Objective To study the difference of meat quality between adult and young Xueduo yaks. Methods Healthy adult and young castrated Xueduo yaks were selected to analyze the composition and content of conventional nutrients, edible meat quality, amino acids, fatty acids, vitamins and minerals in longissimus dorsi muscle. Results The moisture content of adult Xueduo yak meat was 72.89%, which was lower than that of young Xueduo yak meat. The crude protein, crude fat and crude ash contents of adult Xueduo yak meat were 4.99%, 86.67% and 38.54% higher than those of young Xueduo yak meat, respectively. Therefore, adult Xueduo yak meat had higher nutritional value. The edible meat quality of young Xueduo yak meat was excellent, and the color, taste and tenderness were better than those of adult Xueduo yak meat. Both adult and young Xueduo yak meat were high-quality protein sources. The contents of essential amino acids and total amino acids in young Xueduo yak meat were 5.95% and 14.00%, which were significantly higher than those in adult Xueduo yak meat, but adult Xueduo yak meat tastes more delicious. The fatty acid composition of young Xueduo yak meat was slightly better than that of adult Xueduo yak meat. In terms of vitamin and mineral composition and content, vitamin A in young Xueduo yak meat was significantly higher than that in adult Xueduo yak meat. The iodine content of adult Xueduo yak meat was significantly higher than that of young Xueduo yak meat. Conclusion The meat quality of young Xueduo yak is better than that of adult Xueduo yak. The growth cycle of young Xueduo yak is short, which can create higher economic value under limited forage resources. At the same time, it can improve the market competitiveness of yak meat and provide scientific and effective guidance for yak breeding in pastoral areas.

目的 研究成年和幼龄雪多牦牛肉品质差异。方法 选择健康的成年和幼龄阉割雪多牦牛, 对其背最长肌常规营养成分、食用肉品质、氨基酸、脂肪酸、维生素和矿物质构成与含量进行分析。结果 成年雪多牦牛肉水分含量为72.89%, 低于幼龄雪多牦牛肉; 成年雪多牦牛肉粗蛋白质、粗脂肪和粗灰分含量分别比幼龄雪多牦牛肉高4.99%、86.67%、38.54%, 因此成年雪多牦牛肉具有更高的营养价值。幼龄雪多牦牛肉食用肉品质极佳, 色泽、口感、嫩度等都优于成年雪多牦牛肉。成年和幼龄雪多牦牛肉都是优质蛋白质来源, 幼龄雪多牦牛肉中必需氨基酸和总氨基酸含量为5.95%和14.00%, 均显著高于成年雪多牦牛肉, 但成年雪多牦牛肉味更鲜美。幼龄雪多牦牛肉的脂肪酸构成略优于成年雪多牦牛肉。在维生素和矿物质组成和含量上, 幼龄雪多牦牛肉的维生素A显著高于成年雪多牦牛肉。成年雪多牦牛肉的碘含量显著高于幼龄雪多牦牛肉。结论 幼龄雪多牦牛肉的脂肪酸和氨基酸构成更加合理健康, 色泽、口感和嫩度更好, 因此幼龄雪多牦牛肉品质比成年雪多牦牛肉好。幼龄雪多牦牛生长周期短, 能够在有限的牧草资源下创造更高的经济价值, 同时可以提高牦牛肉的市场竞争力, 对牧区的牦牛养殖提供科学有效的指导。

