Objective To investigate the differences in mineral element content between sole fish and basa fish and establish an authenticity identification method. Methods Inductively coupled plasma mass spectrometry were employed to determine 25 kinds of mineral elements in 29 sole fish samples and 24 basa fish samples. Principal component analysis were performed using SPSS 25.0 software on the measurement results, and comparative analysis were conducted on mineral element contents among 3 kinds of different genera of sole fish. Results The total mineral element content in sole fish were significantly higher than that in basa fish. Na, K and Ca were identified as the primary mineral elements in both species, with Sr, As, Mn, Fe and Zn serving as secondary elements. Notably, Sr, As, Mn and Fe were detected exclusively in sole fish but absent in basa fish. Significant differences were observed in 9 kinds of elements: Mg, Ti, Zn, As, K, Ca, Na, Mn, and Fe. Principal components of sole fish within the same genus exhibited relative clustering, with marked differences in 7 kinds of elements: V, Mn, B, Ba, Fe, As and Sr. Conclusion Mineral element analysis can effectively distinguish sole fish from basa fish for authenticity verification.

目的 探讨龙利鱼和巴沙鱼矿物元素含量差异, 并对其进行真伪鉴定。方法 以电感耦合等离子体质谱法测定29份龙利鱼和24份巴沙鱼中25种矿物元素含量, 对测定结果采用SPSS 25.0软件进行主成分分析, 并对3种不同属的龙利鱼矿物元素含量进行比较分析。结果 龙利鱼中矿物元素总含量明显高于巴沙鱼, Na、K和Ca是两类鱼中主要的矿物元素, 次要元素主要有5种Sr、As、Mn、Fe、Zn, 其中Sr、As、Mn、Fe在龙利鱼中均有检出, 在巴沙鱼中均未检出, Mg、Ti、Zn、As、K、Ca、Na、Mn、Fe 9种元素上存在显著差异; 相同属间龙利鱼主成分相对集中, V、Mn、B、Ba、Fe、As、Sr 7种元素存在显著差异。结论 通过矿物元素分析可以有效用于龙利鱼和巴沙鱼的真伪鉴定。

Objective To investigate the differences in mineral element content between sole fish and basa fish and establish an authenticity identification method. Methods Inductively coupled plasma mass spectrometry were employed to determine 25 kinds of mineral elements in 29 sole fish samples and 24 basa fish samples. Principal component analysis were performed using SPSS 25.0 software on the measurement results, and comparative analysis were conducted on mineral element contents among 3 kinds of different genera of sole fish. Results The total mineral element content in sole fish were significantly higher than that in basa fish. Na, K and Ca were identified as the primary mineral elements in both species, with Sr, As, Mn, Fe and Zn serving as secondary