 甲醇 (Fisher Chemical)、乙腈 (Sigma-Aldrich), 均为色谱纯, 磷酸 (天津市恒兴化学试剂) 为分析纯, 水为娃哈哈纯净水。
取桂枝肌瘤丸供试品适量,剪碎,混匀,取约 ${0.66}\sim {0.8}\mathrm{g}$ , 精密称定,置具塞锥形瓶中,精密加入甲醇 ${25}\mathrm{\;{mL}}$ ,密塞,称定重量,超声处理 (功率 ${250}\mathrm{w}$ ,频率 ${50}\mathrm{{kHz}}$ ) ${30}\mathrm{\;{min}}$ ,放冷, 再称定重量, 用甲醇补足减失的重量, 摇匀, 滤过, 再用微孔滤膜 ( ${0.45\mu }\mathrm{m}$ ) 滤过,作为供试品溶液。
精密称取芍药苷对照品约 ${18}\mathrm{{mg}}$ ,桂皮醛对照品约 ${18}\mathrm{{mg}}$ , 丹皮酚对照品约 ${10}\mathrm{{mg}}$ ,分别置于 ${50}\text{、}{100}\text{、}{50}\mathrm{\;{mL}}$ 容量瓶中, 用甲醇溶解并稀释至刻度, 摇匀, 作为对照品储备液; 再精密量取三种对照品储备液各 $2\mathrm{\;{mL}}$ ,置于同一 ${10}\mathrm{\;{mL}}$ 容量瓶中,用甲醇稀释至刻度,摇匀,用微孔滤膜 $\left({{0.45\mu }\mathrm{m}}\right)$ 滤过,作为混合对照品溶液。
因桂皮醛为桂枝主要成分, 芍药苷、丹皮酚为赤芍、牡丹皮共有成分 [ 7 - 9 ] ,故按处方比例制成缺桂枝阴性样品,缺赤芍、牡丹皮阴性样品,其余操作同 1.3.1,作为阴性对照溶液。
色谱柱: ES Epic ${\mathrm{C}}_{18}$ ( ${4.6}\mathrm{\;{mm}}\times {250}\mathrm{\;{mm}},{5\mu }\mathrm{m}$ ),以乙腈 (A)-0.1% 磷酸水溶液 (B) 梯度洗脱, 0~19 min,15%A; 19~23 min,45%A;23~40 min,35%A;40~51 min,15%A,流 速为 ${1.0}\mathrm{\;{mL}}/\mathrm{{min}}$ ,柱温为 ${25}^{\circ }\mathrm{C}$ ,检测波长为 ${230}\mathrm{\;{nm}}\left({0 \sim {25}\mathrm{\;{min}}}\right)$${290}\mathrm{\;{nm}}\left({{25}\sim {41}\mathrm{\;{min}}}\right)\text{、}{274}\mathrm{\;{nm}}\left({{41}\sim {51}\mathrm{\;{min}}}\right)$ ,进 样 量 为 ${20\mu }\mathrm{L}$ ,按外标法以峰面积分别计算芍药苷、桂皮醛及丹皮酚三种成分含量。
取 1.3 项下的溶液,照 1.4 项下方法进行测定,结果表明, 供试品溶液中芍药苷、桂皮醛、丹皮酚保留时间与混合对照品溶液一致,阴性对照溶液对各成分测定无干扰,见 图 1
取 1.3.2 项下的三种对照品储备液,精密量取各 ${0.1}\mathrm{\;{mL}}$ ,置于同一 ${10}\mathrm{\;{mL}}$ 容量瓶中,用甲醇稀释至刻度,摇匀,备用。然后精密量各取 ${0.5}\text{、}1\text{、}{1.5}\text{、}2\text{、}{2.5}\mathrm{\;{mL}}$ ,同法操作。