Objective To establish a fingerprint spectrum for the volatile compounds of A brand ready-to-eat bird's nest by solid-phase microextraction-gas chromatography-mass spectrometry (SPME-GC-MS), and conduct differential analysis on different brands and types of ready-to-eat bird's nest products. Methods Using commercially available ready-to-eat bird's nest as the research object, SPME-GC-MS was used to analyze the volatile substances in 37 batches of A brand ready-to-eat bird's nest samples with different storage periods. The volatile substances were analyzed through the database of NIST 20 and the method of peak area normalization. The “TCM chromatographic fingerprint similarity evaluation system” was used to establish the volatile substances fingerprint spectrum of A brand ready-to-eat bird's nest. Results Under the service of NIST 20 database, 26 kinds of characteristic volatile components were identified in brand A's ready-to-eat bird's nest, mainly including unsaturated compounds such as 5-hydroxymethylfurfural, nonanol, lauryl alcohol, tetrahydro lavender alcohol, benzaldehyde, and so on. A fingerprint spectrum of the volatile substances in this ready-to-eat bird's nest product was established. The evaluation criteria were based on a total of 26 matching peaks, the similarity was greater than or equal to 65.8%, which could be well applied to evaluate the quality of different commercially available ready-to-eat bird's nests. Conclusion By analyzing the composition of volatile substances in ready-to-eat bird's nest and establishing a fingerprint and similarity evaluation method of volatile components for ready-to-eat bird's nest, it is possible to effectively distinguish different quality levels of ready-to-eat bird's nest, providing a scientific basis for quality control and authenticity identification of ready-to-eat bird's nest.

目的 基于固相微萃取-气相色谱-质谱法(solid-phase microextraction-gas chromatography-mass spectrometry, SPME-GC-MS)构建A品牌即食燕窝挥发性物质指纹图谱, 并对不同品牌和不同类型的即食燕窝产品进行差异分析。方法 以市售即食燕窝为研究对象, 采用SPME-GC-MS分析不同储存期共计37批次A品牌即食燕窝样品的挥发性物质, 通过NIST 20数据库和峰面积归一化法对挥发性物质进行分析研究, 并使用“中药色谱指纹图谱相似度评价系统”进行A品牌即食燕窝挥发性成分指纹图谱的建立。结果 利用NIST 20数据库在品牌A即食燕窝中共鉴定出26种特征挥发性成分, 主要包括5-羟甲基糠醛、壬醇、月桂醇、四氢熏衣草醇、苯甲醛等化合物, 并建立了该款即食燕窝产品的挥发性物质指纹图谱。评价标准为: 以相匹配26个共有峰为基础, 相似度≥65.8%, 可较好地应用于不同市售即食燕窝差异分析。结论 通过分析即食燕窝挥发性物质的组成, 建立即食燕窝挥发性成分指纹图谱和相似度评价方法, 可有效区分不同类别的即食燕窝产品, 为即食燕窝的产品鉴别和分类提供了科学依据。

目的 基于固相微萃取-气相色谱-质谱法(solid-phase microextraction-gas chromatography-mass spectrometry, SPME-GC-MS)构建A品牌即食燕窝挥发性物质指纹图谱, 并对不同品牌和不同类型的即食燕窝产品进行差异分析。方法 以市售即食燕窝为研究对象, 采用SPME-GC-MS分析不同储存期共计37批次A品牌即食燕窝样品的挥发性物质, 通过NIST 20数据库和峰面积归一化法对挥发性物质进行分析研究, 并使用“中药色谱指纹图谱相似度评价系统”进行A品牌即食燕窝挥发性成分指纹图谱的建立。结果 利用NIST 20数据库在品牌A即食燕窝中共鉴定出26种特征挥发性成分, 主要包括5-羟甲基糠醛、壬醇、月桂醇、四氢熏衣草醇、苯甲醛等化合物, 并建立了该款即食燕窝产品的挥发性物质指纹图谱。评价标准为: 以相匹配26个共有峰为基础, 相似度≥65.8%, 可较好地应用于不同市售即食燕窝差异分析。结论 通过分析即食燕窝挥发性物质的组成, 建立即食燕窝挥发性成分指纹图谱和相似度评价方法, 可有效区分不同类别的即食燕窝产品, 为即食燕窝的产品鉴别和分类提供了科学依据。

Objective To establish a fingerprint spectrum for the volatile compounds of A brand ready-to-eat bird's nest by solid-phase microextraction-gas chromatography-mass spectrometry (SPME-GC-MS), and conduct differential analysis on different brands and types of ready-to-eat bird's nest products. Methods Using commercially available ready-to-eat bird's nest as the research object, SPME-GC-MS was used to analyze the volatile substances in 37 batches of A brand ready-to-eat bird's nest samples with different storage periods. The volatile substances were analyzed through the database of NIST 20 and the method of peak area normalization. The “TCM chromatographic fingerprint similarity evaluation system” was used to establish the volatile substances fingerprint spectrum of A brand ready-to-eat bird's nest. Results Under the service of NIST 20 database, 26 kinds of characteristic volatile components were identified in brand A's ready-to-eat bird's nest, mainly including unsaturated compounds such as 5-hydroxymethylfurfural, nonanol, lauryl alcohol, tetrahydro lavender alcohol, benzaldehyde, and so on. A fingerprint spectrum of the volatile substances in this ready-to-eat bird's nest product was established. The evaluation criteria were based on a total of 26 matching peaks, the similarity was greater than or equal to 65.8%, which could be well applied to evaluate the quality of different commercially available ready-to-eat bird's nests. Conclusion By analyzing the composition of volatile substances in ready-to-eat bird's nest and establishing a fingerprint and similarity evaluation method of volatile components for ready-to-eat bird's nest, it is possible to effectively distinguish different quality levels of ready-to-eat bird's nest, providing a scientific basis for quality control and authenticity identification of ready-to-eat bird's nest.

