Objective To establish ultra performance liquid chromatography-tandem quadrupole electrostatic field orbital trap mass spectrometry (UPLC-Q-Orbitrap) and use various data analysis methods to quickly identify the chemical composition of Liubao tea. Methods A Thermo Scientific Hypersil GOLD aQ (2.1 mm×100 mm, 1.9 μm) counm was used with 0.1% formic acid-acetonitrile solution (A) and 0.1% formic acid-water solution (B) as the mobile phase in a gradient elution mode. Detection of chemical components in Liubao tea by positive and negative ion mode of electrostatic spray ion source combined with electrostatic field orbital trap detector. Based on accurate mass to charge ratio data from primary high-resolution mass spectrometry, fragment ions from secondary high-resolution mass spectrometry, Thermo Scientific mzCloud network database, Thermo Scientific high-resolution mass spectrometry database for traditional Chinese medicine components, and relevant literature reports, identify the chemical components. Results Rapid analysis was conducted on the chemical components of Liubao tea, and a total of 36 kinds of compounds were identified, included 3 kinds of amino acid compounds, 3 kinds of phenolic compounds, 16 kinds of flavonoids, 1 kinds of lignin compound, 4 kinds of alkaloids, 1 kinds of terpenoid compound, 2 kinds of coumarins, 4 kinds of organic acid compounds, and 2 kinds of other compounds. Conclusion This study apply UPLC-Q-Orbitrap database to analyze the composition of Liubao tea, which can provide scientific basis for further study on the efficacy and flavor substance basis of Liubao tea.

目的 建立超高效液相色谱-串联四极杆静电场轨道阱质谱法(ultra performance liquid chromatography-tandem quadrupole electrostatic field orbital trap mass spectrometry, UPLC-Q-Orbitrap)与多种数据解析方式对六堡茶化学成分进行快速鉴定。方法 采用Thermo Scientific Hypersil GOLD AQ (2.1 mm× 100 mm, 1.9 μm)色谱柱, 在流动相A: 0.1%甲酸-水; 流动相B: 0.1%甲酸-乙腈中进行梯度洗脱; 以电喷雾离子源搭配静电场轨道阱检测器正负离子模式对六堡茶中的化学成分进行检测。根据一级高分辨率质谱精确质荷比数据、二级高分辨率质谱碎片离子、Thermo Scientific mzCloud网络数据库、Thermo Scientific中药成分高分辨质谱数据库以及相关文献报道, 对化学成分进行鉴定。结果 对六堡茶化学成分进行快速分析, 共鉴定出36种, 包括3种氨基酸, 3种酚, 16种黄酮, 1种木质素, 4种生物碱, 1种萜, 2种香豆素, 4种有机酸, 其他2种。结论 本研究应用UPLC-Q-Orbitrap结合数据库对六堡茶进行成分分析, 可为进一步研究六堡茶的功效、风味物质基础提供了科学依据。

目的 建立超高效液相色谱-串联四极杆静电场轨道阱质谱法(ultra performance liquid chromatography-tandem quadrupole electrostatic field orbital trap mass spectrometry, UPLC-Q-Orbitrap)与多种数据解析方式对六堡茶化学成分进行快速鉴定。方法 采用Thermo Scientific Hypersil GOLD AQ (2.1 mm× 100 mm, 1.9 μm)色谱柱, 在流动相A: 0.1%甲酸-水; 流动相B: 0.1%甲酸-乙腈中进行梯度洗脱; 以电喷雾离子源搭配静电场轨道阱检测器正负离子模式对六堡茶中的化学成分进行检测。根据一级高分辨率质谱精确质荷比数据、二级高分辨率质谱碎片离子、Thermo Scientific mzCloud网络数据库、Thermo Scientific中药成分高分辨质谱数据库以及相关文献报道, 对化学成分进行鉴定。结果 对六堡茶化学成分进行快速分析, 共鉴定出36种, 包括3种氨基酸, 3种酚, 16种黄酮, 1种木质素, 4种生物碱, 1种萜, 2种香豆素, 4种有机酸, 其他2种。结论 本研究应用UPLC-Q-Orbitrap结合数据库对六堡茶进行成分分析, 可为进一步研究六堡茶的功效、风味物质基础提供了科学依据。

Objective To establish ultra performance liquid chromatography-tandem quadrupole electrostatic field orbital trap mass spectrometry (UPLC-Q-Orbitrap) and use various data analysis methods to quickly identify the chemical composition of Liubao tea. Methods A Thermo Scientific Hypersil GOLD aQ (2.1 mm×100 mm, 1.9 μm) counm was used with 0.1% formic acid-acetonitrile solution (A) and 0.1% formic acid-water solution (B) as the mobile phase in a gradient elution mode. Detection of chemical components in Liubao tea by positive and negative ion mode of electrostatic spray ion source combined with electrostatic field orbital trap detector. Based on accurate mass to charge ratio data from primary high-resolution mass spectrometry, fragment ions from secondary high-resolution mass spectrometry, