In the economic shellfish farming on coastal mudflats in China, the Sinovovacula constricta occupies an important position. In recent years, due to the rapid urbanization and industrialization in China, marine pollution has threatened the living environment of Sinovovacula constricta. Polluted Sinovovacula constricta can accumulate toxins in their bodies, which can be transmitted through the food chain and ultimately endanger human health. Therefore, the food safety issue of Sinovovacula constricta has received widespread attention. With the continuous improvement of the transportation network and the gradual maturity of the live-keeping technology system, more and more Sinovovacula constricta are being sent to inland areas, expanding the consumer market. Therefore, Sinovovacula constricta must undergo purification treatment before entering the circulation process. As consumers' demands for the taste and quality of fresh Sinovovacula constricta continue to increase, the purification and live-keeping processes of Sinovovacula constricta have become particularly important. This article reviewed the current research progress in the purification methods of Sinovovacula constricta in China, the advantages and limitations of different purification processes, the influences of environmental parameters on purification, and live-keeping technologies, providing a reference for in-depth research on the purification and live-keeping technologies of Sinovovacula constricta.

在我国沿海滩涂养殖的经济贝类中, 缢蛏占据重要地位。近年来, 由于我国城市化和工业化发展迅速, 海洋污染威胁缢蛏的生存环境, 受污染的缢蛏会在体内蓄积毒素, 由食物链传导, 最终危害人体健康, 所以缢蛏的食品安全问题受到广泛关注。随着交通运输网的不断完善和保活技术体系逐渐成熟, 越来越多的缢蛏被送往内陆地区, 消费市场扩大。因此, 缢蛏在进入流通环节前必须进行净化处理。随着消费者对鲜活缢蛏口感品质的要求不断提高, 缢蛏的净化和保活环节显得尤为重要。本文综述了当前国内缢蛏的净化方式、不同净化工艺的优势和局限性、环境参数对净化的影响、保活技术等方面的研究进展, 为深入开展缢蛏净化与保活技术相关研究提供参考。

在我国沿海滩涂养殖的经济贝类中, 缢蛏占据重要地位。近年来, 由于我国城市化和工业化发展迅速, 海洋污染威胁缢蛏的生存环境, 受污染的缢蛏会在体内蓄积毒素, 由食物链传导, 最终危害人体健康, 所以缢蛏的食品安全问题受到广泛关注。随着交通运输网的不断完善和保活技术体系逐渐成熟, 越来越多的缢蛏被送往内陆地区, 消费市场扩大。因此, 缢蛏在进入流通环节前必须进行净化处理。随着消费者对鲜活缢蛏口感品质的要求不断提高, 缢蛏的净化和保活环节显得尤为重要。本文综述了当前国内缢蛏的净化方式、不同净化工艺的优势和局限性、环境参数对净化的影响、保活技术等方面的研究进展, 为深入开展缢蛏净化与保活技术相关研究提供参考。

In the economic shellfish farming on coastal mudflats in China, the Sinovovacula constricta occupies an important position. In recent years, due to the rapid urbanization and industrialization in China, marine pollution has threatened the living environment of Sinovovacula constricta. Polluted Sinovovacula constricta can accumulate toxins in their bodies, which can be transmitted through the food chain and ultimately endanger human health. Therefore, the food safety issue of Sinovovacula constricta has received widespread attention. With the continuous improvement of the transportation network and the gradual maturity of the live-keeping technology system, more and more Sinovovacula constricta are being sent to inland areas, expanding the consumer market. Therefore, Sinovovacula constricta must undergo purification treatment before entering the circulation process. As consumers' demands for the taste and quality of fresh Sinovovacula constricta continue to increase, the purification and live-keeping processes of Sinovovacula constricta have become particularly important. This article reviewed the current research progress in the purification methods of Sinovovacula constricta in China, the advantages and limitations of different purification processes, the influences of environmental parameters on purification, and live-keeping technologies, providing a reference for in-depth research on the purification and live-keeping technologies of Sinovovacula constricta.

