Objective To explore the effects of common food preservatives on the abundance and flora structure of antimicrobial-resistant bacteria in Crassostrea gigas. Methods The control group was refrigerated at 4 ℃, and the experimental group was refrigerated at 4 ℃ after soaking with potassium sorbate, chitosan, Nisin and ɛ-polylysine, respectively. The change of antimicrobial-resistant bacteria content in Crassostrea gigas treated with different food preservatives was analyzed by antimicrobial-resistant plate counting method. The structural changes of antimicrobial-resistant bacteria were analyzed by high-throughput sequencing of 16S rRNA amplicon. Results Potassium sorbate and chitosan could significantly reduce the number of resistant bacteria of single and multiple antimicrobials; Nisin significantly reduced the number of resistant bacteria to co-trimoxazole, chloramphenicol, gentamicin, and bacteria resistant to 2, 3, or 4 antimicrobials simultaneously; ε-polylysine significantly decreased the number of ciprofloxacin resistant bacteria, co-trimoxazole resistant bacteria, chloramphenicol resistant bacteria, gentamicin resistant bacteria and resistant bacteria resistant to 3 or 4 kinds of antimicrobials at the same time. Potassium sorbate and chitosan had a significant effect on the composition of tetracycline and ciprofloxacin resistant bacteria, and Nisin and ε-polysine had a significant effect on the composition of co-trimoxazole resistant bacteria. Conclusion The research results show that the content of antimicrobial-resistant bacteria in Crassostrea gigas can be controlled by applying appropriate food preservatives, food preservatives can affect the composition of culturable antimicrobial-resistant bacteria in Crassostrea gigas and different preservatives can inhibit specific antimicrobial-resistant bacteria, which provides a new idea for the establishment of antimicrobial-resistant bacteria control techniques in Crassostrea gigas.

