Objective To explore the factors affecting the accuracy of sulfur dioxide detection in preserves and its quality control. Methods Adopt internal quality control measures such as blank experiments, negative matrix testing, negative matrix labeling, quality control sample testing, and personnel parallelism and external quality control to conduct sulfur dioxide testing on preserved fruits. Verifyed the experimental effect with Z value, in depth discussion and analysed of the problems encountered in the process of sulfur dioxide detection and their solutions. Propose technical points for controlling and eliminating experimental deviations. Results The laboratory had passed measurement review and proficiency testing by the series of quality control measures formulated. Conclusion GB 5009.34—2022 National standards for food safety-Determination of sulfur dioxide in food acid-base titration method for detecting sulfur dioxide in food is prone to result deviation. This study proposes the development of various quality control measures, problems encountered during the detection process and their solutions, technical points for controlling and eliminating experimental deviation, which can help improve the accuracy of sulfur dioxide detection, and provide experimental references for other testing institutions to carry out this project.

目的 探究蜜饯中二氧化硫检测准确度的影响因素及质量控制。方法 采取空白实验、阴性基质测试、阴性基质加标、质控样测试、人员平行等内部质量控制和测量审核、能力验证外部质量控制对蜜饯食品开展二氧化硫检测, 以Z值验证实验效果, 深入讨论分析检测过程中遇到的问题及解决思路, 提出控制和消除实验偏离的技术要点。结果 实验室通过制定的系列质量控制措施通过测量审核、能力验证, 评价结果为合格满意。结论 GB 5009.34—2022《食品安全国家标准 食品中二氧化硫的测定》酸碱滴定法检测食品中二氧化硫时容易产生结果偏离, 本研究提出各种质量控制措施、检测过程中遇到的问题及解决思路、控制和消除实验偏离的技术要点等可有助于二氧化硫检测准确性的提高, 也为其他检测机构在该项目的开展提供实验参考。

目的 探究蜜饯中二氧化硫检测准确度的影响因素及质量控制。方法 采取空白实验、阴性基质测试、阴性基质加标、质控样测试、人员平行等内部质量控制和测量审核、能力验证外部质量控制对蜜饯食品开展二氧化硫检测, 以Z值验证实验效果, 深入讨论分析检测过程中遇到的问题及解决思路, 提出控制和消除实验偏离的技术要点。结果 实验室通过制定的系列质量控制措施通过测量审核、能力验证, 评价结果为合格满意。结论 GB 5009.34—2022《食品安全国家标准 食品中二氧化硫的测定》酸碱滴定法检测食品中二氧化硫时容易产生结果偏离, 本研究提出各种质量控制措施、检测过程中遇到的问题及解决思路、控制和消除实验偏离的技术要点等可有助于二氧化硫检测准确性的提高, 也为其他检测机构在该项目的开展提供实验参考。

Objective To explore the factors affecting the accuracy of sulfur dioxide detection in preserves and its quality control. Methods Adopt internal quality control measures such as blank experiments, negative matrix testing, negative matrix labeling, quality control sample testing, and personnel parallelism and external quality control to conduct sulfur dioxide testing on preserved fruits. Verifyed the experimental effect with Z value, in depth discussion and analysed of the problems encountered in the process of sulfur dioxide detection and their solutions. Propose technical points for controlling and eliminating experimental deviations. Results The laboratory had passed measurement review and proficiency testing by the series of quality control measures formulated. Conclusion GB 5009.34—2022 National standards for food safety-Determination of sulfur dioxide in food acid-base titration method for detecting sulfur dioxide in food is prone to result deviation. This study proposes the development of various quality control measures, problems encountered during the detection process and their solutions, technical points for controlling and eliminating experimental deviation, which can help improve the accuracy of sulfur dioxide detection, and provide experimental references for other testing institutions to carry out this project.

