Objective To investigate the residues characteristics of veterinary drug in Gallus gallus domesticus from Hubei Province and assess the dietary exposure risks. Methods The 34 types of veterinary drug residues were detected in 100 randomly collected samples of Gallus gallus domesticus from farms, agricultural markets, wholesale markets and slaughterhouses in Hubei Province in 2024 by ultra performance liquid chromatography-tandem mass spectrometry (UPLC-MS/MS). The dietary exposure risk assessment of detected veterinary drug residues was conducted using the veterinary drug residue risk ranking matrix of the United Kingdom Veterinary Drug Residue Committee and the index of food safety (IFS) method. Results The 12 kinds of veterinary drugs were detected in the samples, with a residue detection rate of 49%. Among these, 2 kinds of drugs were prohibited for use in food animals, and 1 kind of drug was approved for therapeutic use but prohibited from detection in animal-derived foods. The risk assessment revealed that the IFS values of all detected veterinary drugs were significantly less than 1, indicating a low dietary exposure risk from residues. Conclusion This suggests that the quality and safety of Gallus gallus domesticus in Hubei Province are satisfactory, with minimal health risks posed by residual veterinary drugs to humans.

目的 探讨湖北地区乌骨鸡肉中兽药残留特征以及膳食暴露风险。方法 使用超高效液相色谱-串联质谱法(ultra performance liquid chromatography-tandem mass spectrometry, UPLC-MS/MS)对2024年在湖北地区养殖场、农贸市场、批发市场及屠宰场随机采集的100份乌骨鸡肉样品中的34种兽药残留进行分析, 参考英国兽药残留委员会兽药残留风险排序矩阵和食品安全指数(index of food safety, IFS)法对检出的兽药残留进行膳食暴露风险评估。结果 本次抽检的乌骨鸡肉共检出12种兽药, 药物残留检出率为49%, 其中2种为食品动物中停止使用药物, 1种为允许作治疗用, 但不得在动物性食品中检出。根据风险评估结果显示, 检出兽药的IFS值均远小于1, 兽药残留的膳食暴露风险处于较低水平。结论 本次监测的湖北乌骨鸡肉质量安全状况较好, 残留兽药对人体健康的危害风险较低。

目的 探讨湖北地区乌骨鸡肉中兽药残留特征以及膳食暴露风险。方法 使用超高效液相色谱-串联质谱法(ultra performance liquid chromatography-tandem mass spectrometry, UPLC-MS/MS)对2024年在湖北地区养殖场、农贸市场、批发市场及屠宰场随机采集的100份乌骨鸡肉样品中的34种兽药残留进行分析, 参考英国兽药残留委员会兽药残留风险排序矩阵和食品安全指数(index of food safety, IFS)法对检出的兽药残留进行膳食暴露风险评估。结果 本次抽检的乌骨鸡肉共检出12种兽药, 药物残留检出率为49%, 其中2种为食品动物中停止使用药物, 1种为允许作治疗用, 但不得在动物性食品中检出。根据风险评估结果显示, 检出兽药的IFS值均远小于1, 兽药残留的膳食暴露风险处于较低水平。结论 本次监测的湖北乌骨鸡肉质量安全状况较好, 残留兽药对人体健康的危害风险较低。

Objective To investigate the residues characteristics of veterinary drug in Gallus gallus domesticus from Hubei Province and assess the dietary exposure risks. Methods The 34 types of veterinary drug residues were detected in 100 randomly collected samples of Gallus gallus domesticus from farms, agricultural markets, wholesale markets and slaughterhouses in Hubei Province in 2024 by ultra performance liquid chromatography-tandem mass spectrometry (UPLC-MS/MS). The dietary exposure risk assessment of detected veterinary drug residues was conducted using the veterinary drug residue risk ranking matrix of the United Kingdom Veterinary Drug Residue Committee and the index of food safety (IFS) method. Results The 12 kinds of veterinary drugs were detected in the samples, with a residue detection rate of 49%. Among these, 2 kinds of drugs were prohibited for use in food animals, and 1 kind of drug was approved for therapeutic use but prohibited from detection in animal-derived foods. The risk assessment revealed that the IFS values of all detected veterinary drugs were significantly less than 1, indicating a low dietary exposure risk from residues. Conclusion This suggests that the quality and safety of Gallus gallus domesticus in Hubei Province are satisfactory, with minimal health risks posed by residual veterinary drugs to humans.