目的 探究常用食品防腐剂对牡蛎中耐药共生菌的影响作用。方法 以4 ℃冷藏为对照组, 以分别经山梨酸钾、壳聚糖、乳酸链球菌素、ɛ-聚赖氨酸浸泡处理后进行4 ℃冷藏为实验组, 通过耐药平板计数法分析不同食品防腐剂处理后牡蛎中耐药菌含量的变化情况, 并通过16S rRNA扩增子高通量测序分析耐药菌群结构的变化。结果 山梨酸钾与壳聚糖能显著降低单种与多重抗菌药的耐药菌数; 乳酸链球菌素能显著降低复方新诺明耐药菌数、氯霉素耐药菌数、庆大霉素耐药菌数和同时耐受2、3、4种药物的耐药菌数; ε-聚赖氨酸能显著降低环丙沙星耐药菌数、复方新诺明耐药菌数、氯霉素耐药菌数、庆大霉素耐药菌数和同时耐受3、4种药物的耐药菌数。山梨酸钾与壳聚糖对四环素与环丙沙星耐药菌群的组成影响较大, 乳酸链球菌素与ε-聚赖氨酸对复方新诺明耐药菌群的组成影响较大。结论 本研究表明通过应用合适的食品防腐剂可以控制牡蛎中耐药菌的含量, 食品防腐剂可以影响牡蛎中可培养耐药菌群的组成且不同防腐剂能抑制特定的抗菌药耐药菌, 为建立牡蛎中耐药菌的控制措施提供了新思路。

目的 探究常用食品防腐剂对牡蛎中耐药共生菌的影响作用。方法 以4 ℃冷藏为对照组, 以分别经山梨酸钾、壳聚糖、乳酸链球菌素、ɛ-聚赖氨酸浸泡处理后进行4 ℃冷藏为实验组, 通过耐药平板计数法分析不同食品防腐剂处理后牡蛎中耐药菌含量的变化情况, 并通过16S rRNA扩增子高通量测序分析耐药菌群结构的变化。结果 山梨酸钾与壳聚糖能显著降低单种与多重抗菌药的耐药菌数; 乳酸链球菌素能显著降低复方新诺明耐药菌数、氯霉素耐药菌数、庆大霉素耐药菌数和同时耐受2、3、4种药物的耐药菌数; ε-聚赖氨酸能显著降低环丙沙星耐药菌数、复方新诺明耐药菌数、氯霉素耐药菌数、庆大霉素耐药菌数和同时耐受3、4种药物的耐药菌数。山梨酸钾与壳聚糖对四环素与环丙沙星耐药菌群的组成影响较大, 乳酸链球菌素与ε-聚赖氨酸对复方新诺明耐药菌群的组成影响较大。结论 本研究表明通过应用合适的食品防腐剂可以控制牡蛎中耐药菌的含量, 食品防腐剂可以影响牡蛎中可培养耐药菌群的组成且不同防腐剂能抑制特定的抗菌药耐药菌, 为建立牡蛎中耐药菌的控制措施提供了新思路。

Objective To explore the effects of common food preservatives on the abundance and flora structure of antimicrobial-resistant bacteria in Crassostrea gigas. Methods The control group was refrigerated at 4 ℃, and the experimental group was refrigerated at 4 ℃ after soaking with potassium sorbate, chitosan, Nisin and ɛ-polylysine, respectively. The change of antimicrobial-resistant bacteria content in Crassostrea gigas treated with different food preservatives was analyzed by antimicrobial-resistant plate counting method. The structural changes of antimicrobial-resistant bacteria were analyzed by high-throughput sequencing of 16S rRNA amplicon. Results Potassium sorbate and chitosan could significantly reduce the number of resistant bacteria of single and multiple antimicrobials; Nisin significantly reduced the number