Objective To analyze the content characteristics of 5 kinds of heavy metals including lead (Pb), cadmium (Ca), total arsenic (As), total chromium (Cr), and total mercury (Hg) in Arachis hypogaea L. sold in Anhui Province, and to establish risk assessment of dietary exposure based on consumption data. Methods A total of 80 Arachis hypogaea L. samples were collected from 16 cities in Anhui Province. The content of Pb, Ca, As, Cr and Hg in Arachis hypogaea L. samples were determined using inductively coupled plasma mass spectrometry (ICP-MS). The Nemerow comprehensive pollution index were applied to evaluate the heavy metal pollution status in Arachis hypogaea L.. Combined with the consumption survey data on Arachis hypogaea L. consumption of residents in Anhui Province from 2015 to 2017, to analyze the exposure of various population groups in Anhui Province to heavy metals via Arachis hypogaea L. consumption, and the health effects were evaluated. Results Among the 80 Arachis hypogaea L. samples, the detection rates of Pb, Cd, As, Cr and Hg were 92.5%, 100.0%, 100.0%, 93.8% and 7.5%, respectively. Except for Cr, which had an over-limit rate of 5.0%, all other elements remained within the acceptable limits. The difference was statistically significant (P<0.05). The single-factor pollution index of samples collected from Huainan City and Chuzhou City was greater than 0.7. Males aged 18-50 years had the highest Cr EDI [0.0622 μg/(kg·d), P97.5=0.413 μg/(kg· d)]. The margin of exposure (MOE)/margin of safety (MOS) values of the 5 kinds of heavy metal elements in Arachis hypogaea L. were all greater than 1. The MOS value of the Cr element from 18-50 aged was lowed than 10, while the MOS values of Cd in were the lowest. Conclusion The detection rates of 5 kinds of heavy metals in Arachis hypogaea L. in Anhui Province are high, but the over-limit rates are low. Cr levels exceeds standards in certain localized areas, while Cd pollution indices varied across different sampling stages. The health risks from heavy metal exposure through Arachis hypogaea L. consumption are determined to be at acceptably low levels for local residents.

目的 分析安徽省市售散装生花生中铅(Pb)、镉(Cd)、总砷(As)、铬(Cr)、总汞(Hg) 5种重金属含量特征, 结合消费量数据开展膳食暴露风险评估。方法 随机采集安徽省16地市不同环节花生样品, 使用电感耦合等离子体质谱法(inductively coupled plasma mass spectrometry, ICP-MS)测定样品中5种重金属元素含量。使用内梅罗污染指数评价花生中的重金属污染状况, 结合2015—2017年安徽省居民花生消费量调查数据, 分析不同人群暴露重金属情况, 评估健康风险。结果 80份花生样品中Pb、Cd、As、Cr、Hg检出率依次为92.5%、100.0%、100.0%、93.8%、7.5%, 其中Cr超标率为5.0%。结果差异具有统计学意义(P<0.05)。淮南市、滁州市采集的样品单因子污染指数PCr>0.7。男性重金属暴露量高于女性, 18~50岁男性Cr元素暴露量最高[0.0622 μg/(kg· d), P97.5=0.413 μg/(kg·d)]。不同性别、年龄的人群花生中5种重金属元素的膳食暴露限值(margin of exposure, MOE)和安全限值(margin of safety, MOS)均大于1, Cd的MOS值较其他元素最低, 18~50岁人群膳食Cr元素MOS值较低(<10)。结论 安徽省花生中重金属检出率高, 超标率低, Cr元素在个别地区存在超标现象。Cd元素在不同采样环节的样品中污染指数存在差异, 居民食用花生暴露重金属的健康风险较低。

