Objective To establish an analytical method for determining γ-amino butyric acid content in foods of plant origin by high performance liquid chromatography. Method Foods samples were pre-column derivatized with 4-dimethylamino-azobenzene 4-sulfonyl chloride, orthogonal experiments were designed and methodological verification was carried out based on extraction solvent, solid-liquid ratio, extraction method and extraction time. The results of γ-amino butyric acid were analyzed by detecting different plant-derived foods and comparing with instruments. Results The results showed that the optimal conditions were determined as 60% ethanol solution, which was extracted by water bath (40 ℃) or ultrasonic for 30 min at 1:25 (g:mL) solid-liquid ratio. The separation of γ-amino butyric acid was achieved on C18 column (250 mm×4.6 mm, 5 μm) using acetonitrile+sodium acetate solution (20:80, V:V) as the mobile phase at a flow rate of 1.0 mL/min. The absorption wavelength of γ-amino butyric acid was 436 nm and column temperature was set at 30 ℃, respectively. The linear range of γ-amino butyric acid was 1.092-131.064 μg/mL. The relative standard deviation of γ-amino butyric acid was all less than 5.0% in pitaya (white heart), tomato, and malt. The average recovery rate of γ-amino butyric acid was between 95.80%-104.36%, indicating that the method had good precisions and high recoveries. Conclusion The method established in this study has good repeatability and high accuracy, and can accurately quantify the content of γ-amino butyric acid in foods of plant origin.

目的 建立高效液相色谱法测定植物源性食品中γ-氨基丁酸含量的分析方法。方法 以4-二甲基胺基偶氮苯-4-磺酰氯为柱前衍生试剂, 以提取溶剂、料液比、提取方式和提取时间为考察因素, 设计正交实验并进行方法学验证。通过检测不同植物源性食品、仪器比对测定, 分析γ-氨基丁酸测定结果。结果 确定提取γ-氨基丁酸最优条件为60%乙醇溶液, 按1:25 (g:mL)的料液比, 通过水浴振荡(40 ℃)或超声提取30 min。色谱条件: C18色谱柱(250 mm×4.6 mm, 5 μm), 柱温30 ℃, 流动相为乙腈+乙酸钠溶液(20:80, V:V), 流速1.0 mL/min, 检测波长为436 nm。γ-氨基丁酸在1.092~131.064 μg/mL范围内与峰面积具有良好的线性关系。火龙果(白心)、番茄和麦芽中γ-氨基丁酸精密度的相对标准偏差均小于5.0%, 加标回收率均在95.80%~104.36%之间, 表明方法精密度良好, 添加回收率高。结论 本研究建立的方法重复性好、准确度高, 可以准确定量植物源性食品中γ-氨基丁酸的含量。

目的 建立高效液相色谱法测定植物源性食品中γ-氨基丁酸含量的分析方法。方法 以4-二甲基胺基偶氮苯-4-磺酰氯为柱前衍生试剂, 以提取溶剂、料液比、提取方式和提取时间为考察因素, 设计正交实验并进行方法学验证。通过检测不同植物源性食品、仪器比对测定, 分析γ-氨基丁酸测定结果。结果 确定提取γ-氨基丁酸最优条件为60%乙醇溶液, 按1:25 (g:mL)的料液比, 通过水浴振荡(40 ℃)或超声提取30 min。色谱条件: C18色谱柱(250 mm×4.6 mm, 5 μm), 柱温30 ℃, 流动相为乙腈+乙酸钠溶液(20:80, V:V), 流速1.0 mL/min, 检测波长为436 nm。γ-氨基丁酸在1.092~131.064 μg/mL范围内与峰面积具有良好的线性关系。火龙果(白心)、番茄和麦芽中γ-氨基丁酸精密度的相对标准偏差均小于5.0%, 加标回收率均在95.80%~104.36%之间, 表明方法精密度良好, 添加回收率高。结论 本研究建立的方法重复性好、准确度高, 可以准确定量植物源性食品中γ-氨基丁酸的含量。

Objective To establish an analytical method for determining γ-amino butyric acid content in foods of plant origin by high performance liquid chromatography. Method Foods samples were pre-column derivatized with 4-dimethylamino-azobenzene 4-sulfonyl chloride, orthogonal experiments were designed and methodological verification was carried out based on extraction solvent, solid-liquid ratio, extraction method and extraction time. The results of γ-amino butyric acid were analyzed by detecting different plant-derived foods and comparing with instruments. Results The results showed that the optimal conditions were determined as 60% ethanol solution, which was extracted by water bath (40 ℃) or ultrasonic for 30 min at 1:25 (g:mL) solid-liquid ratio. The separation of γ-amino butyric acid was achieved on C18 column (250 mm×4.6 mm, 5 μm) using acetonitrile+sodium acetate solution (20:80, V:V) as the mobile phase at a flow rate of 1.0 mL/min. The absorption wavelength of γ-amino butyric acid was 436 nm and column temperature was set at 30 ℃, respectively. The linear range of γ-amino butyric acid was 1.092-131.064 μg/mL. The relative standard deviation of γ-amino butyric acid was all less than 5.0% in pitaya (white heart), tomato, and malt. The average recovery rate of γ-amino butyric acid was between 95.80%-104.36%, indicating that the method had good precisions and high recoveries. Conclusion The method established in this study has good repeatability and high accuracy, and can accurately quantify the content of γ-amino butyric acid in foods of plant origin.

