Penicillin antibiotics are widely used in the field of animal and plant disease prevention and control. However, due to improper or even illegal use, there are residues of penicillin antibiotics in food. The molecular structure of penicillin is unstable and undergoes varying degrees of degradation under different conditions. The degradation products, such as penicilloic acid, are the primary contributors to its allergenicity and cytotoxicity. And once penicillin degrades, conventional detection technologies for it will not accurately reflect the actual situation. At present, the potential risks of degradation products of penicillin antibiotics in food safety have not been given sufficient attention both domestically and internationally. This article summarized the degradation processes of penicillin antibiotics under different conditions, summarized the specific applications of high performance liquid chromatography, liquid chromatography-tandem mass spectrometry, immunoassay and surface-enhanced Raman spectroscopy in the detection of degradation products of penicillin antibiotics. Additionally, this paper analyzed the applicability characteristics and limitations of these methods to raise awareness of food safety issues caused by degradation products of penicillin antibiotics and provide references for further research and development of related detection technologies.

青霉素类抗生素被广泛应用于动植物疾病防控领域, 但由于使用不规范甚至违规使用, 导致食品中青霉素的残留。青霉素的分子结构不稳定, 不同条件下会产生不同程度的降解, 其降解产物如青霉噻唑酸等是其致敏性和细胞毒性的主要因素。且一旦发生降解, 原来针对青霉素原药的检测技术将无法准确反映实际情况。目前, 国内外对于青霉素类抗生素降解产物在食品安全方面的潜在风险尚未给予足够重视。本文总结了青霉素类抗生素在不同条件下的降解过程; 重点综述了高效液相色谱法、液相色谱-串联质谱法、免疫分析法以及表面增强拉曼光谱法在青霉素类抗生素降解产物检测中的具体应用; 同时分析了各类方法所具备的适用性特征以及存在的局限性, 以期提高对青霉素类抗生素降解产物残留所引发的食品安全问题的重视, 并为相关检测技术的进一步研究与发展提供参考。

青霉素类抗生素被广泛应用于动植物疾病防控领域, 但由于使用不规范甚至违规使用, 导致食品中青霉素的残留。青霉素的分子结构不稳定, 不同条件下会产生不同程度的降解, 其降解产物如青霉噻唑酸等是其致敏性和细胞毒性的主要因素。且一旦发生降解, 原来针对青霉素原药的检测技术将无法准确反映实际情况。目前, 国内外对于青霉素类抗生素降解产物在食品安全方面的潜在风险尚未给予足够重视。本文总结了青霉素类抗生素在不同条件下的降解过程; 重点综述了高效液相色谱法、液相色谱-串联质谱法、免疫分析法以及表面增强拉曼光谱法在青霉素类抗生素降解产物检测中的具体应用; 同时分析了各类方法所具备的适用性特征以及存在的局限性, 以期提高对青霉素类抗生素降解产物残留所引发的食品安全问题的重视, 并为相关检测技术的进一步研究与发展提供参考。

Penicillin antibiotics are widely used in the field of animal and plant disease prevention and control. However, due to improper or even illegal use, there are residues of penicillin antibiotics in food. The molecular structure of penicillin is unstable and undergoes varying degrees of degradation under different conditions. The degradation products, such as penicilloic acid, are the primary contributors to its allergenicity and cytotoxicity. And once penicillin degrades, conventional detection technologies for it will not accurately reflect the actual situation. At present, the potential risks of degradation products of penicillin antibiotics in food safety have not been given sufficient attention both domestically and internationally. This article summarized the degradation processes of penicillin antibiotics under different conditions, summarized the specific applications of high performance liquid chromatography, liquid chromatography-tandem mass spectrometry, immunoassay and surface-enhanced Raman spectroscopy in the detection of degradation products of penicillin antibiotics. Additionally, this paper analyzed the applicability characteristics and limitations of these methods to raise awareness of food safety issues caused by degradation products of penicillin antibiotics and provide references for further research and development of related detection technologies.