of resistant bacteria to co-trimoxazole, chloramphenicol, gentamicin, and bacteria resistant to 2, 3, or 4 antimicrobials simultaneously; ε-polylysine significantly decreased the number of ciprofloxacin resistant bacteria, co-trimoxazole resistant bacteria, chloramphenicol resistant bacteria, gentamicin resistant bacteria and resistant bacteria resistant to 3 or 4 kinds of antimicrobials at the same time. Potassium sorbate and chitosan had a significant effect on the composition of tetracycline and ciprofloxacin resistant bacteria, and Nisin and ε-polysine had a significant effect on the composition of co-trimoxazole resistant bacteria. Conclusion The research results show that the content of antimicrobial-resistant bacteria in Crassostrea gigas can be controlled by applying appropriate food preservatives, food preservatives can affect the composition of culturable antimicrobial-resistant bacteria in Crassostrea gigas and different preservatives can inhibit specific antimicrobial-resistant bacteria, which provides a new idea for the establishment of antimicrobial-resistant bacteria control techniques in Crassostrea gigas.

抗菌药能够抑制或杀灭微生物, 广泛用于治疗和控制由细菌感染引发的疾病。然而, 抗菌药物的反复使用会导致细菌耐药性的出现[1]。细菌耐药性已对全球人类公共卫生安全造成严重威胁[2-6], 据2015年欧洲国家的数据统计, 每年有数万人因耐药菌感染而死亡[7]。据估计, 到2050年, 全球因细菌耐药性导致的死亡人数将达到1000万[8]。目前, 防治水产细菌性疾病的手段仍以抗菌药为主[9-10], 虽然开放海域养殖的水生动物基本不会用药, 但由于环境污染[11]以及耐药基因可以在细菌间水平转移等原因, 也会导致水生动物体内细菌产生耐药性[12-13]
牡蛎是中国近海养殖的重要海产品, 其产量位居中国海水养殖产品之首, 具有味道鲜美、营养价值高的特点。作为滤食性动物, 牡蛎的移动能力较差, 当生长的水环境受到污染时, 牡蛎体内极易富集包括微生物在内的多种有害物质[12]。前期研究报道显示牡蛎中含有耐药菌且多重耐药现象较为严重[14-19], 生食或食用未完全煮熟的牡蛎可能会导致这些耐药菌进入人体, 如果耐药菌为致病菌, 则直接导致人体发生疾病, 如果耐药菌为非致病菌, 其含有的耐药基因也可能转移到肠道菌群中使其产生耐药性[20-22], 危害人体健康。因此有必要探究牡蛎中耐药菌的控制措施。
水产品由于含水量较高, 如果储存不当, 其中的微生物极易生长繁殖, 导致产品的腐败及食用安全风险。防腐剂能够有效抑制微生物生长, 阻止微生物繁殖, 是水产品常用的保鲜方式之一, 但防腐剂是否具有抑制耐药菌的作用不得而知。山梨酸钾、乳酸链球菌素、ε-聚赖氨酸与壳聚糖是我国允许使用且常用于水产品保鲜的防腐剂, 本研究将分析这4种食品防腐剂在牡蛎保鲜过程中对耐药菌的影响作用, 并探讨不同防腐剂对耐药菌的作用是否具有种属特异性, 为建立牡蛎中耐药菌的控制措施奠定基础。