elements. Notably, Sr, As, Mn and Fe were detected exclusively in sole fish but absent in basa fish. Significant differences were observed in 9 kinds of elements: Mg, Ti, Zn, As, K, Ca, Na, Mn, and Fe. Principal components of sole fish within the same genus exhibited relative clustering, with marked differences in 7 kinds of elements: V, Mn, B, Ba, Fe, As and Sr. Conclusion Mineral element analysis can effectively distinguish sole fish from basa fish for authenticity verification.

龙利鱼通常指鲽亚目(Pleuronectoidei)舌鳎科(Cynoglossidae)或鳎科(Soleidae)物种的统称[1], 其共同特征为身体扁平, 呈舌状, 双眼位于身体同一侧(类似比目鱼), 属海水鱼类, 栖息于近海底层。巴沙鱼通常指鲶亚目(Siluroidei)芒鲇科(Pangasiidae)的低眼无齿芒(Pangasianodon hypophthalmus)或博氏巨鲶(Pangasius bocourti)[2], 其特征为体型类似普通鲶鱼, 无鳞, 有须, 属淡水鱼类, 原产于东南亚湄公河流域, 多为人工养殖。
龙利鱼和巴沙鱼作为两种常见的食用鱼类, 在居民消费结构中占有一席之地。龙利鱼, 作为一种海水鱼类, 质细腻、无刺, 不仅蛋白质含量高, 而且含有丰富的多不饱和脂肪酸, 这些脂肪酸对于降低人体胆固醇含量、预防心血管疾病等方面具有重要作用[3-4]。相比之下, 巴沙鱼作为一种淡水养殖品种, 同样具有生长迅速、个体大、产量高、易饲养、刺少、易加工等诸多优点[5], 但其营养价值和市场价格相对较低。由于外观和口感的相似性, 消费者对巴沙鱼和龙利鱼认知混淆, 水产市场、餐饮行业将“巴沙鱼”冠名为“龙利鱼”进行销售[6]。在进出口申报和消费环节容易出现高值低报和以次充好的可能。
为了追逐更高经济效益, 冒牌、贴标等虚假销售现象时有发生, 迫切需要完善的监管体系和适宜的检测技术对食品真伪进行甄别, 以保障食品的真实性。水产品产地溯源与真伪鉴别技术一直是食品安全领域关注的热点问题[7]。目前对水产品真实性和溯源的研究还处于起步阶段, 其主要原因是此前我国进口水产品尚处于量增阶段, 主要关注点放在水产品的兽药残留等问题方面, 而对产品本身真实性的关注度相对不高, 技术热度较低。因此, 开发广泛、快速、廉价和可靠的水产品溯源方法极为重要。目前水产品的溯源技术发展迅速, 如稳定同位素分析法[7-12]、矿物元素分析法[13-16]、光谱分析法[17-20]、核磁共振法[21]、核酸检测法[22-25]和代谢组学法[26-29]等, 正逐渐成为水产品质量安全方向的研究热点。其中, 矿物元素分析技术其原理基于生物体无法自身合成矿物元素, 必须从周围环境中摄取, 因能够反映水产品产地环境特征和自身饮食特征, 且具有灵敏度高、可靠性强等优点在水产品溯源研究中应用广泛、效果突出。目前矿物元素溯源技术主要运用于不同区域土壤种植的植物源性产品, 水产品因水域变化性较大, 真伪鉴别案例相对较少。本研究通过测定巴沙鱼和龙利鱼中25种矿物元素(B、Mg、Al、Ti、V、Cr、Mn、Fe等)含量, 并对测定结果进行主成分分析, 筛选出具有显著差异的元素作为鉴别因子, 建立进行巴沙鱼和龙利鱼水真伪鉴别模式, 能够为龙利鱼和巴沙鱼真伪鉴别提供一种快速的鉴定方法, 满足消费者真实的购物需求。
龙利鱼和巴沙鱼购自广西东兴市国贸市场, 龙利鱼(编号1~29, 共计29个样本)、巴沙鱼(编号1~24, 共计24个样本)。每份样本约500 g。其中龙利鱼共3种不同属(条鳎属1个、宽箬鳎属1个、舌鳎属27个)。
65%硝酸(优级纯, 德国Merck公司); 多种元素混合标准溶液(B、Mg、Al、Ti、V、Cr、Mn、Fe、Co、Ni、Cu、Zn、Ga、As、Sr、Cd、Sn、Sb、Ba、Hg、Tl、Pb, 质量浓度1000 μg/mL)、单一元素标准溶液(Na、Mg、Al、K、Ca、Mn、Fe、Cu、Zn、Sr, 质量浓度1000 μg/mL)、内标混标溶液(Bi、Ge、In、Re、Rh、Sc, 质量浓度100 μg/mL)(国家有色金属及电子材料分析测试中心); 质谱混合调谐标准溶液(Li、Be、In、U, 质量浓度10 μg/mL)(美国Agilent公司)。
Agilent 7800电感耦合等离子体质谱仪(美国Agilent公司); Exceed-Ad-EDI-16超纯水机(成都唐氏康宁科技发展有限公司); MARS6微波消解仪(美国CEM公司); BHW-09C控温赶酸器(上海博通化学科技有限公司); Puiverisette11刀式研磨仪(德国飞驰公司)。
将每一份样本中鱼的可食部分, 用刀式混合研磨仪磨碎, 将制备好的试样装入洁净的容器内, 密封并标明标记, 并冷冻保存于-18 ℃冰箱。
样品前处理方法以及仪器参数, 主要参照GB 5009.268—2016《食品安全国家标准 食品中多元素的测定》的标准执行, 称取鱼肉样品0.5~1.0 g(精确至0.001 g), 置于消解内罐中, 加入5 mL硝酸, 将消解管置于120 ℃赶酸电热板上预消解20~30 min[30]进行预消解, 将消解内罐加外盖旋紧放入微波消解装置中, 按设定参数进行消解, 微波消解程序见表1。消解后取出微波消解管, 于通风橱内小心打开内外盖开罐排气, 用少量水冲洗内盖, 将微波消解管放至赶酸仪中, 于100 ℃加热30 min, 冷却后, 倒出样液后再用水洗消解管至少3次, 合并于刻度准确的50 mL塑料离心管中, 用水定容至50 mL刻度, 混匀待测, 同时做空白试验。
使用He碰撞模式, 减少干扰。通过测试质谱调谐液优化电感耦合等离子体质谱法的仪器采集方法参数。优化后的采集方法参数为: RF功率: 1550 W; 等离子体气体: 15.0 L/min; 辅助气流量: 1.0 L/min; 雾化气流量: 1.0 L/min; 雾化室温度: 2 ℃; 碰撞模式: He模式; 碰撞池氦气流量: 3.5 mL/min。采样深度: 8 mm; 采样锥孔径: 1.0 mm, 截取锥孔径: 0.4 mm; 采样时间: 40 s; 重复次数: 3次。
对样品中B、Mg、Al、Ti、V、Cr、Mn、Fe、Co、Ni、Cu、Zn、Ga、As、Sr、Cd、Sn、Sb、Ba、Hg、Tl、Pb、Na、K、Ca 25种矿物元素含量按照1.3.3 样品前处理方法进行测定, 对于高含量的的元素Na、Mg、Al、K、Ca、Mn、Fe、Cu、Zn、Sr元素还需要根据初测结果稀释50~100倍进行检测。
采用SPSS 25.0统计软件对结果进行主成分分析(principal component analysis, PCA)。
对29种龙利鱼和24种巴沙鱼进行了25种矿物元素的检测。结果显示龙利鱼的矿质元素总含量为(4409.88±1058.83) mg/kg, 巴沙鱼的矿质元素总含量(3088.87±606.22) mg/kg。龙利鱼矿物元素的总含量明显高于巴沙鱼的含量。其中Na、K、Ca均是两类鱼中主要的矿质元素, 龙利鱼中Na、K、Ca含量分别为(806.37± 387.03)、(2040.80±578.75)、(1169.92±638.74) mg/kg; 巴沙鱼中钠、钾和钙含量分别为(2182.92±751.86)、(779.12±390.85)、(47.21±16.15) mg/kg。次要元素主要有5种, 分别为Sr、As、Mn、Fe、Zn, 其中Sr、As、Mn、Fe在龙利鱼中有检出, 在巴沙鱼中均未检出, Zn元素龙利鱼大于巴沙鱼, 见表2。9种元素Pb、TI、Sb、Ga、Ni、Cr、Cd、Sn、Co在两种鱼种基本均未检出。
采用SPSS 25.0软件对龙利鱼和巴沙鱼矿物元素含量数据进行PCA, 结果见图1, 可看出龙利鱼和巴沙鱼矿物元素含量差异显著, 说明两种鱼能够较好的分离。将B、Mg、Al、Ti、V、Cr、Mn、Fe、Co、Ni、Cu、Zn、Ga、As、Sr、Cd、Sn、Sb、Ba、Hg、Tl、Pb、Na、K、Ca共25种元素分别按序号1~25编号。通过VIP scores分析, 每个矿物元素可以得出一个VIP值, 即变量重要性投影, VIP值大小代表该物质对于区分组间差异的贡献大小, 在判别过程中, 一般认为VIP值>1表示该变量具有重要作用, VIP值越大, 贡献率越大, 说明该变量在不同处理组间的差异越显著, 通常VIP>1为常见的差异物质筛选标准[31]。从图1可以看出Mg、Ti、Zn、As、K、Ca、Na、Mn、Fe 共9种元素VIP>1, 是龙利鱼和巴沙鱼的主要的差异元素。
29种龙利鱼按照水产品分类学可以细分3种不同属(A条鳎属1个、B宽箬鳎属1个、C舌鳎属27个), 进行PCA, 见图2, 可看出舌鳎属27个样本主成分相对比较集中, 说明相同属间龙利鱼矿物元素含量具有比较相似的规律, 因条鳎属和宽箬鳎属样本较少, 不具有统计学意义, 但可以看出不同属的龙利鱼A、B与C之间还是有一定的差异性, 下一步需要收集更多的条鳎属和宽箬鳎属的龙利鱼样本。从VIP得分图可以看出, V、Mn、B、Ba、Fe、As、Sr共7种元素VIP>1, 是龙利鱼的主要差异元素。
本研究提出了一种通过检测25种矿物元素来鉴定龙利鱼和巴沙鱼真伪的方法。通过分析发现, 龙利鱼矿物元素总含量高于巴沙鱼, Na、K和Ga是两类鱼中主要的矿质元素, 通过PCA, 发现龙利鱼和巴沙鱼在矿物元素含量差异显著, Mg、Ti、Zn、As、K、Ca、Na、Mn、Fe 共9种元素上存在显著差异。次要元素主要有5种分别为Sr、As、Mn、Fe、Zn, 其中Sr、As、Mn、Fe在龙利鱼中有检出, 在巴沙鱼中均未检出, Zn元素龙利鱼大于巴沙鱼, 9种元素Pb、Tl、Sb、Ga、Ni、Cr、Cd、Sn、Co在两种鱼种基本均未检出。相同属间龙利鱼矿物元素含量具有比较相似的规律, 因其他2种属样本量比较少, 不具有统计学意义, 但可以看出不同属的龙利鱼之间还是有一定的差异性。矿物元素分析是一种有效的鱼类真伪鉴定手段, 能够为食品安全监管和消费者权益保护提供技术支持。
下一步可以扩大样本类型, 包括不同地区、不同生长阶段的龙利鱼和巴沙鱼, 以增强研究结果的普适性和代表性, 并建立更为全面的矿物元素数据库。同时, 根据不同水产品的特点, 可以结合形态学鉴定、分子生物学方法等, 建立综合鉴定技术, 以实现更全面的水产品真伪鉴定, 从而更好地保护消费者权益和提升食品安全水平。