Pollution characteristics of 5 kinds of heavy metals and dietary exposure risk assessment in Arachis hypogaea L. in Anhui Province

目的 分析安徽省市售散装生花生中铅(Pb)、镉(Cd)、总砷(As)、铬(Cr)、总汞(Hg) 5种重金属含量特征, 结合消费量数据开展膳食暴露风险评估。方法 随机采集安徽省16地市不同环节花生样品, 使用电感耦合等离子体质谱法(inductively coupled plasma mass spectrometry, ICP-MS)测定样品中5种重金属元素含量。使用内梅罗污染指数评价花生中的重金属污染状况, 结合2015—2017年安徽省居民花生消费量调查数据, 分析不同人群暴露重金属情况, 评估健康风险。结果 80份花生样品中Pb、Cd、As、Cr、Hg检出率依次为92.5%、100.0%、100.0%、93.8%、7.5%, 其中Cr超标率为5.0%。结果差异具有统计学意义(P<0.05)。淮南市、滁州市采集的样品单因子污染指数PCr>0.7。男性重金属暴露量高于女性, 18~50岁男性Cr元素暴露量最高[0.0622 μg/(kg· d), P97.5=0.413 μg/(kg·d)]。不同性别、年龄的人群花生中5种重金属元素的膳食暴露限值(margin of exposure, MOE)和安全限值(margin of safety, MOS)均大于1, Cd的MOS值较其他元素最低, 18~50岁人群膳食Cr元素MOS值较低(<10)。结论 安徽省花生中重金属检出率高, 超标率低, Cr元素在个别地区存在超标现象。Cd元素在不同采样环节的样品中污染指数存在差异, 居民食用花生暴露重金属的健康风险较低。

Objective To analyze the content characteristics of 5 kinds of heavy metals including lead (Pb), cadmium (Ca), total arsenic (As), total chromium (Cr), and total mercury (Hg) in Arachis hypogaea L. sold in Anhui Province, and to establish risk assessment of dietary exposure based on consumption data. Methods A total of 80 Arachis hypogaea L. samples were collected from 16 cities in Anhui Province. The content of Pb, Ca, As, Cr and Hg in Arachis hypogaea L. samples were determined using inductively coupled plasma mass spectrometry (ICP-MS). The Nemerow comprehensive pollution index were applied to evaluate the heavy metal pollution status in Arachis hypogaea L.. Combined with the consumption survey data on Arachis hypogaea L. consumption of residents in Anhui Province from 2015 to 2017, to analyze the exposure of various population groups in Anhui Province to heavy metals via Arachis hypogaea L. consumption, and the health effects were evaluated. Results Among the 80 Arachis hypogaea L. samples, the detection rates of Pb, Cd, As, Cr and Hg were 92.5%, 100.0%, 100.0%, 93.8% and 7.5%, respectively. Except for Cr, which had an over-limit rate of 5.0%, all other elements remained within the acceptable limits. The difference was statistically significant (P<0.05). The single-factor pollution index of samples collected from Huainan City and Chuzhou City was greater than 0.7. Males aged 18-50 years had the highest Cr EDI [0.0622 μg/(kg·d), P97.5=0.413 μg/(kg· d)]. The margin of exposure (MOE)/margin of safety (MOS) values of the 5 kinds of heavy metal elements in Arachis hypogaea L. were all greater than 1. The MOS value of the Cr element from 18-50 aged was lowed than 10, while the MOS values of Cd in were the lowest. Conclusion The detection rates of 5 kinds of heavy metals in Arachis hypogaea L. in Anhui Province are high, but the over-limit rates are low. Cr levels exceeds standards in certain localized areas, while Cd pollution indices varied across different sampling stages. The health risks from heavy metal exposure through Arachis hypogaea L. consumption are determined to be at acceptably low levels for local residents.