γ-氨基丁酸(γ-amino butyric acid), 化学名称为4-氨基丁酸, 别名叫γ-氨基丁酸、氨酪酸、哌啶酸, 广泛分布于动植物体内[1-3]。在植物中, 如豆属、参属、中草药等的种子、根茎和组织液中都含有γ-氨基丁酸[4-7]; 在动物体内, γ-氨基丁酸几乎只存在于神经组织中, 其中脑组织中的含量大约为0.1~0.6 mg/g组织, 浓度最高的区域为大脑中黑质[8-10]
γ-氨基丁酸是研究较为深入的一种重要的抑制性神经递质, 可以抑制动物的活动, 减少能量的消耗[11-13]。此外, 它还参与多种代谢活动, 具有镇静[14-17]、催眠[18-22]、抗惊厥[23-25]、降血压[26-28]、治疗癫痫和改善记忆力[29-31]等生理活性。因此, 研究γ-氨基丁酸的检测方法对指导生产生活具有重要意义。
NY/T 2890—2016《稻米中γ-氨基丁酸的测定 高效液相色谱法》、QB/T 5633.7—2022《氨基酸、氨基酸盐及其类似物 第7部分: γ-氨基丁酸》等国内相关γ-氨基丁酸检测方法中, NY/T 2890—2016只规定了稻米中γ-氨基丁酸的测定, 不适用于谷物、蔬菜、水果等植物源性食品中γ-氨基丁酸含量的测定, 采用此方法测定的植物源性食品中γ-氨基丁酸含量, 测定结果不能满足现阶段检测需求。国外方面, 国际标准化组织(International Organization for Standardization, ISO)、政府农业化学师学会(Association of Official Agricultural Chemists, AOAC)、美国食品和药物管理局(Food and Drug Administration, FDA)及欧洲食品安全局(European Food Safety Authority, EFSA)等相关检测标准, 均未涉及到γ-氨基丁酸测定方法。
γ-氨基丁酸对电化学、紫外和可见光不灵敏, 用直接方法测定比较困难。目前, 国内外测定γ-氨基丁酸的检测方法主要有: 氨基酸分析仪法、酶法、色谱-质谱法、柱层析荧光测定法、纸电泳比色法、毛细管气相色谱法、放射性受体法、毛细管电泳法、高效液相色谱法等[32-37]。氨基酸分析仪法由于仪器设备价格昂贵, 仪器应用领域也仅限制于氨基酸相关项目检测等方面检测, 不适用于大批量定量分析; 色谱-质谱法虽操作简便、分离效果良好, 但其成本较高; 比色法样液中其他游离氨、色素以及显色液稳定性等对测定结果有影响; 柱层析荧光测定法操作步骤相对复杂, 荧光信号容易受到环境因素, 如温度、光线、污染物等的影响。因此, 本研究建立高效液相色谱法检测植物源性食品中γ-氨基丁酸含量的分析方法, 通过检验不同的植物源性样品, 分析γ-氨基丁酸的含量, 为研究食品质量、营养价值评价和技术开发利用提供支持。
香菇、平菇、油菜、蒜苔、黄瓜、西葫芦、西兰花、大白菜、菜花、菠菜、架豆角、胡萝卜、紫洋葱、丝瓜、莲藕、青萝卜、冬瓜、番茄、土豆、南瓜、麦芽、紫甘蓝、芹菜、大葱、绿豆芽、圆白菜、李子、绿香瓜、蜜瓜、猕猴桃、杏子、蓝莓、荔枝、香蕉、火龙果(白心)、水蜜桃、梨、橙子、柠檬、芒果、油桃、木瓜、杨梅、樱桃、草莓、苹果、小米、大豆样品, 市售。
γ-氨基丁酸标准品(纯度≥99.9%, 美国Sigma公司); 甲醇、乙腈、乙醇(色谱纯, 上海Anpel公司); 碳酸氢钠、三水合乙酸钠(分析纯, 北京国药集团化学试剂有限公司); 4-二甲基胺基偶氮苯-4-磺酰氯(上海源叶生物公司)。
