Objective To evaluate the toxicological safety of xylooligosaccharides. Methods According to the national food safety standards, the safety of xylooligosaccharides was evaluated through acute oral toxicity test in mice, micronucleus tests in mammalian bone marrow red blood cells, chromosomal aberration tests in spermatocytes, Bacterial reverse mutation (Ames) test, and 28 day oral toxicity test. Results The acute maximum tolerated dose (MTD) of xylooligosaccharides in male and female mice was greater than 10000 mg/(kg·bw), which was classified as practically non-toxic; the micronucleus test of mammalian bone marrow red blood cells, chromosome aberration test of spermatocytes, and Ames test were all negative; set up 3 dose groups: 0 (basic feed control), 4167, 8333 and 16667 mg/(kg·bw) (equivalent to 25 times, 50 times and 100 times the maximum recommended intake), orally administered once a day for 28 consecutive days. The high-dose group of animals showed a decrease in total weight gain related to the test sample, changes in biochemical indicators such as alanine aminotransferase, alkaline phosphatase, glucose and urea nitrogen, and a decrease in organ weight and visceral to body ratio in male rats. However, there were no corresponding changes observed in the medium and low-dose groups (equivalent to 50 times and 25 times the maximum recommended intake) and the Weixing group. Conclusion Xylooligosaccharides do not show acute toxicity or genetic toxicity; the no observed adverse effect level (NOAEL) of the sample for 28 days of oral toxicity in female and male rats is 8333 mg/(kg·bw).