三倍体长牡蛎, 采自威海乳山牡蛎养殖场, 低温保存并于当天运回实验室后立即进行实验。
2216E培养基(青岛海博生物技术有限公司); 琼脂粉(北京索莱宝生物科技有限公司); 四环素(tetracycline, TE)、环丙沙星(ciprofloxacin, CIP)、复方新诺明[甲氧苄啶:磺胺甲恶唑(sulphamethoxazole:trimethoprim)=1:19, SXT]、氯霉素[(chloramphenicol, C), 美国药典级]、庆大霉素(gentamicin, CN)、山梨酸钾(纯度99%)、壳聚糖[生工生物工程(上海)股份有限公司]; 乳酸链球菌素、ɛ-聚赖氨酸(浙江新银象生物工程有限公司); E.Z.N.A™Mag-Bind Soil DNA Kit(美国Omega Bio-Tek公司); Qubit dsDNA HS分析试剂盒(美国Thermo Fisher Scientific公司); 2×Hieff®Robust PCR Master Mix、Hieff NGS™DNA Selection Beads[翌圣生物科技(上海)股份有限公司]。
BT 4202S电子分析天平[精度0.01 g, 北京赛多利斯公司]; NU-425-600E型ClassⅡ生物安全柜(美国Nuaire公司); MIR-254型生化培养箱(日本SANYO公司); Pico-21型台式离心机、Q33238型Qubit®4.0荧光计[赛默飞世尔科技(中国)有限公司]; DYY-6C型电泳仪(北京市六一仪器厂); FR-1000型凝胶成像系统(上海复日科技有限公司); ETC 811型PCR仪(北京东胜创新生物科技有限公司)。
流水冲净牡蛎表面, 用灭菌刀具撬取牡蛎所有可食用部分用于实验。将牡蛎肉置于无菌袋中, 4 ℃保存10 d, 作为空白组。实验组则将牡蛎肉在4 ℃条件下分别浸泡于0.1%的山梨酸钾溶液、0.6%的壳聚糖溶液、0.5%的乳酸链球菌素溶液、0.25%的ɛ-聚赖氨酸溶液中30 min, 取出沥干后, 置于无菌袋中4 ℃储存。储存2、4、7和10 d时采集样品进行耐药菌的测定, 每个时间点测定3份样品, 实验重复3次。
每份样品取25 g加入225 g无菌磷酸盐缓冲液, 充分匀浆后进行10倍梯度稀释, 选择适宜浓度的稀释液0.1 mL分别涂布单抗平板(2216E培养基中分别含药物终浓度为TE 16 mg/mL (TE16)、TE 32 mg/mL (TE32)、CIP 4 mg/mL、SXT 80 mg/mL[甲氧苄啶:磺胺甲恶唑=4:76 (mg/mL)]、C 32 mg/mL、CN 16 mg/mL、多抗平板[2216E培养基分别含TE 16 mg/mL+SXT 80 mg/mL (TS)、TE 16 mg/mL+SXT 80 mg/mL+CIP 4 mg/mL (TSC)、TE 16 mg/mL+SXT 80 mg/mL+CIP 4 mg/mL+CN 16 mg/mL (TSCCN)]以及空白平板(不含药物, K), 每个稀释度涂布3个平板, 对于低浓度的耐药菌, 则取1 mL涂布3个耐药平板(检出限为10 CFU/g)。将平板置于30 ℃恒温培养箱中培养72 h±3 h。对各个平板上的菌落进行计数, 并计算出样品中各类抗菌药耐药菌的数量(以lg CFU/g计)。当样品中菌落数低于本方法的检出限10 CFU/g时, 以检出限的二分之一计算。
选择1.4.2中对照组和经防腐剂处理7 d的实验组涂布后, 选择含有适宜菌数的耐药平板。向平板中加入适量磷酸盐缓冲液, 用无菌接种环轻轻刮取菌落, 将菌液混匀后转移至无菌离心管中。重复上述步骤2次, 合并菌液, 于8000 r/min离心20 min, 弃去上清液, 沉淀用E.Z.N.A™ Mag-Bind Soil DNA Kit提取试剂盒提取基因组DNA, 并通过电泳检测确定DNA浓度。
