Objective The application of quality control chart in the quantitative detection of Staphylococcus aureus in milk was discussed by collecting the routine detection process of Staphylococcus aureus in milk and testing the results of Staphylococcus aureus in quality control samples simultaneously. Methods Monitoring the detection data by utilizing the center line of the single value control chart and the moving range control chart, an upper warning limit, a lower warning limit, an upper control limit and a lower control limit, standard error and data point inside and outside the boundary of the range control chart are used to monitor and evaluate the laboratory detection process, and the status of the personnel, instruments and reagents in the experimental detection process is judged. Results By observing the range of test data and analyzing the distribution characteristics and changes of data, the results showed that single value control chart and moving range control chart could effectively monitor the detection process of Staphylococcus aureus in milk, and the data results were reliable. Conclusion The construction of the quality control chart provides an effective means of food hygiene quality control for laboratory testing of the project, and helps to promote the smooth development of the quality control work in the laboratory testing process.

目的 通过收集牛奶中金黄色葡萄球菌日常检测过程,以及同步测试质控样品中的金黄色葡萄球菌检测结果,探讨质控图在牛奶中金黄色葡萄球菌定量检测过程中的应用。方法 利用构建单值控制图和移动极差控制图中心线、上警示限、下警示限、上控制限、下控制限对检测数据进行监控,采用标准差和移动极差控制图界限内和界限线外的数据点对实验室检测过程进行监控和评估,判断实验检测过程人员、仪器、试剂的状态。结果 通过观察实验检测数据所在范围,分析数据分布特点和变化,表明单值控制图和移动极差控制图可以有效监控牛奶中金黄色葡萄球菌检测过程,数据结果可靠。结论 该质控图的构建为实验室检测该项目提供了一种有效的食品卫生质量监控手段,有助于促进实验室检测过程的质量控制工作的顺利开展。

Objective The application of quality control chart in the quantitative detection of Staphylococcus aureus in milk was discussed by collecting the routine detection process of Staphylococcus aureus in milk and testing the results of Staphylococcus aureus in quality control samples simultaneously. Methods Monitoring the detection data by utilizing the center line of the single value control chart and the moving range control chart, an upper warning limit, a lower warning limit, an upper control limit and a lower control limit, standard error and data point inside and outside the boundary of the range control chart are used to monitor and evaluate the laboratory detection process, and the status of the personnel, instruments and reagents in the experimental detection process is judged. Results By observing the range of test data and analyzing the distribution