缢蛏(Sinovovacula constricta)是我国南北沿海滩涂地区养殖的主要经济贝类[1]。由于缢蛏肉质鲜美, 营养价值高, 富含丰富的不饱和脂肪酸、氨基酸和脂质等营养成分, 深受消费者的喜爱[2], 所以我国缢蛏年产量超80万t, 具有极高的经济价值[3]。近年来, 随着我国城市化和工业化快速发展, 养殖产业迅速增长, 导致海域污染日益严重, 对缢蛏的养殖环境构成威胁。由于缢蛏属于滤食性生物且迁移能力差, 其生存环境一旦受到污染, 污染物不仅会附着在体表, 还会通过呼吸和摄食活动进入缢蛏体内蓄积形成生物毒素, 最终通过食物链危害人体健康[4]。因此, 缢蛏净化是降低致病风险的关键环节。目前, 关于缢蛏的净化技术可归纳为海水暂养净化法、水体消毒灭菌法、化学试剂脱除法和生物代谢法等, 不同的净化技术均有各自的优点和局限性[5]
由于缢蛏死亡后, 会滋生细菌, 导致缢蛏腐败变质; 同时不饱和脂肪酸氧化酸败, 蛋白质变质产生胺类物质, 均会危害人体健康。所以通常采用保活配送的方式来保障在流通环节中缢蛏的风味和品质[6]。不同的保活方式, 对缢蛏的保活效果不同。保活过程中的温度、盐度、溶氧、pH、二氧化碳浓度、浊度等关键因子的变化, 也会引起缢蛏风味和品质的变化[7]。现今, 常用的保活技术有化学保活法、模拟生态环境保活法、低温保活法和生态冰温保活法等[8]
缢蛏净化是将其放在合适盐度的干净盐水或海水中, 通过其正常的摄食活动以减少体内的砂石等污染物, 起到净化体内生物、化学和物理等危害物的效果[4]。贝类常见的净化模式有暂养净化、流水净化和循环水净化。针对于缢蛏净化, 最常用的方式是暂养净化。缢蛏在经过干净海水暂养和净化, 可以降低大肠埃希氏菌和菌落总数的数量, 而且随着时间的延长, 其大肠埃希氏菌数量由11000 MPN/g降低至30 MPN/g, 符合相关标准[9]。虽然通过暂养净化可以显著降低缢蛏体内致病菌等污染物至可安全食用水平, 但由于耗时长(一般净化24~48 h)、费劳力、经济效益水平不高, 且存在因暂养导致缢蛏品质下降、污染其他水体等潜在风险[10]
缢蛏净化工艺主要针对泥沙去除、减菌、重金属去除、贝类毒素、有机污染物去除、改善贝肉口感品质等方面开展。泥沙去除是缢蛏净化工艺最易实现的目标, 可在较短时间内完成, 如在适宜盐度的海水或盐水中, 通过正常生理活动会将体内的泥沙尽数排出, 通常缢蛏吐沙净化在10 h内均可完成[11]
微生物去除净化是目前净化工艺升级的重点方向, 其是利用消毒杀菌后的洁净水体养殖缢蛏, 通过缢蛏自身的呼吸和摄食活动将细菌排出体外。当前使用较多的水体消毒工艺包括紫外线、氯制剂、臭氧等技术。紫外线水体净化可显著降低大肠杆菌和副溶血性弧菌的含量, 同时对海水中的病毒也有较好的杀灭效果[12]。氯制剂由于其强氧化性和杀菌效率高被广泛应用于净化水、饮用水的杀菌。氯制剂的类型也由二氧化氯、次氯酸等逐渐发展出含氯电解水。有研究报道, 30 mg/L的电解水可显著降低贝类体内的副溶血性弧菌和创伤弧菌[13], 但过高浓度的电解水会导致贝类的死亡。臭氧技术因其广谱、便捷、杀菌效果良好等优势而应用于海水净化, 50 mg/L的臭氧净化海水44 h后可分别降低大肠杆菌、霍乱弧菌和副溶血弧菌99.8%、90%和85.1%[14], 但臭氧浓度过高(大于0.5 mg/L)会对缢蛏产生毒性。单一的杀菌净化手段具有局限性, 多组合的杀菌净化技术可以实现快速且显著的杀菌效果, 如臭氧结合紫外线杀菌可促使水体中产生氧化性极强的羟基自由基而达到高效杀菌, 缩短杀菌时间75%[15]; 而紫外线与氯制剂联合可完全去除贝类中的染伤寒沙门氏菌[16]
