Objective In order to ensure the quality control of enterprise production products and improve detection efficiency, a high-performance liquid chromatography method for Benzyl Propionate Oxadiazon Granules is established. Methods The HPLC method was used to separate and determine the contents of Benzyl Propionate Oxadiazon Granules which contained the active ingredients mentioned above. In the method, the column ZORBAX Eclipse Plus${\mathrm{C}}_{18}$(${4.6}\mathrm{\;{mm}}\times {50}\mathrm{\;{mm}},{1.8}\mathrm{\;{\mu m}},{600}\mathrm{\;{Bar}}$) was used and the mixture of acetonitrile, isopropanol and 0.05% phosphoric acid solution was used as the mobile phase, and${245}\mathrm{\;{nm}}$was the selected wavelength in the determination. Results It has been found that bensulfuron methyl, pretilachlor, and oxadiazon not only have high chromatographic peak purity under this analytical method, but also have a good linear relationship. The peak purity of bensulfuron methyl in this product is 992, with a recovery rate of 98.82%. The peak purity of pretilachlor is 993.3, with a recovery rate of 99.88%. The peak purity of oxadiazon is 999.3, and the recovery rate is 99.22%. Conclusion This chromatographic method is stable and suitable for quality control analysis of this product.

目的 为了做好企业生产产品的质量把控,提高检测效率,特建立苄嘧磺隆·丙草胺·噁草酮颗粒剂的高效液相色谱方法。方法 采用高效液相色谱法,使用色谱柱 ZORBAX Eclipse Plus${\mathrm{C}}_{18}$(${4.6}\mathrm{\;{mm}}\times {50}\mathrm{\;{mm}},{1.8\mu }\mathrm{m}$,${600}\mathrm{{Bar}}$),以乙腈、异丙醇和${0.05}\%$磷酸水溶液为流动相,在${245}\mathrm{\;{nm}}$波长下对苄嘧磺隆·丙草胺·噁草酮颗粒剂进行液相色谱分析。结果 经检测苄嘧磺隆、丙草胺和噁草酮在此分析方法下不仅具有较高的色谱峰纯度, 同时也具有良好的线性关系。该产品中苄嘧磺隆峰纯度 992,回收率 98.82%;丙草胺峰纯度 993.3 ,回收率 99.88%;噁草酮峰纯度 999.3,回收率 99.22%。结论 该色谱方法稳定,适合该产品的质量控制分析。

目的 为了做好企业生产产品的质量把控,提高检测效率,特建立苄嘧磺隆·丙草胺·噁草酮颗粒剂的高效液相色谱方法。方法 采用高效液相色谱法,使用色谱柱 ZORBAX Eclipse Plus${\mathrm{C}}_{18}$(${4.6}\mathrm{\;{mm}}\times {50}\mathrm{\;{mm}},{1.8\mu }\mathrm{m}$,${600}\mathrm{{Bar}}$),以乙腈、异丙醇和${0.05}\%$磷酸水溶液为流动相,在${245}\mathrm{\;{nm}}$波长下对苄嘧磺隆·丙草胺·噁草酮颗粒剂进行液相色谱分析。结果 经检测苄嘧磺隆、丙草胺和噁草酮在此分析方法下不仅具有较高的色谱峰纯度, 同时也具有良好的线性关系。该产品中苄嘧磺隆峰纯度 992,回收率 98.82%;丙草胺峰纯度 993.3 ,回收率 99.88%;噁草酮峰纯度 999.3,回收率 99.22%。结论 该色谱方法稳定,适合该产品的质量控制分析。

Objective In order to ensure the quality control of enterprise production products and improve detection efficiency, a high-performance liquid chromatography method for Benzyl Propionate Oxadiazon Granules is established. Methods The HPLC method was used to separate and determine the contents of Benzyl Propionate Oxadiazon Granules which contained the active ingredients mentioned above. In the method, the column ZORBAX Eclipse Plus${\mathrm{C}}_{18}$(${4.6}\mathrm{\;{mm}}\times {50}\mathrm{\;{mm}},{1.8}\mathrm{\;{\mu m}},{600}\mathrm{\;{Bar}}$) was used and the mixture of acetonitrile, isopropanol and 0.05% phosphoric acid solution was used as the mobile phase, and${245}\mathrm{\;{nm}}$was the selected wavelength in the determination. Results It has been found that bensulfuron methyl, pretilachlor, and oxadiazon not only have high chromatographic peak purity under this analytical method, but also have a good linear relationship. The peak purity of bensulfuron methyl in this product is 992, with a recovery rate of 98.82%. The peak purity of pretilachlor is 993.3, with a recovery rate of 99.88%. The peak purity of oxadiazon is 999.3, and the recovery rate is 99.22%. Conclusion This chromatographic method is stable and suitable for quality control analysis of this product.