花生是我国广泛种植的重要油料作物之一, 在安徽省油料作物播种面积中排名第二, 仅次于油菜籽[1], 花生也是安徽省重要的经济作物。花生仁中含有丰富的脂肪酸、维生素、氨基酸、矿物质元素等人体必需营养素, 《中国居民膳食指南(2022)》推荐每周摄入坚果50~70 g, 适量食用花生有益于心脏健康[2-3]
食品中的重金属主要来源于土壤、水、化肥、杀虫剂等[4], 有研究表明, 花生能富集多种重金属元素, 镉元素含量与土壤中镉含量呈正相关[5]。重金属经食物链进入人体后, 难以被人体分解代谢, 产生累积性。长期、低剂量摄入重金属, 会对人体肾脏、肝脏、神经系统、骨骼组织等造成严重的健康危害[6]。我国在GB 2762—2022《食品国家安全标准食品中污染物限量》和NY/T 1067—2006《中华人民共和国农业行业标准 食用花生》中对花生中重金属的最大残留量分别作了规定, 依次是铅(Pb)≤0.2 mg/kg、镉(Cd)≤0.5 mg/kg、无机砷(As)≤0.2 mg/kg、铬(Cr)≤1.0 mg/kg、汞(Hg)≤0.02 mg/kg。
目前, 我国对于本地食品中重金属的污染研究大部分集中在粮食、水果、蔬菜和肉类等食品[7-9], 花生中重金属的分析少有报道[10]。安徽省是我国十大花生种植主产区之一[11], 单产水平常年居全国领先水平[12]。调查污染物水平, 关注种植质量, 将提高我国花生产业可持续发展能力。本研究使用电感耦合等离子体质谱(inductively coupled plasma mass spectrometry, ICP-MS)法对花生样品中重金属元素Pb、Cd、As、Cr和Hg的含量进行测定, 分析安徽省花生中这5种重金属元素的污染特征。评估花生中的重金属污染对本地居民产生的健康风险, 为相关卫生与健康政策的制定与优化提供数据支持, 确保居民饮食安全与健康。
采集安徽省16地市的的花生样品, 从每个市随机抽取不少于3个采样点, 每个采样点采集样品1~2份, 每份样品不少于1 kg, 均为散装干花生。采样环节涵盖种植环节和流通环节(商场超市、农贸市场、个体商店), 共采集80份, 其中74份样品产地为安徽省, 6份样品分别产自河南、河北、辽宁、吉林和黑龙江省。样品来源分布见图1
将采集的花生去除杂、除壳, 不脱花生衣, 经实验室专用粉碎机粉碎, 过筛, 储存于洁净的塑料保鲜盒中, 于2~8 ℃保存。
多元素混合溶液标准物质[GBW(E)082429, 浓度10 mg/kg]、汞单元素溶液标准物质(GBW08617, 质量浓度1000 mg/L)(中国计量科学研究院); 内标元素混合标准溶液(GNM-M043583-2013, 质量浓度100 mg/L, 国家有色金属及电子材料分析测试中心); 调谐溶液(质量浓度1 μg/L, 美国PerkinElmer公司); 硝酸(色谱级, 苏州晶瑞化学有限公司); 黄豆粉成分分析标准物质(GBW10190, 坛墨质检科技股份有限公司)。
NexLON 300D电感耦合等离子体质谱仪(美国PerkinElmer公司); ultraCLAVE超级微波消解仪(北京莱伯泰科仪器股份有限公司); XPE204万分之一天平[梅特勒-托利多仪器(上海)有限公司]; Milli-Q IQ element超纯水机(美国Millipore公司)。
称取0.2 g样品于消解管中, 加入3 mL硝酸, 将消解管放入装有440 mL纯水和10 mL硝酸的内衬桶内, 经超级微波消解仪消解。消解完成后, 用纯水转移并定容至25 mL, 混匀待测, 同时进行空白实验。
根据《国家食品污染和有害因素风险监测工作手册》中食品中多元素分析的标准操作程序和GB 5009.268—2016《食品安全国家标准 食品中多元素的测定》, 采用ICP-MS法同时测定Pb、Cd、As、Cr、Hg 5种元素含量, 测定结果以mg/kg表示。
选用黄豆粉成分分析标准物质为质量控制样品, 进行6次平行测定试验; 随机选取10%的样品进行平行样品测定, 对结果超标或异常样品进行复测试验。