目的 评价低聚木糖的毒理学安全性。方法 依据食品安全国家标准, 通过小鼠急性经口毒性试验、哺乳动物骨髓红细胞微核试验、精母细胞染色体畸变试验、细菌回复突变(Ames)试验、28 d经口毒性试验对其进行安全性评价。结果 低聚木糖对雌雄小鼠急性经口最大耐受剂量(maximum tolerated dose, MTD)均大于10000 mg/(kg·bw), 属实际无毒级; 哺乳动物骨髓红细胞微核试验、精母细胞染色体畸变试验和细菌回复突变试验试验均为阴性; 设0(基础饲料对照)、4167、8333、16667 mg/(kg·bw) 3个剂量组(相当于最大推荐摄入量的25倍、50倍和100倍), 每日经口灌胃一次, 连续28 d。高剂量组动物出现与受试样品相关的总增重降低, 丙氨酸氨基转移酶、碱性磷酸酶、血糖及尿素氮等生化指标改变, 雄性大鼠部分脏器重量及脏体比减少等变化, 而中、低剂量组(相当于最大推荐摄入量的50倍和25倍)以及卫星组未见相应变化。结论 低聚木糖未显示急性毒性、遗传毒性; 该样品对雌、雄大鼠28 d经口毒性未观察到有害作用剂量(no observed adverse effect level, NOAEL)为8333 mg/(kg·bw)。

目的 评价低聚木糖的毒理学安全性。方法 依据食品安全国家标准, 通过小鼠急性经口毒性试验、哺乳动物骨髓红细胞微核试验、精母细胞染色体畸变试验、细菌回复突变(Ames)试验、28 d经口毒性试验对其进行安全性评价。结果 低聚木糖对雌雄小鼠急性经口最大耐受剂量(maximum tolerated dose, MTD)均大于10000 mg/(kg·bw), 属实际无毒级; 哺乳动物骨髓红细胞微核试验、精母细胞染色体畸变试验和细菌回复突变试验试验均为阴性; 设0(基础饲料对照)、4167、8333、16667 mg/(kg·bw) 3个剂量组(相当于最大推荐摄入量的25倍、50倍和100倍), 每日经口灌胃一次, 连续28 d。高剂量组动物出现与受试样品相关的总增重降低, 丙氨酸氨基转移酶、碱性磷酸酶、血糖及尿素氮等生化指标改变, 雄性大鼠部分脏器重量及脏体比减少等变化, 而中、低剂量组(相当于最大推荐摄入量的50倍和25倍)以及卫星组未见相应变化。结论 低聚木糖未显示急性毒性、遗传毒性; 该样品对雌、雄大鼠28 d经口毒性未观察到有害作用剂量(no observed adverse effect level, NOAEL)为8333 mg/(kg·bw)。

Objective To evaluate the toxicological safety of xylooligosaccharides. Methods According to the national food safety standards, the safety of xylooligosaccharides was evaluated through acute oral toxicity test in mice, micronucleus tests in mammalian bone marrow red blood cells, chromosomal aberration tests in spermatocytes, Bacterial reverse mutation (Ames) test, and 28 day oral toxicity test. Results The acute maximum tolerated dose (MTD) of xylooligosaccharides in male and female mice was greater than 10000 mg/(kg·bw), which was classified as practically non-toxic; the micronucleus test of mammalian bone marrow red blood cells, chromosome aberration test of spermatocytes, and Ames test were all negative; set up 3 dose groups: 0 (basic feed control), 4167, 8333 and 16667 mg/(kg·bw) (equivalent to 25 times, 50 times and 100 times the maximum recommended intake), orally administered once a day for 28 consecutive days. The high-dose group of animals showed a decrease in total weight gain related to the test sample, changes