对基因组DNA进行聚合酶链反应(polymerase chain reaction, PCR)扩增, 采用2轮PCR扩增方法。第一轮采用上游引物(Nobar_341F: 5’-CCTACGGGNG GCWGCAG-3’)和下游引物(Nobar_805R: 5’-GACTACHV GGGTATCTA ATCC-3’)对16S rRNA基因的V3~V4可变区进行扩增。第一轮PCR扩增体系: 2×Hieff®Robust PCR Master Mix 15 μL, 上下引物各1 μL, 模板10~20 ng, 超纯水补充至30 μL。PCR反应条件为95 ℃预变性3 min, 94 ℃变性20 s, 55 ℃退火20 s, 72 ℃延伸30 s, 进行5个循环, 72 ℃延伸5 min, 最后10 ℃保温。
第二轮扩增引入Illumina桥式PCR兼容引物进行扩增。第二轮PCR扩增体系: 2×Hieff®Robust PCR Master Mix 15 μL, 上下引物各1 μL, 模板20~30 ng, 超纯水补充至30 μL。反应条件为95 ℃预变性3 min, 94 ℃变性20 s, 55 ℃退火20 s, 72 ℃延伸30 s, 进行5个循环, 72 ℃延伸5 min, 最后10 ℃保温。扩增结束后, 通过2%琼脂糖凝胶电泳检测文库大小, 并使用Qubit 4.0荧光定量仪测定文库浓度。所有样本按照1:1等量混合后依托生工生物工程(上海)股份有限公司的Illumina MiSeq测序平台进行高通量测序。
测序后获得双端序列数据, 使用Cutadapt[23]软件去除引物接头序列, 然后依据PE reads间的overlap关系使用PEAR[24]软件将成对的reads拼接(merge)成一条序列。根据barcode标签序列区分各样本数据, 通过质控过滤得到用于后续分析的有效数据。采用RDP classifier[25]软件对97%相似度水平的操作分类单元(operational taxonomic unit, OUT)代表序列进行分类学分析, 并在属水平统计各个样本的菌群组成。
实验结果以平均值±标准偏差表示, 使用GraphPad Prism 9进行绘图, 通过SPSS 25进行t检验分析, P<0.05表示具有显著性差异, P<0.01表示具有极显著性差异。
冷藏条件下牡蛎中耐药菌含量的变化见图1。在10 d的储存过程中, 空白平板(K)的细菌总数以及TE耐药菌数整体上呈上升趋势, 且尤以空白平板表现的最为明显, 这表明细菌在冷藏条件下仍能生长。储存10 d后, K、TE16和TE32的耐药菌数分别增加了1.69、1.31和1.35 lg CFU/g, CIP耐药菌数、C耐药菌数、同时耐受TE和SXT双重抗菌药(TS)的耐药菌数以及同时耐受TE、SXT和CIP三重抗菌药(TSC)的耐药菌数没有显著变化, SXT耐药菌数、CN耐药菌数、同时耐受TE、SXT、C和CN四重抗菌药(TSCCN)的耐药菌数与初始相比极显著下降(P<0.01), 分别降低了0.92、1.12和0.86 lg CFU/g。TSCCN的耐药菌数在储存7 d后降低至检出限以下。由此可见, 冷藏过程只能显著降低牡蛎中SXT、CN与TSCCN的耐药菌数。
山梨酸钾处理后牡蛎中耐药菌含量的变化见图2。经过山梨酸钾处理后, 牡蛎中的细菌总数整体上不断下降, 这表明山梨酸钾能抑制细菌的生长。两种浓度的TE耐药菌数变化趋势相似, 在储存2 d菌数下降的最为明显, 在4~10 d菌数下降缓慢。储存10 d后, TE16、TE32的耐药菌数显著降低(P<0.05), 其他单种和多重药物耐药菌数降低更为明显(P<0.01), CIP、SXT、C、CN、TS、TSC耐药菌数分别降低了1.52、2.08、1.02、1.16、1.42和1.93 lg CFU/g。TSCCN的耐药菌数在储存4 d后降低至检出限以下。与空白组相比, 山梨酸钾还可显著降低TE、CIP、C、TS与TSC的耐药菌数。
乳酸链球菌素处理后牡蛎中耐药菌含量的变化见图3。储存10 d后, 牡蛎中的细菌总数与各耐药菌数均有所下降, 但TE32耐药菌数和CIP耐药菌数变化并不显著(P>0.05), SXT、C、CN、TS、TSC耐药菌数分别降低了1.78、1.84、1.18、1.30和1.92 lg CFU/g。TSCCN的耐药菌数在储存7 d后降低至检出限以下。与空白组相比, 乳酸链球菌素还可显著降低C、TS与TSC的耐药菌数。