重金属是缢蛏净化工艺中较难去除的一种污染物。缢蛏出现重金属富集, 主要富集于消化腺。由于短时间暂养, 缢蛏经自身代谢无法排尽重金属, 通常通过脱除剂进行脱离, 包括乙二胺四乙酸(ethylene diamine tetraacetic acid, EDTA)类、壳聚糖类、硒化物类、微生物制剂等。研究表明, 通过EDTA净化的贻贝, 镉去除率可达到40%以上[17]; 壳聚糖联合海藻可以降低牡蛎体中的镉和铅至符合国家标准[18]; 硒化卡拉胶可净化贻贝体内的镉, 脱除率达38%[17]。不同的重金属脱除方式不尽相同, 主要受到金属元素的生物半衰期影响, 因此需要有针对性的评估缢蛏体内的重金属类型, 筛选适宜、高效的脱除手段。
贝类毒素是贝类摄食有毒藻类后, 在其内脏团和消化腺中蓄积形成的一类有毒化合物, 该毒素会经食物链传递引起人中毒。因此在缢蛏净化工艺中, 去除贝类毒素至关重要。活体贝类可通过贝类净化工艺降低体内的毒素含量。有学者研究发现, 采用紫外循环消毒水净化人工感染脱氨甲酰基新石房蛤毒素(decarbamoylneosaxitoxin, dcNEO)的紫贻贝, 净化10 d后, 脱除率达57.8%[19]; 通过臭氧消毒水联合饵料投喂的净化模式净化人工感染麻痹性贝毒(paralytic shellfish poisoning, PSP)的文蛤, 净化15 d后, 脱除率达47.2%[20]; 向海水中添加0.05 g/L壳聚糖, 净化7 d, 可使近江牡蛎中的PSP降至1.41 MU/g[21]。贝类毒素去除难度大, 净化成本高昂且净化方法存在局限性。因此, 选择安全无污染的海域进行养殖生产, 同时严格把关安全检测, 是获得无毒贝类的最优方法。
缢蛏净化工艺以及优缺点见表1
缢蛏净化速率与环境参数密切相关。缢蛏的最适生长环境条件与其净化速率最快的环境参数并不相符。根据去除目标污染物的不同, 选择最优的环境参数来提高缢蛏的净化效率。为达到这一目的, 往往选择人工可调控的环境参数, 比如饵料、温度、盐度和水流速度等。
温度是影响缢蛏净化速率的重要环境因素。有研究表明, 适宜的净化温度有助于去除贝类中的微生物。以牡蛎的微生物去除净化为例, 在去除病毒的过程中,当暂养水温度超过20 ℃时, 3~4 d即可下降一个对数值[22]。在去除细菌的过程中, 将暂养水温度控制在7~15 ℃内, 净化5 d后, 长牡蛎中的副溶血性弧菌数减少3个对数值, 牡蛎死亡率为0[23]。明显可见, 净化温度偏低有利于去除贝类中的细菌, 而净化温度偏高有利于去除贝类中的病毒。另外, 当暂养水温度过高时, 会对缢蛏的生命活动构成威胁; 当暂养水温度过低时, 缢蛏呼吸代谢活动降低, 抗氧化能力下降, 都会使净化速率减缓。因此, 在缢蛏净化时, 需根据要去除的目标污染物来选择合适的净化温度, 以加快净化速率。