蜜饯是一种以果蔬等为主要原料, 添加(或不添加)食品添加剂和其他辅料, 经糖、蜂蜜或食盐腌制(或不腌制)等工艺制成的制品[12]。蜜饯在加工储存过程中容易生虫、发霉和变色, 因此生产者多采用硫磺熏蒸进行加工和养护, 以延长其保存期限[23]。GB 2760—2014《食品安全国家标准 食品添加剂使用标准》规定蜜饯凉果的二氧化硫(包括焦亚硫酸钾、焦亚硫酸钠、亚硫酸钠、亚硫酸氢钠等)和硫磺重熏的最大使用量为0.35 g/kg(以二氧化硫残留量计), 其主要作用是漂白、防腐和抗氧化[46]。而过量硫熏导致二氧化硫残留过量会对人体产生一定危害, 使食用者出现呕吐、恶心等不适症状[79], 所以各地监管部门都会严控蜜饯食品中二氧化硫残留量的超标[7]。因此准确高效地开展蜜饯食品中食品添加剂二氧化硫残留量的监测, 确保蜜饯食品的质量稳定性具有重要的现实意义。
GB 5009.34—2022《食品安全国家标准 食品中二氧化硫的测定》内含3个方法, 其中第一法为酸碱滴定法, 该法适用于所有食品, 仪器装置简便, 操作相对简单, 是各检测机构和企业检测食品中二氧化硫最常用的方法, 也是各实验拥有食品二氧化硫检测能力的必认方法[1011]。但是研究发现该法在实际检测过程中容易受不同因素的影响导致结果出现正向偏离或负向偏离, 且有不同研究者先后发现该法在测定不同食品时出现一定程度的不适用性, 并偶会出现假阳性现象, 都会对检测结果的正确研判带来不少干扰[10,12]。因此如何用酸碱滴定法准确地检测食品中二氧化硫的残留量成了实验室的一道难题[13]
质量控制分内部质量控制和外部质量控制[1415], 内部质量控制是指实验室为保证检测结果的准确性和可靠性, 采取一系列措施, 对检测过程进行全方位监控和管理, 以达到质量控制的目的[14,16]。外部质量控制可以反向推动检测机构深入学习研究标准检测方法, 促进检测机构与同行业其他机构对比, 识别检测过程中存在的问题, 促进检测机构提升检验能力水平, 同时与内部质量控制手段一起配合开展, 共同为检测机构的质量控制提供保障体系[17]。本研究通过内部质量控制包括空白实验、阴性基质测试、阴性基质加标、质控样测试、人员平行等与外部质量控制包括测量审核、能力验证等相结合[15,1819], 对蜜饯食品中二氧化硫的残留量深入开展检测, 以Z值验证执行情况和效果, 同时深入剖析二氧化硫第一法酸碱滴定法检测蜜饯食品中二氧化硫准确性的影响因素, 深入分析检测结果正向偏离、负向偏离产生的原因以及控制和消除实验偏离的技术要点, 并提出了最终的解决方案。本研究的开展有助于蜜饯食品中二氧化硫检测准确性的提高, 同时为其他食品二氧化硫检测准确性的提高提供了借鉴方案, 也为其他检测机构进一步促进该项目检测能力的提升拓宽了思路。
BSA223S电子天平(精度0.01 g, 德国赛多利斯公司); ST106-1RW二氧化硫测定仪(由原ST106-1RW改造, 济南盛泰电子科技有限公司)。
36%~38%盐酸(分析纯, 浙江中星化工试剂有限公司); 30%过氧化氢(hydrogen peroxide, H2O2)(分析纯, 西陇科学股份有限公司); 0.1002 mol/L氢氧化钠标准滴定溶液[GBW(E)082913, 上海安谱实验科技股份有限公司]; 亚硫酸根标准物质(1000 μg/L, 坛墨质检科技股份有限公司)。
测量审核基质样品为菠萝蜜干、山楂干, 厂家均为大连中食国实检测技术有限公司。
盲样为陈皮基质, 用GB 5009.34—2022第一法 酸碱滴定法, 建议样品最小称样量为20 g。
阴性样品为米粉基质, 超市购买。其他样品来自食品生产企业抽样、菜市场抽样等。
取0.1 mol/L氢氧化钠标准滴定溶液10 mL, 用无二氧化碳的水稀释至100.0 mL, 配制成浓度为0.01 mol/L的氢氧化钠标准滴定溶液。
配制3%过氧化氢溶液50 mL作为吸收液, 在吸收液中加入3滴2.5 g/L甲基红乙醇溶液指示剂, 并用0.01 mol/L氢氧化钠标准滴定溶液滴定至黄色终点。
升温功率: 500 W, 升温时间: 10 min, 缓冲时间: 3 min, 保温功率: 300 W, 时间设定: 120 min, 循环冷却水温设置: 10~15 ℃, 氮气流量1.2 L/min左右。
取固体20 g以上(精确至0.01 g), 将称取好的试样置于蒸馏瓶中, 加水300 mL, 置于二氧化硫测定仪, 将玻璃导管插入1.3.2吸收液液面以下。打开循环水泵, 打开氮气, 打开分液漏斗, 使提前加入的10 mL 6 mol/L盐酸溶液快速流入蒸馏瓶, 立即开启仪器按1.3.3设置自动运行至结束。