ε-聚赖氨酸处理后牡蛎中耐药菌含量的变化见图4。储存10 d后, 细菌总数、TE耐药菌数与TS耐药菌变化不显著(P>0.05), CIP、SXT、C、CN耐药菌数显著降低, 分别减少了1.08、1.33、0.74和2.15 lg CFU/g。TS与TSC耐药菌数在7 d时达到最低后, 10 d时的耐药菌数反而升高, 可能是由于有些对ε-聚赖氨酸耐受的菌得到了进一步的增长。TSCCN的耐药菌数在储存4 d后降低至检出限以下。与空白组相比, ε-聚赖氨酸还可显著降低CIP、C与TSC的耐药菌数。
壳聚糖处理后牡蛎中耐药菌含量的变化见图5。不同类型的抗菌药耐药菌数在2 d后都显著下降。细菌总数与TE耐药菌数的变化趋势相似, 且10 d后耐药菌数均降低了2.00 lg CFU/g以上。CIP、SXT、C、CN、TS、TSC的耐药菌数在10 d后均降低了1.60 lg CFU/g以上, 其中以C耐药菌数较低的最明显, 降低了1.97 lg CFU/g。TSCCN的耐药菌数在储存4 d后降低至检出限以下。壳聚糖可显著降低细菌总数与各药物耐药菌数, 表明壳聚糖的抑菌效果最好。
经过防腐剂的处理并冷藏7 d后, 牡蛎中耐药菌数发生显著变化, 因此测定了7 d后耐药菌群的组成变化情况, 结果见图6
未经防腐剂处理的对照组样品初始菌群以黏着杆菌(Tenacibaculum)和弧菌(Vibrio)为主, 占比分别为40.27%与45.13%。冷藏7 d后, 细菌菌群(K7)的相对丰度有明显不同, 其中黏着杆菌下降了6.26%, 而假交替单胞菌(Pseudoalteromonas)与弧菌的相对丰度分别上升了13.26%与23.77%。防腐剂处理后, 细菌组成与相对丰度发生了很大变化, 其中山梨酸钾组(PS7)以弧菌为主, 占比超过59.79%, 黄杆菌(Flavobacterium)的相对丰度较对照组有所上升, 乳酸链球菌组(N7)以假交替单胞菌、希瓦氏菌(Shewanella)和弧菌为主, ε-聚赖氨酸组(PL7)以假交替单胞菌和希瓦氏菌为主, 壳聚糖组(C7)以黏着杆菌占绝对比重。由此可见, 经不同防腐剂处理后, 牡蛎中细菌菌群组成差异明显, 说明防腐剂的抑菌作用具有种属特异性。
对于牡蛎中的TE耐药菌, 对照组初始样本以弧菌、希瓦氏菌、假交替单胞菌为主, 占比分别为59.34%、12.94%、7.47%, 冷藏7 d后仍以这3种菌为主, 占比分别为44.25%、18.32%、32.26%。经防腐剂处理后, TE耐药菌群的相对丰度发生变化; 山梨酸钾处理后(PS7-TE32), 仍以弧菌占比最大, 假交替单胞菌所占比重大大降低, 黄杆菌的相对丰度有所增加; 乳酸链球菌素处理后(N7-TE32), 以假交替单胞菌占绝对优势, 弧菌与希瓦氏菌比重下降; ε-聚赖氨酸处理后(PL7-TE32)的物种组成与N7-TE32相似; 壳聚糖处理后(C7-TE32)则以不动杆菌(Acinetobacter)、希瓦氏菌和黄杆菌为主。由此可见, 山梨酸钾与壳聚糖对TE耐药菌群的组成有较大影响。
对于CIP耐药菌, 对照组初始样本以未分类的黄杆菌(unclassified-Flavobacteriaceae)为主, 冷藏7 d后, 菌群结构变化较大, 以假交替单胞菌、嗜冷杆菌(Psychrobacter)和弓形杆菌(Arcobacter)为主。经4种防腐剂处理后, 与K7-CIP相比, 嗜冷杆菌的相对丰度下降最明显; N7-CIP与PL7-CIP均以假交替单胞菌占比最大, 而PS7-CIP与C7-CIP以黄杆菌为主, 相对丰度均在80%以上。由此可见, 山梨酸钾与壳聚糖对CIP耐药菌群的组成有较大影响。
对于SXT耐药菌, 对照组初始样本以未分类的红杆菌(unclassified-Rhodobacteraceae)为主, 冷藏7 d后, 假交替单胞菌和弧菌的比率有所增加。经不同防腐剂处理后, N7-SXT以假交替单胞菌占绝对优势, 其占比为75.99%; PL7-SXT主要以黏着杆菌、不动杆菌和假单胞菌(Pseudomonas)为主; PS7-SXT与C7-SXT组成相似, 均以不动杆菌、鲁杰氏菌(Ruegeria)和乳酸杆菌(Lactobacillus)为主, 说明乳酸链球菌素与ε-聚赖氨酸对SXT耐药菌群的组成有较大影响。
无论是否采用防腐剂处理, 牡蛎中CN耐药菌群均以黏着杆菌占绝对优势, 相对丰度超过85%, 说明防腐剂对CN耐药菌群的组成无较大影响。