盐度是贝类生存环境中必不可少的要素之一。暂养水中的盐度高低所引起水中渗透压的变化会直接影响贝类的摄食代谢活动。有学者研究发现, 贝类的净化过程受到盐度的影响。例如, 在净化去除长牡蛎中的副溶血性弧菌时, 盐度为10的净化效果要劣于盐度为20~30的净化效果[24]; 当盐度低于7.4时, 牡蛎的摄食活动缓慢下降甚至暂停[25-26]。在净化去除缢蛏中的泥沙时, 设置盐度范围为10~35。在盐度为10时, 出现其吐沙率的最小值为35.1%。当盐度升高至25时, 缢蛏吐沙率达到最大值为50.6%, 净化4 h后, 盐度为25的缢蛏吐沙率达到92.4%[27]。可见, 在相同时间内, 当盐度偏低, 会抑制缢蛏摄食代谢活动, 延长净化时间。
水流速度对缢蛏净化有间接影响。在净化过程中, 缢蛏会通过其呼吸代谢和摄食活动将体内污染物排出。若不及时处理这些排泄物, 那么排泄物中包含的泥沙、微生物和重金属等污染物可能会对缢蛏构成二次污染。当水流速度过小时, 暂养水体易缺氧导致缢蛏摄食代谢行为减弱, 降低净化效率; 当水流速度过大会冲击已沉淀的排泄物, 使水体浑浊, 影响杀菌效果[28]。因此, 在选择水流速度时, 需要考虑缢蛏的呼吸代谢和摄食活动, 有利于缢蛏排污最大化并完成暂养水体的消毒, 达到净化的要求。
选择合适的饵料种类可以加快缢蛏的净化速率。比如在去除泥沙环节中, 向循环水系统或者流水净化设施内, 按蛏料比与一定的时间间隔, 分批投喂扁藻、小球藻、金藻和三角褐指藻等藻类饲料, 基本可以在十小时内, 完成去除泥沙的净化过程[11,25]; 优质的浮游生物饵料可以促进缢蛏滤食活动, 提高其净化效率; 人工饵料的营养更加均衡, 可根据缢蛏自身特性制定适宜的营养成分的配比。饵料的质量, 包括新鲜度、营养成分配比、颗粒粗细等因素都对缢蛏的净化塑料厂有一定的影响。
不同的堆放方式也会影响缢蛏的净化效果。采用单层堆放方式, 缢蛏可充分接触净化水, 有利于呼吸代谢活动的进行, 从而提高净化效果。此外, 单存堆放可以更好地观察缢蛏的形态, 及时发现和处理异常情况。缺点是单存堆放所占空间较大, 在实际生产利用时易受空间的限制。多层堆积方式可以节省空间, 提高场地利用率, 但可能会导致部分缢蛏不能与净化水充分接触, 失水死亡, 或者存放在底层的缢蛏会受到挤压作用, 导致贝体破裂死亡。因此, 选择合适的堆放密度对缢蛏的净化有关键作用。环境参数对缢蛏净化的影响见表2
缢蛏产区主要集中在沿海地区, 保活配送是其产业拓展、农业增效的必由之路。目前贝类的保活技术已逐渐形成体系, 众多学者围绕保活技术与品质维持等方面做了大量的基础性研究, 主要包括化学保活法、模拟生态环境保活法、低温保活法等。不论哪种方法, 缢蛏保活技术必须要考虑到温度、盐度、溶氧、pH、氨氮、污染物等关键环境因子的影响。
