Objective To establish a rapid detection model for the nutritional components of Oncorhynchus mykiss based on near-infrared spectroscopy (NIR). Methods Firstly, near-infrared spectral data of 200 Oncorhynchus mykiss were collected. Then, the content of 3 kinds of nutritional components (moisture, fat and protein) of each Oncorhynchus mykiss were determined by national standard methods. The near-infrared spectral data and nutritional component data were matched one by one. The near-infrared rapid detection model was established by combining NIR with partial least squares (PLS), and the best detection model was screened out. Results The pretreatment method of the moisture content detection model was multiple scattering correction (MSC), and the optimal model was obtained when the wavelength range was 4000-10000 cm-1. The pretreatment method of the fat content detection model was standard normal variable (SNV), and the optimal model was obtained when the wavelength range was 5000-7144 cm-1 and 7404-10000 cm-1. The pretreatment method of the protein detection model was second derivative (ds2)+SNV+Savitzky-Golay smoothing (sg9), and the optimal model was obtained when the wavelength range was 4100-5100 cm-1 and 5400-9000 cm-1. The Q value, the correlation coefficient of the correlation coefficients, and the correlation coefficient of prediction of the optimal model were all relatively large, and the standard deviation of square error corrected and standard error of cross validation were close to each other, which met the best modeling principle. Conclusion The best model is verified by the prediction set that do not participate in the modeling. The absolute deviation between the predicted value and the true value (determined by the national standard method) is no more than 5.7%. This indicates that the model can be used for the detection of three nutritional components of Oncorhynchus mykiss, and can achieve non-destructive and rapid detection of the nutritional components of Oncorhynchus mykiss, saving detection costs and shortening the detection cycle.

目的 基于近红外光谱(near-infrared spectroscopy, NIR)技术构建虹鳟鱼营养成分快速检测模型。方法 先采集200条虹鳟鱼NIR数据, 再用国标方法测定每条虹鳟鱼3种营养成分(水分、脂肪和蛋白质)的含量, 将NIR数据与营养成分数据一一对应, 利用NIR结合偏最小二乘法(partial least squares, PLS)建立近红外快速检测模型, 并筛选出最佳检测模型。结果 水分含量检测模型的预处理方法为多元散射校正(multiple scatter correction, MSC), 波段为4000~10000 cm-1时, 模型最优; 脂肪含量检测模型预处理方法为标准正态变量变换(standard normal variable transformation, SNV), 波段为5000~7144 cm-1, 7404~10000 cm-1时模型最优; 蛋白质含量检测模型的预处理方法为二阶导数(ds2)+SNV+Savitzky-Golay平滑(sg9), 波段为4100~5100 cm-1, 5400~9000 cm-1时模型最优, 最优模型的Q值、校正模型相关系数、交互验证集相关系数均较大, 校正集标准差和预测集标准误差互相接近, 满足最佳建模原则。结论 使用未参与建模的预测集对最佳模型进行验证, 跌出结果为预测值与真实值(国标方法测定值)的绝对偏差均不超过5.7%, 说明该模型可用于虹鳟鱼3种营养成分的检测, 可实现虹鳟鱼营养成分的无损、快速检测, 节约检测成本, 缩短检测周期。

