Objective To evaluate the edible safety of HuangqiSangyeYuzhu granules. Methods The edible safety of HuangqiSangyeYuzhu granules was comprehensively evaluated by acute oral toxicity test, 3 genotoxicity tests, and 28 d oral toxicity test. Results The results of acute oral toxicity test showed no abnormality in the test animals, and the maximum tolerated dose of HuangqiSangyeYuzhu granules was greater than 30.0 g/kg·BW, which was practically non-toxic substance. The results of bacterial reverse mutation test, mammalian erythrocyte micronucleus test and spermatogonial chromosome aberration test were negative, indicating that the HuangqiSangyeYuzhu granules had no genotoxicity. During the 28 d oral toxicity test, the rats growed well, and no signs of toxicity were observed, and no adverse effects associated with HuangqiSangyeYuzhu Granules were seen in blood routine, blood biochemistry, and pathology indices, and the no observed adverse effect level was 10.0 g/kg·BW. Conclusion HuangqiSangyeYuzhu granules have edible safety within the dosage range of this experiment.

目的 评价黄芪桑叶玉竹颗粒的食用安全性。方法 通过小鼠急性经口毒性试验、3项遗传毒性试验、28 d经口毒性试验综合评估黄芪桑叶玉竹颗粒的食用安全性。结果 小鼠急性经口毒性试验结果显示受试动物未见异常, 且黄芪桑叶玉竹颗粒的最大耐受剂量大于30.0 g/kg·BW, 属实际无毒性物质。细菌回复突变试验、哺乳动物红细胞微核试验及精原细胞染色体畸变试验结果均为阴性, 表明黄芪桑叶玉竹颗粒无遗传毒性。28 d经口毒性试验期间大鼠生长状况良好, 未见中毒体征, 血常规、血生化及病理学等检查指标均未见与黄芪桑叶玉竹颗粒相关的不良影响, 未观察到有害作用剂量为10.0 g/kg·BW。结论 黄芪桑叶玉竹颗粒在本研究剂量范围内具有食用安全性。

目的 评价黄芪桑叶玉竹颗粒的食用安全性。方法 通过小鼠急性经口毒性试验、3项遗传毒性试验、28 d经口毒性试验综合评估黄芪桑叶玉竹颗粒的食用安全性。结果 小鼠急性经口毒性试验结果显示受试动物未见异常, 且黄芪桑叶玉竹颗粒的最大耐受剂量大于30.0 g/kg·BW, 属实际无毒性物质。细菌回复突变试验、哺乳动物红细胞微核试验及精原细胞染色体畸变试验结果均为阴性, 表明黄芪桑叶玉竹颗粒无遗传毒性。28 d经口毒性试验期间大鼠生长状况良好, 未见中毒体征, 血常规、血生化及病理学等检查指标均未见与黄芪桑叶玉竹颗粒相关的不良影响, 未观察到有害作用剂量为10.0 g/kg·BW。结论 黄芪桑叶玉竹颗粒在本研究剂量范围内具有食用安全性。

Objective To evaluate the edible safety of HuangqiSangyeYuzhu granules. Methods The edible safety of HuangqiSangyeYuzhu granules was comprehensively evaluated by acute oral toxicity test, 3 genotoxicity tests, and 28 d oral toxicity test. Results The results of acute oral toxicity test showed no abnormality in the test animals, and the maximum tolerated dose of HuangqiSangyeYuzhu granules was greater than 30.0 g/kg·BW, which was practically non-toxic substance. The results of bacterial reverse mutation test, mammalian erythrocyte micronucleus test and spermatogonial chromosome aberration test were negative, indicating that the HuangqiSangyeYuzhu granules had no genotoxicity. During the 28 d oral toxicity test, the rats growed well, and no signs of toxicity were observed, and no adverse effects associated with HuangqiSangyeYuzhu Granules were seen in blood routine, blood biochemistry, and pathology indices, and the no observed adverse effect level was 10.0 g/kg·BW. Conclusion HuangqiSangyeYuzhu granules have edible safety within the dosage range of this experiment.