目的 研究成年和幼龄雪多牦牛肉品质差异。方法 选择健康的成年和幼龄阉割雪多牦牛, 对其背最长肌常规营养成分、食用肉品质、氨基酸、脂肪酸、维生素和矿物质构成与含量进行分析。结果 成年雪多牦牛肉水分含量为72.89%, 低于幼龄雪多牦牛肉; 成年雪多牦牛肉粗蛋白质、粗脂肪和粗灰分含量分别比幼龄雪多牦牛肉高4.99%、86.67%、38.54%, 因此成年雪多牦牛肉具有更高的营养价值。幼龄雪多牦牛肉食用肉品质极佳, 色泽、口感、嫩度等都优于成年雪多牦牛肉。成年和幼龄雪多牦牛肉都是优质蛋白质来源, 幼龄雪多牦牛肉中必需氨基酸和总氨基酸含量为5.95%和14.00%, 均显著高于成年雪多牦牛肉, 但成年雪多牦牛肉味更鲜美。幼龄雪多牦牛肉的脂肪酸构成略优于成年雪多牦牛肉。在维生素和矿物质组成和含量上, 幼龄雪多牦牛肉的维生素A显著高于成年雪多牦牛肉。成年雪多牦牛肉的碘含量显著高于幼龄雪多牦牛肉。结论 幼龄雪多牦牛肉的脂肪酸和氨基酸构成更加合理健康, 色泽、口感和嫩度更好, 因此幼龄雪多牦牛肉品质比成年雪多牦牛肉好。幼龄雪多牦牛生长周期短, 能够在有限的牧草资源下创造更高的经济价值, 同时可以提高牦牛肉的市场竞争力, 对牧区的牦牛养殖提供科学有效的指导。

Objective To study the difference of meat quality between adult and young Xueduo yaks. Methods Healthy adult and young castrated Xueduo yaks were selected to analyze the composition and content of conventional nutrients, edible meat quality, amino acids, fatty acids, vitamins and minerals in longissimus dorsi muscle. Results The moisture content of adult Xueduo yak meat was 72.89%, which was lower than that of young Xueduo yak meat. The crude protein, crude fat and crude ash contents of adult Xueduo yak meat were 4.99%, 86.67% and 38.54% higher than those of young Xueduo yak meat, respectively. Therefore, adult Xueduo yak meat had higher nutritional value. The edible meat quality of young Xueduo yak meat was excellent, and the color, taste and tenderness were better than those of adult Xueduo yak meat. Both adult and young Xueduo yak meat were high-quality protein sources. The contents of essential amino acids and total amino acids in young Xueduo yak meat were 5.95% and 14.00%, which were significantly higher than those in adult Xueduo yak meat, but adult Xueduo yak meat tastes more delicious. The fatty acid composition of young Xueduo yak meat was slightly better than that of adult Xueduo yak meat. In terms of vitamin and mineral composition and content, vitamin A in young Xueduo yak meat was significantly higher than that in adult Xueduo yak meat. The iodine content of adult Xueduo yak meat was significantly higher than that of young Xueduo yak meat. Conclusion The meat quality of young Xueduo yak is better than that of adult Xueduo yak. The growth cycle of young Xueduo yak is short, which can create higher economic value under limited forage resources. At the same time, it can improve the market competitiveness of yak meat and provide scientific and effective guidance for yak breeding in pastoral areas.