目的 基于近红外光谱(near-infrared spectroscopy, NIR)技术构建虹鳟鱼营养成分快速检测模型。方法 先采集200条虹鳟鱼NIR数据, 再用国标方法测定每条虹鳟鱼3种营养成分(水分、脂肪和蛋白质)的含量, 将NIR数据与营养成分数据一一对应, 利用NIR结合偏最小二乘法(partial least squares, PLS)建立近红外快速检测模型, 并筛选出最佳检测模型。结果 水分含量检测模型的预处理方法为多元散射校正(multiple scatter correction, MSC), 波段为4000~10000 cm-1时, 模型最优; 脂肪含量检测模型预处理方法为标准正态变量变换(standard normal variable transformation, SNV), 波段为5000~7144 cm-1, 7404~10000 cm-1时模型最优; 蛋白质含量检测模型的预处理方法为二阶导数(ds2)+SNV+Savitzky-Golay平滑(sg9), 波段为4100~5100 cm-1, 5400~9000 cm-1时模型最优, 最优模型的Q值、校正模型相关系数、交互验证集相关系数均较大, 校正集标准差和预测集标准误差互相接近, 满足最佳建模原则。结论 使用未参与建模的预测集对最佳模型进行验证, 跌出结果为预测值与真实值(国标方法测定值)的绝对偏差均不超过5.7%, 说明该模型可用于虹鳟鱼3种营养成分的检测, 可实现虹鳟鱼营养成分的无损、快速检测, 节约检测成本, 缩短检测周期。

Objective To establish a rapid detection model for the nutritional components of Oncorhynchus mykiss based on near-infrared spectroscopy (NIR). Methods Firstly, near-infrared spectral data of 200 Oncorhynchus mykiss were collected. Then, the content of 3 kinds of nutritional components (moisture, fat and protein) of each Oncorhynchus mykiss were determined by national standard methods. The near-infrared spectral data and nutritional component data were matched one by one. The near-infrared rapid detection model was established by combining NIR with partial least squares (PLS), and the best detection model was screened out. Results The pretreatment method of the moisture content detection model was multiple scattering correction (MSC), and the optimal model was obtained when the wavelength range was 4000-10000 cm-1. The pretreatment method of the fat content detection model was standard normal variable (SNV), and the optimal model was obtained when the wavelength range was 5000-7144 cm-1 and 7404-10000 cm-1. The pretreatment method of the protein detection model was second derivative (ds2)+SNV+Savitzky-Golay smoothing (sg9), and the optimal model was obtained when the wavelength range was 4100-5100 cm-1 and 5400-9000 cm-1. The Q value, the correlation coefficient of the correlation coefficients, and the correlation coefficient of prediction of the optimal model were all relatively large, and the standard deviation of square error corrected and standard error of cross validation were close to each other, which met the best modeling principle. Conclusion The best model is verified by the prediction set that do not participate in the modeling. The absolute deviation between the predicted value and the true value (determined by the national standard method) is no more than 5.7%. This indicates that the model can be used for the detection of three nutritional components of Oncorhynchus mykiss, and can achieve non-destructive and rapid detection of the nutritional components of Oncorhynchus mykiss, saving detection costs and shortening the detection cycle.

虹鳟鱼(Oncorhynchus mykiss)属于太平洋鲑冷水性鱼类, 因其鱼体两侧有紫红色的彩带故名虹鳟[1], 其肉质鲜美、营养价值丰富且与三文鱼相似, 因此被称为“淡水三文鱼”[2]。其原产自北美洲和太平洋, 上世纪中叶传入中国, 经过几十年的繁育, 现已成为我国主要淡水养殖鱼类[3-7]。虹鳟鱼还是联合国粮农组织向世界推广的品质优良的4大淡水鱼种之一[8-10], 其肉质鲜美, 无小骨刺, 蛋白含量较高, 且含有丰富的氨基酸、矿物质、维生素、二十二碳六烯酸(docosahexaenoic acid, DHA)和二十碳五烯酸(eicosapentaenoic acid, EPA)等[11-17], 现已成为人们生活中补充蛋白质不可或缺的一种食品。
近几年, 对于虹鳟鱼的研究多集中在生物信息学、遗传育种、保鲜贮藏和品质评价等领域[18-22]。目前, 虹鳟鱼的品质评价方法多采用传统方法检测其营养成分, 存在实验过程烦琐、周期较长等劣势[23-26]。因此, 研究快速、准确且操作简单的虹鳟鱼营养成分测定方法是十分有必要的。随着交叉学科的兴起, 近红外光谱(near-infraredred spectroscopy, NIR)定量分析技术已广泛应用于食品领域[27-29]。王若楠等[30]采用NIR技术, 建立了谷子淀粉多样性预测模型; 赵培栋等[31]采用NIR分析仪WINISI软件系统, 构建出的油菜未成熟种子含水量近红外模型得出的预测值与真实值拟合度高, 预测性较好; 潘发明等[32]利用NIR技术结合数学和化学计量学构建出全株玉米青贮营养成分含量近红外预测模型; LIU等[33]采用NIR结合化学计量学, 建立了大米中淀粉、蛋白质等营养成分的快速无损检测模型。
本研究以200批次虹鳟鱼为样品, 利用NIR技术结合偏最小二乘法(partial least squares, PLS), 建立虹鳟鱼中水分、脂肪、蛋白质3种营养成分的快速检测模型, 并通过改变建模条件筛选最佳模型, 为实现高效、无损测定虹鳟鱼营养成分提供了理论基础和技术支持。
虹鳟鱼样品: 由新疆天蕴有机农业有限公司养殖, 新疆鳟贵鲜食科技有限公司负责加工, 共200批次样品。
石油醚、无水乙醇(分析纯, 天津市鑫铂特化工有限公司); 硫酸、盐酸(分析纯, 成都市科隆化学品有限公司)。
NIR Master近红外光谱仪(配有温控InGaAs检测器、钨卤灯及NIRCal 5.6.6000数据处理软件, 步琦实验室设备贸易有限公司); 8420全自动凯氏定氮仪(丹麦福斯公司); AL204-IC电子天平(万分位, 瑞士梅特勒-托利多仪器有限公司); B811全自动索氏提取仪(瑞士步琦有限公司); 101-3EBS电热鼓风干燥箱(中国永光明医疗仪器有限公司)。
虹鳟鱼由企业工作人员去皮、剔骨, 切去头尾, 只留鱼中段, 真空包装, 冷链运输至实验室。打开真空包装后, 为保证实验结果的准确性, 先用纸巾擦干鱼肉表面水分等杂质, 避免对实验结果造成影响。
虹鳟鱼肉经NIR采集后, 切成3×3 cm小方块, 放入洗净、擦干的辅食机中粉碎成质地均匀的鱼糜, 放入自封袋中待测。
精密称取制备好的虹鳟鱼样品, 按照国标方法分别测定3种营养成分的含量。蛋白质参照 GB 5009.5—2016《食品安全国家标准 食品中蛋白质的测定》第一法 凯氏定氮法; 脂肪参照GB 5009.6—2016《食品安全国家标准 食品中脂肪的测定》第一法 索氏抽提法; 水分参照GB/T 5009.3—2016《食品安全国家标准 食品中水分的测定》第一法 直接干燥法。
根据虹鳟鱼样品3种品质含量测定数据分布情况, 按照各参数含量测定值均匀分布原则, 将200批次样品分成校正集和验证集, 其中校正集样品与验证集样品共150份, 二者之比为2:1, 预测集样品为50份。校正集和验证集样品用于定量预测模型的建立, 预测集不参与模型建立, 只用于外部验证定量预测模型的实用性和稳健性。
为保证仪器的稳定性, 开机后预热40 min, 将虹鳟鱼样品置于NIR仪采集系统样品盘里, 以仪器内置背景(白板)为参比, 采集4000~10000 cm-1波长范围内的样品NIR。分辨率为8 cm-1。扫描过程中实验室温度保持在室温。为避免样品放置引起的误差, 每个样品重复扫描4次, 求出4次扫描光谱的平均光谱为该样品最终光谱数据, 供建模使用。
实验采用NIR仪自带的NIRCal 5.6.6000软件, 首先将每个虹鳟鱼样品的平均光谱数据与每个样品的3种营养成分测定值一一对应, 采用系统自带的数据处理软件进行自动建模, 系统自动对选中的光谱进行标准化处理和微分计算, 从而建立最佳的定量预测模型。分别建立虹鳟鱼3种营养成分(蛋白质、脂肪、水分)含量的同时定量预测的最佳定量预测模型集, 该模型可以用于虹鳟鱼中3种营养成分的同时定量预测分析。定量预测模型的优劣由Q值模型评价参数进行综合评价。一般认为, 当所建模型的Q值越高, 说明所建立校正模型的预测性能和稳健性越好, 精度越高。
将虹鳟鱼3种营养成分含量已知的预测样品集(未参与建模)NIR扫描数据带入到已建立的虹鳟鱼最佳定量预测模型集中, 同时对预测集样品的3种成分含量进行快速定量预测, 根据预测集样品3种品质含量测定值与定量预测模型预测值的偏差判断模型的稳健性和适应性。