青霉素类抗生素属于β-内酰胺类抗生素中的一种[1], 因其具有抗菌能力强、高效低毒、价格低廉等特点, 被广泛应用于动植物的疾病防控, 但由于使用不规范甚至违规使用, 导致青霉素类抗生素及其降解产物在食品中残留[2-3]。青霉素类抗生素的分子结构中均包含β-内酰胺环、噻唑环结构[4-6], 其中β-内酰胺环张力较大, 结构不稳定, 在不同的条件下易发生不同程度的降解, 生成青霉噻唑酸、青霉烯酸等降解产物。青霉素类抗生素易引起过敏反应已为大众所熟知, 但近年来研究表明, 青霉素类抗生素本身相对较为安全, 其致敏性、细胞毒性等危害主要是由其降解产物如青霉噻唑酸所引起[7-8]
由于对青霉素类抗生素降解产物的安全性认识不足, 目前报道的检测技术主要针对食品中青霉素类抗生素的残留, 而对青霉噻唑酸等降解产物的检测报道较少。现今国内外也仅规定了食品中青霉素类抗生素的最大残留限量标准[9-12], 而对其降解产物尚未有相关规定。然而, 青霉素类抗生素在食品中一旦发生降解, 原先针对青霉素原药的检测技术将无法准确反映实际情况。鉴于此, 有必要提高对青霉素类抗生素降解产物在食品中残留危害的重视度并建立其检测技术。
本文总结阐述了青霉素类抗生素在不同条件下的降解途径, 重点综述了国内外关于青霉素类抗生素降解产物的检测技术, 分析了每种方法的优缺点以及用于青霉素类抗生素降解产物残留检测的适用性, 以期提高对青霉素类抗生素降解产物的关注度, 以及为合理选择检测技术提供参考。
食品中残留的青霉素类抗生素降解产物主要来源于两方面, (1)动植物用药后, 一部分原药发生降解, 进而导致食品中降解产物的残留; (2)食品中残留的原药在加工、储藏过程中发生了不同程度的降解。
以青霉素G为例, 常温(25 ℃)下青霉素G遇强酸(pH<4.0)或氯化高汞会降解生成青霉噻唑酸和青霉醛酸, 后者不稳定, 会继续脱去羧基生成青霉醛[7,13]。而在37 ℃的酸性(pH 2.0~4.0)或氯化高汞溶液中, 青霉素G的降解情况有所不同, 其β-内酰胺环结构会重排生成不稳定的中间体化合物, 如青霉烯酸、青霉二酸和青霉噻唑酸等[14-16]; 青霉烯酸随后转化为青霉烷酸和青霉二酸, 青霉二酸和青霉噻唑酸进一步降解与脱羧反应, 生成稳定的代谢产物脱羧青霉噻唑酸[17]; 也有文献报道, 酸性环境下青霉二酸会继续分解, 产生青霉醛与青霉胺[18]。FADI等[19]通过树干注射法, 用青霉素G处理被柑橘黄龙病感染的柑橘树, 170 d后在其果实中检测出青霉噻唑酸和青霉二酸等降解产物, 未检测出青霉素G, 推测可能是由于果实自身的酸性环境(pH约为4.0)促使青霉素G发生了降解。碱性条件(pH 7.5~9.0)下, 碱性基团会攻击青霉素G的β-内酰胺环, 导致其迅速降解为青霉噻唑酸[20]; FADI等[19]的研究也发现青霉素G在强碱性条件(pH 12.0)下不稳定, 易降解为青霉噻唑酸、脱羧青霉噻唑酸等降解产物。高温条件下, 青霉素G容易开环生成青霉噻唑酸及脱羧青霉噻唑酸[21-23]。崔成[23]对青霉素G钠水溶液进行100 ℃热处理, 模拟青霉素G钠在动物源性食品中经过烹饪的环境, 发现10 min后基本无青霉素G钠检出, 其主要产物青霉噻唑酸在进一步加热后转化为脱羧青霉噻唑酸; 若此时遇到氯化高汞, 脱羧青霉噻唑酸将进一步分解成青霉胺与青霉醛。故若畜禽肉等动物源性食品中残留有青霉素类抗生素, 其煮沸后的汤汁便含有青霉噻唑酸、脱羧青霉噻唑酸等青霉素G的热解产物。