Objective To explore the application of the combination of real-time fluorescence quantitative polymerase chain reaction (qPCR) and droplet digital polymerase chain reaction (ddPCR) in the quantitative detection of meat adulteration. Methods This study employed qPCR and ddPCR technologies to develop a detection system with specific primers and probes for beef, lamb and pork samples, aiming to assess meat adulteration in the Beijing market. Results The experimental results revealed that the sample “Chang 5” contained adulteration of lamb and pork at a ratio of 17:10. qPCR demonstrated high sensitivity and specificity in qualitative detection, while ddPCR enabled absolute quantification of adulteration proportions. The combination of these 2 methods significantly improved detection accuracy. Conclusion The established qPCR-ddPCR combined method exhibits high sensitivity and specificity, making it suitable for rapid and precise detection of meat adulteration. This approach provides reliable technical support for food safety regulation.

目的 探究实时荧光定量聚合酶链反应(real-time fluorescence quantitative polymerase chain reaction, qPCR)与微滴式数字聚合酶链反应(droplet digital polymerase chain reaction, ddPCR)结合在肉类掺假定量检测中的应用。方法 本研究采用qPCR和ddPCR技术, 针对牛、羊、猪等肉类样品建立特异性引物和探针检测体系, 以评估北京市肉类市场的掺假情况。结果 “昌5”样品存在羊肉和猪肉的掺杂, 且掺杂比例为17:10。qPCR在定性检测中表现出高灵敏度和特异性, 而ddPCR实现了掺假比例的绝对定量, 两者结合显著提高了检测准确性。结论 本研究建立的qPCR-ddPCR联用方法具有高灵敏度和特异性, 适用于肉类掺假的快速精准检测, 为食品安全监管提供了可靠的技术支持。

Objective To explore the application of the combination of real-time fluorescence quantitative polymerase chain reaction (qPCR) and droplet digital polymerase chain reaction (ddPCR) in the quantitative detection of meat adulteration. Methods This study employed qPCR and ddPCR technologies to develop a detection system with specific primers and probes for beef, lamb and pork samples, aiming to assess meat adulteration in the Beijing market. Results The experimental results revealed that the sample “Chang 5” contained adulteration of lamb and pork at a ratio of 17:10. qPCR demonstrated high sensitivity and specificity in qualitative detection, while ddPCR enabled absolute quantification of adulteration proportions. The combination of these 2 methods significantly improved detection accuracy. Conclusion The established qPCR-ddPCR combined method exhibits high sensitivity and specificity, making it suitable for rapid and precise detection of meat adulteration. This approach provides reliable technical support for food safety regulation.