in biochemical indicators such as alanine aminotransferase, alkaline phosphatase, glucose and urea nitrogen, and a decrease in organ weight and visceral to body ratio in male rats. However, there were no corresponding changes observed in the medium and low-dose groups (equivalent to 50 times and 25 times the maximum recommended intake) and the Weixing group. Conclusion Xylooligosaccharides do not show acute toxicity or genetic toxicity; the no observed adverse effect level (NOAEL) of the sample for 28 days of oral toxicity in female and male rats is 8333 mg/(kg·bw).

低聚木糖, 是指以玉米秸秆(玉米芯)或小麦秸秆为原料, 通过高压蒸煮法或者蒸汽爆破法[1-3], 经木聚糖酶酶解生产而成, 主要成分为木二糖~木七糖[4-6]。低聚木糖作为一种天然来源的益生元, 近年来因其独特的生理活性和潜在的健康益处, 在食品工业及保健食品领域受到了广泛关注[7-8]。低聚木糖在合适的剂量下不仅能够促进肠道有益菌群生长、改善肠道环境、增强营养吸收[9-11], 还具有调节免疫、血糖(glucose, GLU)以及抗氧化等多种生物活性, 是一类具有广泛应用前景的功能食品添加剂[12-15]。随着人们对益生元功能需求的增加, 更高剂量的低聚木糖在特殊人群或特定疾病中的应用潜力逐渐受到关注。更高的剂量可以带来更强的功能效应, 但其安全性和潜在风险尚未得到充分研究, 尤其是在长期使用的情况下。
目前, 关于低聚木糖的大剂量研究仍较为有限, 且缺乏系统的剂量-效应关系和安全性评估数据。已有少数研究表明, 过量摄入低聚木糖可能导致胃肠道不适(如腹胀、腹泻)以及代谢异常, 但其对器官功能(如肝脏、肾脏)的潜在影响尚未明确[16]。此外, 大剂量低聚木糖是否会对动物或人体的生长发育、代谢平衡及免疫系统产生负面影响, 仍需进一步探索。因此, 本研究按照GB 15193—2014《食品安全国家标准 食品安全性毒理学评价程序与方法》, 通过小鼠急性经口毒性试验、哺乳动物骨髓红细胞(red blood cell, RBC)微核试验、精母细胞染色体畸变试验、Ames试验和28 d经口毒性试验对低聚木糖进行毒理学安全性评价[16-22]。系统研究不同剂量低聚木糖的安全性对于指导其合理应用、制定安全剂量标准以及开发新型功能性食品具有重要意义。
低聚木糖, 由河南益常青生物技术有限公司委托检验。样品推荐食用量: 小于等于6.0 g/d(以低聚木糖计)。根据委托方提供的检验报告, 样品呈黏稠状透明液体, 干物质(固形物)含量为73.5%, 期中低聚木糖含量81.5%(以干基计), 低聚木糖2-4含量71.5%(以干基计)。折算成送检样品中低聚木糖含量为60%, 送检样品的人体每日推荐摄入量: 小于等于10.0 g/d, 成人体重按60 kg计, 即样品每日最大推荐摄入量为167 mg/(kg·bw)。
健康SPF级SD大鼠, 浙江维通利华试验动物技术有限公司提供, 生产许可证号: SCXK(浙)2019-0001号。健康SPF级ICR小鼠, 南京医科大学提供, 生产许可证号: SCXK(苏)2016-0002号。饲料和灭菌水自由食用。
MS204TS型电子天平[精度1 mg, 梅特勒-托利多仪器(上海)有限公司]; 蔡司AXIOSTAR生物显微镜[蔡司光学仪器(上海)国际贸易有限公司]; LRH-400型生化培养箱(韶关市泰宏医疗器械有限公司); Scan 500型尿分析仪(德国科宝公司); AU640型全自动生化分析仪[日本奥林巴斯(OLYMPUS)公司]; ADVIA2120型血细胞分析仪(德国SIEMENS公司); Coatron1800型全自动血凝仪[德国德高(TECO)公司)]; HISTECHPANNORAMIC MIDI数字病理扫描分析仪(匈牙利3DHISTECH有限公司); RM 2245型半自动石蜡切片机[德国徕卡(LEICA)公司]。