在溶液中的Cu2+、Hg2+、Zn2+、Pb2+、Ni2+等离子的催化作用下, 青霉素G的β-内酰胺环被打开, 生成青霉噻唑酸以及脱羧青霉噻唑酸[24]。另外, 对硫原子有特殊亲和力的Cu2+、Hg2+、Zn2+等离子, 还会对青霉素G的噻唑环产生作用, 使其开环转化为最终的稳定产物[25]。比如, 在Hg2+或Cu2+的中性水溶液中时, 青霉素G降解为青霉烯酸等产物[26]; 再如, 在Zn2+的水溶液中, 青霉素G则会降解为青霉烷酸[23-24]。在β-内酰胺酶的作用下, 青霉素G会酶解开环生成无抗菌活性的青霉噻唑酸、6-氨基青霉烷酸等产物[27]。基于此, 一些不法分子向牛乳中加入β-内酰胺酶分解青霉素G, 人为制造“无抗奶”, 以逃避乳品厂对牛乳中残留青霉素类抗生素的检测[28-30], 从而导致一些乳制品中青霉噻唑酸等降解产物的残留。此外, 自然条件下的青霉素G也会有不同程度的降解。LEVINE[20]发现室温条件下的青霉素G在水溶液中容易发生分子重排, 生成青霉烯酸, 并进一步转化为青霉噻唑酸。周杰[9]通过实验也佐证了该结论, 实验发现在室温条件下, 青霉素G标准溶液的降解速率随其浓度的增加而增加, 100 mg/L的青霉素G在3 d内降解了接近30%, 主要降解产物为脱羧青霉噻唑酸和青霉二酸, 后者可进一步转化为脱羧青霉噻唑酸。图1整理了青霉素G在不同条件下的降解途径。
目前, 针对青霉素类抗生素降解产物的检测手段主要有: 高效液相色谱法(high performance liquid chromatography, HPLC)、液相色谱-串联质谱法(liquid chromatography-tandem mass spectrometry, LC-MS/MS)、免疫分析法(immunoassay, IA)、表面增强拉曼光谱法(surface-enhanced Raman spectroscopy, SERS)等。
HPLC依据混合物中各组分分子大小、结构性质不同, 在固定相与流动相间的分配系数不同而实现有效分离, 逐一进入检测器实现定性、定量检测, 适用于高沸点、热稳定性差有机物的分析检测[31-33]。但由于结构类似物和同分异构体的性质相似, 在色谱行为上不易分离, 导致无法准确定性定量, 通常需要采用更复杂的分离模式和方法[34-35]
范筱京等[36]采用冷冻离心脱脂、乙腈沉淀蛋白的样品前处理方法, 建立了牛奶中青霉噻唑酸残留检测的高效液相色谱-二极管阵列检测法, 检出限为5 mg/L, 回收率为93.0%~97.7%。高颀等[10]利用HPLC技术定量检测原料乳中青霉噻唑酸钾残留, 该方法采用紫外检测器有效提高了检测灵敏度, 其检出限(S/N=3)为0.2 mg/L, 方法回收率为89.7%~92.7%。该团队对6份盲样进行检测, 其中2份样品呈阳性, 青霉噻唑酸钾残留量分别为0.4 mg/L、0.5 mg/L。
LC-MS/MS技术, 结合了液相色谱高分离能力和质谱强结构鉴定能力, 具有高特异性、高准确性、高灵敏度等优点, 被广泛应用于食品安全检测、环境监测、疾病筛查、治疗药物监测等领域[37-41]