乌骨鸡, 又称乌鸡、竹丝鸡等, 是一种具有滋补、药用和观赏价值的珍禽。近年来, 随着乌骨鸡市场需求量的增加, 养殖规模的扩大, 使用兽药防治疾病, 促进动物生长提高成活率, 增加了兽药残留的风险[1-3]。其兽药残留水平的高低直接决定了乌骨鸡食用的安全性, 因此开展乌骨鸡肉中兽药残留分析, 并进行兽药残留膳食风险评估对于保障人体健康尤为重要。
目前, 国内关于猪肉、鸡蛋、鸡肉等动物性食品中兽药残留分析及膳食暴露风险评估的研究报道较多[4-6], 兽药残留污染及健康风险评估[7-9]一直广受关注。如张大文等[10]在2013年1—12月对江西省的两个区域的农贸市场长期监测, 对采集的265份猪肉样品中四环素类药物进行残留风险分析, 发现强力霉素检出率为21.89%。武玉平等[11]对山东部分地区140份鸡蛋进行15种兽药及禁用药物监测, 检出率为16.43%。马洁等[12]对天津市2016—2022年市售的418份鸡肉样品进行检测, 发现兽药残留检出率为46.65%, 不合格率为8.37%。这些兽药残留的食品安全指数(index of food safety, IFS)均小于1, 暴露风险较低, 不会对人体健康产生不良作用。刘峰等[13]对宁夏市2016—2020年市售鸡肉和鸡蛋中23种兽药残留及健康风险评估, 发现鸡肉中兽药残留检出率为38.0%, 鸡蛋中兽药残留检出率为26.6%。鸡肉中强力霉素的IFS, 鸡蛋中强力霉素、氟甲喹和氟苯尼考的IFS均远大于1, 存在健康风险隐患。这些监测数据表明兽药残留现象较为普遍, 如果长期食用存在兽药残留风险的动物性食品可能会对人体健康造成潜在危害[14-16], 因此对动物性食品中兽药残留进行定期监测及膳食暴露风险评估十分必要, 然而目前针对乌骨鸡肉中的兽药残留及膳食暴露风险评估研究报道较少。
为了解湖北省乌骨鸡肉中兽药残留特征, 掌握兽药残留对人体健康风险[17-19], 本研究通过前期查询文献资料, 调研湖北地区乌骨鸡养殖场兽药使用情况, 初步对乌骨鸡肉中的风险因子进行了摸底排查; 对养殖过程中使用过的7类兽药进行监测, 采用英国兽药残留委员会兽药残留风险排序矩阵[20-21]和IFS法进行兽药残留的膳食暴露风险评估[22-24], 为保障湖北乌骨鸡肉质量安全以及产业持续绿色发展提供技术支撑。
2024年以湖北省13个地区(黄石、咸宁、襄阳、荆州、十堰、孝感、随州、仙桃、潜江、天门、神农架、恩施、黄冈)作为采样区域, 随机选择养殖场、农贸市场、批发市场及屠宰场作为采样点, 共采集100份乌骨鸡肉样品, 每份约200 g, 采集后进行匀浆处理并冷冻保存。
SCIEX TRIPLE QUAD 6500+超高效液相色谱-串联质谱仪(美国SCIEX公司); PRiME HLB固相萃取小柱(200 mg/6 mL, 美国Waters公司); 固相萃取装置、0.22 µm微孔尼龙滤膜、Eclipse Plus C18 RRHD色谱柱(2.1 mm× 150 mm, 1.8 μm)(美国Agilent公司)。
乙腈、甲醇(质谱级, 美国Merk公司); 甲酸(质谱级, 美国赛默飞世尔科技公司); 红霉素(纯度91.4%)、金霉素(纯度93.5%)、土霉素(纯度88.8%)、甲砜霉素、氟苯尼考、恩诺沙星、环丙沙星、氧氟沙星、替米考星、四环素、磺胺氯哒嗪、磺胺二甲嘧啶、磺胺甲噁唑、磺胺喹噁啉、甲氧苄啶(纯度≥95%)(中国兽医药品监察所); 氯霉素、氟苯尼考胺、司帕沙星、氟罗沙星、二氟沙星、氟甲喹、阿奇霉素、罗红霉素、克拉霉素、多西环素、磺胺甲氧嘧啶、甲硝唑及代谢物、地美硝唑及代谢物、金刚烷胺、阿昔洛韦、奥司他韦、咪喹莫特(纯度≥95%, 德国Dr公司)。
根据前期乌骨鸡养殖环节的用药种类调研, 检测了34种兽药及其代谢物, 包括4种酰胺醇类药物(氯霉素、甲砜霉素、氟苯尼考及代谢物氟苯尼考胺)、7种氟喹诺酮类药物(司帕沙星、氟罗沙星、二氟沙星、氟甲喹、恩诺沙星、环丙沙星、氧氟沙星)、5种大环内酯类药物(阿奇霉素、罗红霉素、克拉霉素、红霉素、替米考星)、4种四环素类药物(四环素、多西环素、金霉素、土霉素)、5种磺胺类药物(磺胺氯哒嗪、磺胺二甲嘧啶、磺胺甲氧嘧啶、磺胺甲噁唑、磺胺喹噁啉)、4种硝基咪唑类药物(甲硝唑及代谢物、地美硝唑及代谢物)、4种抗病毒类药物(金刚烷胺、阿昔洛韦、奥司他韦、咪喹莫特)和甲氧苄啶。