食用燕窝是一种天然食品, 为雨燕科(Apodidase)金丝燕及多种同属燕类分泌的唾液与其绒羽混合凝结而筑成的巢窝, 其主要产自东南亚地区, 具有丰富的营养和药用价值[1-2]。在中国古代, 燕窝被认为是一种高端的滋补食品[3]。现代研究表明, 食用燕窝的主要营养成分包括蛋白质(56.34%~69.5%)、碳水化合物(17.12%~31.68%)、唾液酸(7.64%~12.52%)、多种常量和微量元素、维生素、表皮生长因子等[4-10], 具有诸多生物功效, 例如抗氧化、延缓衰老、促进表皮生长、滋阴养肺、保护软骨组织等[11-16]。随着人们生活水平的提高和生活方式的改变, 即食燕窝与人工炖煮比较, 一方面可以节省炖煮时间, 方便消费者食用, 另一方面即食燕窝通过严格的工艺生产, 风味品质更优于人工炖煮, 使得即食燕窝越来越受到消费者的青睐。目前对即食燕窝的风味研究已有相应的文献报道[17-18], 但通过研究即食燕窝的风味变化来控制产品的生产和评价品质鲜少有相应的文献报道。
随着即食燕窝市场不断扩大, 即食燕窝的品类也呈现多种多样。由于燕窝化学组成复杂, 对燕窝进行品质检测、营养评价、真假鉴别等方面分析往往需要复杂的前处理过程, 同时需要对多个指标进行测定后评价[19-21], 该方式耗时耗力, 而且常常涉及到有害试剂与试剂浪费的问题, 增加成本。随着指纹图谱分析技术和现代仪器分析技术的快速发展, 风味指纹图谱已经被广泛应用于水果的香气成分、肉类风味、酒类挥发性物质及香精香料风味组分的鉴定[22-24]。在现代仪器分析技术中, 气相色谱-质谱法(gas chromatography-mass spectrometry, GC-MS)是较为常用的方法之一, 与固相微萃取(solid-phase microextraction, SPME)联用具有操作时间短、样品量少、无需萃取溶剂、重现性好等优点[25], 适用于食品中易挥发性成分的检测, 应用范围包括食品种类、产地溯源及食品质量评价等[26-27]。基于此, 结合SPME-GC-MS可全面准确分析即食燕窝风味组成, 并根据分析结果筛选对即食燕窝风味起决定作用的特征成分, 建立其指纹图谱中的特征指纹信息, 构建即食燕窝挥发性成分指纹图谱, 可为即食燕窝产品的开发和差异分析提供坚实的理论依据。
挥发性指纹图谱基于食品的固有品质特性, 通过解析食品中挥发性成分的组成与变化, 能够间接反映关键营养物质的含量、加工稳定性及功能活性[28]。本研究以A品牌碗装燕窝为主要研究对象, 通过SPME-GC-MS对不同批次即食燕窝产品的挥发性物质进行检测分析, 利用NIST 20质谱库进行定性分析, 结合“中药色谱指纹图谱相似度评价系统”建立即食燕窝挥发性成分指纹图谱相似度评价方法, 以期提供高效、直观的分析手段应用于快速检测、工艺优化和产品溯源。
品牌A (37批不同货架期碗装即食燕窝)(厦门市燕之屋丝浓生物科技有限公司); 品牌B、C(市售碗装即食燕窝样品); 品牌D、E、F、G、H、I(市售瓶装即食燕窝样品)。
GC/MS-TQ8040气相色谱-质谱联用仪(日本岛津公司); 57347-U固相微萃取手动进样手柄、75 μm CAR/PDMS固相微萃取柱(美国Supelco公司); SHJ-4数显恒温磁力水浴锅(常州金坛宏华仪器厂); T18DS25均质器(德国IKA公司); JSC-THB-600电子天平(精度0.01 g, 台衡精密测控股份有限公司); DB-wax石英毛细管柱(30 m×0.32 mm, 1.2 μm)(美国Agilent公司)。
参照本实验室前期建立的即食燕窝中挥发性成分测定方法[17-18], 并作适当调整, 具体实验条件如下。