路杨等[42]采用高效液相色谱-串联三重四极杆质谱法(high performance liquid chromatography-triple quadrupole tandem mass spectrometry, HPLC-QQQ-MS)定量检测牛奶中4种青霉素类抗生素及其代谢产物脱羧噻唑酸。牛奶经0.2%氨水乙腈超声提取, 经固相萃取柱净化, 8种目标分析物的区分度良好, 采用基质匹配标准溶液校准法消除基质效应, 外标法定量, 对青霉素G脱羧噻唑酸、青霉素V脱羧噻唑酸、氨苄西林脱羧噻唑酸、阿莫西林脱羧噻唑酸的检出限分别为1.0、1.0、5.0、10.0 μg/kg。LI等[43]建立了一种HPLC-QQQ-MS法快速、高通量定量检测废水中青霉素、头孢菌素、喹诺酮类等23类共109种抗菌药物及其代谢产物, 方法采用内标法定量, 对阿莫西林降解产物(CAS 42947-63-7)、青霉酸V降解产物(CAS 1049-84-9)的检出限分别为0.030 μg/L、0.029 μg/L。刘红河等[44]建立了一种超高效液相色谱-四极杆串联线性离子阱质谱法(ultra performance liquid chromatography-hybrid triple quadrupole linear ion trap-mass spectrometry, UPLC-QqLIT-MS)定量测定乳制品中8种青霉素类抗生素及其相应的噻唑酸代谢产物。通过碱水解法制备每种抗生素对应的噻唑酸代谢产物标准品, 采用多反应监测方式定量, 8种青霉素类抗生素对应的噻唑酸代谢产物在固体乳粉中的检出限为0.01~0.04 μg/kg, 在液体奶中的检出限为0.002~0.008 μg/kg。实验通过增强子离子全扫描建立了8种青霉素类抗生素及其相应的噻唑酸代谢产物的碎片谱库, 采用信息依赖采集扫描功能结合增强子离子扫描模式精确定性, 在65份乳制品盲样中筛查出一份青霉噻唑酸阳性样品。
近年来迅速发展的液相色谱-串联高分辨质谱技术(liquid chromatography-high resolution mass spectrometry, LC-HRMS)具有高质量精确度、高分辨率、高灵敏度等优点。LC-HRMS采用一级母离子全扫描和自动触发的二级子离子扫描, 可采集待测物的一级母离子和二级子离子的高质量精度的质谱图谱(质量精确度小于5×10-6)可实现待测物的准确定性, 结合自建或商品化二级碎片离子谱图数据库的使用及数据分析, 可以实现复杂样品基质中已知目标物的靶向筛查和未知目标物的非靶向筛查, 在食品安全、环境监测、代谢组学等领域的多组分物质检测中具有巨大应用潜力[45-48]。鉴于青霉素类抗生素降解过程的复杂性、降解产物种类的丰富性及相应标准品的缺乏, LC-HRMS技术为相对全面、精确地识别与测定所有青霉素类抗生素降解产物提供了可能。赵凤娟等[49]对牛奶中羟氨苄青霉素、氨苄青霉素、邻氯青霉素、苄青霉素4种青霉素类抗生素经β-内酰胺酶水解后的未知降解产物进行定性分析, 采用高效液相色谱-四极杆/静电场轨道阱高分辨质谱法(high performance liquid chromatography coupled to quadrupole orbitrap high-resolution mass spectrometry, HPLC-Q-Orbitrap HRMS)获得降解产物的全扫描图谱和高分辨二级质谱信息, 根据碎片离子的精确质量数和同位素丰度等质谱信息, 结合对其可能酶解产物裂解机制的理论推测, 分析确定4种青霉素类抗生素的降解产物分别为相应的噻唑酸和脱羧噻唑酸, 对人工“无抗奶”的鉴别浓度低限为10 μg/kg。牛奶中抗生素残留是乳制品行业中一个严重的健康和技术问题, CHIESA等[50]采用HPLC-Q-Orbitrap HRMS结合Compound Discoverer™软件, 对生鲜乳中不同种类的抗生素(66种)及其非靶向代谢物进行筛查, 实际样品中筛查出代谢物青霉噻唑酸阳性。