BS224S分析天平[精度0.1 mg, 德国赛多利斯科学仪器(北京)有限公司]; Centrifuge 5420高速冷冻离心机(德国艾本德公司); KQ-500DE型数控超声波清洗器(舒美昆山市超声仪器有限公司); DIONEX ULTRAMATE U-3000高效液相色谱仪(配有二极管阵列检测器或性能相当的检测器)[赛默飞世尔科技(中国)有限公司]。
标准储备液(1000 μg/mL): 称取γ-氨基丁酸标准品10.0 mg(精度0.1 mg), 用无水乙醇溶解后, 转移到10 mL棕色容量瓶中, 用无水乙醇定容至刻度。标准储备液贮存于密闭棕色玻璃瓶中, 于-18 ℃中冷冻保存, 有效期3个月。
4-二甲基胺基偶氮苯-4-磺酰氯溶液(2.0 mg/mL): 称取4-二甲基胺基偶氮苯-4-磺酰氯20.0 mg, 用乙腈溶解后, 转移至10 mL棕色容量瓶中, 用乙腈定容, 混匀, 现配现用。
乙醇水溶液: 无水乙醇+水(6:4, V:V), 取无水乙醇600 mL和水400 mL混匀。
碳酸氢钠溶液(0.04 g/mL): 称取碳酸氢钠0.40 g, 用水溶解后, 转移到10 mL容量瓶中, 用水定容, 混匀, 现配现用。
乙酸钠溶液(6.8 mg/mL): 称取三水合乙酸钠3.40 g, 用水溶解并稀释至500 mL, 经微孔滤膜(0.45 μm)过滤。
称取干样品1.0~2.0 g, 鲜样品2.0~5.0 g(样品精确至0.001 g)于50 mL离心管中, 加入乙醇水溶液10 mL, 超声提取30 min后, 静置5 min, 于5000 r/min离心5 min, 将上清液转入25 mL容量瓶中, 样品残渣再用10 mL提取溶液提取一次, 合并2次提取液, 用乙醇水溶液定容至25 mL, 摇匀, 待衍生化。
准确吸取1 mL试样溶液或标准工作溶液于具塞试管中, 加入0.2 mL碳酸氢钠溶液和0.4 mL 4-二甲基胺基偶氮苯-4-磺酰氯衍生试剂, 混匀后在70 ℃水浴中衍生反应20 min, 用微孔滤膜(0.45 μm)过滤, 待测。
确定色谱条件为: C18柱(250 mm×4.6 mm, 5 μm), 或具同等性能的色谱柱; 柱温: 30 ℃; 流动相: 乙腈+乙酸钠溶液(20:80, V:V); 流速: 1.0 mL/min; 检测波长: 436 nm; 进样量: 10 μL。
分别称取待测样品于50 mL离心管中, 加入提取试剂10 mL, 超声提取30 min后, 在涡旋混匀器上振荡2 min, 静置5 min, 于5000 r/min离心5 min, 将上清液转入25 mL容量瓶中, 样品残渣再用10 mL提取溶液提取一次, 合并2次提取液, 用提取液定容至25 mL, 摇匀, 待衍生化。
准确吸取1 mL试样溶液或标准工作溶液于具塞试管中, 加入0.2 mL碳酸氢钠溶液和0.4 mL 4-二甲基胺基偶氮苯-4-磺酰氯衍生试剂, 混匀后在70 ℃水浴中衍生反应20 min, 用微孔滤膜(0.45 μm)过滤, 待测。
(1)提取溶剂对提取率的影响
选取大豆、小米、菜花、火龙果(白心)、麦芽、绿香瓜和番茄样品, 分别按以下12种提取试剂溶液: a: 40%甲醇溶液; b: 60%甲醇溶液; c: 80%甲醇溶液; d: 甲醇溶液; e: 40%乙醇溶液; f: 60%乙醇溶液; g: 80%乙醇溶液; h: 乙醇溶液; i: 40%乙腈溶液; j: 60%乙腈溶液; k: 80%乙腈溶液; l: 乙腈溶液10 mL加入提取, 其他操作步骤同1.2.4。
