Objective Establish a ultra high performance liquidchromatography (UPLC) method for the simultaneous determination of sevenginsenosides Rg1, Re, Rf, Rb1, Rc, Rb2, and Rd in ginseng medicinalmaterials. Methods Using CAPCELL PAK ${\mathrm{C}}_{18}\mathrm{{MG}}\left({{2.0}\mathrm{\;{mm}}\times {150}\mathrm{\;{mm}},{2.0\mu}\mathrm{m}}\right)$ Chromatographic column with gradientelution of acetonitrile-0.05% phosphoric acid solution as mobile phase,flow rate of ${0.38}\mathrm{\;{mL}}\cdot {\mathrm{{min}}}^{-1}$ ,detection wavelength of ${203}\mathrm{\;{nm}}$ , column temperatureof ${30}^{\circ }\mathrm{C}$ , andinjection volume of ${1\mu}\mathrm{L}$ . Results All 7 ginsenosides can be completelyseparated and have a good linear relationship $\left({{r}^{2}> {0.999}}\right)$ withina certain linear concentration range, with an average recovery rate of ${97.22}\%\sim {104.56}\%$ ; Therelative standard deviation (RSD) is ${0.4}\%\sim {1.8}\%\left({n = 9}\right)$ ,and the precision RSD is ${0.57}\%\sim {1.42}\%$ . Conclusion Thismethod has strong specificity, excellent precision, good accuracy, andmore efficient detection process, and can be used for the determinationof seven ginsenosides in ginseng medicinal materials.

目的 建立同时测定人参药材中 7 种人参皂苷Rg1、Re、Rf、Rb1、Rc、Rb2、Rd 含量的超高效液相色谱法(Ultra PerformanceLiquid Chromatography, UPLC) 方法。方法 采用 CAPCELL PAK ${\mathrm{C}}_{18}\mathrm{{MG}}\left({{2.0}\mathrm{\;{mm}}\times {150}\mathrm{\;{mm}},{2.0\mu}\mathrm{m}}\right)$ 色谱柱,流动相为乙腈 $-{0.05}\%$ 磷酸溶液体系梯度洗脱,流速为 ${0.38}\mathrm{\;{mL}}\cdot {\mathrm{{min}}}^{-1}$ ,检测波长均为 ${203}\mathrm{\;{nm}}$ ,柱温均为 ${30}^{\circ }\mathrm{C}$ ,进样量 ${1\mu }\mathrm{L}$结果 7种人参皂苷均能完全分离,在一定的线性浓度范围内具有良好线性关系 ( ${r}^{2}> {0.999}$ ), 平均回收率为97.22%~104.56%;相对标准偏差(RSD)为 0.4%~1.8%( $n = 9$ ),精密度 RSD 为 0.57%~1.42%。 结论 该方法的专属性强,精密度极佳,准确度好,检测过程更加高效,可用于人参药材中 7种人参皂苷含量的测定。关键词:中药制剂;人参;人参皂苷;超高效液相色谱法

Objective Establish a ultra high performance liquidchromatography (UPLC) method for the simultaneous determination of sevenginsenosides Rg1, Re, Rf, Rb1, Rc, Rb2, and Rd in ginseng medicinalmaterials. Methods Using CAPCELL PAK ${\mathrm{C}}_{18}\mathrm{{MG}}\left({{2.0}\mathrm{\;{mm}}\times {150}\mathrm{\;{mm}},{2.0\mu}\mathrm{m}}\right)$ Chromatographic column with gradientelution of acetonitrile-0.05% phosphoric acid solution as mobile phase,flow rate of ${0.38}\mathrm{\;{mL}}\cdot {\mathrm{{min}}}^{-1}$ ,detection wavelength of ${203}\mathrm{\;{nm}}$ , column temperatureof ${30}^{\circ }\mathrm{C}$ , andinjection volume of ${1\mu}\mathrm{L}$ . Results All 7 ginsenosides can be completelyseparated and have a good linear relationship $\left({{r}^{2}> {0.999}}\right)$ withina certain linear concentration range, with an average recovery rate of ${97.22}\%\sim {104.56}\%$ ; Therelative standard deviation (RSD) is ${0.4}\%\sim {1.8}\%\left({n = 9}\right)$ ,and the precision RSD is ${0.57}\%\sim {1.42}\%$ . Conclusion Thismethod has strong specificity, excellent precision, good accuracy, andmore efficient detection process, and can be used for the determinationof seven ginsenosides in ginseng medicinal materials.