近年来, 随着经济的快速发展和人们生活水平的不断提高, 食品安全问题日益受到广泛关注。肉类作为人们日常饮食中的重要组成部分, 其质量和安全性直接关系到消费者的健康和权益[1]。然而, 由于利益驱动, 市场上肉类掺假、肉制品污染现象时有发生[2-4]。2024年4月, 国务院食安办等部门部署各地开展严厉打击肉类产品违法犯罪专项整治行动[5], 截至目前已查办一批肉类产品违法犯罪案件, 截至11月底, 全国共接到肉类产品相关举报1.4万起、查实举报4000余起; 行政执法立案1.4万起, 查获涉案物品9000余t; 查办假冒牛(羊、驴)肉制品违法案件1000余起; 公安机关刑事立案2400余起、抓获犯罪嫌疑人6000余人。
肉类掺假问题不仅对消费者的健康构成威胁, 还可能引发清真食品的信仰问题[6], 进而影响民族团结和社会稳定[7-8]。 欧盟定量成分声明强制要求肉制品需要标识出所有成分及其净含量, 并规定肉的主要成分为骨骼肌, 结缔组织和脂肪等成分需单独列出且规定了最高含量[9]。因此, 建立一种快速、准确、灵敏的肉类掺假检测方法, 对于保障食品安全、维护消费者权益具有重要意义。
当前, 肉类掺假的检测手段主要包括: 感官鉴定方法, 光谱分析技术[10-11], 基于免疫学原理的检测技术[12], 生物质谱技术[13], 基于核酸层面的DNA扩增分析技术[14-16]等。实时荧光定量聚合酶链式反应(real-time fluorescence quantitative polymerase chain reaction, qPCR)和微滴式数字聚合酶链式反应(droplet digital polymerase chain reaction, ddPCR)是目前常用的核酸定量检测技术。qPCR技术具有灵敏度高、特异性强、操作简便等优点, 能够实现对目标核酸的定性检测[17-18]。然而, qPCR的定量检测需要依赖标准曲线, 存在一定的局限性[19], ddPCR技术则克服了这一缺点, 其通过将反应体系分割成数万个微滴, 实现对目标核酸的绝对定量检测, 目前已应用于基因表达量[20]、微生物丰度[21]、转基因产品含量[22]及食品成分的定量检测[23-24], 此技术具有更高的准确性和可靠性[25-26]。将qPCR和ddPCR相结合, 可以充分发挥两种技术的优势, 实现对肉类掺假的快速、准确检测。
北京市作为中国的首都, 人口众多, 民族成分复杂, 清真食品需求量大。根据第七次全国人口普查数据, 北京市少数民族户籍人口数量为104.8万, 其中回族27.4万, 占北京市少数民族的26.1%[27]。在某些区域, 如牛街、教子胡同、花市、长营、马甸、于家务等地, 以及大兴、昌平、顺义、密云等远郊区县, 清真居民相对集中。因此, 保障北京市肉类产品的质量和安全性, 对于维护清真食品的信仰和民族团结具有重要意义。近年来, 北京市市场监督管理局在打击食品领域违法行为, 特别是肉类掺假案件方面采取了严厉措施, 根据国务院食安办、公安部、农业农村部、市场监管总局的通知, 从2024年4月至11月, 全国将开展严厉打击肉类产品违法犯罪专项整治行动, 以生猪、肉牛、肉羊、肉鸡及其肉类产品为重点, 深挖违法犯罪源头线索, 对全链条开展专项整治。然而, 由于检测技术的局限性, 部分肉类掺假行为仍难以被及时发现和查处。因此, 建立一种适用于北京市肉类掺假检测的高效方法, 对于提高食品安全监管水平、保障消费者权益具有重要意义。
本研究旨在采用qPCR和ddPCR相结合的方法, 对北京市肉类样品中的掺假情况进行检测。通过对不同肉类样品的定性与定量分析, 揭示肉类掺假的现状和规律, 为食品安全监管部门提供技术支持和参考依据, 进一步推动食品安全监管工作的深入开展。
实际样品为北京市区内被市民举报掺假的5类肉类样品, 含牛、羊、猪等肉类样品以及羊肉卷、牛肉卷市售, 分别标注为“房7”“房6”“昌5”“丰7”“丰10”, 其中“房7”“丰7”标识为纯牛肉, “房6”“昌5”“丰10”标识为纯羊肉。
4×浓度即用型预混液、微滴生成专用油、微滴生成卡(苏州思纳福医疗科技有限公司); 血液、细胞、组织基因组DNA提取试剂盒[天根生化科技(北京)有限公司]; 引物、探针[生工生物工程(上海)股份有限公司]。
ABI7500 qPCR仪(美国Thermo Fisher Scientific公司); DQ-24液滴式数字PCR仪(苏州思纳福医疗科技有限公司); 5420微量高速离心机(德国Eppendorf公司); QSJ-C04B1绞肉机(广东小熊电器有限公司); ME204E万分之一电子分析天平(瑞士Mettler Toledo公司)。
将送检的样品随机取样, 用绞肉机搅碎所取的样品, 并分别从5类样品中称取8小份(每份20 mg)的样品放入离心管中, 10000 r/min离心1 min, 倒尽上清, 加入200 μL裂解缓冲液, 振荡至彻底悬浮, 后续操作参照DNA提取试剂盒说明书进行DNA提取。
从已经发表的文章和报告中查找对应的引物、探针序列, 并在美国国家生物技术信息中心​(National Center for Biotechnology Information​, NCBI)(https://www.ncbi.nlm.nih.gov/)网站上通过BLAST将序列进行比对, 最终确定牛肉、羊肉和猪肉的特异性引物和探针, 如表1所示, 交由生工生物工程(上海)股份有限公司合成, 引物序列见表1
在本研究中, 选取了羊肉、猪肉和牛肉的基因组DNA作为模板, 利用表1中列出的特定引物和探针进行PCR扩增反应。为了确保实验的准确性, 设置了无菌水作为阴性对照。实验过程中, 使用ABI 7500 qPCR仪进行扩增反应。通过该仪器的高灵敏度和特异性检测功能, 对所筛选的引物和探针的特异性进行了详细验证。这一过程有助于确保引物和探针在不同肉类样本中的特异性扩增能力, 为后续的实验可靠的基础。
qPCR扩增反应体系见表2。反应条件为95 ℃预变性10 min; 95 ℃循环反应15 s, 60 ℃退火及延伸1 min; 45个循环。
ddPCR扩增反应体系见表3, 反应体系充分混匀后加入微滴生成卡中, 油包水微滴生成按照厂家的操作说明进行。
温控反应程序见表4
肉的质量与样品DNA溶液中RPA1的基因拷贝数及单位质量的基因拷贝数有关, 根据公式(1)计算。