牦牛主要分布在青藏高原地区, 是高原特有种畜资源, 有“雪域之舟”的美称[1]。作为高原牧民的重要生活资产, 其肉、乳、毛、粪等均在高原牧民的生产生活中占据重要地位, 对实现高原地区的生态价值、社会价值以及经济价值等都发挥着不可替代的作用[2]。在2021版《国家畜禽遗传资源品种名录》中, 总计收录地方牦牛品种18个, 培育品种2个, 其中, 雪多牦牛于2017年接受国家畜禽遗传资源委员会办公室鉴定, 在2018年进入国家畜禽遗传资源目录[3]。雪多牦牛来自青海省河南蒙古自治县海拔3 km以上的山地高寒草甸地区, 属于偏肉用的肉乳兼用型牦牛。雪多牦牛是经过长期自然选择和人工选育而逐渐形成的地方遗传资源, 因其品质优良, 雪多牦牛肉素有“牛中鲜”之称[4-5]。由于雪多牦牛品种认定比较迟, 目前只是从分子水平上探究了雪多牦牛的母系遗传多样性及遗传背景, 而对雪多牦牛肉品质的研究较少, 缺乏年龄对雪多牦牛肉品质影响的相关研究, 因此本研究选择青海省黄南州河南县塞尔龙乡及其周边地区不同年龄的雪多牦牛肉, 分析其常规营养品质、食用肉品质、矿物质含量、维生素含量、氨基酸及脂肪酸组成, 对不同年龄雪多牦牛肉品质进行评估, 为雪多牦牛肉的推广提供理论基础。
在青海省黄南州河南县塞尔龙乡及其周边地区, 随机选取放牧条件下发育正常、健康无病的4岁和1岁阉割雪多牦牛各10头。屠宰前禁食12 h, 停水2 h。集中于雪多肉类加工车间屠宰, 宰后进行部分指标的测定并采集每头牛的背最长肌4 kg, 分块置于洁净保鲜袋中密封, 于-20 ℃冷冻保存待测。另取50 g背最长肌切成1 cm3方丁, 装入100 mL小瓶封口冻结待测。
石油醚(沸点40~60 ℃, 济南世纪通达化工有限公司); 纯盐酸(37%)、纯硫酸(98%)、柠檬酸钠、甲醛(10%)(青海利宝商贸有限公司); Ca标准溶液、Se标准溶液、Mg、I、Zn、Fe、Mn、Cu (1000 μg/mL, 北京有色金属研究总院)。
Testo205便携式肉质pH计(德国Testo公司); NS800分光测色仪(广东深圳3nH公司); DT-607肉品压力仪、DT-609肉品剪切力仪(山东济南德天公司); 7890A-5975C气相色谱-质谱联用仪、G1701EA质谱工作站(美国Agilent公司); Synergy 2多功能酶标仪(美国BioTek公司); 岛津835型氨基酸分析仪(日本岛津公司); 原子吸收分光光度计(北京TAS-990公司)。
水分、粗灰分、粗脂肪、粗蛋白质分别按照GB 5009.3—2016《食品安全国家标准 食品中水分的测定》、GB 5009.4—2016《食品安全国家标准 食品中灰分的测定》、GB 5009.6—2016《食品安全国家标准 食品中脂肪的测定》、GB 5009.5—2016《食品安全国家标准 食品中蛋白质的测定》进行测定。干物质计算如公式(1):
干物质/%=(烘干前样品重-烘干后样品重)/烘干前样品重×100%
使用CR-10色差仪, 在屠宰后10 min内将校正好的色差仪镜头置于背最长肌新鲜横切面, 镜口紧扣肉面不漏光, 随机在每个样品3个不同位置各测定1次, 测定亮度(L*)、红色度(a*)和黄色度(b*)值, 取平均值。
使用美式标准图谱, 将第1腰椎处的背最长肌, 在0~4 ℃条件下冷却24 h, 取新鲜横切面与美式标准图谱对照打分。只有痕迹为1分, 微量2分, 少量3分, 适量4分, 过量5分, 3分和4分说明肉品质好, 过低过高均差。
