目的 改进水中叶绿素a检测过程中存在的丙酮易挥发、叶绿素a易降解、750 nm 处吸光值偏高等操作问题,降低实验误差,提升测定结果的准确度和精确度。方法 根据环境保护部发布的《水质叶绿素a的测定 分光光度法》(HJ897—2017)标准方法,优化改进实验操作中存在的叶绿素a易降解、丙酮易挥发和待测液的浊度影响吸光值等导致分析误差的操作条件。结果 优化改进后,降低了操作危险性,实验结果提升26.3%,加标回收率在 10 µg/L和20 µg/L 加标浓度下分别提升10.8%、2.9%。结论 本文研究了用分光光度法检测水质叶绿素a的实验操作条件,通过对操作条件的改变,证明减少了叶绿素a的降解和丙酮的挥发,降低了待测液的浊度,提升了叶绿素a浓度,为提高测定结果的准确性提供参考。