GB 2762—2022中对花生中的Pb和Cd的限量标准作了规定, As、Cr、Hg则依据农业部标准NY/T 1067—2006《食用花生》的卫生指标进行评价, 详见表1
使用单因子污染指数法和内梅罗综合污染指数法分别对花生中5种重金属的污染程度展开评价[13]。单因子污染指数计算见公式(1)。

式中: Pi为污染元素i的污染指数; Ci为污染元素i在样品中的浓度, mg/kg; Si为污染元素的最高残留限量值, mg/kg。
内梅罗综合污染指数计算见公式(2)。

式中: P为内梅罗综合污染指数; Pi为各污染元素i的污染指数的平均值; Pi,max为各污染元素i污染指数的最大值。参考CHENG等[14]对重金属污染划分为5个等级: P≤0.7, 0.7<P≤1.0, 1.0<P≤2.0, 2.0<P≤3.0, P>3.0, 分别对应安全等级、警戒限、轻度污染、中度污染和重度污染。
花生消费量数据来自2015—2017年安徽省居民营养与健康状况监测, 在全省共设置12个监测点, 运用多阶段整群随机抽样的方法选择调查对象, 开展3 d 24 h膳食回顾调查, 收集各调查对象的个体花生消费量数据。共调查18岁及以上人群7548人, 其中男性3588人, 女性3960人, 18~50岁组3066人, 50~65岁组2733人, 65岁以上组1749人。
为匹配检测样品类型, 本研究只针对花生仁(花生米)的消费量进行统计, 对花生制品的消费量不作统计。带壳花生与花生仁的质量转换系数以0.7计。
结合消费量调查数据与花生样品中Pb、Cd、As、Cr、Hg的含量, 采用点评估模型[15], 根据公式(3)计算人群每日每公斤体重的膳食暴露量(estimated daily intake, EDI)。

式中: F为花生的人群每日消费量, g/d; X为花生中重金属i的平均含量, mg/kg; BW为人群体重, kg。
联合国粮农组织和世界卫生组织食品添加剂联合专家委员会(the Joint FAO/WHO Expert Committee on Food Additives, JECFA)指出1.2 μg/kg·bw·d的Pb暴露水平, 将引起人群收缩压升高1 mmHg[16], 因此将1.2 μg/kg·bw·d作为Pb元素的基准剂量下限值(benchmark dose limit, BMDL); 推荐Cd元素的每月可耐受摄入量(provisional tolerable monthly intake, PTMI)为25 μg/kg·bw, 则每日可耐受摄入量(tolerable daily intake, TDI)为0.833 μg/kg·bw[16]; JECFA通过评估, 给出无机As元素引起肺癌的基准剂量下限(95%可信区间, BMDL0.5)为3 μg/kg·bw·d[17]; 在2018年, 美国环保署(U.S. Environmental Protection Agency, USEPA)提出的每日安全摄入六价Cr限值为3 μg/kg· bw[18]; JECFA建议除鱼类和贝类以外总Hg对于成人的每周可耐受摄入量(provisional tolerable weekly intake, PTWI)为1 μg/kg·bw, 儿童为4 μg/kg·bw[17]
使用暴露限值法(margin of exposure, MOE)评估通过食用花生摄入Pb和As的暴露风险情况[19]。无机As毒性较强, 在总As含量占比约70%[20], 保守选择总As作为无机As的评价指标。MOE=BMDL/EDI, BMDL为Pb和无机As的基准剂量下限, EDI为对应元素的膳食暴露量。若MOE>1, 认为健康风险较低; MOE≤1, 认为存在一定健康风险, 需予以关注。
使用安全限值(margin of safety, MOS)法评价Cd、Hg和Cr的暴露风险[15]。MOS=TDI/EDI, 将PTMI、PTWI换算成每日可耐受摄入量TDI, Cd元素TDI为0.833 μg/kg·bw, Hg元素为0.14 μg/kg·bw。六价Cr为致癌物, 而三价Cr为营养元素[21], 保守认为Cr元素TDI为3 μg/kg·bw。当MOS≤1时, 认为存在暴露风险。MOS值越大, 风险越低。