生化试剂盒(美康生物科技股份有限公司); 血细胞仪试剂(德国西门子公司); 凝血检测试剂盒[德国德高(TECO)公司)]; 科宝11SYS尿分析试纸条(德国科宝公司); 秋水仙素(纯度≥98%, 国药集团化学试剂有限公司); Ames试验相关菌株(美国Moltox公司); S9(齐氏生物科技有限公司); 环磷酰胺(纯度≥97.0%, 美国Sigma公司); 丝裂霉素(浙江海正药业股份有限公司)。
选用健康SPF级ICR小鼠20只(雌雄各半), 体重18.1~21.4 g。限量法, 称取样品16700 mg加纯净水至20 mL混合均匀后作为受试物, 剂量16700 mg/(kg·bw)。小鼠禁食6 h后, 受试物按20 mL/(kg·bw)一次灌胃, 观察期14 d。记录中毒表现及死亡情况。
选用健康SPF级ICR小鼠50只(雌雄各半), 按性别体重随机分为5组, 每组10只, 分别为溶剂对照组(纯净水)、16700、8350、4175 mg/(kg·bw) 3个剂量组及阳性对照组[环磷酰胺40 mg/(kg·bw)]。30 h内两次灌胃, 灌胃量为20 mL/kg, 末次灌胃6 h后处死小鼠, 取股骨骨髓制片染色, 每鼠镜检2000个嗜多染红细胞(polychromatic erythrocytes, PCE), 计数含微核PCE数, 同时观察200个PCE并计数正染红细胞(normochromatic erythrocytes, NCE), 计算PCE/总RBC的比值。
选用健康雄性SPF级ICR小鼠25只。按体重随机分5组, 每组5只, 分别为溶剂对照组(纯净水)、16700、8350、4175 mg/(kg·bw) 3个剂量组、阳性对照组(丝裂霉素2 mg/kg)。阳性组动物仅于试验首日腹腔注射1次, 注射量为10 mL/kg, 其余各组以20 mL/kg灌胃容量连续灌胃5 d, 每日1次。试验第14 d, 腹腔注射秋水仙素4 mg/kg, 注射量为10 mL/kg。4 h后处死动物, 分离曲细精管, 低渗后固定两次, 用60%冰乙酸软化后, 制片, 阅片。
采用平板掺入法, 使用TA97a、TA98、TA100、TA102和TA1535菌株, S9为β-萘黄酮和苯巴比妥诱导的大鼠肝匀浆(10%浓度)。称取样品500 mg定容至10 mL, 121 ℃灭菌20 min作为最高剂量(5000 μg/皿)。设5个剂量组: 第一次试验5倍稀释(1000、200、40、8 μg/皿), 第二次试验4倍稀释(1250、312.5、78.1、19.5 μg/皿)。另设2个溶剂对照组[纯水、二甲基亚砜(dimethyl sulfoxide, DMSO)]、1个自发回变组, 2个阳性对照组。每测试点3个平行样, 相同条件重复两次。
选用雌、雄各半的健康SPF级SD大鼠100只, 体重77~98 g。分性别按体重分层随机分入, 主试验每组10只, 卫星组中对照组和高剂量组每组5只。样品的人体每日最大推荐摄入量为167 mg/(kg·bw), 设0(基础饲料对照)和4167、8333、16667 mg/(kg·bw) 3个样品剂量组(相当于最大推荐摄入量的25倍、50倍和100倍), 分别称取样品41.67、83.33和166.67 g, 加入纯净水至150 mL, 混合均匀作为受试物。每日经口灌胃一次, 灌胃量为15 mL/(kg·bw)。连续28 d。每周称重1次, 记录摄食量2次, 调整喂食量并计算食物利用率, 计算各组体重增长量、总摄食量、总食物利用率。28 d后对主试验组动物进行各项指标检测, 卫星组动物不再给予受试物, 继续观察14 d后进行各项指标检测。尿液指标, 包括尿糖(glucose, GLU)、蛋白(protein, PRO)、相对密度(specific gravity, SG)、pH和潜血(erythrocyte, ERY)等。