characteristics and changes of data, the results showed that single value control chart and moving range control chart could effectively monitor the detection process of Staphylococcus aureus in milk, and the data results were reliable. Conclusion The construction of the quality control chart provides an effective means of food hygiene quality control for laboratory testing of the project, and helps to promote the smooth development of the quality control work in the laboratory testing process.

金黄色葡萄球菌(Staphylococcus aureus)广泛存在于自然界中, 能引起人类和动物的感染, 是引起食物中毒的重要致病菌之一。在我们日常饮食当中,接触最多的肉蛋奶制品等是金黄色葡萄球菌易存在的主要介质,肠毒素是金黄色葡萄球菌的主要次级代谢产物, 是造成食品安全事故的重要元凶, 食品是否受到金黄色葡萄球菌的污染问题不容轻视, 因此金黄色葡萄球菌一直是食品质量卫生检验检疫中重要检测项目, 对预防和控制食物中毒具有实际意义 [ 1 - 2 ]
在 UHT 牛奶的食品卫生标准中, 对金黄色葡萄球菌的定量检测是判断 UHT 牛奶质量是否合格的一项重要检测指标, 但在实验室对 UHT 牛奶中金黄色葡萄球菌的检测过程中,实验人员、 检测设备、试剂材料、实验方法、环境等因素都可以影响实验检测结果的准确性、可靠性和稳定性 [ 3 - 4 ] ,因此,对实验室检测过程的质量控制就显得尤为重要。
在实验室质量控制评价方法中, 控制图方法由美国沃特 · 阿曼德·休哈特于 1924 年提出 [ 5 ] ,该方法基于统计显著性原则进行过程控制, 可通过区分 “偶然原因” 和 “可查明原因”[ 6 - 8 ], 及时发现实验或生产过程中的异常现象, 并通过识别异常程度和分析产生原因, 实现实验室质量控制。因此, 被食品检测实验室广泛应用的质量控制图是监视和分析检测数据质量的有效措施, 也是各个实验室内部质量控制和提升实验室检测能力的重要手段 [ 9 - 11 ] 。本文旨在利用单值控制图和移动极差控制图构建应用于牛奶中金黄色葡萄球菌定量检测的质量控制, 以确保实验室检测结果的准确性、可靠性和公正性 [ 12 - 13 ] ,为实验室质量管理和检测能力评价提供参考依据。
GB 4789.10-2016 第二法规定的培养基和试剂。
Baird-Parker 琼脂平板 (北京陆桥技术股份有限公司); 血琼脂平板(北京陆桥技术股份有限公司);脑心浸出液肉汤(BHI)(北京陆桥技术股份有限公司);兔血浆(北京陆桥技术股份有限公司);营养琼脂小斜面 (北京陆桥技术股份有限公司);革兰氏染色液 (北京陆桥技术股份有限公司)。
GB 4789.10-2016 要求所用的器具。
设备:MIR-H263L-PC 恒温培养箱(普和希健康医疗器械(上海)有限公司)、ME2002 电子天平(0.01 g,梅特勒托利多科技 (中国)有限公司)、MLS-830L 高压蒸汽灭菌锅(普和希健康医疗器械(上海)有限公司)、XT5118-OV240 电热鼓风干燥箱(杭州雪中炭恒温技术有限公司)、1374 生物安全柜(赛默飞世尔科技(中国)有限公司)。
材料: 市售超高温瞬时灭菌(UHT)奶、商业定量冻干菌株颗粒。
(1)将 $9\mathrm{\;{mL}}$ 预热至 $\left({{36}\pm 1}\right){}^{\circ }\mathrm{C}$ 的磷酸盐缓冲液加到 ${12}\mathrm{\;{cm}}\times {20}\mathrm{\;{cm}}$ 匀质袋中或者 ${50}\mathrm{\;{mL}}$ 无菌离心管中。
(2)从冰箱中取出定量装有冻干菌株颗粒的小瓶,在打开样品瓶之前, 让样品瓶中的菌粒平衡至室温 (在室温下放置, 至少 30 min)。
注: 如果打开冷冻的管子, 瓶中可能形成冷凝水。这种水分和氧气的混合物会产生有害的自由基, 进而降低冻干细胞的活性。
(3)用无菌镊子将菌株颗粒转移到装有 $9\mathrm{\;{mL}}$ 磷酸盐缓冲液的匀质袋中或者无菌离心管中。
(4)让小球在稀释液中静置 ${60}\mathrm{\;s}$ 后,用手轻揉匀质袋,或者在使用离心管时, 使用涡旋振荡器轻柔振荡, 直至菌株颗粒完全分散, 此为初级菌悬液。
(5)将菌悬液置于 $\left({{36}\pm 1}\right){}^{\circ }\mathrm{C}$ 复苏 ${30}\mathrm{\;{min}}$
(6)根据计划制备质控样品数量和单个样品体积,计算需要 UHT 牛奶总量。本案例每一颗冻干菌株颗粒需要 ${600}\mathrm{\;{mL}}$ UHT 牛奶。在接种前, UHT 牛奶需要放至室温。
(7)准备一个无菌容器瓶,并放入无菌磁转子。往容器瓶中加入 ${600}\mathrm{\;{mL}}$ 的 UHT 牛奶,标记。
(8)从相应标记的容器瓶中吸取大约 ${25}\mathrm{{mL}}$ UHT 牛奶,加入装有初级菌悬液的匀质袋中或者无菌离心管中。封好袋口或者拧紧离心管盖子,轻摇混合,为次级菌悬液。
(9)将以上约 ${34}\mathrm{\;{mL}}$ 次级菌悬液,加到所标记的容器瓶中 (UHT 的总体积为 ${600}\mathrm{\;{mL}}$ ), ${30}\mathrm{\;{min}}$ 之内完成操作。