参照世界卫生组织(World Health Organization, WHO)关于低水平污染物检测数据可信度统计规则的建议[22], 若检测结果低于检出限(limit of detection, LOD), 记为未检出。当未检出率(未检出样品在所有检测样品中的比例)小于60%时, 所有未检出数据以LOD的一半(1/2 LOD)作为替代值; 当未检出率大于等于60%时, 所有未检出数据直接用LOD替代。
采用Excel 2007和SPSS 20软件进行数据统计分析。对计量资料进行正态性检验(Kolmogorov-Smirnov检验)后, 发现Pb、As、Cr、Hg元素含量不符合正态分布(P<0.05), Cd元素含量符合正态分布(P=0.20), 采用均值与中位数对数据进行分布描述。用Kruskal-Wallis 检验分析不同重金属元素检测结果之间的差异性。检出率和超标率的差异采用χ2检验, 检验水准α=0.05。以P<0.05为差异有统计学意义。
Cr元素含量最高, 平均值为0.334 mg/kg, 其次是Cd, 平均值为0.209 mg/kg, Pb和As含量平均水平小于0.1 mg/kg, Hg元素水平最低(<0.001 mg/kg)。不同元素的检出率差异具有统计学意义(χ2=286.882, P<0.05)。Cd和As元素检出率100.0%, Hg检出率(7.5%) 最低, 仅有6份检出, 另外Pb和Cr的检出率分别为92.5%和93.8%。不同元素的超标率差异也具有统计学意义(χ2=16.162, P<0.05)。除Cr元素(超标率5.0%)外, 其余4个元素均没有超标现象。见表2
表3可见, 农贸市场和商场/超市采购的花生样品中, 存在Cr元素超标情况, 超标率分别为7.7% (2/26)和9.5% (2/21), 超标样品均为本地生产。Hg元素在种植环节的样品中无检出。不同采样环节的花生中, 同一元素的含量除Cd元素(H=13.28, P<0.05)外, 其他4个元素的含量没有显著性差异(P>0.05)。农贸市场采集的样品中Cd元素平均含量最高, 为0.258 mg/kg, 种植环节(农户家中)采集的花生中Cd元素平均含量最低, 为0.147 mg/kg。
采用单因子污染指数与内梅罗综合污染指数评价法, 对花生中5种重金属元素的污染状况进行评估, 得出单因子污染指数Pi由高到低排列为: Cd (0.42)>Cr (0.29)> Hg (0.21)>Pb (0.14)>As (0.07)。不同采样环节的具体评价结果见图2。农贸市场采集的花生样品Cd污染程度最大(PCd=0.52)。在农贸市场和商场/超市环节, 分别有2份样品处于Cr元素的中度污染和重度污染水平。种植环节存在1份样品出现Pb轻度污染。Hg、As元素在所有样品中均处于安全水平。Cd元素的污染指数在不同采样环节有显著性差异(H=13.29, P<0.05)。不同采样环节的综合污染指数P没有显著性差异(P>0.05)。
分析不同采样地区的花生中的重金属污染程度(表4), 淮南市和滁州市采集的花生样品PCr分别为0.87、2.24。其余采样城市的花生中重金属污染程度较轻, 处于安全水平。
表5可见, 同一年龄组中, 男性的花生消费量大于女性, 有显著差异(P<0.05)。同一性别组人群, 其年龄越大, 体重越低, 花生消费量也呈现降低趋势。18~50岁男性体重最高(68.3 kg), 花生消费量最大(12.7 g/d, P97.5=84.5 g/d)。65岁以上女性体重最低(54.3 kg), 花生消费量最少(1.89 g/d, P97.5=30.0 g/d)。50~65岁男性人群, 最大消费量在105 g/d, 而50~65岁女性人群中, 最大消费量达到200 g/d, 表明在性别整体差异的背景下, 个体间的花生消费行为也存在波动。