取下吸收液接收瓶, 将吸收液放冷后摇匀, 用0.01 mol/L氢氧化钠标准滴定溶液滴定至黄色且20 s不褪色。
同步开展空白试样检测。
质量控制措施分内部质量控制措施和外部质量控制措施, 为确保检测结果准确, 根据GB 5009.34—2022特制定以下质量控制措施: (1)空白实验。清洗仪器6个孔位, 分别进行空白测试, 确保试剂无污染, 仪器无污染、无残留; (2)阴性基质加标。挑选阴性样品, 然后选择2个梯度水平进行加标回收; (3)质控样测试。通过开展测量审核对菠萝蜜干、山楂干质控样进行二氧化硫测试, 根据检测结果计算Z值并分析检测过程和结果; (4)选择不同检测人员同时开展盲样比对测试, 通过盲样测试结果验证质控效果, 并对检测过程和结果进行深入探讨分析; (5)采用不同方法进一步进行验证比对。
各数据参照GB 5009.34—2022方法计算, 结果以平均值表示。所有的统计数据表格采用WPS Office 2019进行统计分析。
根据GB 5009.34—2022酸碱滴定法开展二氧化硫含量空白测试、阴性基质样品确定、加标回收、质控样测试、不同人员盲样比对测试, 不同方法比对等系列质量控制措施, 最终确定实验结果。
对二氧化硫测定仪的的6个孔位进行清洗后用纯水进行空白测试, 6个孔位H2O2吸收液的氢氧化钠滴定量均在0.05 mL以下, 说明仪器所有孔位已彻底清洗干净且并未发现污染现象。
从日常检测过程中筛选出阴性样品基质, 按照1.3进行6次阴性基质实验测试, 检测结果发现阴性基质样品的本底平均值为8.90 mg/kg, 按照GB 5009.34—2022酸碱滴定法结果表示为未检出[以定量限(limit of quantitation, LOQ)表示, LOQ为10 mg/kg], 最终确定该样品可作为阴性基质进行加标测试。
选择2.1.2确定的阴性样品, 选择2个梯度水平进行加标回收测试(详见表1)。加标回收发现梯度1的实测结果分别为265.2、282.7、259.6 mg/kg, 加标1回收率分别为102.5%、103.9%、98.2%, 平均回收值为101.5%。梯度2的实测结果分别为407.4、387.9、399.3 mg/kg, 加标2回收率分别为100.8%、97.4%、98.0%, 平均回收值为98.7%。阴性基质加标回收率的结果满足GB/T 27417—2017《合格评定 化学分析方法确认和验证指南》标准要求。
通过开展测量审核对菠萝蜜干、山楂干质控样进行同步测试, 通过表2可发现质控样(菠萝蜜干)的检测结果分别为0.775、0.750、0.786 g/kg, 根据《作业指导书》提供的Z值计算公式得出, 其Z值分别为‒1.02、‒1.33、‒0.89; 质控样(山楂干)的检测结果为分别为1.602、1.570、1.591 g/kg, 其Z值分别为‒0.17、‒0.38、‒0.24。测量审核质控样(菠萝蜜干、山楂干)的Z值绝对值均<2.0, 测量审核检测结果均为满意。
根据表3中2位经验丰富的检测人员同时开展盲样比对测试发现, 人员Ⅰ组、Ⅱ组检测结果的精密度均符合GB 5009.34—2022标准中规定的两次独立测试结果的绝对差值不得超过算数平均值的10%要求, 但是Ⅰ组人员较Ⅱ组人员相比精密度1.0%<2.0%, 结合前期空白测试、阴性基质样品确定、加标回收、质控样测试等系列质量控制措施综合考虑, 选择最终选择Ⅰ组数据199 mg/kg作为盲样考核结果。
本研究的考核结果为199 mg/kg, Z值为1.01, 评价结果为合格, 本组盲样具体检测结果见表4
该次盲样考核评价方式首先剔除粗大误差, 如抄写错误、样品数据报错、小数点错误等, 然后再对测试数据结果进行直方圆分析, 要求数据在符合近似正态分布的基础上再进行稳健分析, 最后按照GB/T 28043—2019《利用实验室间比对进行能力验证的统计方法》要求, 采用稳健(Rosubt)技术处理测试结果和评价结果。最终以实验室间Z值作为判定检验机构能力的统计参数, 按照公式Z=(xX)/δ进行计算Z值, 其中x为实验室的实测值, X为组织方确认的样品指定值, δ为能力评定标准差[20]。然后对参加实验室的测试结果进行判定, 判定结果分为合格和不合格, 其中|Z|<2.0为满意结果, 判定为合格; 2.0<|Z|<3.0为有问题, 警戒信号, Z≥3.0为不满意结果(离群值), 措施信号, 2者均被判定为不合格。Z值“+、‒”表明结果偏离方向, Z值大小体现了实验室结果与中位值的偏离程度[2123]
