Objective To establish a method for the determination of nitrite content in pickles by nitroso derivative ultra performance liquid chromatography. Methods A novel and sensitive method for the determination of nitrite content was developed. This is a nitroso derivative produced based on the interaction between nitrite and antipyrine This derivative has strong absorption in 0.02 mol/L acetate buffer solution (contains 0.1% acetic acid) at a wavelength of 345 nm the nitrite content was determined by ultra performance liquid chromatography. Results Nitrite in the linear concentration range of 0.05-5.00 mg/L was determined with a limit of detection 0.01 µg/mL. The high, medium and low concentrations were spiked. The extraction recoveries ranged from 88%-96% (n=6). The relative standard deviation was the precision ranged from 2.0%-4.5%. Conclusion The method can effectively eliminate the interference of chloride ions and other anions and pigments on this experiment. The established analytical method has good sensitivity, accuracy, precision and recovery rate, and is suitable for the detection of nitrite in pickles.

目的 建立亚硝化衍生超高效液相色谱法测定酱腌菜中亚硝酸盐含量的方法。方法 在0.02 mol/L乙酸铵缓冲液(内含0.1%乙酸)中, 安替比林与亚硝酸盐相互作用, 生成亚硝基衍生物, 该衍生物在波长345 nm有较强的吸收, 用超高效液相色谱法进行检测分析, 采用ACQUITY UPLC BEH Shield RP18色谱柱(150 mm× 2.1 mm, 1.7 µm)进行分离, 外标法定量。结果 目标物在0.05~5.00 µg/mL质量浓度范围线性良好, 检出限为0.01 mg/L。高、中、低3水平浓度加标, 检测回收率为88%~96% (n=6), 相对标准偏差为2.0%~4.5%。结论 该方法能够有效消除氯离子等阴离子和色素对本研究的干扰, 建立的分析方法灵敏度、准确度、精密度和回收率好, 适用于酱腌菜中亚硝酸盐的测定。

目的 建立亚硝化衍生超高效液相色谱法测定酱腌菜中亚硝酸盐含量的方法。方法 在0.02 mol/L乙酸铵缓冲液(内含0.1%乙酸)中, 安替比林与亚硝酸盐相互作用, 生成亚硝基衍生物, 该衍生物在波长345 nm有较强的吸收, 用超高效液相色谱法进行检测分析, 采用ACQUITY UPLC BEH Shield RP18色谱柱(150 mm× 2.1 mm, 1.7 µm)进行分离, 外标法定量。结果 目标物在0.05~5.00 µg/mL质量浓度范围线性良好, 检出限为0.01 mg/L。高、中、低3水平浓度加标, 检测回收率为88%~96% (n=6), 相对标准偏差为2.0%~4.5%。结论 该方法能够有效消除氯离子等阴离子和色素对本研究的干扰, 建立的分析方法灵敏度、准确度、精密度和回收率好, 适用于酱腌菜中亚硝酸盐的测定。

Objective To establish a method for the determination of nitrite content in pickles by nitroso derivative ultra performance liquid chromatography. Methods A novel and sensitive method for the determination of nitrite content was developed. This is a nitroso derivative produced based on the interaction between nitrite and antipyrine This derivative has strong absorption in 0.02 mol/L acetate buffer solution (contains 0.1% acetic acid) at a wavelength of 345 nm the nitrite content was determined by ultra performance liquid chromatography. Results Nitrite in the linear concentration range of 0.05-5.00 mg/L was determined with a limit of detection 0.01 µg/mL. The high, medium and low concentrations were spiked. The extraction recoveries ranged from 88%-96% (n=6). The relative standard deviation was the precision ranged from 2.0%-4.5%. Conclusion The method can effectively eliminate the interference of chloride ions and other anions and pigments on this experiment. The established analytical method has good sensitivity, accuracy, precision and recovery rate, and is suitable for the detection of nitrite in pickles.