参照中国兽医药品监察所自建动物性食品中多种类药物残留检测[25-26]对乌骨鸡肉样品中34种兽药残留进行分析。称取2.0 g试料于50 mL离心管内, 加入80%乙腈水溶液8.0 mL, 涡旋1 min, 高速振荡提取5 min, 10000 r/min离心10 min, 取上清液4.0 mL直接加载到PRiME HLB固相萃取柱中净化, 保持2 mL/min的流速, 收集全部流出液。于45 ℃下氮吹至体积小于1.0 mL, 用10%甲醇水溶液定容至1.0 mL, 涡旋混匀, 吸取适量过0.22 μm微孔滤膜后供超高效液相色谱-串联质谱法(ultra performance liquid chromatography-tandem mass spectrometry, UPLC-MS/MS)测定[27-29], 禁用药物的定量限为0.5 µg/kg, 限用药物的定量限为1 µg/kg。
禁用药物及兽药残留采用中华人民共和国农业部第2292号公告、农业农村部第250号公告及GB 31650—2019《食品安全国家标准 食品中兽药最大残留限量》中规定的限量值作为标准, 对乌骨鸡肉样品中检出的兽药残留污染特征进行分析和判定, 若检出兽药超过最大残留限量或者检出禁用药物可判定为不合格样品。
参考英国政府官网的Annual Report on Surveillance for Veterinary Residues in Food in the UK 2007(食品中兽药残留监测年度报告)中兽药残留风险矩阵排序, 风险得分公式(1)如下:
风险得分(S)=(A+B)×(C+D+EF
式中: A为毒性(0~6分): 毒理学数据作为兽药评估潜在不良反应, 潜在不良反应越重, 特别是不可逆, 矩阵排名得分越高; B为毒效(0~3分): 参考每日允许摄入量(allowable daily intake, ADI)值, 能引起不良反应的剂量越低, 得分越高; C为膳食比例(0~3分): 来自经过处理的动物的食物比例越高, 得分越高(有些药物只用于单一物种, 而另一些药物则用于多个物种, 从而增加了接触的机会); D为兽药使用频率(0~3分): 给某种特定动物施药的频率; E为高暴露人群(0~3分): 高接触人群, 由于饮食习惯, 一些群体可能会摄入较高数量的特定残留物。如果有证据支持这些群体进行评估, 则得分越高; F为残留水平(0~3分): 检测残留物的评估, 检测到的浓度越高, 与特定物质的最大残留限量(maximum residue limit, MRL)/最低要求执行量(minimum required performance limit, MRPL)相比, 分配的分数越高。
乌骨鸡肉样品中兽药残留风险级别按照风险得分排序, 分为低风险(S<15.0)、中风险(15.0≤S<20.0)和高风险(S≥20.0)。
IFS是将残留结果和膳食暴露相结合来评价化学污染物对人体的健康危害程度[30-32]。本研究采用《食品中化学物风险评估的原则和方法》中的点评估模型进行暴露评估。基本原理为化学物每日估计暴露量(estimated daily intake, EDI)等于食物消费量均值与化学物含量的乘积。公式(2)如下:

式中: EDI为药物每日暴露量估算值, μg/(kg bw d); X为调查人群每日消费量, g/d; 消费量数据参照国家统计局2021年年鉴, 湖北省鸡肉人均年消费为7.7 kg, 换算成人均日平均摄入量为21.1 g/d; N为兽药残留量检测值, µg/kg, 本研究以药物N的最大值进行估计; bw为调查人群平均体重, kg, 取人体平均体重60 kg[33]
采用IFS法对乌骨鸡肉中兽药残留的人群健康风险大小进行评价。当IFS≤1时, 表明通过膳食途径摄入兽药对人体健康影响的风险是可以接受的; 当IFS>1时, 表明通过膳食途径摄入兽药存在发生对健康不利影响的风险, 并且这种风险随着数值的增大而增大。
IFS计算公式(3)如下:
IFS=EDI×f/SI