化学保活法是通过某些化学药物的作用或麻醉作用使得缢蛏进入休眠或麻醉状态, 以降低其对环境胁迫的应激反应, 从而降低其代谢功能与呼吸强度, 以提高运输保活效率。麻醉技术分为物理麻醉和化学麻醉, 化学麻醉包括丁香类、笑气、间氨基苯甲酸乙酯甲基磺酸盐(tricaine methanesulfonate, MS-222)等[29], 其主要弊端是化学药物可能在机体内富集进而影响人体健康, 因此化学麻醉具有较多安全隐患。随着技术的不断发展, 后续出现了一些低毒或无毒麻醉剂(如二氧化碳、丁香酚油等)应用于保活中。徐德峰等[30]用150 mg/L丁香酚成功麻醉凡纳滨对虾, 李宁等[31]证实了丁香酚对黑鲈鱼的应激程度较低, 周翠平[32]用0.6 mg/L二氧化碳浸泡罗非鱼后无水保活时间延长至5 h。目前虽然麻醉技术在鱼类、虾类等水产品品类中有研究报道, 但鲜少将其应用于贝类保活运输。物理麻醉包括低温冷冻法和电击法。低温冷冻法的原理是通过降低水温, 并使用冰水混合物使鱼类减缓呼吸代谢活动, 进入昏迷状态。而电击法则是刺激鱼类的神经中枢系统使鱼类休克, 便于长途运输。物理麻醉法不会在鱼体内残留有害物质, 因此相对安全, 但需要精准把控麻醉时间和操作条件, 避免鱼类产生过强的应激反应, 损伤鱼体。
模拟生态环境保活法是根据贝类的自然生长环境进行模拟运输, 主要调控的环境参数包括温度、湿度、盐度、溶解氧、pH等。目前缢蛏最为常见的模拟生态环境保活的方式是保留部分滩涂泥以延长其保活时间, 但由于滩涂泥不便于运输且易污染包装而存在局限性。有学者通过模拟海湾扇贝的生活温度、湿度和氧气进行无水保活[33], 又或是通过控制水流动性、饵料和水温等因素模拟魁蚶的生长环境条件[34]。模拟生态环境保活技术的关键在于必须依据缢蛏原生环境参数建立生存模型, 并在保活运输过程中控制好关键环境参数, 因此其运输成本大大提高。
低温保活法分为低温有水保活与低温无水保活, 其中低温无水保活是缢蛏主流的保活方式, 可以有效提高水产品的运载量。温度作为缢蛏保活重要的环境因素, 与其代谢功能、抗氧化能力等密切相关。温度的波动会直接影响缢蛏机体的耗氧量增加, 造成体内自由基代谢紊乱并大量积累, 进而损伤了机体的生理机能与免疫能力。有学者对比缢蛏在不同温度条件下[(-4±1)、(0±1)、(5±1)、(10±1)、(25±1) ℃]贮藏的存活率与品质影响, 研究结果表明0 ℃环境下缢蛏的保活效果最好, 其失重率和开壳率较低, 同时带沙组、控湿组的保活效果优于清洗组、不控湿组[35]。方佳琪等[36]证实相较于10~15 ℃条件, 4 ℃环境能够更好的诱导缢蛏机体超氧化物歧化酶(superoxide dismutase, SOD)、过氧化氢酶(catalase, CAT)和还原性谷胱甘肽(glutathione, GSH)活性增加, 以抑制丙二醛和H2O2含量, 同时减缓糖原的消耗与乳酸的积累。类似的结论也在牡蛎、扇贝、蛤仔等的保活研究中发现[37-39]。但胡远辉等[40]发现净化清洗的缢蛏在无水低温保活下活性更佳, 应该是低温降低了缢蛏的新陈代谢。不同的降温方式会极大的影响缢蛏保活效果, 梯度降温和线性降温均比直接降温的方式更有利于缢蛏的保活效果, 缓解低温胁迫导致的氧化应激[41]。缢蛏不同组织部位对温度胁迫的响应是存在差异性的, 相比于外套膜和鳃, 缢蛏消化腺在温度胁迫下维持了较高水平的SOD、CAT、谷胱甘肽过氧化物酶(glutathione peroxidase, GPX)、总抗氧化能力(total antioxidant capacity, T-AOC)活性和GSH含量, 具有更好的抗氧化损伤能力, 说明消化腺能较好地代表缢蛏集体的抗氧化能力的组织部位[42]