宰后1 h和24 h的pH1 h和pH24 h: 按照GB 5009.237—2016《食品安全国家标准 食品pH值的测定》测定。
宰后2 h内, 在第1、2腰椎处的背最长肌上, 垂直截取厚度为1.0 cm的肉片, 在其中央用直径为2.523 cm, 面积为5.0 cm2的取样器在不吸水的硬橡胶板上截取测定样, 称重后(记为W1)在肉样上下各垫16层吸水纸, 置于应变式无侧限压缩平台上, 施加35 kg (68.66 kPa)的压力, 保持5 min, 再称重(记为W2)。每个肉样至少做3次重复, 求平均值。计算见公式(2):

取第1、2腰椎处背最长肌用烘干法测得含水率后计算。计算见公式(3):

切取一定大小的肉样称重(m1), 在80 ℃水浴锅中加热30 min后, 冷却到室温, 用吸水纸吸干表面水分, 再次称质量(m2), 计算如公式(4):
蒸煮损失/%=(m1-m2)/m1×100%
取第1、2腰椎处背最长肌的肌芯, 用10%甲醛溶液固定48 h, 剥离肌纤维制成4 μm的石蜡切片。石蜡切片脱蜡复水后, 按照苏木精-伊红染色试剂盒说明书进行染色。染色结束后于在400倍显微镜下用测微尺测定。每个样品做3个重复, 每个重复测100根肌纤维, 计算平均数。
在第12~13胸椎处截取厚度为3 cm的背最长肌, 装入塑料薄膜袋, 在80 ℃的水浴锅中恒温加热至中心温度75 ℃, 冷却至室温后用直径为1.27 cm的取样器钻取肉柱, 用C-LM型嫩度仪测定剪切力值, 每个样品平行测定3次取平均值。
全氨基酸试样制备: 称取约5 g的样品, 置于水解管中, 加入与试样等量的盐酸溶液, 水解管在真空装置中脱气后, 在水解管A处用煤气灯熔化, 使其密封, 在110 ℃恒温器水解22 h后将水解管开封, 用旋转蒸发器去除盐酸, 干涸后, 用蒸馏水溶解, 再次干涸(在50 ℃以下干燥), 盐酸试样去除, 溶于10 mL柠檬酸钠缓冲液中备用。参照GB 5009.124—2016, 雪多牦牛肉用6 mol/L的盐酸进行水解, 测定肉中16种氨基酸的含量; 再用4.2 mol/L NaOH对牦牛肉进行水解, 测定肉中色氨酸含量; 牦牛肉使用过甲酸氧化法处理, 测定肉中胱氨酸的含量。
氨基酸测定: 水解后的样品用岛津835型氨基酸分析仪测定。
按照GB 5009.168—2016《食品安全国家标准 食品中脂肪酸的测定》第二法外标法测定脂肪酸含量。
维生素A、D、E、B1、B2分别按照GB 5009.82—2016《食品安全国家标准 食品中维生素A、D、E的测定》、GB 5009.84—2016《食品安全国家标准 食品中维生素B1的测定》、GB 5009.85—2016《食品安全国家标 准食品中维生素B2的测定》进行测定。
按照GB 5009.268—2016《食品安全国家标准 食品中多元素的测定》使用原子吸收分光光度法测定矿物质含量。
数据均采用Excel 2016 MSO进行简单统计, 再使用SPSS 27.0进行统计分析, 采用独立样本t检验进行显著性分析, P<0.05表示差异显著, P<0.01表示差异极显著。结果均以“平均值±标准偏差”表示。
表1所示, 成年雪多牦牛肉水分极显著低于幼龄雪多牦牛肉(P<0.01)。成年雪多牦牛肉粗蛋白质、粗脂肪和粗灰分含量分别比幼龄雪多牦牛肉高4.99%、86.67%、38.54%, 差异显著(P<0.05)。