(2)料液比对提取率的影响
选取大豆、小米、菜花、火龙果(白心)、麦芽、绿香瓜和番茄样品, 分别按1:5、1:10、1:25、1:50、1:100和1:250 (g:mL)的料液比加入60%乙醇溶液实验, 超声提取30 min后, 其他操作步骤同1.2.4。
(3)提取方式对提取率的影响
选取大豆、小米、菜花、火龙果(白心)、麦芽、绿香瓜和番茄样品, 加入60%乙醇溶液10 mL, 分别以超声、振荡和水浴振荡(40 ℃) 3种提取方式提取30 min, 其他操作步骤同1.2.4。
(4)提取时间对提取率的影响
选取大豆、小米、菜花、火龙果(白心)、麦芽、绿香瓜和番茄样品, 加入60%乙醇溶液10 mL, 分别超声提取10、30、45、60和90 min, 其他操作步骤同1.2.4。
在参考国内外方法和文献研究的基础上[4,17-21], 以提取溶剂的类型(A)、料液比(B)、提取方式(C)和提取时间(D)为考察因素, γ-氨基丁酸提取率的高低为评价指标, 采用L9(34)正交实验设计对大豆、小米、菠菜、火龙果(白心)、麦芽、绿香瓜和木瓜中γ-氨基丁酸的提取方法进行优化, 不同样品各因素和各水平结果见表1
(1)标准曲线的制作
将系列标准工作液分别注入液相色谱仪中, 测定相应的峰面积, 以标准工作液的浓度为横坐标(X, µg/mL), 以峰面积为纵坐标(Y), 绘制标准曲线。
(2) γ-氨基丁酸含量的测定及含量的计算
γ-氨基丁酸含量的测定及含量的计算如公式(1):

式中, ω为试样中γ-氨基丁酸含量, mg/kg; ρ为根据标准曲线计算得到的γ-氨基丁酸质量浓度, µg/mL; V为试样溶液定容体积, mL; m为测试样品称样量, g; 1000为换算系数。
以3次测定结果的算术平均值表示, 计算结果保留3位有效数字。
采用Microsoft Excel 2010和SPSS 21.0软件对数据进行统计分析, 每组实验进行3次平行, 实验结果以平均值表示。
选取25、30、35、40 ℃不同柱温, 对比不同柱温对γ-氨基丁酸的分离效果, 结果显示, 25、30、35、40 ℃时谱图无明显差异。
选取C18色谱柱、SB-Aq色谱柱和NH2色谱柱, 对比上述3种色谱柱对γ-氨基丁酸的分离效果。
选取6种不同流动相种类[甲醇:水溶液(35:65, V:V)等度洗脱、乙腈:水溶液(35:65, V:V)等度洗脱、乙腈:6.8 g/L乙酸钠溶液(35:65, V:V)等度洗脱、乙腈:3.4 g/L乙酸钠溶液(35:65, V:V)等度洗脱、乙腈:0.68 g/L乙酸钠溶液(20:80, V:V)等度洗脱、乙腈:0.34 g/L乙酸钠溶液(35:65, V:V)等度洗脱], 对比6种不同流动相种类对γ-氨基丁酸的分离效果。
从出峰的时间、峰型对称性、响应程度等因素综合考虑, 由图1可知, C18色谱柱分离效果优于SB-Aq和NH2色谱柱; 乙腈:0.68 g/L乙酸钠溶液(20:80, V:V)等度洗脱效果最佳, 因此, 仪器条件选取柱温30 ℃, 乙腈:0.68 g/L乙酸钠溶液(20:80, V:V)等度洗脱。