(10)将容器瓶放在搅拌器上,打开搅拌开关。选择合适的搅拌速度,保证均质,避免产生泡沫。
(11)在不停搅拌的同时,用 ${10}\mathrm{\;{mL}}$ 移液器从容器瓶中吸取 ${20}\mathrm{\;{mL}}$ 质控样品分装到 ${50}\mathrm{\;{mL}}$ 无菌离心管中。
(12)盖好离心管盖子,立刻将离心管放入 $-{15}^{\circ }\mathrm{C}$$-{20}^{\circ }\mathrm{C}$ 的冰箱。有效期建议不超过一个月。
( 13 )检测客户样品时,从冰箱中拿出一支装有质控样品的离心管, 间隔放置在离心管架上, 连架子一起放入已预热至 (45±1)℃的恒温水浴锅中,快速解冻不超过 15 min。备用。
(1)按照 GB 4789.10-2016 第二法,同步检测客户样品和 UHT 牛奶金黄色葡萄球菌质控样品。UHT 牛奶金黄色葡萄球菌质控样品被视为定量冻干菌株颗粒的 100 倍稀释液,吸出 $1\mathrm{\;{mL}}$ 就是 10~2 的稀释度。
(2)融化后的样品需要立刻进行检测,不能重新冷冻为后续之用。
按照 GB 4789.10-2016 第二法,由 A、B、C、D、E 等五名人员与日常检测同步测试 20 个质控样品 (即 $n ={20}$ ) 中的金黄色葡萄球菌,并记录时间、检测人员和结果。
汇总 20 组金黄色葡萄球菌的结果,并计算对数值 $\left({{\log }_{10}X}\right)$ 、 移动值之和 $\left({\sum {\log }_{10}X}\right)$ 、移动极差 $\left({R}_{m}\right)$ 、移动极差之和 $\left({\sum {R}_{m}}\right)$ , 结果如 表 1
2.2 平均值、标准偏差、移动极差和平均移动极差计算
平均值: $\bar{X}= \frac{\mathop{\sum }\limits_{{i = 1}}^{n}{\log }_{10}X}{n}= \frac{69.49}{20}= {3.47}$ ;
标准偏差: $S =\sqrt{\frac{\mathop{\sum }\limits_{{i = 1}}^{n}{\left({\log }_{10}X -\bar{X}\right)}^{2}}{n - 1}}= {0.09}$ ;
移动极差: ${R}_{m}= \left|{{x}_{j}- {x}_{j - 1}}\right|\left({j = 2,3,\ldots , n}\right)$ ;
平均移动极差: ${\bar{R}}_{m}= \frac{1}{n - 1}\mathop{\sum }\limits_{{j = 2}}^{n}{R}_{m}= \frac{2.18}{19}= {0.11}$
参考 GB/T 4883,使用格拉布斯(Grubbs)检验法判断离群值。
使用标准差 $S$ 计算统计警示限和控制限。
中心线: ${C}_{L}= \bar{X}= {3.47}$ ;
警示限: ${U}_{WL}/{L}_{CL}= \bar{X}\pm {2S}= {3.47}\pm {0.18}$ ;
上/下控制限: ${U}_{CL}/{L}_{CL}= \bar{X}\pm {3S}= {3.47}\pm {0.27}$
注: 上 $/$ 下控制限也可以用公式: ${U}_{CL}/{L}_{CL}= \bar{X}\pm {2.660}\times {\bar{R}}_{m}$ 本案例 图 1 为单值控制图 ( $X$ 控制图)。
2.4.2 移动极差控制图 $\left({R}_{m}\right.$ 控制图)的建立
中心线: ${C}_{L}= {\bar{R}}_{m}= {0.11}$ ;
上控制限: ${U}_{CL}= {3.267}{\bar{R}}_{m}= {3.267}\times {0.11}= {0.36}$ ;
下控制限: ${L}_{CL}= 0$
本案例 图 2 为移动极差控制图 $\left({R}_{m}\right.$ 控制图)。
实验室通过评估决定是否修改控制限和中心线。如果结果的分散性和平均值发生了变化, 则通过统计学的分析决定该变化是否显著。如果变化显著, 且能合理解释变化的原因 (例如, 采用了新的质控样), 则应予以修改。如果不能合理解释变化的原因,则应暂缓修改,展开调查,并采取必要措施。
根据 GB/T 17989.2-2020《控制图:常规控制图》,绘制出 $X$ 控制图和 ${R}_{m}$ 控制图, 图 1 为单值控制图,其中 $\bar{X}$ 为中心线, $\bar{X}\pm {2S}$ 为上下警示限,当实验检测数据在 $\bar{X}\pm {2S}$ 区间内,说明过程处于统计控制状态, 实验数据可靠; 当实验数据超过此区间时, 说明过程虽处于统计控制状态, 但应引起注意, 判断是否要有相应纠正措施。 $\bar{X}\pm {3S}$ 为上下控制限,在此区间内的数据为可接受范围, 当实验数据超过此区间时, 说明过程处于失控状态, 需要采取行动调查原因。
从上面打点图可见, 实验检测数据服从正态分布规律, 点子均落在上下警示限之内表明过程稳定, 处于统计控制状态, 未出现 GB/T 17989.2-2020 中所示的波动可查明原因的几种情形。可知: 该实验过程中, 人员操作稳定规范, 无人为引入干扰因素;检测设备仪器稳定可靠;实验检测系统持续稳定正常, 质控样品数据准确, 实验结果可靠无异常。
本研究通过对牛奶中金黄色葡萄球菌质控样品的定量检测, 利用子组均值 $\bar{X}$ 和子组极差均值 ${\bar{R}}_{m}$ 构建了单值控制图和移动极差控制图, 实现了对该实验程序的有效监控和预警, 确保了实验数据的准确、可靠性。为实验室检测该项目提供了一种有效的食品卫生质量监控手段, 可确保实验室的检测能力, 有利于促进实验室提高对牛奶中金黄色葡萄球菌检测质量控制的认识和结果评价工作 [ 3 ]