表2所示, 不同年龄阶段雪多牦牛肉的L*a*b*和大理石纹等指标差异显著(P<0.05)。其中, L*越高表示肉色色泽越亮, a*越高表示肉色越偏红, b*越高表示肉色越偏黄。幼龄雪多牦牛肉的L*a*b*分别比成年雪多牦牛高2.04、1.55和1.56。而成年雪多牦牛肉的大理石纹评分高于幼龄雪多牦牛肉0.27分。不同年龄阶段雪多牦牛肉的pH1 h存在极显著差异(P<0.01), 成年雪多牦牛肉的pH1 h比幼龄雪多牦牛肉高0.63, 经过时间的推移, 两个年龄阶段的雪多牦牛肉的pH都在不断降低, 幼龄雪多牦牛从pH1 h到pH24 h平均下降了0.39, 而成年雪多牦牛从pH1 h到pH24 h平均下降了1.08, 成年雪多牦牛肉的pH下降更快, 但pH24 h指标在不同年龄阶段雪多牦牛肉上差异不显著(P>0.05)。不同年龄阶段雪多牦牛肉的失水率、蒸煮损失、肌纤维直径和剪切力存在极显著差异(P<0.01)。其中, 幼龄雪多牦牛肉的失水率平均高于成年雪多牦牛肉20.67%, 而成年雪多牦牛肉的蒸煮损失、肌纤维直径和剪切力分别高于幼龄雪多牦牛肉6.82%、162.88%和61.14%。
成年雪多牦牛肉中色氨酸(tryptophan, Trp), 甘氨酸(glycine, Gly), 丙氨酸(alanine, Ala)均极显著高于幼龄雪多牦牛肉(P<0.01), 而缬氨酸(valine, Val), 异亮氨酸(isoleucine, Ile), 组氨酸(histidine, His)含量均极显著低于幼龄雪多牦牛肉(P<0.01), 苯丙氨酸(phenylalanine, Phe)含量显著低于幼龄雪多牦牛肉(P<0.05)。其他氨基酸在幼龄雪多牦牛肉和成年雪多牦牛肉中含量差异不显著(P>0.05)。非必需氨基酸(nonessential amino acid, NAA)的含量在两个年龄阶段雪多牦牛肉中差异不显著(P<0.05), 幼龄雪多牦牛肉中的必需氨基酸(eessential amino acid, EAA)、氨基酸总量(total amino acid, TAA)以及EAA/NAA和EAA/TAA均显著高于成年雪多牦牛肉(P<0.05), 其中EAA和TAA分别高出0.70和0.71, 如表3所示。
成年雪多牦牛肉中十八碳-9,12-二烯酸(亚油酸)极显著高于幼龄雪多牦牛肉(P<0.01), 而十八碳-11-烯酸、十八碳-9,12,15-三烯酸(α-亚麻酸)、二十碳-5,8,11-三烯酸、二十碳-7,10,13-三烯酸、二十碳-5,8,11,14-四烯酸(花生四烯酸)、二十碳-5,8,11,14,17-五烯酸均极显著低于幼龄雪多牦牛肉(P<0.01)。整体来看, 成年雪多牦牛肉饱和脂肪酸(saturated fatty acid, SFA)含量极显著高于幼龄雪多牦牛肉(P<0.01), 单不饱和脂肪酸(monounsaturated fatty acid, MUFA)含量与幼龄雪多牦牛肉差异不显著(P>0.05); 成年雪多牦牛肉中多不饱和脂肪酸(polyunsaturated fatty acid, PUFA)、PUFA/SFA (P:S)、n-3系列不饱和脂肪酸含量均极显著低于幼龄雪多牦牛肉, 而n-6系列不饱和脂肪酸、n-6/n-3及致动脉粥样硬化指数(arteriosclerosis index, AI)均显著高于幼龄雪多牦牛肉(P<0.05), 如表4所示。