式中: f为药物安全摄入量的校正因子; SI为药物的安全摄入量, 若SI采用ADI值, f取1。毒理学参考值参照国际食品法典委员会(Codex Alimentarius Commission, CAC)规定的ADI表示。兽药残留ADI值参考GB 31650—2019, 部分药物尚未设定ADI值, 因此未进行统计。
采用Excel 2016软件分析数据和绘制图表[34], 对100份乌骨鸡肉样品中的兽药检出率和不合格率进行统计, 运用风险排序方法进行风险排序, IFS进行膳食暴露风险评估。
在100份样品中, 检出12种兽药残留样品49份, 检出率为49%, 其中不合格样品6份, 不合格率为6%。检出样品中有44份检出1种兽药; 有5份同时检出2种兽药, 占10.2%。检出率最高的为恩诺沙星(以恩诺沙星与环丙沙星之和计), 占30.61%; 其次为氟苯尼考(以氟苯尼考及代谢物氟苯尼考胺之和计), 占18.37%; 后面依次是替米考星、多西环素、甲氧苄啶、四环素。不得检出药物中, 氧氟沙星、金刚烷胺和羟基地美硝唑均有检出(表1), 其中2种为食品动物中停止使用药物, 1种为允许作治疗用, 但不得在动物性食品中检出。
不同采样环节的兽药残留情况如表2所示。养殖场的检出率为43.55%, 不合格率为6.45%。农贸市场的检出率为47.06%, 不合格率为2.94%。因为批发市场和屠宰场的样品量少, 可能导致结果分析存在一定的不确定性, 所以分析结果不具有代表性。本研究根据不合格样品的采样来源发现, 养殖环节监测出的不合格药物有氧氟沙星、金刚烷胺、羟基地美硝唑和甲氧苄啶。说明养殖户法律意识淡薄, 缺少科学用药意识, 存在违规用药和未严格遵守休药期的现象。
根据1.3.4的风险排序方法, 每种药物的毒性得分(A)、毒效得分(B)可通过各种兽药半数致死剂量(lethal dose 50%, LD50)、ADI来赋值, 其他根据相关文献资料和检测残留物水平来赋值, 金刚烷胺、氧氟沙星、羟基地美硝唑、甲氧苄啶、恩诺沙星(以恩诺沙星与环丙沙星之和计)和氟苯尼考(以氟苯尼考与氟苯尼考胺之和计)为高风险药物, 需要引起关注; 多西环素、四环素、磺胺甲噁唑为中风险药物, 替米考星为低风险药物。具体结果见表3
采用点评估法计算了检出兽药EDI和IFS。结果显示, 所有检出兽药的IFS值均远小于1, 表明当前乌骨鸡肉中兽药残留的健康风险较低(表4)。然而, 甲氧苄啶的IFS值为0.1845, 需进一步关注。不得检出药物金刚烷胺和羟基地美硝唑尚未设定ADI值, 未进行风险评价。
本研究抽取了100份湖北地区乌骨鸡肉样品, 采用UPLC-MS/MS对乌骨鸡肉中34种兽药残留进行分析[35-36], 兽药残留检出率为49%, 不合格率为6%。检出样品中有44份检出1种兽药, 5份同时检出2种兽药, 占10.2%, 可能存在联合用药。在检出的12种兽药中2种为食品动物中停止使用药物氧氟沙星和金刚烷胺, 1种为允许作治疗用, 但不得在动物性食品中检出的药物地美硝唑代谢物羟基地美硝唑, 采用英国兽药残留委员会兽药残留风险矩阵进行残留风险排序, 氧氟沙星、金刚烷胺和羟基地美硝唑的风险得分也最高。建议监管部门加大禁限药的宣传力度, 同时加强日常监管。
本次检出率位列前4的药物为恩诺沙星(以恩诺沙星与环丙沙星之和计)、氟苯尼考(以氟苯尼考及代谢物氟苯尼考胺之和计)、替米考星、多西环素, 有研究表明恩诺沙星联合替米考星、氟苯尼考联合多西环素能增强抗菌能力的效果[37-38]。检出率排列第5的药物是甲氧苄啶, 甲氧苄啶在兽医临床是作为磺胺类药物的增效剂, 检出率远高于磺胺类药物, 有研究表明甲氧苄啶与氟苯尼考等其他兽用抗菌药联合使用也能增强抗菌药的活性[39-40], 说明养殖中可能存在联合用药的情况。联合用药会影响药物的代谢和排泄过程, 改变药物在体内的休药期, 这可能是以上这些允许使用药物容易被检出的原因之一。
根据膳食暴露风险评估结果, 检出的12种兽药, 除了金刚烷胺和羟基地美硝唑尚未设定ADI值, 未进行风险评价。其他药物的IFS值均远小于1, 说明本次监测的乌骨鸡肉样品的兽药残留状况对人体产生的膳食摄入风险较低。结合本研究建议兽医行业主管部门按照农业农村部发布的《乌鸡养殖安全用药管控技术性指导意见》要求, 指导养殖人员科学合理用药, 保障乌骨鸡产业健康发展。