糖尿病已经成为对人类健康构成严重威胁的主要慢性疾病, 这种疾病常导致严重的代谢疾病和并发症。近年来糖尿病患病率不断上升, 因此, 如何有效地预防和治疗糖尿病变得至关重要[1-2]。中医理念讲究药食同源, 相较于药物治疗, 适当使用药食同源的原料可以改善病症, 提高生存质量[3]。黄芪桑叶玉竹颗粒是一种结合中医“辨证保健”理论和营养学研究成果, 创新研制的功能性食品, 结合了黄芪、桑叶、葛根、玉竹、蜂胶等药食同源物质, 可通过协同作用将各药材的功效发挥到最佳, 达到益气养阴、清热止渴的效果。其中黄芪、桑叶为君药, 黄芪归肺、脾经, 具有健脾补气, 止汗利尿之功效, 可提高免疫力、抗氧化能力, 降血糖等[4-7]。桑叶归肺、肝经, 主功效为清肺润燥、平肝明目, 可通过多种途径发挥降血糖作用[8-11]。葛根、玉竹为臣药, 葛根归脾、胃、肺经, 玉竹归肺、胃经, 二者均有降血糖功效[12-15]。蜂胶为补药, 归脾、胃经, 可提升免疫力, 调节血糖[16-17]。以往研究表明这些中药成分具有较高的食用安全性, 冯文燕等[18]对甘肃道产黄芪的食用毒理学进行研究, 结果表明甘肃道产黄芪提取物无一般毒性及繁殖毒性, 具有较高的安全性。胡志航等[19]对黄芪进行安全性评价, 未见急性毒性、遗传毒性及亚慢性毒性, 未观察到有害作用剂量为8.0 g/kg·BW。LI等[20]通过急性、亚急性毒性和遗传毒性试验评估桑叶提取物的毒理学特征, 认为桑叶提取物是安全的, 支持其用作新型食品成分。詹键等[21]研究表明葛根食用安全, 不会引起基因突变。CHIVAPAT等[22]研究表明长期食用泰国葛根会影响血细胞产生。
尽管多项研究对黄芪桑叶玉竹颗粒中的这些中药成分进行了安全性评价, 但中药复方保健食品的安全性评价要考虑多方面因素[23]。某些中药材中会存在一些有害物质, 可能来源于其自身的化学成分, 也可能来自外源性物质, 例如残留的杀虫剂、包装材料、有害元素及真菌毒素污染等[24-26], 此外传统中药之间的配伍禁忌及产品的制作工艺均可能对中药复方保健食品的安全性产生影响。因此, 对于创新的中药复方保健食品需要进行安全性评价, 才能确保其在实际应用中的安全性, 为其在养生保健领域的推广提供可靠的试验数据支持。本研究通过急性经口毒性试验、3项遗传毒性试验及28 d经口毒性试验等一系列毒理学评价程序对黄芪桑叶玉竹颗粒进行安全性评价, 以期为保健食品黄芪桑叶玉竹颗粒的食用安全性提供试验依据。
黄芪桑叶玉竹颗粒(批号: 20220601, 山西一果健康科技有限公司)。
试验菌株TA100、TA1535, TA97a、TA98、TA102(美国MOLTOX公司); 2-氨基芴、环磷酰胺(分析纯, 美国Sigma-Aldrich公司); 大鼠肝微粒体酶(S9)溶液(北京安宝迪科技有限公司); 敌克松(分析纯, 美国AccuStandard公司); 叠氮钠(分析纯, 美国Amresco公司); 1,8-二羟基蒽醌(分析纯, 北京百灵威科技有限公司)。
ADVIA2120i血细胞分析仪(西门子医疗系统有限公司); TBA-120FR全自动生化仪(日本东芝株式会社); PUN-2048A半自动血凝分析仪(北京普朗新技术有限公司); URIT-150VET动物尿液分析仪(桂林优利特医疗电子有限公司); MEDICA EaysLyte-PLUS电解质分析仪(美国麦迪卡公司); ME203E/02电子天平(精度0.001 g, 瑞士梅特勒托利多公司); ST40R离心机(美国赛默飞世尔科技公司); CXL31RTSF显微镜(日本奥林巴斯株式会社); SVE-6A1垂直流超净工作台(新加坡艺思高科技有限公司)。