表5所示, 成年雪多牦牛肉与幼龄雪多牦牛肉中维生素B1、维生素B2、维生素E含量差异均不显著(P>0.05)。幼龄雪多牦牛肉中维生素A含量是成年雪多牦牛肉的13倍, 差异显著(P<0.05)。
表6所示, 不同年龄阶段雪多牦牛肉的硒、锌、铁、锰、镁、钙、铜等矿物质元素含量差异不显著(P>0.05), 但成年雪多牦牛肉中锌的含量相比幼龄雪多牦牛高7.96%, 有显著差异趋势。不同年龄雪多牦牛肉碘的含量存在极显著差异, 成年雪多牦牛肉的碘含量平均比幼龄雪多牦牛肉高出12.17%。
牦牛肉中水分占比最高, 其次是粗蛋白质和粗脂肪, 营养成分的含量受牦牛品种、饲养方式、年龄和性别等因素影响[6]。徐瑛等[7]研究发现随着放牧牦牛年龄增长, 牦牛肉中水分含量呈显著下降趋势。本研究中, 幼龄雪多牦牛肉水分极显著高于成年雪多牦牛肉, 该结果与上述研究结果一致。蛋白质是牦牛肉养分中的第二大营养素, 本研究中成年雪多牦牛肉中蛋白质含量显著高于幼龄雪多牦牛肉(P<0.05), 该结果与WAN等[8]报道的结论一致。脂肪与牦牛肉的嫩度相关, 是影响牦牛肉口感的重要指标之一。本研究中, 成年雪多牦牛肉脂肪含量极显著高于幼龄雪多牦牛肉(P<0.05)。SOJI等[9]认为动物在青年时期生长发育较快, 前期主要是蛋白质沉积, 性成熟过后生长速度变缓, 逐渐由原先蛋白质沉积转为脂肪沉积为主, 因此在放牧条件下随着年龄增长, 牦牛肉中肌内脂肪含量逐渐增加; 徐瑛[10]在研究年龄对牦牛肉肉用品质的实验中指出, 随着放牧牦牛年龄的增长, 牦牛肉中水分的含量显著降低, 而粗脂肪和粗蛋白质含量显著增加(P<0.05); 以上研究结果均与本研究结果一致。综上所述, 成年雪多牦牛肉虽然水分含量较低, 但其蛋白质、脂肪水平显著高于幼龄雪多牦牛肉, 具有更高的营养价值, 其常规营养成分更佳。
肉色和大理石花纹是消费者在购买牛肉产品时最直观的评价标准。本研究对不同年龄阶段雪多牦牛肉进行色度评价, 结果表明幼龄雪多牦牛肉的色泽更亮, 且颜色更深。大理石花纹是因脂肪沉积在横断面上而表现出来的纹理, 主要受肌间脂肪含量和分布的影响[11]。本研究中, 成年雪多牦牛肉的大理石花纹评分显著高于幼龄雪多牦牛肉, 主要原因可能是幼龄雪多牦牛肉中脂肪含量较低, 肌间脂肪沉积较少, 但两者的大理石花纹评分均在3分以上, 处于理想的评分区间内。pH是判断牛肉新鲜程度的重要指标, 通常以pH1 h<6.0作为白肌肉(pale, soft, exudative meat, PSE)肉的判定依据, 以pH24 h>6.5作为判定黑干肉(dark, firm, dry meat, DFD)的依据[12]。本研究中, 成年雪多牦牛肉的pH1 h显著高于幼龄雪多牦牛肉, 但两者的pH24 h差异不显著, 两者的pH1 h和pH24 h均处于正常范围, 说明不同年龄阶段雪多牦牛肉的肉质不属于PSE肉和DFD肉, 这与何文斌等[13]研究一致。
失水率、蒸煮损失和剪切力等物理特性都是评断牛肉食用品质的重要指标。失水率可以较为直接地反映牦牛肉的保水能力, 失水率过高会使牦牛肉中总色素含量流失过多, 最终导致肉色变淡。在本研究中, 幼龄雪多牦牛肉的失水率显著升高, 主要原因可能是幼龄雪多牦牛肉中水分含量较高, 更容易失去水分。蒸煮损失也能直接反映肉产品保水能力, 蒸煮损失越高, 其熟制之后所流失的水分越少, 就能使咀嚼时更好地呈现多汁状态。本研究中成年雪多牦牛肉的蒸煮损失显著高于幼龄雪多牦牛肉, 这与常规养分中其水分也较高相对应。