气相色谱仪是用高纯气体作为流动相,待测样品或标准试样在气化室气化后被载气带入色谱柱,各组分在色谱柱内分离后,被检测器检测。基线是气相色谱仪运行中检测器信号的直接表现,也是仪器性能的综合表现,组成仪器的各部分发生故障、操作条件和外界条件的变化等因素都会反映到基线上,因此根据基线异常状况可以判断故障的原因和部位。

随着现代检测技术的发展, 越来越多的分析仪器被开发与应用。气相色谱仪作为常见的分析仪器, 其具有的高灵敏度、 高选择性、分析速度快、所需试样量少、应用范围广等优点, 已经被大量应用于医药卫生、石油化工、环境监测、生物化学等领域 [ 1 ] 。色谱法是以高纯气体或高纯液体为流动相的分析方法, 用于分离分析各类物质, 已成为极为重要的分离分析方法之一 [ 2 ] 。通过统计梳理各类基线异常问题,查找原因,归纳总结, 以期通过分析色谱基线问题, 为排除气相色谱仪故障查明原因提供帮助。
色谱仪是以气体作为流动相 (载气), 样品用定量环或者微量注射器 “注射” 进入气化室后, 被载气携带进入相应能分离所需物质的填充柱或毛细管色谱柱 [ 3 ] 。用已知浓度的标准样品来定性和定量, 通过各组分在色谱柱分配系数差异, 在载气的带动下,各组分在色谱中得到分离,然后用接在柱后的各类检测器(例如氢火焰离子化检测器 FID (Flame Ionization Detector)、热导检测器 TCD (Thermal Conductivity Detector) 等) 将各组分按顺序检测出来 [ 4 ] 。气相色谱仪的种类多,功能有所差异。一般由载气和温度控制部分、进样部分、分离部分 (色谱柱)、检测部分、数据处理部分组成 [ 5 ] ,如 图 1
载气问题,气源压力低,钢瓶气源压力低后污染物大量流出, 排气流量有波动, 载气流量不稳定; 净化器失活, 仪器内设置的净化器受污染后的影响;气路系统漏气;色谱柱污染或失效等。
检查气路是否有泄漏或污染, 更换新气源; 核实更正或更换净化器;重新检漏;活化再生或更换色谱柱。对色谱仪载气气瓶减压阀进行更换,更换后对色谱仪进行分析,仪器波动消失, 分析结果正常 [ 6 ]
基线缓慢上涨或减小, 在对气路检查未发现泄漏和压力控制不稳定的问题,排除载气问题后,有可能是环境温度、电压等环境条件的大范围的波动, 导致温度控制偏移。还有可能是 TCD 检测器故障,热导池内热丝老化,检测器信号持续上涨。 进样口、色谱柱污染, 进样口堵塞, 色谱柱污染, 污染物随时间持续流出色谱柱, 导致的基线持续走高或走低, 但这种状态一般不会持续太久。
检查其环境或电压等环境条件是否有变化。更换新的热导丝或 TCD 检测器模块。清洁进样口、更换进样口衬管、更换玻璃棉; 高温老化色谱柱,切除进样口端色谱柱 10-50cm,如若问题还未得到改善,可以使用溶剂清洗色谱柱或更换新的色谱柱 [ 7 ]
色谱柱连接检测器一端不当,进检测器端色谱柱长度过短或过长;未按仪器要求尺寸裁切色谱柱长短,进样口端色谱柱裁切部位有毛刺; 玻璃衬管破损, 玻璃碎屑掉在气化室或分流平板上导致分流不均匀;分析过程中气体泄漏;燃气选择或流量不当;进样口隔垫漏气或长时间未更换, 老化或有碎屑产生, 在分析过程中产生不规则变化; 色谱柱污染或柱流失, 污染物或色谱柱流动相随时间持续流出色谱柱,导致基线不规则变化: 检测器污染,有高沸点物质流出色谱柱后附着在检测器上 [ 8 ]
重新按仪器要求连接色谱柱与检测器端, 连接长度应和色谱仪要求长度一致; 进样口端色谱柱应切面平整, 无毛刺; 检查色谱衬管是否破损, 隔垫是否老化或漏气, 否则应及时更换, 并清理气化室内污染物; 色谱柱污染时需及时高温老化色谱柱或切取被污染部分;检测器被污染时需及时在高于使用温度下老化检测器,老化无效时拆下清洗检测器 [ 9 ]