急性经口毒性试验和小鼠精原细胞染色体畸变试验所用SPF级癌症研究所小鼠(Institute of Cancer Research mouse, ICR)购自斯贝福(北京)生物技术有限公司, 许可证号: SCXK(京)2019-0010。哺乳动物红细胞微核试验所用SPF级ICR小鼠购自北京维通利华试验动物技术有限公司, 许可证号: SCXK(京)2021-0006。28 d经口毒性试验所用SPF级SD大鼠购自北京维通利华试验动物技术有限公司, 许可证号: SCXK(京)2021-0011。饲养环境设定温度为20~26 ℃, 相对湿度设定在40%~70%区间, 正常饮食, 预饲3~5 d。
选取SPF级小鼠20只, 体重18~22 g, 雌雄各半, 在给予受试物当天进行编号称重。试验开始前禁食4 h, 饮水自由。采用限量法, 通过一次性经口灌胃的方式给予小鼠供试液, 灌胃剂量为30.00 g/kg·BW, 灌胃后2 h恢复其正常饮食。连续14 d密切观察小鼠表现, 记录小鼠的中毒体征变化、消失情况以及死亡时间。试验周期结束后, 对所有小鼠进行解剖检查, 记录病理学变化。如存在明显解剖病理改变的小鼠, 进一步进行病理组织学观察。
将受试物终浓度分别设置为50、158、500、1581及5000 µg/皿, 依据GB 15193.4—2014《食品安全国家标准 细菌回复突变试验》进行细菌回复突变试验。将处理好的培养皿放置于37 ℃环境下孵育48 h, 然后记录每皿回复突变菌落数, 每个剂量组均设有3个平行的培养皿, 以提高结果的准确性和可重复性。初次试验结果显示为阴性, 采用5倍组距进行第二次验证, 试验剂量设定依次为5000.0、1000.0、200.0、40.0和8.0 µg/皿, 试验重复1次。
选用SPF级小鼠50只, 体重25~30 g, 在给予受试物当天进行编号称重。将小鼠随机分为5组, 3个剂量组、溶剂对照组和环磷酰胺阳性对照组, 每组10只, 雌雄各半。剂量组和溶剂对照组灌胃量为20 mL/kg·BW, 阳性对照组灌胃量为40 mg/kg·BW。分两次灌胃, 时间间隔为24 h。给予受试物结束后6 h, 将小鼠处死取股骨骨髓, 制备为涂片后用甲醇进行固定, Giemsa进行染色, 置于显微镜下阅片, 计算嗜多染红细胞数(polychromatic erythrocyte, PCE)所占比例及微核发生率。
选用SPF级雄性小鼠30只, 体重25~35 g, 在给予受试物当天进行编号称重。将小鼠随机分为5组, 3个剂量组、溶剂对照组和环磷酰胺阳性对照组, 其中高剂量组10只, 其余每组5只。剂量组和溶剂对照组灌胃量为20 mL/kg·BW, 阳性对照组灌胃量为40 mg/kg·BW。高剂量组小鼠在给予受试物结束后的24 h和48 h处死进行采样, 其余小鼠统一在24 h处死采样。处死前4小时给小鼠腹腔注射秋水仙素(5 mg/kg·BW)。处死小鼠取两侧睾丸, 按照GB 15193.8—2014《食品安全国家标准 小鼠精原细胞或精母细胞染色体畸变试验》制作精原细胞Giemsa染色标本后, 显微镜下观察染色体改变。