剪切力是直接将嫩度指标数值化的一项指标, 通常和嫩度指标呈现负相关, 即剪切力越低, 嫩度越好。DESTEFANIS等[14]通过研究嫩度与剪切力之间的关系, 认为剪切力小于42.72 N时肉质较嫩, 小于52.63 N时嫩度在可接受范围内。本研究中, 幼龄雪多牦牛剪切力平均可达3.5 kg·f(约34.3 N), 而成年雪多牦牛的剪切力可达5.64 kg·f(约55.27 N), 表明幼龄雪多牦牛肉的嫩度较佳, 而成年雪多牦牛肉的嫩度超出了可接受范围, 这与赵洪文等[15]研究结果一致。肌纤维直径的粗细对嫩度有着一定的影响, 一般而言, 肌纤维直径越细, 肌肉的嫩度越好[16]。本研究中, 成年雪多牦牛肉的肌纤维直径平均可达60.62 μm, 而幼龄雪多牦牛肉肌纤维直径平均只有23.06 μm。这说明幼龄雪多牦牛肉的肌纤维更细, 其嫩度更好, 这与徐瑛等[7]和田帅等[17]结果一致。
综上所述, 虽然幼龄雪多牦牛的蒸煮损失显著低于成年雪多牦牛肉, 但仍具有良好的加工性能和生产效益。幼龄雪多牦牛肉食用肉品质极佳, 色泽、口感、嫩度等都优于成年雪多牦牛肉, 表明随着年龄增长, 雪多牦牛肉的食用肉品质逐渐下降。
蛋白质是构成人体生命活动的物质基础, 与人体健康密切相关, 蛋白质品质主要由氨基酸组成决定。本研究从牦牛肉中共检测到18种氨基酸, 其中EAA 8种, NAA 10种。成年雪多牦牛肉和幼龄雪多牦牛肉中EAA/TAA、EAA/NAA分别为0.39、0.43和0.66、0.74, 均符合联合国粮食及农业组织(Food and Agriculture Organization of the United Nations, FAO)和世界卫生组织(World Health Organization, WHO)标准(EAA/TAA推荐值40%左右; EAA/NAA推荐值60%以上), 这与董瑷榕等[18]和洪晶等[19]的研究结果基本一致, 说明雪多牦牛肉是优质蛋白质来源。刘海珍[20]研究表明, 牦牛肉中氨基酸的含量非常稳定, 与部位和品种无关。鲜味氨基酸由天冬氨酸、精氨酸、谷氨酸、丙氨酸和甘氨酸组成, 它的含量能够影响肉的滋味[21]。本研究中成年雪多牦牛肉中鲜味氨基酸含量为5.57%, 而幼龄雪多牦牛肉中鲜味氨基酸仅为5%, 由此可见, 成年雪多牦牛肉的鲜味更浓, 这与马骋等[22]的研究中随着年龄的增长雪多牦牛肉中的鲜味氨基酸也随之增长的这一结果一致。此外, 本研究中幼龄雪多牦牛肉中EAA和TAA含量均显著高于成年雪多牦牛肉, 这表明幼龄雪多牦牛肉氨基酸组成更为平衡, 与董瑷榕等[18]研究中1.5岁牦牛肉氨基酸含量高于4.5岁牦牛肉的结果一致。
综上所述, 幼龄雪多牦牛肉在氨基酸组成和含量上优于成年雪多牦牛肉, 而成年雪多牦牛肉的鲜味氨基酸显著多于幼龄雪多牦牛肉, 即有更好的风味。
脂肪酸是人体必需的营养素, 最新研究也表明适宜的脂肪酸数量和组成对调节人体健康的肠道菌群平衡具有重要作用, 可以提高肠道益生菌数量, 增强人体免疫力[21]。MUFA有降血糖、保护心脏、调节血脂等生理功能, PUFA具有保护视力、降胆固醇、抗炎等作用[23]。本研究对不同年龄雪多牦牛肉分析, 共检测出了21种脂肪酸, 其中包括油酸、亚油酸、α-亚麻酸、花生四烯酸、二十碳五烯酸(eicosapentaenoic acid, EPA)等在内的PUFA含量均较高, PUFA有着积极的生物学功能, 以n-6系和n-3系PUFA为主的必需脂肪酸调节着机体内的许多代谢过程[24]n-3和n-6系列脂肪酸为典型的功能性脂肪酸, 对于人体健康有积极作用。