表2可知, 按不同浓度的甲醇溶液(40%、60%、80%, V:V)、乙醇溶液(40%、60%、80%, V:V)和乙腈溶液(40%、60%、80%, V:V)进行实验, 结果表明, 不同浓度提取大豆、小米、菜花、火龙果(白心)、麦芽、绿香瓜和番茄中γ-氨基丁酸, 其含量较高, 表明提取效果较好。但是测得的γ-氨基丁酸含量之间没有明显差异, 所以选60%甲醇、60%乙醇和40%乙腈作为正交实验的提取试剂。
表3可知, 按1:5、1:10、1:25、1:50、1:100和1:250 (g:mL)的料液比进行实验, 结果表明, 1:5和1:10 (g:mL)料液比提取大豆、小米、菜花、火龙果(白心)、麦芽、绿香瓜和番茄样品中的γ-氨基丁酸, 由于提取不够充分, 其含量较低; 但是, 1:25、1:50、1:100和1:250 (g:mL)料液比提取的不同样品, 其之间无明显差异, 考虑到扩大料液比会带来实验经济成本的增加、实验耗材的浪费以及对环境保护的影响。综合考虑, 以1:25、1:50和1:100 (g:mL)的料液比作为正交实验的提取因素。
表4可知, 以振荡、超声和水浴振荡(40 ℃) 3种提取方式进行实验, 结果表明, 振荡提取大豆、小米、菜花、火龙果(白心)、麦芽、绿香瓜和番茄样品中的γ-氨基丁酸, 其含量较低, 以水浴振荡(40 ℃)和超声的含量要高一些, 且超声和水浴振荡(40 ℃)提取方式之间无明显差异, 这可能是因为单纯的振荡提取效果不够充分, 水浴振荡(40 ℃)和超声两种方式在振荡的同时, 也提供了适合提取γ-氨基丁酸的温度条件, 导致其含量升高。
表5可知, 以10、30、45、60和90 min的提取时间进行实验, 结果表明, 提取时间为60 min时, 大豆、小米、菜花、火龙果(白心)、麦芽、绿香瓜和番茄样品中的γ-氨基丁酸含量较高, 这可能是因为提取时间低于60 min时, 提取效果不够充分, 其中的γ-氨基丁酸没有提取完全, 随着提取时间的继续延长, 样品提取结果已趋于稳定, 且各提取时间之间无明显差异, 综合考虑, 以30、45和60 min作为正交实验的提取因素。
对大豆、小米、菜花、火龙果(白心)、麦芽、绿香瓜和番茄进行正交实验, 由表6方差分析结果可知, 提取溶剂类型对小米、菜花、火龙果(白心)、绿香瓜和番茄中γ-氨基丁酸含量存在显著性影响(P<0.05); 料液比对麦芽和绿香瓜中γ-氨基丁酸含量存在显著性影响(P<0.05); 提取方式对大豆、菜花和麦芽中γ-氨基丁酸含量存在显著性影响(P<0.05); 提取时间对麦芽中γ-氨基丁酸含量存在显著性影响(P<0.05)。综合考虑极差分析和方差分析结果, 确定最优条件为A2B1C1D1A2B1C2D1, 即60%乙醇溶液, 按1:25的料液比, 通过水浴振荡(40 ℃)或超声提取30 min。
将标准系列工作液分别注入液相色谱仪中, 测定相应的峰面积, 以标准工作液的浓度为横坐标, 以峰面积为纵坐标, 绘制标准曲线, 结果表明, 在质量浓度1.092~131.064 μg/mL时, γ-氨基丁酸标准曲线的相关系数(r2)为0.9999, 线性关系良好, 可以满足检测要求。
本研究中方法的检出限和定量限分别以3倍和10倍的信噪比计算。当样品取样量为5.0 g, 定容体积为50 mL时, γ-氨基丁酸的检出限为0.03 mg/kg, 定量限为0.10 mg/kg, 能满足植物源性食品中γ-氨基丁酸含量的准确定量。
选取含有γ-氨基丁酸的代表性食品, 以火龙果(白心)、番茄和麦芽为研究对象别进行精密度实验, 平行测定6次, 考察检测方法的精密度。