选用SD大鼠80只, 50~100 g, 在给予受试物当天进行编号称重。将大鼠随机分为4组, 3个剂量组和溶剂对照组, 每组20只, 雌雄各半。受试物规格及食用方法为3 g/袋, 每日2次, 每次1袋, 合人体推荐剂量为0.1 g/kg·BW。采用经口灌胃的方式进行, 试验分别设置2.5、5.0、10.0 g/kg·BW作为低、中、高剂量组, 分别相当于人体推荐摄入量的25倍、50倍、100倍。连续28 d在同一时间段进行灌胃, 并记录其体重摄食。试验结束时, 于腹主动脉采集血液, 测定其血液学及血液生化指标, 并选取关键组织进行病理组织学检查。
本研究数据采用Excel 2016进行整理, 统计学处理均采用SPSS 24进行分析。除微核发生率和精原细胞畸变率使用χ2检验外, 其他指标均使用单因素方差分析。结果以平均值±标准偏差进行表示, P<0.05, 差异显著(*), P<0.01, 差异极显著(**)。
急性毒性试验是安全性评价的必要指标, 经过14 d的连续观察, 小鼠的粪便形态、体毛状况及活动行为均未发现异常变化。同时, 体重增长趋势保持正常, 未显现任何与受试物直接相关的毒性症状或死亡案例, 解剖检查各组织均未发现异常病理改变。结果表明该受试物对小鼠的急性口服毒性耐受剂量均大于30.00 g/kg·BW, 属无毒性物质。
细菌回复突变试验通过受试物诱导组氨酸营养缺陷型菌株恢复合成组氨酸能力的突变频率, 评估物质的致突变性[27-28]。本研究中各剂量组平皿中菌苔生长状况无异常变化, 观察发现所有受试物剂量组测试菌株的回变菌落数量均低于对照组菌落数的2倍, 且不同剂量组之间, 并未观察到明确的剂量与反应之间的相关性。无论在有或无S9代谢活化系统的条件下, 均未见受试物对细菌有致突变作用, 提示受试物在该试验条件下不会引起受试菌株的基因突变。
哺乳动物红细胞微核试验是评价遗传毒性的重要工具[29], 通过检测受试物诱导的红细胞微核率, 可以反映染色体断裂或纺锤体功能损伤等遗传损伤效应[30-31]。由表1可知, 受试物各剂量组的PCE百分比均属于正常范围, 并且大于溶剂对照组的20%, 表明受试物在试验剂量范围内未对红细胞的生成产生影响。阳性对照微核率与溶剂对照组相比差异具有统计学意义(P<0.01), 阳性对照成立。受试物各剂量组的微核率与溶剂对照组相比不具有统计学意义(P>0.05), 表明受试物对小鼠骨髓细胞未产生毒性作用, 未影响其微核发生率, 未引起遗传损伤效应。
小鼠精原细胞染色体畸变试验可以通过精原细胞有丝分裂中期染色体结构异常情况直接反映受试物对雄性生殖系细胞的致突变性。本研究精原细胞染色体畸变的统计结果见表2, 阳性对照组的细胞畸变率与溶剂对照组之间相比差异具有统计学意义(P<0.01), 表明阳性对照有效。进一步对比各受试物剂量组与溶剂对照组之间的关系, 结果显示小鼠精原细胞的染色体畸变数量、类型以及发生畸变的细胞比例差异不具有统计学意义(P>0.05)。此外, 不同剂量组之间也并未观察到明显的剂量-反应关系, 表明该受试物不会诱导小鼠精原细胞染色体产生畸变。
连续28 d经口灌胃SD大鼠受试物后, 大鼠生长状况无异常表现, 各剂量组动物体重、摄食量等情况如表3所示。受试物各剂量组与对照组相比差异不具有统计学意义(P>0.05), 体重呈增长状态, 摄食量与食物利用率均表现正常, 表明受试物未对大鼠体重及摄食量等产生不良影响。