血液学指标, 包括凝血酶原时间(prothrombin time, PT)、活化部分凝血活酶时间(activated partial thromboplastin time, APTT)、RBC、白细胞(white blood cell, WBC)、血小板(platelet, PLT)、血红蛋白(hemoglobin, HGB)、红细胞压积(hematocrit, HCT)、中性粒细胞(neutrophils, NE)、淋巴细胞(lymphocyte, LY)、单核细胞(monocyte, MO)、嗜碱性粒细胞(basophil granulocyte, BA)、嗜酸性粒细胞(eosinophils, EO)。血清生化指标, 包括丙氨酸氨基转移酶(alanine aminotransferase, ALT)、碱性磷酸酶(alkaline phosphatase, ALP)、γ-谷氨酰转肽酶(γ-gamma-glutamyl transferase, γ-GT)、天门冬氨酸氨基转移酶(aspartate aminotransferase, AST)、总蛋白(total protein, TP)、白蛋白(albumin, ALB)、GLU、尿素氮(urea nitrogen, URE)、肌酐(creatinine, CRE)、总胆固醇(total cholesterol, T-CH)、甘油三酯(triglyceride, TG)、氯(Cl-)、钾(K+)、钠(Na+)。组织病理学检查, 包括脑、胸腺、甲状腺、心脏、肝、脾、肾、肾上腺、胃、十二指肠、结肠、肠系膜淋巴结、胰、睾丸、卵巢、膀胱。
试验数据以均值±标准偏差表示。骨髓RBC微核试验、精母细胞染色体畸变试验结果均采用卡方检验分析。28 d经口毒性试验中, 用SPSS 19.0软件对计量资料进行单因素方差分析或独立样本t检验; 若方差不齐, 先进行变量转换, 仍不齐则改用秩和检验; 用秩和检验对等级资料进行分析。
灌胃后小鼠未见明显中毒症状, 无死亡。观察期结束大体解剖未见明显异常。表1显示, 样品对雌雄小鼠的急性经口MTD均>16700 mg/(kg·bw)(推荐摄入量的100倍), 根据急性经口毒性分级标准, 属实际无毒级。
表2显示, 阳性对照组微核细胞率显著高于溶剂对照组(P<0.01)。其他各组微核细胞率与溶剂对照组无显著差异(P>0.05), 且无剂量-反应关系, PCE比例(PCE/RBC总数)均大于20%, 表明骨髓RBC增殖未受明显抑制, 低聚木糖哺乳动物RBC微核试验结果为阴性。
表3显示, 阳性对照组的染色体畸变率均显著高于溶剂对照组(P<0.01)。样品各剂量组动物初级精母细胞染色体畸变率与溶剂对照组无显著差异(P>0.05)。该剂量下, 样品显示对小鼠初级精母细胞的无致染色体畸变作用。
表4显示, 剂量达5000 μg/皿时, 加或不加S9条件下, 样品对TA97a、TA98、TA100、TA102和TA1535标准菌株均未显示明显诱变活性。
试验期间每日观察, 动物外观、排便、进食及活动均无异常, 未出现中毒体征或死亡。
表5显示, 主试验各剂量组雌雄大鼠各观察点体重与对照组无显著差异(P>0.05), 但是总增重均低于对照组, 其中雄性高剂量组总增重显著低于对照组(P<0.05, P=0.035)。表6显示, 恢复期卫星组中, 高剂量组雌雄大鼠各观察点体重与对照组无显著差异(P>0.05), 雌雄高剂量组42 d总增重低于对照组, 但差异不显著(P>0.05)。
表78显示, 雌雄大鼠各观察点及总摄食量、食物利用率与对照组无显著差异(P>0.05)。表910显示, 恢复期卫星组中, 高剂量组雌性大鼠第3、4周摄食量及雄性大鼠第1周摄食量偏低(P<0.05, P分别为0.037, 0.045, 0.047); 雄性大鼠第2周食物利用率偏高, 雌雄大鼠第4周食物利用率偏低, 差异显著(P<0.05, P分别为0.038, 0.036, 0.026)。其余观察点及42 d总摄食量、食物利用率与对照组无显著差异(P>0.05)。
表11显示, 与对照组相比, 雌性高剂量组WBC偏低, 中剂量组WBC、RBC、HGB、HCT偏低; 雄性低剂量组PLT偏低、LY偏高, 差异显著(P<0.05, P<0.01), 但均在试验室历史正常值范围内, 且无剂量-反应关系; 雄性中、高剂量组PT偏高, 差异显著(P<0.05, P<0.01), 但仍在正常范围内, 且雌雄变化趋势不一致, 结合其他指标分析, 认为无生物学及毒理学意义。其余血液学指标与对照组无显著差异(P>0.05)。