根据GB 5009.295—2023《食品安全国家标准 化学分析方法验证通则》的要求, 被测组分含量大于100 mg/kg (10 mg/100 g)时, 实验室内变异系数为3.8%。由表7可知, 样品中的γ-氨基丁酸6次平行实验测定结果的RSDs均小于3.8%, 表明该方法的精密度良好, 适合定量分析。
分别准确称取含有γ-氨基丁酸的代表性食品火龙果(白心)、番茄和麦芽样品, 向样品本底添加低、中、高3个浓度水平的标准溶液, 进行三水平三平行加标回收实验, 计算γ-氨基丁酸的回收率和相对标准偏差。
根据GB 5009.295—2023的要求, 被测组分含量大于100 mg/kg (10 mg/100 g)时, 回收率范围在95%~105%之间。由表8知, 火龙果(白心)、番茄和麦芽样品中的γ-氨基丁酸平均加标回收率分别在99.67%~104.36%、95.80%~97.59%和96.25%~99.30%之间, 加标回收率的相对标准偏差均小于5.0%, 各组分回收率结果满足GB 5009.295—2023的要求, 表明方法回收率高, 适用于植物源性食品中γ-氨基丁酸含量的测定。
选取香菇、平菇、油菜、蒜苔、黄瓜、西葫芦、西兰花、大白菜、菜花、菠菜、架豆角、胡萝卜、紫洋葱、丝瓜、莲藕、青萝卜、冬瓜、番茄、土豆、南瓜、麦芽、紫甘蓝、芹菜、大葱、绿豆芽、圆白菜、李子、绿香瓜、蜜瓜、猕猴桃、杏子、蓝莓、荔枝、香蕉、火龙果(白心)、水蜜桃、梨、橙子、柠檬、芒果、油桃、木瓜、杨梅、樱桃、草莓、苹果46种样品。按本研究建立的方法, 计算不同样品中γ-氨基丁酸的含量和相对标准偏差。结果显示, 各样品在重复性条件下获得的两次独立测定结果的绝对差值均小于平均值的10%, 见表9, 说明本研究建立的方法可适用于植物源性食品中γ-氨基丁酸含量的测定。
选取酒糟原料、酵素、谷物粉、安心蛋白肽固体饮料、陈醋、青稞、银耳芦笋汁、发芽糙米、发芽大豆、青稞苗粉、小麦芽苗菜粉、荞麦芽苗菜粉、小麦籽粒、麦芽、番茄、土豆、南瓜、紫甘蓝、豌豆芽苗菜粉、蚕豆芽苗菜粉、萌芽豆浆、γ-氨基丁酸大豆酸奶、荔枝和木瓜样品, 按本研究建立的方法, 分别计算高效液相色谱仪和氨基酸分析仪测定不同样品中γ-氨基丁酸的含量和相对标准偏差。结果显示, 各样品在重复性条件下获得的两次独立测定结果的绝对差值均小于平均值的10%, 见表10。由于氨基酸分析仪方法处理样品时间较长, 设备维护成本偏高, 仪器应用领域也仅限制于氨基酸相关项目检测等方面, 本研究建立的方法能满足植物源性食品中γ-氨基丁酸含量的测定。
本研究建立了高效液相色谱法测定植物源性食品中γ-氨基丁酸含量的分析方法, 以大豆、小米、菜花、火龙果(白心)、麦芽、绿香瓜和番茄为代表性样品, 进行了单因素实验和正交实验, 确定了最优提取条件为: 60%乙醇溶液, 按1:25 (g:mL)的料液比, 通过水浴振荡(40 ℃)或超声提取30 min。其中, 又以代表性样品火龙果(白心)、番茄、麦芽对线性范围、检出限和定量限、精密度、准确度等指标进行考察, 结果表明, 该方法操作简单、快速, 准确度高, 重现性好, 特异性强, 适用于植物源性食品中γ-氨基丁酸含量的测定。选取多个实际样品测定, 以及仪器比对结果, 解决了仪器方法处理样品时间较长、设备维护成本偏高, 仪器应用领域受限、操作复杂且烦琐、数据结果易受环境因素影响等方面问题。综上所述, 本研究为植物源性食品中γ-氨基丁酸分析与研究提供了方法支持。