对于小分子化合物IA方法的建立, 分析其三维结构特征及抗原决定簇, 设计并合成具有待测物结构特征的半抗原, 进而制备高质量特异性抗体是关键[51-53]。目前报道的IA方法主要是对青霉噻唑酸快速测定的酶联免疫吸附测定(enzyme linked immunosorbent assay, ELISA)和胶体金免疫层析技术(colloidal gold immunochromatography assay, GICA)等[54-55]
ZHANG等[54]鉴于青霉噻唑酸与其原药青霉素G结构的相似性, 直接以青霉素G作为半抗原, 采用活泼酯法将青霉素G分子中噻唑环上的羧基活化后与‌钥孔血蓝蛋白偶联制备针对青霉噻唑酸的人工免疫原, 通过动物免疫, 成功制备出了针对青霉噻唑酸的特异性抗体, 并建立了其直接竞争ELISA检测技术。方法与青霉素G存在弱交叉反应性(交叉反应率1.5%), 与其他青霉素类抗生素及其降解产物的无交叉反应。生牛乳、奶粉(先用水溶解)样品经高速离心脱脂, 用缓冲液稀释40倍、320倍消除基质效应进行ELISA检测, 方法对生牛乳、奶粉中青霉噻唑酸的检出限分别为1.2 μg/kg、9.6 μg/kg, 回收率在72.75%~93.25%之间, 与液相色谱-质谱法的测定结果相关性良好, 相关系数(r²)为0.9977。王莹[55]基于ZHANG等[54]制备的青霉噻唑酸特异性抗体, 建立了GICA方法与免疫亲和凝胶检测柱两种快速检测技术, 不借助仪器, 通过目视法实现了青霉噻唑酸的定性检测。GICA方法对牛奶、奶粉样品中青霉噻唑酸残留检出限分别为10 μg/L和10 μg/kg; 免疫亲和凝胶检测柱分析方法在牛奶中的检出限为10 μg/L。实验采用间接竞争ELISA实验验证了两种方法对青霉噻唑酸的检测结果准确性, 检测结果一致性良好。
SERS是利用特定纳米结构引发的局域等离子体共振效应增强拉曼散射信号的分析技术, 独特的“指纹”光谱特性使其具有准确、灵敏、快速和可实现多残留检测的特点。与上述分析方法相比, SERS的样品前处理简单, 或者不需要对样品进行前处理, 大大缩短了分析检测的时间, 简化了样品检测的过程, 同时得益于微流控、智能传感等技术的发展, SERS检测设备更加便携化、智能化, 使其在现场快速筛查中有极大应用前景[56-58]
张丽颖[59]针对青霉素药品残留的微量青霉噻唑酸在给药后易引起过敏反应的问题, 构建了核壳型金属溶胶和二维分子印记单层两种基底, 对青霉素药品中青霉噻唑酸的含量进行SERS检测。实验制备了核壳型Au@Ag纳米粒子, 因青霉噻唑酸分子具有两个不在同一平面的羧基, 可同时连接不同的Au@Ag纳米粒子, 引起粒子团聚形成“热点”增强SERS信号; 而青霉素分子仅有一个羧基难以形成“热点”效应, 但会与青霉噻唑酸竞争Au@Ag纳米粒子的活性位点形成抑制效应, 当提高Au@Ag纳米粒子的浓度时, 抑制作用降低, 对青霉噻唑酸在青霉素药品中的检出限可以低至0.1% (g/g)。