生态冰温保活法是通过低温诱导使得贝类进入半休眠或者完全休眠状态, 降低呼吸代谢强度同时维持生命体所必须的生存条件, 减少或避免贝类在运输过程中因环境温度骤变而导致其代谢功能紊乱, 造成贝体应激胁迫损伤, 以达到保活效果[43]。针对贝类保活过程, 常用的降温方式有3种, 分别是直接降温、均匀降温和梯度降温。有学者研究发现, 降温速率快慢与代谢功能紊乱密切相关。梯度降温相较于其他两者, 更能显著降低贝类由于温度变化而导致应激胁迫损伤的发生率, 极大限度地减少温度变化所带来的不良反应, 延长贝类的保活时间, 从而提高经济效益[44]。该项技术最早来源于日本学者, 其关键控制点是始终保持生命体处于生态冰温温度区。生态冰温温度区是指从冻结点到能够区分生死的临界温度之间的范围, 且不同生命体的生态冰温温度区域是不相同的。例如, 皱纹盘鲍的生态冰温区为-1.5~0℃[45]; 太平洋牡蛎生态冰温区为-1.7~0℃[46]; 毛蚶生态冰温区为-1.6~0℃[47], 目前, 利用生态冰温技术, 已在牡蛎、扇贝、贻贝、蛤仔、鲍鱼等贝类中得以实现。倪锦等[45]利用冷海水喷淋结合充氧与生态冰温保存, 显著提高了皱纹盘鲍的存活率。高加龙等[48]利用生态冰温技术使得香港牡蛎保活9 d且存活率达90%以上。因此在开展缢蛏生态冰温保活前需要根据缢蛏在不同温度下的活动状况和呼吸频率, 将缢蛏开始出现无规律的呼吸频率, 同时观察不到腮的呼吸运动, 且缢蛏尚未死亡时的温度, 作为缢蛏的临界温度; 根据缢蛏在0℃以下, 体内组织细胞水分开始冻结时的温度, 作为缢蛏的冻结点。最后根据冻结点到临界温度的范围来确定其生态冰温区。近年来, 随着贝类的营养价值被不断挖掘, 电商消费的不断发展和消费者对生鲜水产品品质的要求不断提高, 市场对贝类的需求量正在逐渐增加。生态冰温技术因其可避免在运输环节中由于水体变质、温度骤变等因素导致贝类品质恶化以及优化传统运输方式运输不便等优势, 将成为未来贝类运输的主流技术。
缢蛏保活技术以及优缺点见表3
缢蛏作为我国独特的双贝类软体动物, 其独特风味与丰富营养, 深受消费者喜爱。随着海域环境风险的不断提升以及电商消费模式的转变, 缢蛏的安全与运输保活问题越来越受关注。我国缢蛏净化与保活技术起步较晚, 没有体系化的净化与保活工艺且其工艺参数缺乏量化依据。因此需要重点开展以下几方面的基础性研究: (1)构建兼顾风味保持与品质提升的体系化净化工艺研究。缢蛏净化期间所使用的净化海水、投喂的饵料均会影响其净化后的风味与品质, 探究净化方式与工艺对缢蛏风味与品质的影响, 构建兼顾风味保持与品质提升新型净化技术是目前产业的迫切需求点; (2)缢蛏快递物流保活技术标准化。随着电商与互联网+的不断发展, 缢蛏快递物流保活技术需求迫切, 虽然目前缢蛏低温无水保活的研究已逐步开展, 但适应于不同运输条件、环境与距离的保活技术有待进一步细化探究, 同时应建立以市场需求为导向的标准化缢蛏物流保活工艺。(3)开展缢蛏生物毒素的净化研究。受生物毒素污染的缢蛏无法用肉眼直接判断, 常规的烹调加工方式也无法彻底破坏生物毒素。目前对缢蛏中生物毒素去除的净化研究有待更深一步的探究。因此, 迫切需要开展相关研究, 建立有效的贝类食品安全控制法。