式中: M为肉的质量; Q为基因拷贝数; C为单位质量肉的基因拷贝数。通过数字PCR测得的基因拷贝数(Q)数据统计分析使用DQ-24液滴式数字PCR自带系统进行。
实验中5份样本分别做3个平行重复, 每个样品都使用干净的仪器, 以防止交叉污染, 取平均值带入式(1)计算。
以牛、羊、猪、鸡、鸭、马、驴等7种畜禽肉中提取的基因组DNA作为模板, 设置无菌水作为空白对照, 进行qPCR检测, 以确保实验的准确性和可靠性。如图1实验结果显示, 空白对照和非靶标肉中均未出现微滴扩增, 特异性扩增仅针对靶标肉, 能够满足实验的检测需求。
通过qPCR对5种肉类样品进行DNA扩增分析, 如图2结果显示, 标识为纯牛肉的“房7”和“丰7”样品仅对牛的特异性引物产生扩增反应, 表明这两种样品符合其标识的纯牛肉特性。而标识为纯羊肉的“房6”“昌5”和“丰10”样品均对羊的特异性引物产生扩增反应, 显示其含有羊肉成分。“昌5”样品还对猪的特异性引物产生了扩增反应, 这一结果提示“昌5”样品存在肉类掺假的可能性。而“房6”“丰10”样品虽对猪的特异性引物产生了扩增反应, 但扩增Ct值较靠后, 接近35, 据此基本可判定这两个样品在生产或销售环节与猪肉存在一定程度的沾染, 而非故意掺假。
采用本研究建立的ddPCR检测方法对有肉类掺假可能性的“昌5”样品中羊肉和猪肉的相对含量进行定量分析。如图3图4结果表明, 经DNA提取和ddPCR扩增后, 单位质量的羊肉与猪肉阳性液滴比值为850:913, 按照公式(2)进行计算[22]

式中: Mp和Mm分别为猪肉和羊肉的质量; Qp和Qm分别为混合样品中猪肉和羊肉的基因拷贝数; Cp和Cm分别为单位质量猪肉和羊肉的基因拷贝数。
鉴于同一肉种的细胞密度和基因组大小保持恒定, 因此对于任意肉种, 其Cm/Cp比值均为固定值。任君安等[28]利用数字PCR技术对含有1%、10%、50%、60%和80%猪肉含量的羊肉样品分别进行定量分析, 检测其中的RPA1基因拷贝数, 并计算Cm/Cp比值及其相对标准偏差(relative standard deviation, RSD)。结果显示, 5种不同猪肉含量样品的Cm/Cp平均值为1.84, RSD为3.50%, 表明Cm/Cp具有较高的稳定性。此外, 反应生成的总微滴数均超过10000个, 满足泊松分布的统计学要求。因此, 可以计算得出“昌5”样品中羊肉和猪肉的掺杂比例为17:10。
本研究采用qPCR和ddPCR相结合的方法对北京市肉类样品中的掺假情况进行检测。qPCR方法在定性检测中显示出较高的灵敏度和特异性, 能够有效区分不同种类的肉类成分, 满足了公安机关对案件快速检测定性的需求。KÖPPEL等[25]的研究显示实时荧光qPCR方法在最低含量为1%时的相对误差最高为65%, 而ddPCR方法在定量检测中具有显著优势, 其绝对定量能力能够直接测量目标核酸的绝对数量, 不受标准曲线的影响, 从而为肉类掺假的定量分析提供了更为精确的数据支持。
在对“昌5”样品的检测中, qPCR结果显示该样品中存在猪肉成分, 而ddPCR定量分析经过计算得出了羊肉和猪肉的掺杂比例为17:10。这一结果表明, 该样品中羊肉和猪肉的掺假程度较为严重, 可能对消费者的健康和权益造成影响。同时, 这也反映出市场上部分肉类产品的质量监管存在漏洞, 需要进一步加强肉类产品的质量控制和监管力度。
本研究建立的qPCR和ddPCR检测方法具有较高的灵敏度和特异性, 能够有效应用于肉类掺假的检测。该方法不仅适用于实验室环境, 还具有一定的现场快速检测潜力, 为食品安全监管部门提供了有力的技术支持。然而, 该方法在实际应用中也存在一些局限性。例如, 对于复杂的肉类混合样品, 可能存在多种肉类成分的相互干扰, 影响检测结果的准确性[29-30]。此外, 实验过程中对DNA提取和扩增条件的要求较高, 操作不当可能导致假阳性或假阴性结果。因此, 在未来的研究中, 可以进一步优化实验条件和方法, 提高其在复杂样品中的检测性能, 同时探索与其他检测技术的联合应用, 以提高检测的准确性和可靠性。
本研究的结果为食品安全监管部门提供了重要的参考依据。首先, 监管部门应加强对肉类产品的质量监管, 特别是对高风险的肉类掺假行为进行重点监控[31]。其次, 应加大对肉类生产、加工和销售环节的监督检查力度, 确保肉类产品的质量和安全。此外, 还应加强与科研机构的合作, 推广先进的检测技术和方法, 提高食品安全检测的水平和效率。通过多方面的努力, 共同维护消费者的食品安全和合法权益, 促进食品市场的健康发展。