同时, 他们将二维分子印记技术与拉曼光谱相结合, 设计并优化了一种二维分子印记单层SERS基底。将卤素离子引入到银镜反应制备高均一性和重现性Ag膜, 在此基础上将模板分子青霉噻唑酸对苯二胺复合物修饰在Ag膜的表面, 制备了对青霉噻唑酸具有高选择性的二维分子印记单层基底, 能够很好地降低青霉素分子对检测的干扰, 对青霉素类药品中青霉噻唑酸的检出限约为0.01% (g/g), 比采用Au@Ag纳米粒子作为基底时的灵敏度提高了10倍。El-ZAHRY等[60]采用银纳米离子作为基底, 监测了青霉素G在酸性和碱性条降解过程的SERS光谱变化, 建立了氨苄西林、青霉素G、羧苄西林和青霉噻唑酸的SERS检测技术。
当前研究已确切证实, 青霉噻唑酸具备致敏特性与细胞毒性。现阶段, 针对青霉素类抗生素降解产物的检测工作, 重点聚焦于青霉噻唑酸的检测分析, 而对于青霉二酸、青霉烯酸等其他降解产物的研究尚显不足, 其潜在危害也尚未得到清晰界定。此外, 明确所有降解产物的结构是实现对这些物质全面检测的关键前提。除青霉素G外, 目前针对其他青霉素类抗生素降解途径的研究相对匮乏。鉴于此, 有必要进一步加强对这一系列降解产物的关注与研究力度。
食品中青霉素类抗生素降解产物的残留问题呈现出迁移性、累积性及多样性等显著特征。在此背景下, 通过一次实验尽可能多地涵盖目标分析物已成为当前研究的发展趋势, 这一趋势不仅包括对降解产物的检测, 还涵盖了对原药含量的同步检测。由于部分青霉素类抗生素降解产物的结构相似度高, 使得其采用HPLC检测时完全分离难度较大, 给目标分析物的准确定性和定量提出挑战。另外HPLC对青霉噻唑酸的检测灵敏度较低, 对动物源性食品中青霉素类抗生素降解产物做痕量分析时, 需要结合离线或者在线富集手段, 以满足检测灵敏度需求。
HPLC-QQQ-MS采用的多反应监测模式具有优良的定量性能, UPLC-QqLIT-MS在HPLC-QQQ-MS准确定量的基础上, 可获得目标分析物高质量二级碎片离子, 进一步提高了定性的准确性。这两种方法适用于已知目标分析物的靶向筛查, 均需依赖标准品, 逐个优化目标分析物的子离子和质谱条件, 而大部分青霉素类抗生素代谢产物均无商品化标准品, 严重限制了方法的应用。LC-HRMS具有出色的质量精度、分辨率和灵敏度, 还可以弥补低效分辨质谱HPLC-QQQ-MS、UPLC-QqLIT-MS对标准品的依赖。后续可通过建立一个全面、准确的青霉素类抗生素降解产物数据库, 在标准品种类有限的情况下, 实现其定向及半定量筛查。但LC-HRMS设备昂贵、维护成本高, 对操作人员专业技术要求高, 一定程度上限制了其推广应用。IA操作简便、经济、灵敏度高, 但在实际应用一般最多能同时检测2~3种目标分析物, 对更多种目标分析物的同时检测存在一定难度。目前, SERS对青霉素类抗生素降解产物的检测, 仅停留于个别常见目标物, 未来应尝试覆盖更多目标分析物, 可以基于标准化检测流程建立青霉素类抗生素降解产物的SERS光谱数据库, 并结合化学计量学方法以实现在没有相应标准物质的情况下快速定性筛查。另外, 鉴于食品基质的复杂性, 必要的样品预处理步骤是保障检测准确性、灵敏度的必要条件, 如何找到一种适合大部分或者全部青霉素类抗生素降解产物的样品预处理方法, 并能与SERS技术联用也是一项重要工作[61-62]。综上所述, 检测工作者在选择检测技术时, 可综合考虑实验室条件、检测需求等选择合适的检测技术。