安徽省经食用花生摄入5种重金属元素的暴露量结果见图3。男性通过食用花生摄入5种重金属的暴露量均高于女性群体。Cr元素暴露量最高[0.0116~0.0622 μg/(kg·d)], 其次是Cd元素[0.0073~0.0389μg/(kg·d)]、Pb元素[0.0009~0.0051 μg/(kg·d)], As元素与Hg元素暴露水平较低。18~50岁男性Cr元素暴露量最高[0.0622 μg/(kg·d), P97.5=0.413 μg/(kg·d)]。
根据各地区花生重金属含量的平均值水平和1.7.2中描述方法, 进行不同性别、年龄人群的花生膳食暴露风险评估。5种元素在不同组别人群中的MOE/MOS值均大于1, 表明我省居民花生中重金属暴露对健康的潜在风险较低。高端消费人群Cd的MOE值均在10以下, 表明Cd较其他元素经花生膳食的暴露风险更高。此外, 18~50岁人群Cr元素高端暴露的MOS值也低于10。见表6
本研究对安徽省本地花生中5种重金属(Pb、Cd、As、Cr、Hg)的含量特征及其人群膳食暴露风险进行分析。结果显示, 80份花生样品中重金属的检出率较高, 但超标率较低, 仅Cr元素出现5.0%超标率的情况。安徽省花生在种植和销售过程中, 整体重金属污染情况处于可控范围。
不同地区采集的花生中重金属污染程度存在差异。花生中的Cd含量与土壤中的Cd含量呈现显著相关, 也可能有农药、肥料等其他重金属来源[23]。与其他坚果相比, 因花生的果实于土壤生长, 其检出率及检出含量也相对其他种类坚果较高[24]。本研究发现淮南市和滁州市采集的当地生产花生样品中Cr元素的污染指数较高。分别达到警戒线和中度污染水平, 表明这些地区生产的花生可能存在较高的Cr污染风险, 需引起重视。周启武等[25]也发现了云南省西双版纳州采集的红花生样品中存在Cr元素轻度污染(PCr=1.138)。此外, 芜湖市的花生样品中Cd元素的单因子污染指数为0.71, 处于警戒限, 表明芜湖市花生中Cd元素污染需给予关注, 结果与文献报道芜湖市蔬菜与粮食中Cd元素超标情况一致[26]
安徽省花生成人平均消费量为6.05 g/d, 与《第五次中国总膳食研究》中辽宁省花生平均消费量为6.06 g/d最接近[27]。受饮食习惯影响, 18~50岁男性花生消费量最大, 一方面此年龄段属于坚果摄入量最高人群[28], 与胡国党等[29]对北京市居民食用花生的年龄分布描述一致, 另一方面与成年男性喜爱以花生作为伴酒菜有关[30]。综上所述, 在进行健康风险评估时, 需要综合考虑人群差异, 以提高评估的准确性和针对性。
基于MOE与MOS的计算结果, 表明花生中的5种重金属暴露对安徽省居民健康的潜在风险较低, 而不同元素也存在风险大小的差异。黄发源等[31]调查表示安徽省部分地区土壤中镉超标现象较严重, 王晓等[32]发现花生中Cd是对人体的危害风险最大的重金属元素。本研究中Cd元素的MOS相对其他元素低, 因而安徽省花生中Cd的潜在健康风险也值得关注。另外, 18~50岁高端消费者存在Cr元素暴露风险较高的情况, 黄彪等[33]对福建省连城县种植花生的风险评估也出现此Cr元素健康风险较其他元素高的情况。而由于铬形态不同, 毒性不同, 对样品中的铬形态进一步测定以评估风险, 将更具科学性。本次未对儿童进行暴露风险评估, 更精确的评估工作需要进一步对人群类别进行划分。
本评估存在一定的不确定性: (1)大部分样品的Hg元素检测结果都小于LOD, 在评估时全部使用LOD代替将造成一定程度的风险高估。(2)评估过程多关注单一元素, 对不同元素累积暴露风险考虑欠缺。(3)依据3 d内的花生消费调查数据来评估人群的长期花生摄入量, 较短的时间范围难以代表全年不同季节的实际消费习惯, 如罗宝章等[34]调查发现上海市居民在夏季坚果、籽类食品摄入量较其他季节更低。
安徽省各流通环节的花生中重金属元素的污染情况总体呈良好态势, 重金属的膳食暴露量显著低于健康指导标准, 人群摄入适量坚果不会引起重金属蓄积导致的健康危害。但仍有必要开展针对消费者合理均衡饮食的科普工作, 重点关注高端消费人群的健康风险。因采样环节分析时发现农贸市场的花生样品中Cd元素污染指数较其他流通环节高, 还建议相关部门重点关注污染源调查工作, 助力我省农作物高质量种植和发展, 保障居民饮食安全与健康。