6% 苄嘧磺隆·丙草胺·噁草酮颗粒剂是由苄嘧磺隆、丙草胺及噁草酮作为有效成分, 与其他辅料助剂作为载体配制而成的三元复配的颗粒型除草剂。苄嘧磺隆是一种防治对象广泛的可喷雾法可药土法除草剂;丙草胺属 2- 氯化乙酰替苯胺类除草剂, 是选择性芽前处理剂。可通过植物下胚轴、中胚轴和胚芽鞘吸收, 根部略有吸收, 直接干扰杂草体内蛋白质合成, 噁草酮用于防除多种一年生单子叶和双子叶杂草, 主要用于水田除草, 对旱田的花生、棉花、甘蔗等亦有效, 是一种触杀性芽前、 芽后除草剂。田间试验表明, 该产品以抛洒施药的方式, 亩用量在${350}\sim {450}\mathrm{\;g}$之间,通过对杂草的根部、叶片吸收,对水稻田一年生杂草有良好的防治效果。
产品是企业的基石, 符合标准要求的高质量产品更是企业前进发展的动力, 6% 苄嘧磺隆·丙草胺·噁草酮颗粒剂作为企业典型的三元复配颗粒剂, 质量把关控制工作不容有失。只有建立稳定的检测方法, 严格控制质量, 做到产品合格才能出厂, 只有这样, 才能让农民用上放心的药, 对作物负责, 为作物量产做保障。为此,针对 6% 苄嘧磺隆·丙草胺·噁草酮颗粒剂组成多填料多, 色谱检测复杂的情况, 特此情况研究并开发建立了高效液相色谱法, 为企业其他的颗粒剂产品的生产与检测提供参考价值。
液相色谱仪 (型号 1260 ,具有 DAD 检测器及四元梯度系统, 美国安捷伦)、色谱柱 ZORBAX Eclipse Plus${\mathrm{C}}_{18}({4.6}\mathrm{\;{mm}}\times {50}\mathrm{\;{mm}}$ ,${1.8\mu }\mathrm{m},{600}\mathrm{{Bar}}$ ,美国安捷伦)、电子天平(CPA225D,天平精度 0.01 mg,德国 Sartorius)、超声波清洗器 (KS-150D,宁波海曙科生超声设备)、超纯水机 (Smart-S15, 上海和泰仪器)。
甲醇、乙腈和异丙醇均为色谱纯度; 磷酸为分析纯度。标准品: 苄嘧磺隆含量是 98.4%,采购于沈阳沈化院测试技术有限公司;丙草胺含量是 98.3%,采购于沈阳沈化院测试技术有限公司;噁草酮含量是 99.2%,采购于上海市农药研究所。待分析产品(有效成分苄嘧磺隆占比 0.5%、丙草胺占比 4.0%、噁草酮占比 1.5%)为公司产品。
分别准确称取 3 个标准品于${100}\mathrm{\;{mL}}$的容量瓶中,其中苄嘧磺隆${20}\mathrm{{mg}}$、丙草胺${160}\mathrm{{mg}}$、噁草酮${60}\mathrm{{mg}}$,用移液管移入${10}\mathrm{\;{mL}}$乙腈充分混匀后,再用量筒添加甲醇接近刻度,最后将容量瓶放置于超声波清洗器水浴中超声辅助溶解${10}\mathrm{\;{min}}$(每隔$2\mathrm{\;{min}}$将容量瓶取出剧烈摇晃 10 次,以保证试样有效成分溶解完全), 超声溶解完成后取出容量瓶降温到室温, 最后用甲醇补加,定容到刻度线,摇匀过滤后装到进样瓶,待用。
将混匀的$1\mathrm{\;{kg}}$样品进行充分研磨混匀,向${100}\mathrm{\;{mL}}$的容量瓶中准确加入研磨后的样品$4\mathrm{\;g}$,先加$5\mathrm{\;{mL}}$水超声打散,保证样品化开,再加${10}\mathrm{\;{mL}}$乙腈后混匀,最后补加甲醇近刻度,将容量瓶至于超声波水浴中超声溶解${10}\mathrm{\;{min}}$(每隔$2\mathrm{\;{min}}$将容量瓶取出剧烈摇晃 10 次,以保证试样有效成分溶解完全),超声溶解完成后取出容量瓶降温到室温, 最后用甲醇补加, 定容到刻度线, 摇匀过滤后装到进样瓶, 待用。