表12显示, 恢复期卫星组中, 雄性高剂量组RBC、HGB、HCT偏低, 差异显著(P<0.01), 但均在正常指范围内。其余血液学指标与对照组无显著差异(P>0.05)。
表13显示, 与对照组相比, 雌雄大鼠高剂量组ALT、ALP、GLU升高, URE降低, 雌性中、低剂量组GLU偏高, 差异显著(P<0.01, P均为0); 雌中剂量组URE偏低, 差异显著(P<0.05, P=0.013)。雌雄大鼠生化指标变化趋势一致, 提示高剂量样品可能引起肝脏轻度损伤及糖代谢变化。雌雄低剂量组AST偏低, 雌性中、低剂量组γ-GT、CRE偏低, 差异显著(P<0.01, P<0.05), 但无生物学及毒理学意义。其余生化指标及电解质浓度无显著差异(P>0.05)。
表14显示, 恢复期卫星组中, 高剂量组雌性大鼠γ-GT偏高, 雄性大鼠CRE偏低, 差异显著(P<0.05, P<0.01), 但均在试验室历史正常值范围内。高剂量组雌雄大鼠其余生化指标及电解质浓度与对照组无显著差异(P>0.05)。
表15显示, 各剂量组大鼠尿液无明显异常, 雌雄大鼠尿液检测指标与对照组无显著差异(P>0.05)。表16显示, 恢复期卫星组中, 大鼠尿液外观无明显异, 高剂量组雌雄大鼠尿液检测指标与对照组无显著差异(P>0.05)。
表17显示, 高剂量组雌雄大鼠心脏、肾脏湿重偏小, 其中雄性高剂量组肾脏和心脏湿重及心脏/体重比显著低于对照组(P<0.05, P<0.01, P分别为0.045, 0.005, 0.009)。其余剂量组脏器重量及脏体比与对照组无显著差异(P>0.05)。表1819显示, 恢复期卫星组中, 雌性高剂量组脑湿重及雄性高剂量组心脏湿重仍偏小, 差异显著(P<0.05, P分别为0.046, 0.025), 其余各剂量组的脏器重量及脏体比与对照组无显著差异(P>0.05)。
试验结束时对所有动物进行大体解剖检查, 均未发现明显异常, 先对高剂量组和对照组进行组织病理学检查。与对照组相比, 高剂量组大鼠脏器未发现与低聚木糖相关的特异性病理改变; 仅高剂量组1例雌鼠肝、脾出现局部灶状坏死, 加做中剂量组、卫星对照组和卫星高剂量组肝、脾切片, 均未发现类似不可逆病变。
在本研究条件下, 低聚木糖小鼠急性经口MTD值均大于10000 mg/(kg·bw), 属于实际无毒级。哺乳动物骨髓RBC微核试验、Ames试验、精母细胞染色体畸变试验结果均为阴性, 显示样品无遗传毒性。28 d经口毒性试验高剂量组动物出现总增重降低, ALT、ALP、GLU及URE等生化指标改变, 雄性大鼠部分脏器重量及脏体比减少等变化。故该样品对雌、雄大鼠28 d经口毒性NOAEL值取中剂量值, 为8333 mg/(kg·bw) (相当于最大推荐摄入量的50倍)。该样品低聚木糖含量为60%, 成人体重按60 kg计, 动物毒性试验结果外推到人的安全系数取100, 所以该样品推荐食用量应小于等于3.0 g/d(以低聚木糖计), 与国家卫生健康委2014年发布的第20号文规定低聚木糖食用量为小于等于3.0 g/d(以木二糖-木七糖计)相吻合。同时也与欧洲食品安全局(European Food Safety Authority, EFSA)所规定成人每天食用量1.9~3.7 g范围相吻合[23]。而本次试验样品推荐食用量: 小于等于6.0 g/d(以低聚木糖计), 远超出国家卫生健康委与欧盟规定的标准。相对应的就是本次动物试验的高剂量, 试验结果显示高剂量出现安全性问题。
低聚木糖作为一种天然来源益生元可以明显促进双歧杆菌和乳杆菌的增殖[24-26], 具有润肠通便和改善免疫功能等作用[27-30]。随着低聚木糖剂量的增加, 其对肠道菌群的调节作用增强, 可以在更短的时间内改善患者症状, 缩短某些与肠道健康相关疾病的病程。在人群调查中发现, 适量增加低聚木糖的摄入可以更快的通便, 解除便秘症状。然而, 任何功能性成分的使用必须考虑安全性因素, 要实现功能性与安全性之间的平衡, 需要探寻出合适的剂量。低聚木糖作为一种有益的膳食补充剂, 其使用需谨慎权衡剂量与功能之间的关系。本研究中高剂量的低聚木糖可能对大鼠的消化系统产生了不良影响, 营养缺乏影响了大鼠的生长发育, 导致其增重下降、生化指标的异常, 而中、低剂量组均未出现异常。未来, 随着研究的深入, 希望找到更加精准的摄入剂量和摄入方式, 使得低聚木糖的应用更加安全高效。