医学研究表明, n-6和n-3的比值应在4:1或者更低[25]。本研究中成年雪多牦牛肉中n-6系列脂肪酸含量显著高于幼龄雪多牦牛肉(P<0.05), 而n-3系列脂肪酸含量极显著低于幼龄雪多牦牛肉(P<0.01), 成年和幼龄雪多牦牛肉的n-6/n-3分别为2.90和1.47, 均在理想范围内。张明等[26]的研究表明, 随着反刍动物年龄增长, 氢化能力也随之加强, 会将体内更多的不饱和脂肪酸转换成SFA, 从而导致肉中SFA含量升高。本研究中, 幼龄雪多牦牛肉中PUFA含量极显著高于成年雪多牦牛肉, 这与上述研究相符, 罗毅皓[27]在实验中也有相同结果。PUFA:SFA是衡量膳食中脂肪酸营养价值的重要指标, WHO推荐的PUFA:SFA值要高于0.4[28], 而本研究中成年和幼龄雪多牦牛肉中PUFA:SFA值分别为0.27和0.36, 这低于推荐值, 与董瑷榕等[18]研究果一致。致动脉粥样硬化指数(AI)能反映患心血管疾病, 动脉粥样硬化的风险, 本研究中成年雪多牦牛肉AI极显著高于幼龄雪多牦牛肉, 再次说明幼龄雪多牦牛肉更为健康。
综上所述, 雪多牦牛肉中脂肪酸构成与人体膳食均衡脂肪酸比例存在一定差距, 虽未达到最佳比例, 但仍处在正常范围内, 幼龄雪多牦牛肉略健康于成年雪多牦牛肉, 但差异不大。
牦牛肉是高原牧民获取维生素的主要途径之一, 而维生素与人体健康密切相关, 如维生素A与人眼视力, 维护上皮细胞, 增强机体免疫功能等有关[29]; 维生素E具有抗氧化、促进性腺激素分泌的功能[30]。维生素B1参与能量代谢, 在维持神经、肌肉特别是心肌的正常功能, 以及维持正常食欲、胃肠蠕动和消化分泌等方面也具有重要作用[31]。维生素B2缺乏时, 会影响机体的生物氧化, 使代谢发生障碍[32]。本研究中成年雪多牦牛肉中维生素A显著低于幼龄雪多牦牛肉(P<0.05), 而维生素B1、维生素B2、维生素E均与幼龄雪多牦牛肉差异不显著, 而刘勇[33]研究结果表明, 犊牦牛肉中维生素B1和维生素B2显著高于成年牦牛肉, 与本研究不一致, 这可能是牦牛饲喂方式不同导致, 本研究是自由放牧, 刘勇的研究则是半放牧半饲养。
年龄对雪多牦牛肉中大部分矿物质元素含量的影响不显著, 本研究对硒、碘、锌、锰、镁、钙等矿物质元素进行了测定, 结果表明成年雪多牦牛肉中碘元素含量显著高于幼龄雪多牦牛肉, 碘可以通过摄食消化吸收从而储存在甲状腺中参与甲状腺素的合成, 成年雪多牦牛通过进食沉积了更多的碘, 而其他矿物质元素在不同年龄阶段雪多牦牛肉中均不存在显著差异[34]。一般认为, 牦牛肉中矿物质元素含量的差异与牦牛的生存环境和采食种类有关, 邱翔等[35]对多种主要黄牛肉中的矿物质元素进行测定, 与其结果对比, 雪多牦牛肉中多种矿物质元素含量均高于普通牛肉, 这可能与青藏高原地区土地和牧草多含矿物质相关。
成年雪多牦牛肉尽管营养价值相对较高, 但养殖周期较长, 且肉质嫩度较低, 使牦牛肉生产环节的经济效益较差, 无法充分发挥资源优势。相比成年雪多牦牛肉, 幼龄雪多牦牛肉的脂肪酸和氨基酸构成更加合理健康, 色泽、口感和嫩度更好。幼龄雪多牦牛饲养周期短, 饲喂成本低, 经济效益更好, 解决了成年牦牛肉市场的诸多局限性, 为牦牛肉提升市场竞争力提供了合理途径和理论基础。