酱腌菜是一种以蔬菜为主要原料, 经过腌制、调味等工艺加工而成的食品。酱腌菜的制作过程包括精选、清洗、切分(或不切分)、脱盐、添加调味料和食品添加剂(如辣椒油、食用盐、酿造酱油、味精、白砂糖及香辛料等), 再经过调配或炒制、分装等工序。酱腌菜的种类多样, 包括‌酱渍菜、‌盐渍菜和‌酱油渍菜等方法对根茎类蔬菜进行制作, 味鲜可口, 深受人们的喜爱。现代科学研究发现, 在腌制过程中, 蔬菜中的硝酸盐会被大量还原, 产生酱腌菜的亚硝酸盐积累问题, 亚硝酸盐作为常见的致癌物质, 严重威胁人们的身体健康, 也因此受到人们的广泛关注。在GB 2714—2003《食品安全国家标准 酱腌菜》酱腌菜卫生标准中, 明确规定酱腌菜中亚硝酸盐含量不得高于20 mg/kg。目前已发展多种亚硝酸盐检测方法, 诸如紫外可见分光光度法[1-3]、动力学分光光度法[4-6]、极谱法[7-8]、荧光分析法[9-12]、离子色谱法[13-15]、气相色谱法[16-19]、流动注射[20-22]和液相色谱法[23-27]等, 食品中亚硝酸盐的国家检测方法为GB 5009.33—2016《食品安全国家标准 食品中亚硝酸盐与硝酸盐的测定》, 该检测方法中有两种: 第一是离子色谱法, 通过电导检测器直接检测亚硝酸根, 该方法极易受到其他阴离子的干扰, 容易造成假阳性结果; 第二是分光光度法, 此方法检测灵敏度较低, 且分光光度计本身抗干扰能力差, 易受提取液糊化、本底颜色等影响。酱腌菜因工艺过程复杂, 蔬菜本身就含有各种复杂色素, 部分食品还添加淀粉、酱油等的, 提取液透光性较差, 加入活性炭并不能完全吸附有色的物质, 导致有色的物质对比色时有严重干扰, 出现结果偏差的缺点, 而高效液相法能有效解决这一问题。
目前, 应用高效液相色谱仪来检测食品中的亚硝酸盐含量的文献报道较少, 之所以亚硝酸盐检测无法在高效液相色谱仪上得到应用, 主要原因在于缺少合适的衍生试剂。亚硝酸盐自身不具有响应信号, 需要通过与检测试剂反应后得到准确且高灵敏的检测信号。目前, 已有标准及国内外文献报道的衍生试剂主要为Griess试剂和2,3-二氨基萘试剂[28-29], 并应用于紫外和荧光分光光度计的检测。陈文生等[30]采用高效液相色谱法在204 nm波长下测定亚硝酸根离子, 此法前处理较简单, 但分析的基质比较单一, 且低波长下检测可能存在干扰。沈䶮鸣等[31]用2,3-二氨基萘衍生剂采用高效液相色谱法在233 nm波长下测定亚硝酸盐, 该方法也是在低波长下检测, 干扰较多。本研究通过探究衍生试剂的反应活性和反应性质, 筛选出高性能的亚硝酸盐衍生试剂, 利用亚硝酸根离子与衍生剂生成的化合物(目标物)进行测定, 采用C18色谱柱对目标物进行分离, 在高波长(345 nm)下进行检测, 可为含高盐和色素等复杂基质中亚硝酸盐含量的测定提供技术支持和参考。
ACQUITY UPLC H-Class超高效液相色谱仪(配备二极管阵列检测器)、ACQUITY UPLC BEH Shield RP18色谱柱(150 mm×2.1 mm, 1.7 µm)(美国Waters公司); BSA223S万分之一电子天平(德国赛多利斯公司); DK-S28水浴箱(上海精宏实验设备有限公司); TG16-WS台式高速离心机(上海卢湘仪离心机仪器有限公司); FW80型高速万能粉碎机(天津泰斯特仪器有限公司); 0.22 µm微孔滤膜(上海安谱实验科技股份有限公司)。
萝卜类酱腌菜、黄瓜类酱腌菜、榨菜类酱腌菜和白菜类酱腌菜各2件, 市售散装。