选取不同样品进行影响因子实验, 该些样品采用GB 5009.34—2022第一法酸碱滴定法严格按照上述质量控制措施进行检测并经GB 5009.34—2022第三法离子色谱对检测结果进行验证, 发现检测结果一致。对该些样品进一步进行准确度影响因子实验后发现, 不同影响因子的变化最终影响实验结果的准确度, 详见表5
表4可发现陈皮中二氧化硫残留量的负偏差最低的为110 mg/kg, 正偏差最高的值为216 mg/kg, 严重偏离中位值, 实际检测过程也发现类似结果偏离。该实验包括空白测试、加热蒸馏、酸碱滴定等操作步骤, 本研究对出现正向偏离、负向偏离的可能性进行影响因子深入探究。
当检测结果偏离中位值或真值的方向为正时即为正向偏离, 该实验检测结果过高的可能原因有: 样品原因、仪器原因、操作原因等方面因素影响。样品原因具体为未按照标准、作业指导书要求进行制样、取样, 导致样品中二氧化硫存在一定不均匀且局部偏高情况, 其他仪器原因、操作原因等方面影响因素详见表6
当检测结果偏离中位值或真值的方向为负时即为负向偏离, 该实验检测结果过低的可能原因有: 样品原因、仪器原因、操作原因等方面因素影响。样品原因具体为未按照标准、作业指导书要求进行制样, 导致样品中二氧化硫存在一定不均匀且局部偏低情况, 其他诸如仪器原因、操作原因等方面影响因素详见表7
此外数据处理引入的误差, 氮气流量计、氢氧化钠标准滴定溶液、滴定管的不确定度, 空白与每个样品间滴定起点颜色和终点颜色未按统一标准判定等都会影响检测结果的准确性[27]
酸碱滴定作为二氧化硫检测现行有效的国标第一法, 仪器简单、操作简便, 是检测食品二氧化硫最常用的方法, 在各级实验室被广泛应用, 然而实验过程有许多细节措施、技术要点需要实验人员把控来提高实验结果准确性。
为防止结果正向偏离过大, 在进行日常检测、测量审核和能力验证时需从样品角度、仪器角度、操作角度等方面进行逐一排查和监控, 样品角度需做到均匀制样取样; 仪器角度需做到控制氮气流速和冷却水温, 蒸馏装置的硫化橡胶部分改用全玻璃, 控制好加热功率等; 操作角度需做好空白校正, 排查假阳性干扰等, 详见表6提及的控制和消除正向偏离的技术要点。
为防止结果负向偏离过大, 在进行日常检测、测量审核和能力验证时需从样品角度、仪器角度、操作角度等方面进行逐一排查和监控, 样品角度需做到均匀制样并捣碎防止结块等; 仪器角度需做到防止蒸馏装置漏气, 控制氮气纯度、流速和加热温度等; 操作角度需控制好酸量、酸浓度、控制好吸收液的初始颜色和配制时间、样品提取时间等, 详见表7提及的控制和消除负向偏离的技术要点。
为避免引入更多额外的误差, 在该项目进行日常检测、测量审核和能力验证时还需注意: (1)根据自身仪器设备的特点, 按照GB/T 27417—2017做低、中、高至少3个浓度水平的加标回收, 根据回收结果选择合适的加热速率、加热温度、氮气流量等, 以防止加热温度、加热速度、氮气流量过高过快过大或过低过慢过小造成实验误差[28]; (2)按照GB/T 27417—2017做阴性样品加标回收、检出限、LOQ、测量范围、准确度、精密度等; (3)用其他方法如离子色谱法进行结果比对和验证。部分食品由于基质特性, 用酸碱滴定法测试可能存在不适性, 可用离子色谱法验证并出具实验结果等[10,12,24]; (4)空白、每个样品间滴定起点颜色和终点颜色要按统一的判断标准等。
酸碱滴定法作为食品中二氧化硫最常用且应用最广的方法, 其检测结果的准确性对监管部门、生产销售商家等而言都尤为重要, 因此提高食品中二氧化硫检测的准确度意义重大。本研究通过研究建立质量控制措施以防二氧化硫检测的准确度产生偏离, 并详细列出了影响二氧化硫检测准确度的可能因素, 同时提出控制和消除实验偏离的技术要点。本研究建立的质量控制措施可靠, 以Z值验证内部质量控制与外部质量控制的执行情况和效果良好, 不同方法验证实验结果一致。本研究的开展有助于蜜饯食品中二氧化硫检测准确性的提高, 也可将其应用在其他食品中二氧化硫残留量的检测上, 建立规范的质量控制措施, 排查影响结果准确性的可能性因素, 在提高检测结果准确性、可靠性的同时, 也提升了实验室自身的检测技术能力[17,2930], 可为其他实验室参与该项目日常检测、测量审核、能力验证提供参考和思路。