样品开启后于均质机中迅速完成均质, 称取均质后的40.0 g样品于250 mL的SPME瓶中, 迅速盖上瓶盖, 将样品瓶放入到60 ℃磁力搅拌恒温水浴锅中, 恒温平衡5 min, 将预先于230 ℃老化5 min的SPME萃取头(75 μm CAR/PDMS)插入样品瓶中, 推出纤维头, 顶空平衡吸附30 min。吸附后, 立即收回纤维头, 将SPME移至GC中, 在230 ℃条件下解析5 min。
色谱柱: DB-wax石英毛细管柱(30 m×0.32 mm, 1.2 μm); 进样口温度: 230 ℃; 柱升温程序: 初温60 ℃保持0.5 min, 以10 ℃/min的速率升温到200 ℃, 保持10 min, 再以20 ℃/min的速率升温到230 ℃, 保持6 min; 载气: 高纯氦气(99.999%); 柱流量: 1.61 mL/min; 不分流模式。
电子轰击源(electron impact, EI); 离子源温度: 230 ℃; 接口温度: 230 ℃; 溶剂延时: 2.5 min; 扫描方式: Q3 SCAN; 质量扫描范围m/z 20~500。
在岛津GC/MS Solution工作站中导出经NIST 20数据库定性后的GC-MS数据文件, 运用Excel 2019软件进行指纹图谱数据处理转换为“中药色谱指纹图谱相似度评价系统(2012版)”格式文件, 将转换格式后的数据文件导入“中药色谱指纹图谱相似度评价系统(2012版)”进行分析, 以S1号样品的色谱图作为参照图谱, 时间窗宽度设定0.1, 运用多点校正法手工标注进行Mark峰匹配, 以平均数法生成指纹图谱。
每个样品进行3次独立平行实验, 结果以平均值作为指纹图谱建立所需数据, 通过《中药色谱指纹图谱相似度评价软件》对37批A品牌即食燕窝挥发性成分确认共有峰、建立对照图谱并进行相似度评价, 采用Microsoft office excel 2019建立相似度分布图, 以及采用SPSS 23软件对37批A品牌即食燕窝挥发性成分进行聚类分析;
为了保证所抽取的样品具有代表性, 随机抽取不同货架期的正常即食燕窝产品, 按照1.3的方法进行燕窝样品的处理, 测定37批即食燕窝中的挥发性成分, 同时作样品空白。测定结果扣除样品空白后, 对色谱峰逐一以NIST 20与NIST 20s质谱数据库进行检索分析, 结果见表1
由特征挥发性成分可知, A品牌即食燕窝挥发性物质的组成主要为醇类、醛类物质, 亦含少量酚类、酸类等物质。为了验证特征挥发性成分与燕窝风味差异的贡献程度, 采用样品量梯度实验进行考察, 即在样品前处理过程中, 样品取样量分别为10、20、30和40 g, 分析不同样品量对特征挥发性成分的影响。结果发现, 随着取样量的增加, 26种特征挥发性成分存在不同程度的增长。其中, 以5-羟甲基糠醛、壬醇、月桂醇、四氢熏衣草醇、苯甲醛5种挥发性成分增长最为明显, 此类物质具有花香、泥土、蜂蜜等气味, 这可能源于金丝燕的生长环境和广泛的食谱。另一方面, 燕窝中富含蛋白质和碳水化合物, 在热处理过程中, 燕窝中部分氨基化合物与还原糖发生美拉德反应[29], 生成5-羟甲基糠醛等代表性产物, 具有烘焙、杏仁风味, 常常作为风味评价指标[30-31]。综上, 燕窝中挥发性成分较为复杂, 单一的挥发性成分并不能很好地体现燕窝的风味差异, 26种特征挥发性成分均对燕窝风味有着不同程度的贡献。因此, 此次研究以26种作为A品牌即食燕窝挥发性成分指纹图谱构建的基础。
利用“中药色谱指纹图谱相似度评价系统”建立了37批A品牌即食燕窝挥发性成分色谱峰的共有模式, 结果见图1。以S1号样品的色谱图作为参照图谱, 运用多点校正法手工标注26个共有峰进行Mark峰匹配, 以平均数法生成对照图谱, 见图2。统计相似度评价结果, 见表2
指纹图谱能够反映出一类产品的整体性[32], 有利于对燕窝差异作出客观的评价。由表2可知, 37批即食燕窝指纹图谱与标准指纹图谱的相似度范围为0.692~0.962, 结果并不满足指纹图谱的基本要求, 主要由于即食燕窝产品中挥发性特征成分含量与燕窝产地、燕窝种类、加工工艺、配料添加等因素有关, 仅依据相似度并不能很好地区分不同燕窝产品的差异。