出现负峰是检测器的数据处理系统信号极性设置反了, 正极设置为负极或负极设置为正极;圆顶峰大多出现在载气纯度过低或流速不稳定, 进样时未能快速进样; 不规则刺峰可能是由于柱流失或柱污染,检测器严重污染或电路故障导致。
应根据载气与被检测组分的热导系数大小不同进行设置检测器的极性 [ 10 ] 。更换载气或载气纯化器;老化或更换色谱柱. 加大尾吹气量;清洗、维修检测器。
柱温箱内柱温未按程序设定值升温或降温; 载气流速或纯度发生变化;样品使用的溶剂条件发生变化;色谱柱分离能力下降 [ 11 ]
检查柱温箱温度变化是否与程序设定值一致, 不一致时及时排查原因,查看柱温箱是否有破损或泄露; 使用流量计对载气流速进行测定,如果载气流速变化较明显,则需更换载气; 需要样品与标准品使用相同的溶剂,确保保留时间一致 [ 12 ] 。老化色谱柱,更换或裁切色谱柱前端。
进样器或色谱柱对样品吸附能力较强; 进样量大或分流比太小;柱温或气化室温度太低;两个化合物未在色谱柱上完全分离或色谱柱安装不正确; 汽化衬管被污染(内有注射垫碎屑或其他固体颗粒);补充气未开或偏低等 [ 13 ]
更换惰性进样器, 更换合适的色谱柱, 降低或减少拖尾; 不超过色谱柱使用温度的前提下, 提高气化室或柱箱温度, 气化室温度选择应确保比样品所测组分高二十摄氏度; 适当减少进样量或增大分流比使两个化合物分开, 单一组分时适当增大载气流量或升高柱温改善峰形, 选择合适的色谱分离柱, 重新按仪器要求安装色谱柱;重新清洗、填充或更换汽化室衬管; 更换或打开补充气。
进样量太大引起色谱柱过载;样品冷凝,汽化温度偏低, 样品中某些高沸点组分没有迅速气化; 汽化室衬管被污染或对样品产生吸附作用;色谱柱被污染等。
减少进样量或用稀溶液进样; 升高调整气化温度; 采用填充惰性石英棉的惰性衬管和失活进样器;增加载气流量; 更换汽化衬管;适当增加柱温或老化色谱柱 [ 14 ]
样品浓度升高; 色谱柱被污染, 固定相被破坏; 进样失败, 进样不稳定, 形成二次进样, 样品未正常进入色谱仪气化室内。
对样品进行稀释, 减少进样量, 提高分流比; 老化色谱柱或更换新的色谱柱;重新手动进样或采用自动进样器进样 [ 15 ]
(1)基线异常时应先检查色谱仪器外部硬件使用条件是否改变, 是否更换过新的载气或助燃气气瓶或纯化器等设备配件。 如有则要先检查这些外部设备是否连接正确, 工作正常。还应检查载气纯化器纯化后的气体是否满足需求, 流量是否稳定。
(2)还应当检查进样垫是否老化,老化了的隔垫会有碎屑杂质组分进入色谱系统中, 造成基线异常, 遇到这种问题应及时更换进样隔垫。
(3)衬管是否清洁, 衬管内石英棉是否清洁, 是否积碳过多导致样品无法正常气化;造成色谱基线异常波动。清洗衬管时可先用丙酮或乙醇等溶剂充分冲洗浸泡, 用超声波清洗十分钟以上,然后烘干,除去杂质,最后再重新装填专用石英棉使用 [ 16 - 17 ]
(4)色谱柱污染或失效也会导致基线或峰形异常,因此需要不定时对色谱柱进行老化操作, 老化时应选择合适的温度, 一般低于色谱柱的最高使用, 高于所需分析组分的最高沸点温度二十度。有高沸点物质或难气化物质进入到色谱柱内,可选择切掉进样口端色谱柱 1-2 米 (毛细柱)。填充柱可选择增加预分离柱预先进行分离, 保护后分离柱。若色谱柱失效或老化后仍未改善基线或峰形异常,则需更换新的色谱柱。
(5)此外,检测器污染也可能造成基线异常问题,可以通过调高检测器温度 (不超过检测器使用的最高温度), 高温除去附着在检测器上的污染物,或者停机拆下清洗的方法来解决。