流动相比例$\left({V/V/V}\right)$ 为乙腈 : 异丙醇 : 0.05%磷酸水溶液$\left({{15}: {30}: {55}}\right)$ ,数据采集停止时间为${13}\mathrm{\;{min}}$ ; 流动相流速为${1.4}\mathrm{\;{mL}}/\mathrm{{min}}$ ;检测器采用波长${245}\mathrm{\;{nm}}$ ;柱温箱设置柱温为${52}^{\circ }\mathrm{C}$ ; 溶液进样量为${5\mu }{\mathrm{L}}^{\left\lbrack 1 - 2\right\rbrack }$ 。
参照以上分析设置条件情况下,等待仪器预热完成后,谱图基线稳定后(即相邻的同一样品的峰面积相差应在 1.5% 以内), 设置进样顺序依据一针标样溶液、两针试样溶液再一针标样溶液的顺序进行进样测定。产品典型的标样和试样出峰的色谱图见 图 1图 2, 苄嘧磺隆、丙草胺、噁草酮的保留时间分别为 0.7、5.2、9.9 min。
公式如下:

${r}_{1}$ 和${r}_{2}$ 为配制的标样和试样溶液中对应有效成分的峰面积平均值;${m}_{1}$ 和${m}_{2}$ 为对应有效成分的标样和试样质量,$\mathrm{g};p$ 为对应标样品标识的质量分数 %。
选用色谱柱 ZORBAX Eclipse Plus${\mathrm{C}}_{18}$ (${4.6}\mathrm{\;{mm}}\times {50}\mathrm{\;{mm}}$ ,${1.8\mu }\mathrm{m},{600}\mathrm{{Bar}}$ ) 的主要原因与其他同类型色谱柱相比较,该色谱柱有耐高压的特点。因为该色谱方法流动相中的 0.05% 磷酸水相比例占 55%,且在 1.4 mL/min 的流速下,压力较高,在机器未预热完全、柱温未达到${52}^{\circ }\mathrm{C}$ 的稳定方法时间段,压力甚至会激增到 400 Bar 以上, 所以必须采用耐高压高温的色谱柱; 其次该色谱柱相对其他同填料色谱柱, 不仅分离速度较快, 而且分离效果最好, 缩短了数据采集时间的同时, 也保证的有效成分的高分离纯度, 所以选用该色谱柱。
选用该流动相比例的主要原因是乙腈:异丙醇:0.05% 磷酸水溶液$\left({{15}: {30}: {55}}\right)$ 时,能够完全分离出杂质与苄嘧磺隆、 丙草胺和噁草酮三个有效成分, 尤其在丙草胺保留时间附近, 杂质与丙草胺做到了完全分离,且峰型对称稳定,重现性好。
在该色谱分析条件下, 将上述制备的 6% 苄嘧磺隆·丙草胺 - 噁草酮颗粒剂样品溶液进行 DAD 波长 (190~400 nm) 扫描, 得到苄嘧磺隆、丙草胺和噁草酮的吸收光谱图, 测得其峰纯度, 其中苄嘧磺隆峰纯度为 992.0 ;丙草胺峰纯度为 993.3 ;噁草酮峰纯度为 999.3;见图 3~5。结果表明在测定色谱分析条件下, 试样中苄嘧磺隆、丙草胺和噁草酮的峰纯度高,杂质基本无干扰。
分析条件的选择从耐高压耐高温色谱柱的下手,其次到流动相的选择优化比例, 最后 DAD 扫描的特异性的验证, 一步一步做到了 6% 苄嘧磺隆·丙草胺·噁草酮颗粒剂中的苄嘧磺隆、 丙草胺以及噁草酮三个有效成分的分离, 峰形对称稳定、响应合适。有效成分和杂质之间互不干扰, 满足农药产品质量分析方法确认的先决条件。