实验数据均平行测定3次, 以平均值为最终结果, 通过Microsoft Excel 2010软件对数据进行整理与统计; 使用 NIRCal5.6.6000软件进行近红外建模及相关性作图。
虹鳟鱼建模样品集由校正样品集和验证样品集组成。化学值测定值的统计结果见表 1。由表1可知, 虹鳟鱼水分含量在62.27~84.97 g/100 g, 平均值68.90 g/100 g; 脂肪含量在1.07~15.76 g/100 g, 平均值为5.32 g/100 g; 蛋白质含量在19.58~24.14 g/100 g, 平均值21.32 g/100 g。实验样本具有普适性, 满足虹鳟鱼中3种营养成分含量快速检测近红外模型的建立条件要求。
用NIR Master NIR仪对200批次虹鳟鱼样品进行扫描。200批次虹鳟鱼样品在4000~10000 cm-1波长范围内扫描获得的平均光谱原始图见图1图1中纵坐标为NIR吸光度值(A), 横坐标为波长(cm-1)。
图1可知, 200批次虹鳟鱼样品在4000~10000 cm-1范围内近红外光谱吸收峰峰位基本相同, 但每个样品的吸光度值大小有所不同, 由此说明, 虽然200批次样品所含化学成份基本相同, 但是每个样品中化学成分所含有含量有所差异。由图1可知, 虹鳟鱼样品分别在4400~4600, 5800~6200, 7600~8200 cm-1等多处出现明显的NIR吸收峰, 这些特征吸收峰为虹鳟鱼营养成分的快速定量分析数学模型的建立提供了最有利的光谱信息。利用这些特征吸收光谱信息结合多种建模方法, 将光谱特征吸收信息与虹鳟鱼样品中3种营养成分含量之间建立一对应函数关系, 从而建立3种品质含量同时定量预测的近红外预测模型。
根据200批次虹鳟鱼样品3种营养成分国家标准方法含量测定结果, 按照每种品质含量均匀分布的原则, 将200批次样品分成校正集、验证集和预测集3组, 其中校正集数量大约占全部样品的50%, 内部交叉验证集和预测集样品数量均占样品总数量的25%。校正集和验证集用于建立虹鳟鱼3种营养成分含量定量预测模型的建立, 预测集用来检验定量预测模型的外部验证。建模集样品中3种营养成分含量的分布结果见表2
表2可知, 校正集的水分含量在65.62~76.29 g/100 g, 平均值68.86 g/100 g; 脂肪含量在1.07~15.76 g/100 g, 平均值5.23 g/100 g; 蛋白质含量在19.58~24.14 g/100 g, 平均值为21.28 g/100 g; 验证集水分含量在65.77~72.74 g/100 g, 平均值68.93 g/100 g; 脂肪含量在1.89~13.34 g/100 g, 平均值5.52 g/100 g; 蛋白质含量在19.84~24.08 g/100 g, 平均值为21.31 g/100 g; 建模样品集的3种营养成分含量值基本覆盖了虹鳟鱼3种营养成分含量的分布范围, 具有较好的代表性。
在NIRcal里导入采集的虹鳟鱼样品光谱, 采用仪器自带的数据处理软件进行自动建模, 建立相应的数学模型。根据数据处理软件提供的综合评价指标Q值来筛选最优模型, 数据分析处理结果见表3
表3数据可知, 选用5种预处理方法对虹鳟鱼NIR进行预处理, 得出3种营养成分的最佳建模条件如下: 水分含量预测模型的最佳光谱预处理方法为4000~10000 cm-1 光谱范围内进行MSC处理, 此时水分含量预测模型的Q值、RC、RP分别为0.5836、0.8164、0.7792, 校正集标准差(SEC)、验证标准差(SEP)分别为1.0974、1.0880; 脂肪含量预测模型的最佳光谱预处理方法为5000~7144 cm-1, 7404~10000 cm-1光谱范围内进行SNV处理, 此时脂肪含量模型的Q值、RC、RP分别为0.4497、0.6072、0.5562, SEC、SEP分别为2.1183、2.1060; 蛋白质含量预测模型的最佳光谱预处理方法为4100~5100 cm-1, 5400~9000 cm-1光谱范围内进行ds2, SNV, sa9处理, 此时蛋白质含量预测模型的Q值、RC、RP分别为0.5071、0.6665、0.6167, SEC、SEP分别为0.7086、0.7370。以上预处理后的预测模型的Q值更接近于1, 说明建立的模型聚类效果越好, 分离度高, 且SEC、SEP最为接近, 符合最佳模型要求。所建最佳模型的预测效果见图2~图4, 最佳模型评价参数统计结果见表4
图2~4表4的数据可知, 所建立的虹鳟鱼3种营养成分含量同时快速定量测模型的评价参数满足最优模型的评价条件: 模型的Q值、RC、RP均较高, 而且模型预测误差均在虹鳟鱼3种营养成分定量分析允许误差的范围之内, 该模型可以用于未知虹鳟鱼样品水分、蛋白质、脂肪含量的同时定量分析。
为验证虹鳟鱼中3种营养成分含量同时定量预测最佳模型的实际使用效果, 利用3种营养成分含量已知的但未参与建模的50份预测集样品的NIR带入到已建好的虹鳟鱼最佳定量预测模型中对模型的预测性能和可靠性进行检验, 预测集样品中各成分含量的定量预测结果见表5。由表5可知, 虹鳟鱼3种营养成分含量的真值(用国家标准方法测定值)与模型预测值之间的最大偏差均不超过5.7%, 说明该模型可用于虹鳟鱼3种营养成分无损快速检测。
本研究以虹鳟鱼样品为研究对象, 利用NIR技术实现了虹鳟鱼3种营养成分快速、无损定量检测。结合PLS分析, 以所建模型的Q值、RC和RP越大, SEC和SEP越接近为评价标准, 筛选出每个成分最适宜的建模波段、预处理方法, 建立每个成分的最佳定量模型, 成功预测虹鳟鱼水分、脂肪、蛋白质3种营养成分的含量。该模型普适性较好, 可用于所有虹鳟鱼水分、脂肪、蛋白质含量测定, 且该方法克服了传统检测方法步骤烦琐、实验周期长、实时性差等缺陷, 具有无损、易操作、成本低、效率高等优势, 提升检测效率的同时降低了检测难度和成本。虹鳟鱼营养成分近红外快速检测模型的建立, 为NIR技术在食品检测领域的应用提供了理论基础, 本课题组计划以后将该检测方法向企业推广, 让科研成果服务企业, 降低企业检测成本, 缩短检测周期, 实现动态调整养殖方案, 提高产品品质的同时提升国民饮食质量。