取制备好的各溶液,经 ${0.45\mu }\mathrm{m}$ 滤膜过滤,分别取 ${20\mu }\mathrm{L}$ 注入高效液相色谱仪中, 依法测定, 以对照品进样浓度 (C) 为横坐标, 峰面积值 (A) 为纵坐标, 绘制标准曲线, 得芍药苷的线性回归方程为 $A ={2.7848}\times {10}^{4}\mathrm{C}+ {1.2058}\times {10}^{4}, r ={0.9993}$ ; 桂皮醛的线性回归方程为 $A ={1.99715}\times {10}^{5}\mathrm{C}+ {1.1646}\times {10}^{4}, r ={0.9999}$ ; 丹皮酚的线性回归方程为 $A ={1.05861}\times {10}^{5}\mathrm{C}+ {8.0561}\times {10}^{3}, r ={0.9999}$结果 表明,芍药苷、桂皮醛、丹皮酚分别在3.6169~90.4232μg/mL, 1.8388~45.9695 μg/mL,2.0240~50.5994 μg/mL范围内,与峰面积有良好的线性关系。
取 1.3.2 项下的混合对照品溶液, 依法重复测定 6 次, 芍药苷峰面积 RSD 为 0.3%, 桂皮醛峰面积 RSD 为 0.8%, 丹皮酚峰面积 RSD 为 0.5%,精密度良好。
取桂枝肌瘤丸供试品约 0.4g(批号: 2022 0302,已知芍药苷、 共 6 份, 分别置于具塞锥形瓶中, 精密加入芍药苷对照品溶液 (0.4454 mg/mL) 2.0 mL、桂皮醛对照品溶液 (0.0993 mg/mL) 1.0 mL、 丹皮酚对照品溶液 ,挥干溶剂,照 1.3.1 项下方法制备溶液, 并依 1.4 项下方法测定, 计算回收率, 结果见表 1
精密吸取 1.3.1 项下桂枝肌瘤丸供试品溶液 ${20\mu }\mathrm{L}$ ,分别于 0、 $4\text{、}8\text{、}{12}\text{、}{24}\mathrm{\;h}$ ,注入高效液相色谱仪测定峰面积,结果表明, 芍药苷RSD为 1.0%, 桂皮醛 RSD 为 1.2%, 丹皮酚 RSD 为 0.7%, 供试品在 ${24}\mathrm{\;h}$ 内稳定性良好。
取桂枝肌瘤丸(批号: 2022 0302)共 6 份,约 0.8 g,精密称定, 照 1.3.1、1.4 项下方法制备及测定, 计算供试品中三种成分含量。 芍药苷平均含量为 , RSD为 ; 桂皮醛平均含量为 , RSD为 ;丹皮酚平均含量为 , RSD 为 0.9%。该方法重复性良好。
取桂枝肌瘤丸 3 批,照 1.3.1 项下方法制备溶液,照 1.4 项下方法测定三种成分含量, 结果见 表 2
本文通过查阅《中国药典》 [ 10 ] 桂枝、赤芍、牡丹皮、桂枝茯苓丸检测方法及其他参考文献 [ 11 - 15 ] ,试验了甲醇 - 水及乙腈 -0.1% 磷酸水溶液系统,三种待测成分在乙腈 -0.1% 磷酸水溶液系统峰形良好, 且柱压适中, 另通过梯度洗脱调整乙腈与 0.1% 磷酸水溶液比例, 使三个主成分保留时间适中, 且与相邻杂质分离度均大于 1.5 。
用流动相初始比例配制三种对照品溶液, 分别置紫外 - 可见分光光度计中,在 ${190}\sim {600}\mathrm{\;{nm}}$ 波长范围内扫描,芍药苷、桂皮醛、丹皮酚分别在 230、290、274 nm 的波长处有最大吸收, 本文选用波长切换方法使三种成分均在最大吸收波长处检测, 达到较高灵敏度。
通过试验室温、 ${25}^{\circ }\mathrm{C}$${30}^{\circ }\mathrm{C}$ 三种柱温,发现在 ${25}^{\circ }\mathrm{C}$ 条件下, 色谱峰拖尾情况得到改善,且分离度更佳。
本文考察了甲醇超声处理 (功率 ${250}\mathrm{\;W}$ ,频率 ${50}\mathrm{{kHz}}$ ) 供试品 15、30、45 min 三种条件,结合三种成分特性及提取效率, 确定 ${30}\mathrm{\;{min}}$ 为最佳提取条件。
本方法实现复杂样品中多种待检成分同时检测, 灵敏度高, 减少了操作步骤及试剂的消耗, 节省时间, 节约检测成本, 更加具有实用性。为基层医疗机构制剂室、基层药品监管部门全面控制制剂质量提供了科学依据, 并为其他医院制剂含量测定方法建立及质量标准提升工作提供参考。