为了建立适用于即食燕窝的指纹图谱, 基于即食燕窝特征挥发性成分, 根据CNAS-GL027 2018《化学分析实验室内部质量控制指南》中对警戒限(warning limit, WL)和行动限(action limit, AL)的规定, 建立即食燕窝相似度评价质量控制图, 结果见图3。其中, 控制图的CL代表控制值的平均值, 控制图的警戒限为平均值(CL=0.850)的二倍标准偏差(2S)距离处, 行动限为平均值的3倍标准偏差(3S)距离处。在服从正态分布的情况下, 数据落在警戒限(WL=0.722)之内的概率为95%, 落在行动限(AL=0.658)之内的概率为99.7%。综上, 确定即食燕窝指纹图谱质量控制标准为: 以共有峰匹配数量为26个为基础, A品牌即食燕窝指纹图谱相似度≥65.8%。
为了验证本研究所建立的即食燕窝指纹图谱的适用性, 选择2款市售碗装燕窝和6款市售瓶装燕窝作为考察对象, 分别进行相似度分析及聚类分析。
以本研究建立的A品牌即食燕窝产品的指纹图谱为标准指纹图谱, 分别比较市售的即食燕窝与标准指纹图谱共有峰匹配数量与相似度计算结果, 结果见表3。在相似度评价中, 除B品牌(72.1%)与C品牌(74.6%)相似度超过指纹图谱控制线, 其余均不满足最低限。虽然碗装燕窝在峰的匹配与相似度计算更加接近标准指纹图谱, 但所有款式燕窝的共有峰匹配数量在11~22个, 均少于标准指纹图谱共有峰数量的26个, 依旧存在显著性差异。综上所述, 即食燕窝标准图谱的建立不仅要考虑相似度, 还需要关注特征共有成分的种类, 两者结合可应用于即食燕窝差异分析。
以A品牌即食燕窝的26种特征挥发性成分为变量, 在SPSS 23系统软件分别输入9款即食燕窝相应峰面积(n=3), 采取组间平均数联结法计算方法以及使用平方欧式距离法计算样品之间的实际距离, 对即食燕窝聚类展开系统的分析, 如图4所示。
按照分类距离可以看出, 当欧式平方距离为25时, 9款即食燕窝聚成即食燕窝一个大类; 当欧式平方距离为15时, 将即食燕窝产品分类成“碗装燕窝”与“瓶装燕窝”; 进一步的, 当欧式平方距离为5时, 又将A品牌即食燕窝分离出来, 剩余两类包括“碗装燕窝B、C”与“瓶装燕窝D、E、F、G、H、I”。结果表明, 对于市售的即食燕窝产品存在不同差异, 可通过特征挥发性成分进行分类, 与相似度评价结果一致。
结合感官评价分析, 市售的即食燕窝产品通常为冰糖型即食燕窝, 即产品配方仅为冰糖、水及燕窝, 通过调整不同加工工艺得到的产品, 在工艺调整上差异并不显著。例如相似度接近的B品牌(72.1%)与C品牌(74.6%)即食燕窝产品, 外观形态及口感差异并不显著, 仅通过感官很难进行差异区分。因此, 相似度分析与聚类分析反映了挥发性成分与产品风味差异的相关性, 验证了即食燕窝挥发性成分构建指纹图谱的可靠性。
本研究以A品牌即食燕窝为研究对象, 通过SPME-GC-MS对37批次A品牌即食燕窝挥发性物质进行检测分析, 确定了26种特征挥发性成分, 主要包括5-羟甲基糠醛、壬醇、月桂醇、四氢熏衣草醇、苯甲醛等化合物, 并根据CNAS-GL027 2018建立了A品牌即食燕窝挥发性成分指纹图谱和相似度评价标准方法, 评价标准为: 以共有峰匹配数量为26个为基础, A品牌即食燕窝指纹图谱相似度≥65.8%。本研究合理结合化学分析质量控制方法与指纹图谱的应用, 较好地对不同即食燕窝差异评价, 实现快速检测和差异分析。同时, 研究进一步通过聚类分析将即食燕窝分类成碗装燕窝与瓶装燕窝类别, 与相似度评价结果一致, 验证了即食燕窝挥发性成分构建指纹图谱的可靠性。在未来研究中, 可通过长期持续累积研究数据, 建立统一规范的燕窝指纹图谱数据库, 为今后即食燕窝的质量评价和真伪鉴别提供参考和指导。