随着国家对中医药研发的重视和全民中医药热的兴起, 越来越多的药物研发企业加大了对中药新药的研发投入。近年来, 随着分析方法的不断优化和仪器的迭代变更,《中国药典》收录的各类中药材分析方法中,已有部分方法不能够适应目前快节奏、低成本高效益的工业化生产。
《中国药典》一部收录的三宝胶囊、小儿肺咳颗粒等中药制剂中均涉及人参药材、饮片的含量检测。人参中主要含有皂苷、 多糖、挥发油等化学成分 [ 1 ] 。目前多以人参皂苷作为含量的指标性成分进行质量研究, 其中人参皂苷 Rg1、Rb1 被证明具有改善认知功能障碍的作用 [ 2 - 3 ] ; 曹瑀莹等人 [ 4 ] 通过大鼠模型试验确定人参皂苷 Re 能够调节蛋白激酶 A、心肌细胞内环磷酸腺苷的含量,达到干预快速型心律失常的作用。人参中大量的人参皂苷已被鉴定,人参皂苷 $\mathrm{{Rb}}1\text{、}\mathrm{{Rb}}2\text{、}\mathrm{{Rc}}\text{、}\mathrm{{Rd}}\text{、}\mathrm{{Re}}$ 以及 $\mathrm{{Rg}}1$ 的量已成为人参产品的参考标准 [ 5 ]
关于用 HPLC 的方法测定人参中人参皂苷的含量的文献报道有很多 [ 6 - 8 ] ,但采用 UPLC 方法的报道却很少,且多以《中国药典》方法梯度进行检测 [ 9 ] ,单针进样大于 100 分钟耗费时间过长, 不利于快速测定药物中此药材的质量。本研究采用 UPLC 方法同时测定 7 种人参皂苷 Rg1、Re、Rf、Rb1、Rc、 $\mathrm{{Rb}}2\text{、}\mathrm{{Rd}}$ 的含量,单针运行时间仅为 33 分钟且兼具了良好的分离度, 比常用的药典方法单针运行 100 余分钟更为高效, 为快速准确控制人参药材质量提供了参考。
Waters H-class plus 型超高效液相色谱仪 (UPLC), 梅特勒 XSE205DU 电子天平。
人参药材(治佳凯药业股份有限公司,批号:1:220901-01;2:230722-01;3:231106-01)。
乙腈 (色谱级, TEDIA), 甲醇 (色谱级, Sigma-Aldrich), 正丁醇 (色谱级, RMC),氨试液 (分析纯,西陇科学股份有限公司),超纯水 (自制)。
人参皂苷 Rg1、Re、Rf、Rb1、Rb2、Rd 对照品(中国食品药品检定研究院,批号:110703-202235、110754-202129、201806、110704-202230、111715-202104、111818-202104)、人参皂苷 Rc 对照品 (自制工作对照品, 批号: DZP-202205-01)。
采用 CAPCELL PAK ${\mathrm{C}}_{18}\mathrm{{MG}}\left({{2.0}\mathrm{\;{mm}}\times {150}\mathrm{\;{mm}},{2.0\mu }\mathrm{m}}\right)$ 色谱柱,以乙腈 $-{0.05}\%$ 磷酸溶液为流动相进行梯度洗脱( $0 \sim {10.5}\mathrm{\;{min}}$ , 19% 乙腈;10.5~16.5 min,19%~29% 乙腈;16.5~20.8 min,29% 乙腈;20.8~29 min,29%~40% 乙腈;29~30 min,40%~19% 乙腈; ${30}\sim {33}\mathrm{\;{min}},{19}\%$ 乙腈),流速 ${0.38}\mathrm{\;{mL}}/\mathrm{{min}}$ ,柱温 ${30}^{\circ }\mathrm{C}$ ,检测波长 ${203}\mathrm{\;{nm}}$ ,进样量 ${1\mu }\mathrm{L}$
取本品粉末约 $1\mathrm{\;g}$ ,精密称定,置具塞锥形瓶中,精密加入甲醇 ${50}\mathrm{\;{mL}}$ ,密塞,超声 $1\mathrm{\;h}$ ,放冷,摇匀,滤过,用 ${30}\mathrm{\;{mL}}$ 甲醇分两次洗涤滤渣,滤液置蒸发皿中水浴蒸干,加 ${20}\mathrm{\;{mL}}$ 水饱和正丁醇 (1:1 饱和 12 h) 溶解,用氨试液萃取 2 次,每次 ${20}\mathrm{\;{mL}}$ ,弃去氨试液,正丁醇液水浴蒸干,残渣加甲醇溶解, 定容至 ${10}\mathrm{\;{mL}}$ 量瓶中,取续滤液,即得。