亚硝酸盐标准溶液(质量浓度100 mg/L, GSB05-1142-2000, 有效日期: 2027-03-31, 环保标样所); 甲醇(色谱纯, 德国Merck公司); 安替比林、硼砂、亚铁氰化钾、乙酸锌、乙酸钠、乙酸(分析纯, 国药集团化学试剂有限公司); 实验用水均为经Milli-Q超纯水装置过滤的超纯水。
饱和硼砂溶液: 称取5.0 g硼砂钠, 溶于100 mL热水中, 冷却后备用。
亚铁氰化钾溶液: 称取10.6 g亚铁氰化钾, 用水溶解, 并稀释至100 mL。
乙酸锌溶液: 称取22.0 g乙酸锌, 加3 mL冰乙酸溶于水, 并稀释至100 mL。
乙酸铵缓冲溶液: 称取1.54 g乙酸铵, 加水200 mL,加冰乙酸1.0 mL, 加水稀释至1000 mL。
安替比林溶液: 称取安替比林2 g溶于100 mL乙酸铵缓冲溶液中。
临用时, 移取亚硝酸盐标准原液5.00 mL于50 mL容量瓶中, 用乙酸铵缓冲溶液稀释成10.0 mg/L的工作液。于10 mL试管中加入适量10.0 mg/L亚硝酸盐标准使用液, 加乙酸铵缓冲溶液至刻度, 再加入安替比林溶液1.0 mL, 摇匀, 配制成质量浓度为0.05、0.10、0.50、1.00、2.00、5.00 mg/L的系列标准溶液。放置10 min, 过0.22 µm针头滤膜, 上机。
将切碎的样品用四分法取适量, 用粉碎机制成匀浆, 准确称取酱腌菜约5 g(精确到0.001 g)试样于50 mL具塞离心管中, 加水约25 mL, 涡旋混匀, 于50 ℃水浴超声20 min, 冷却至室温后加亚铁氰化钾溶液2 mL和乙酸锌溶液2 mL, 混匀, 于8000 r/min离心5 min, 将水相转移至50 mL容量瓶中, 于残渣中加水20 mL, 涡旋混匀后超声5 min, 于8000 r/min离心5 min, 将水相转移到同一50 mL容量瓶中, 并用水定容至刻度, 混匀, 过滤, 弃掉初滤液, 准确移取适量滤液于10 mL比色管中, 加乙酸铵缓冲液至刻度, 加安替比林溶液1.0 mL, 混匀, 放置10 min, 经0.22 µm滤膜过滤, 上机。
ACQUITY UPLC BEH Shield RP18色谱柱(150 mm× 2.1 mm, 1.7 µm); 流动相: 甲醇:0.02 mol/L乙酸铵缓冲液(内含0.1%乙酸)=20:80 (V:V, 下同); 流速0.30 mL/min; 柱温35 ℃; 检测波长345 nm; 进样体积10 µL。
采用Origin pro 7.5软件对数据进行分析, 并制表、绘制图形。
为选取检测的最佳波长, 对标准溶液在200~400 nm范围内进行光谱扫描, 绘制了吸收曲线, 按实验方法, 见图1可知, 体系在波长345 nm处, 吸光度最大。本研究选用345 nm为测量波长。
安替比林亚硝化后为亚硝化安替比林, 极性适中、分子量不大、可采用反相色谱柱分离。在反相超高效液相色谱法检测中, C18色谱柱对待测物极性的适用范围较大, 对于极性适中、分子量不大的待测物均有较好的保留特性。本研究考察了亚硝化安替比林在C18反相色谱柱上的色谱保留特性, 分别实验了ACQUITY UPLC BEH C18、ACQUITY UPLC BEH Shield RP18和ACQUITY UPLC CSH C18色谱柱, 使用BEH C18色谱柱, 保留时间较短, 使用CSH C18色谱柱, 色谱峰不对称, 有点拖尾。使用BEH Shield RP18色谱柱, 色谱峰对称, 衍生剂对目标分析物无干扰, 分离较好, 因此, 本研究最终采用ACQUITY UPLC BEH Shield RP18 (150 mm×2.1 mm, 1.7 µm)色谱柱。