血常规和血生化检测是评估受试物对造血系统和全身毒性反应的重要指标, 二者互为补充, 共同揭示受试物对造血系统、器官功能及代谢平衡的影响。血常规检测指标结果如表4所示, 与对照组相比, 雌、雄性动物各剂量组差异均无统计学意义(P>0.05), 表明受试物对血常规指标无显著影响。血液生化学检测指标结果如表5所示, 其中雌性动物中、低剂量组谷草转氨酶与对照组相比显著降低, 中剂量组血糖与对照组相比显著升高。雄性动物中剂量组谷草转氨酶、尿素氮、肌酐以及低剂量组肌酐均显著降低, 差异具有统计学意义(P<0.01, P<0.05), 但这些指标的变化均无剂量-反应关系, 不具备生物学意义。此外, 谷草转氨酶常被用于评价肝细胞损伤情况, 给予受试物后, 谷草转氨酶呈现下降趋势, 提示受试物黄芪桑叶玉竹颗粒可能对肝脏具有一定保护能力。其他各指标与对照组相比差异均不具有统计学意义(P>0.05), 表明受试物对血液生化学指标无显著影响。
大鼠大体解剖观察结果表明各剂量组的主要器官均未显示异常。组织病理学检查发现, 个别动物出现轻微空泡变性, 肝细胞索间隙产生轻微的灶性单核细胞浸润, 但并未伴随继发性坏死, 如图1所示。但这些改变均属于动物本身的自发性病理过程, 与试验受试物无直接关联。排除上述轻微且自发的病理变化后, 无论是对照组还是高剂量组的其余雌性、雄性大鼠, 其主要脏器组织均保持了正常的组织学结构, 未显示出与受试物相关的病理学改变, 表明受试物对大鼠主要脏器未造成显著毒性作用。综合分析28 d经口毒性试验全部检测指标, 受试物未观察到有害作用剂量为10.0 g/kg·BW。
中药复方保健食品的研究近年来在慢性病防治和亚健康调理领域备受关注, 其中黄芪、桑叶、葛根、玉竹等药食同源中药因其多靶点、低毒性的等特点成为研究热点。这些药材的协同作用机制及其在代谢综合征、免疫调节等方面的应用进展, 体现了传统中医药理论与现代科学技术的深度融合。黄芪桑叶玉竹颗粒是由黄芪、桑叶、葛根、玉竹和蜂胶为主要原料制备而成一种保健食品, 其依据中医辨证理论, 以黄芪、桑叶为君药, 葛根、玉竹为臣药, 蜂胶为补, 相辅相成, 可以使它们的功效及药理活性得到更好的发挥。本研究对黄芪桑叶玉竹颗粒进行了毒理学安全性研究, 急性毒性试验结果显示黄芪桑叶玉竹颗粒的半数致死剂量大于30.00 g/kg·BW, 属实际无毒级物质。细菌回复突变、红细胞微核及精原细胞染色体畸变试验结果均显示阴性, 表明黄芪桑叶玉竹颗粒无明显遗传毒性。28 d经口毒性试验期间各剂量组大鼠生长状况良好, 未见中毒体征, 各项检测指标均未见与黄芪桑叶玉竹颗粒相关的不良影响。综上所述, 黄芪桑叶玉竹颗粒在本研究条件下未见明显的毒性作用, 未观察到有害作用剂量为10.0 g/kg·BW。这一研究结果表明黄芪桑叶玉竹颗粒符合保健食品“药食同源”的理念, 为其作为保健食品的进一步开发及长期食用的安全性提供试验依据, 为糖尿病前期或早期患者提供非药物干预选择, 对于老年人群或血糖波动轻微需要长期调理的亚健康人群可作为日常保健补充。但现有研究结果仅验证了黄芪桑叶玉竹颗粒在本研究条件下的短期安全性, 多年连续服用带来的潜在影响及不同个体之间存在的响应差异仍需追踪研究。