(1)向${50}\mathrm{\;{mL}}$ 容量瓶中准确称取一定量的苄嘧磺隆、丙草胺以及噁草酮标准品, 然后用量筒向容量瓶中添加乙腈到接近刻度线, 之后将容量瓶放入超声波清洗器水浴中超声辅助溶解${10}\mathrm{\;{min}}$ (每隔$2\mathrm{\;{min}}$ 将容量瓶取出剧烈摇晃 10 次,以保证试样有效成分溶解完全), 再将容量瓶冷却至室温后, 最后用乙腈定容到刻度线,此溶液待用。
(2)使用五根移液管准确移取此溶液 2.0、4.0、6.0、8.0、${10.0}\mathrm{\;{mL}}$ 分别置于 5 个${100}\mathrm{\;{mL}}$ 容量瓶中,用量筒添加乙腈至接近刻度,定容后充分摇匀,此五个浓度容量瓶样品为测试样品。 在上述色谱分析操作条件情况下将不同浓度的三组分有效成分按照浓度从低到高进行进样操作, 用有效成分的质量浓度 (单位为$\mathrm{{mg}}/\mathrm{{mL}}$ )为横坐标$x$ 轴,与之对应的色谱图峰面积为纵坐标$y$ 轴绘制曲线。结果见图 6~8
上述实验说明, 苄嘧磺隆、丙草胺以及噁草酮在上述色谱分析操作条件情况下有较为良好的线性关系。苄嘧磺隆、丙草胺以及噁草酮的线性回归方程分别为: 苄嘧磺隆$y ={7488.1x}+$ ${0.7273}, r ={0.9999}$ ; 丙草胺$y ={717.59x}+ {96.884}, r ={0.9996}$ ; 噁草酮$y ={6707.2x}+ {3.3296}, r ={0.9998}$ 。该试验结果表明,各个有效成分在制备的浓度区间内, 均有较好的线性相关性, 符合样品检测要求。
按标样试样预处理方法对试样产品重复测定 6 次, 精密度测得结果见表 1。根据重复含量的测定, 计算得出苄嘧磺隆的变异系数为 1.04%,丙草胺的变异系数为 0.46%,噁草酮的变异系数为 0.93%。苄嘧磺隆、丙草胺以及噁草酮的相对标准偏差均小于${2}^{\left( 1 -{0.5}\log C\right)} \times {0.67}$ ($C$ 为样品以小数计的有效成分含量), 符合有效成分分析方法确认精密度(重复性)的要求。
在已知质量分数的颗粒剂试样中, 分别加入已知量的苄嘧磺隆、丙草胺以及噁草酮标准品,加入量与已知量比例为$1 : 1$ , 加入量与已知量之和为标准品预处理中有效成分的称样量。采用加标回收率法测定其回收率, 重复测定 5 次, 结果表明回收率高, 符合农药制剂产品中的有效成分分析的回收率要求。(见表 2)。
该色谱检测方法从数据色谱图和峰纯度分析不难得出, 此方法不仅苄嘧磺隆、丙草胺、噁草酮之间的分离效果好, 方法稳定, 峰面积重现好, 而且苄嘧磺隆、丙草胺及噁草酮三个有效成分和杂质峰之间也得到了有效的分离, 基本无干扰。值得一提的是将该颗粒剂产品从老旧的检测方法做了创新, 从苄嘧磺隆和噁草酮(高效液相色谱法检测)、丙草胺(气相色谱法检测) 优化到统一使用高效液相色谱法检测, 大幅缩短了产品检测周期,保证质量的同时提高了检测效率,做到了质量和生产两手抓。该检测方法的不足之处在于压力高、柱温高,接近色谱柱的使用上限,长期的高压高温使用,会缩短该色谱柱的使用寿命, 也会缩短仪器相关耗材的使用寿命, 缩短液相色谱仪的维护保养周期。
在对 6% 苄嘧磺隆·丙草胺·噁草酮颗粒剂的质量方法确认过程中, 通过对分析条件选择的讨论, 线性相关、精密度、准确度的试验结果表明,此色谱分析方法稳定,精密度、准确度高。 适用该颗粒剂产品在生产过程中的质量控制分析。