分别取人参皂苷 Rg1、Re、Rf、Rb1、Rc、Rb2、Rd 对照品适量,精密称定,加甲醇制成每 $1\mathrm{\;{mL}}$ 约含 ${0.2}\mathrm{{mg}}$ 人参皂苷 $\mathrm{{Rg}}1\text{、}{0.2}\mathrm{{mg}}$ 人参皂苷 $\mathrm{{Re}}\text{、}{0.1}\mathrm{{mg}}$ 人参皂苷 $\mathrm{{Rf}}\text{、}{0.2}\mathrm{{mg}}$ 人参皂苷 Rb1、0.2 mg 人参皂苷 Rc、0.2 mg 人参皂苷 Rb2、0.15 mg 人参皂苷 Rd 的混合溶液,摇匀,即得。
分别取空白溶液、对照品溶液、供试品溶液各 ${1\mu }\mathrm{L}$ ,按色谱条件测定, 结果见 图 1 , 各色谱峰出峰顺序依次为人参皂苷 $\mathrm{{Rg}}1\text{、}\mathrm{{Re}}\text{、}\mathrm{{Rf}}\text{、}\mathrm{{Rb}}1\text{、}\mathrm{{Rc}}\text{、}\mathrm{{Rb}}2\text{、}\mathrm{{Rd}}$ ,从色谱图可以看出,空白溶剂对测定无干扰,人参皂苷色谱峰与临近峰分离度均大于 1.5 , 表明其他组分对各目标峰的出峰均无干扰, 分离效果较好。
按照对照品溶液的制备方法, 依次取 50%、100%、150%、 200%、300% 六个浓度作为线性溶液,按 “1.3 色谱条件” 测定, 以各峰面积 - 浓度作图, 考察各组分的线性关系。结果表 1 , 人参皂苷 Rg1、Re、Rf、Rb1、Rc、Rb2、Rd 在一定浓度范围内具有良好的线性,其相关系数 ${r}^{2}$ 均大于 0.999 。
精密称取 6 份人参药材样品各约 $1\mathrm{\;g}$ ,置具塞锥形瓶中,按 “1.4.1 供试品溶液的制备”进行试验, 按 “1.3 色谱条件” 检测。 采用外标法分别计算人参皂苷 Rg1、Re、Rf、Rb1、Rc、Rb2、 $\mathrm{{Rd}}$ 的重复性结果,7 种人参皂苷各组重复性 $\mathrm{{RSD}}$ 结果依次为 1.39%、1.14%、1.37%、0.57%、0.97%、1.42%、1.19%,每组 6 份供试品溶液中各人参皂苷含量的 RSD 均不大于 1.5%,符合 《中国药典》相关要求, 表明重复性良好。
精密称取人参药材样品约 $1\mathrm{\;g}$ ,共9份,置具塞锥形瓶中, 加入一定量对照品溶液, 按 “1.4.1 供试品溶液的制备” 进行试验, 按 “1.3 色谱条件” 检测。采用外标法分别计算人参皂苷 $\mathrm{{Rg}}1$$\mathrm{{Re}}$$\mathrm{{Rf}}$$\mathrm{{Rb}}1$$\mathrm{{Rc}}$$\mathrm{{Rb}}2$$\mathrm{{Rd}}$ 的加样回收率,结果依次为 101.97%、99.34%、101.02%、102.45%、99.74%、100.65%、 100.22%,平均回收率范围为 97.34%~102.45%, RSD 范围为 0.4%~1.8%,表明方法有较高的准确度。
取 3 批样品, 按 “1.3 色谱条件” 进行检测, 7 种人参皂苷含量结果见 表 2
文中采用的 UPLC 方法, 精密度结果极佳, 准确度好, 从色谱图上分析,其专属性强,检测波长在 ${203}\mathrm{\;{nm}}$ 处各目标峰均有良好响应, 图谱可见流动相基线稳定、无明显漂移和杂峰干扰, 符合《中国药典》中《分析方法验证指导原则》的要求 [ 10 ] ,能够满足人参药材质量分析的要求。
《中国药典》收录的人参药材方法单针检测时间过长, 不利于大生产中快速提供检测结果。本文采用的方法与药典方法比较, 该方法更加高效, 节省近 70% 的检测时间, 节省更多试剂成本。
中药材及饮片作为中药制剂的基础,其质量水平的控制在整个制药全生命周期中至关重要,《中国药典》收录的人参药材方法仅含有 3 种人参皂苷检测, 本文根据多批检出情况, 将含量大于 0.05% 的人参皂苷类成分使用同一分析方法进行研究, 确定人参皂苷类 Rg1、Re、Rf、Rb1、Rc、Rb2、Rd 等 7 种人参皂苷含量水平较高,将其作为具有代表性成分研究。但随着中药研究的规范化, 仅对指标性成分的含量进行测定还不能够完整体现药物质量属性, 还应进一步结合指纹图谱、其他理化性质等进行综合考量。