分别用甲醇-水、甲醇-0.1%乙酸、甲醇-0.02 mol/L乙酸铵和甲醇-0.02 mol/L乙酸乙酸铵缓冲液(内含0.1%乙酸)4种体系的流动相分析测定亚硝基衍生物。结果表明, 甲醇-0.1%乙酸乙酸铵缓冲液流动相体系下亚硝化衍生物分离效果最好, 因此甲醇-0.02 mol/L乙酸铵缓冲液(内含0.1%乙酸)体系被选为流动相。调节甲醇的比例, 当甲醇-乙酸铵缓冲液的体积比为20:80时, 亚硝基衍生物出峰时间6.0 min左右, 目标物与样品基质分离, 且分析时间短, 因此确定甲醇-0.02 mol/L乙酸铵缓冲液(内含0.1%乙酸)体积比为20:80。
为保证衍生物的稳定, 根据文献[1], 安替比林与亚硝酸根相互作用分光光度法测定亚硝酸根, 选用乙酸调节乙酸铵溶液pH为3.6、4.6和5.5作对比, 由图2可知, 当添加0.1%乙酸时pH约为4.6, 亚硝基衍生物分离效果最佳, 保留时间最为适宜, 且与亚硝基衍生反应的溶液pH一致, 确保了衍生后物质的稳定, 因此本研究选择内乙酸铵缓冲液含0.1%乙酸作为流动相。
综上所述, 通过实验优化得到了最佳的色谱条件: ACQUITY UPLC BEH Shield RP18色谱柱(150 mm×2.1 mm, 1.7 µm); 流动相: 甲醇-0.02 mol/L乙酸铵缓冲液(内含0.1%乙酸)=20:80; 流速0.30 mL/min; 柱温35 ℃; 检测波长345 nm; 进样体积10 µL。在最佳色谱条件下, 得到的标准溶液色谱图, 如图3所示。
以不含待测成分的酱腌菜作为空白基质, 按照1.3.3前处理方法, 通过3个浓度水平的加标回收实验, 评价方法的准确度和精密度, 根据测定的线性范围设加标浓度为5、10、20 mg/kg, 每个加标水平6次平行, 结果见表1, 回收率为88%~96%, 平均回收率为93%。该方法相对标准偏差为2.0%~4.5%, 相对标准偏差均低于5%, 表明该方法精密度良好, 能够满足对酱腌菜中亚硝酸盐的检测需求。
分别配制质量浓度为0.05、0.10、0.50、1.00、2.00、5.00 mg/L的标准溶液, 在波长345 nm下进行高效液相色谱分析, 以质量浓度为横坐标(X, mg/L), 峰面积为纵坐标(Y), 得出线性回归方程为Y=2.28014×104X-7.34976, r=0.9992, 在0.05~5.00 mg/L质量浓度范围内与峰面积呈现良好的线性关系。以信噪比(S/N≥3)计算方法得出检出限, 检出限为0.01 mg/L, 结果满足检测的要求。
安替比林在紫外233 nm处有较强的吸收, 而在345 nm几乎无吸收, 其他阴离子及有机化合物对测定的影响在本色谱条件下, 由于溶液常见的无机阴离子如氯离子、磷酸根离子、硫酸根离子、碳酸根离子等离子无紫外吸收。即使样品可能存在有机化合物的干扰, 可经BEH Shield RP18色谱柱分离, 虽在色谱图中有杂峰, 但不会造成色谱分离的干扰, 仍可以得到清晰的谱图。
采集8份市售酱腌菜, 按1.3.3样品前处理方法, 对酱腌菜中亚硝酸盐进行测定, 经检测7份样品结果含量均小于2 mg/kg, 其中, 一份黄瓜类酱腌菜样品检测值为26 mg/kg, 高于GB 2762—2017《食品安全国家标准 食品中污染物限量》对蔬菜及其制品(腌渍蔬菜)的亚硝酸盐限量为20 mg/kg。图4为酱腌菜中亚硝酸盐含量测定的样品色谱图。
亚硝酸盐是一种极性很强的化合物, 亚硝酸盐在C18柱上基本无保留, 对亚硝酸盐进行衍生, 通过优化亚硝化衍生条件和色谱条件, 具有较合适的保留和分离作用, 将亚硝化衍生的高效液相色谱技术成功应用于酱腌菜中亚硝酸盐含量的检测, 该方法简便、灵敏、特异性强, 且不必考虑酱腌菜本底色素对检测的干扰。