前期已有牡蛎中耐药菌的相关研究报道, FLORES-VILLASENOR等[14]发现牡蛎中的副溶血性弧菌约73.5%对至少一种抗菌药耐受, 25%的菌株对多种药物具有耐药性; WANG等[15]从牡蛎中分离出的所有菌株对4种或更多抗菌药具有耐药性; ZANGOEI-FARD等[16]从牡蛎中分离出的弧菌属对TE的耐药率最高; 李炳等[17]利用单种抗菌药平板从香港牡蛎消化道分离的310株异养细菌对20种抗菌药存在一定程度耐药性; 王瑞旋等[19]发现来自阳江的香港牡蛎养殖水体与牡蛎体内的异养细菌多重耐药状况严重, 异养菌多重耐药比例较高。本研究采用判定为耐药性的最小抑菌浓度作为药物的使用浓度制备耐药平板, 由于前期发现TE的耐药程度较为严重, 因此增加了二倍最小抑菌浓度的TE耐药平板, 通过这些耐药平板测定样品中的耐药菌数。结果发现牡蛎对TE、CIP、SXT、C、CN均有一定的耐受性, 其中以TE耐药菌含量最高, TE的浓度影响不显著, 其次为CN耐药菌, 多重耐药程度也较为严重, 也验证了牡蛎体内耐药菌污染水平较高。由于耐药基因可以通过质粒或其他可移动基因元件发生水平转移[26-27], 从而造成耐药菌污染范围的扩大, 因此有必要探寻耐药菌的控制技术, 保障牡蛎的食用安全和人类健康。
防腐剂能够有效抑制微生物生长, 是水产品常用的保鲜方式之一。山梨酸钾、乳酸链球菌素、ε-聚赖氨酸与壳聚糖是我国允许使用且常用于水产品保鲜的防腐剂。山梨酸钾能有效抑制鱼、虾、贝等水产品中的霉菌和好氧性细菌[28]; ε-聚赖氨酸抑菌谱广, 对大部分革兰氏阴性菌和阳性菌、酵母菌和霉菌均具有较高的抑菌活性, 是一种安全性高的生物抑菌剂, 对海产品有较好的保鲜作用[29]; 乳酸链球菌素通常能抑制大部分革兰氏阳性菌及其芽孢的生长, 而对革兰氏阴性菌抑菌范围较窄[30], 不仅能有效杀灭单增李斯特菌形成的成熟生物膜[31], 还能够抑制高水分烤虾中腐败菌的生长以延长产品货架期[32]; 壳聚糖对革兰氏阴性菌和革兰氏阳性菌都具有抑制作用, 对革兰氏阳性菌的抑制作用强于革兰氏阴性菌[33]。本研究按照GB 2760—2014《食品安全国家标准 食品添加剂使用标准》中规定的最大使用量来配制防腐剂的浓度, 以期探究防腐剂正常使用过程中对耐药菌的抑制作用。经4种食品防腐剂处理后, 牡蛎中的细菌总数与耐药菌数均有所降低, 进一步验证了以上4种食品防腐剂对水产品中细菌的抑制作用。通过对耐药菌数计数发现, 食品防腐剂能够抑制耐药菌的生长, 不同防腐剂对不同药物耐药菌的抑制作用不同, 其中壳聚糖的抑制效果最好。壳聚糖的抑菌作用有两方面, (1)能渗透进细胞体内, 吸附细胞体内带有阴离子的细胞质并发生絮凝作用, 扰乱细胞正常生理活动, 从而杀灭细菌; (2)壳聚糖能吸附在细胞表面, 形成一层高分子膜, 阻止了营养物质向细胞内的运输, 从而起到抑菌、杀菌作用[34], 因此表现出较强的抑菌活性。防腐剂在抑制微生物的同时, 还具有降低牡蛎中蛋白质的降解、改善牡蛎品质的作用[35]
通过16S扩增子高通量测序分析耐药平板中的菌群结构特征, 结果表明不同防腐剂对不同种类抗菌药耐药菌种属的抑制作用有所不同。山梨酸钾主要抑制耐受TE的假交替单胞菌和黏着杆菌、耐受CIP的假交替单胞菌、弓形杆菌和嗜冷杆菌以及耐受SXT的假交替单胞菌和弧菌。经壳聚糖处理后, TE与SXT耐药菌群中弧菌与假交替单胞菌的相对丰度大大降低, CIP耐药菌群中假交替单胞菌、弓形杆菌与嗜冷杆菌的相对丰度大大降低, 壳聚糖在3种耐药菌群中的抑制作用有所不同, 可能是由不同耐药菌群中优势菌的性质差异造成的。乳酸链球菌素与ε-聚赖氨酸对TE与SXT耐药菌群中弧菌的抑制作用明显, 对CIP耐药菌群中弓形杆菌与嗜冷杆菌的抑制作用明显。但无论是否使用防腐剂, CN耐药菌群中均以黏着杆菌占绝对优势, 其他种属的菌群结构变化不明显, 表明防腐剂对不同种属CN耐药菌群的抑制率相似。从结果可以看出, 假交替单胞菌、弧菌更容易受到防腐剂的抑制。目前并没有防腐剂对耐药菌的抑菌研究报道, 下一步需要深入开展相关研究, 通过分离不同种属耐药菌, 探讨抑菌剂对不同种属耐药菌的抑制作用, 从分子水平解析抑菌的种属特异性机制。
总之, 本研究初步探讨了常用食品防腐剂对牡蛎中耐药共生菌的抑制作用, 表明食品防腐剂能够影响牡蛎中可培养耐药菌群的组成, 通过应用合适的食品防腐剂可以控制牡蛎中耐药菌的含量以提高牡蛎的食用安全性。下一步将探讨更为有效的控制措施对耐药菌的抑制作用及抑制机制, 以期建立高效的耐药菌防控技术。
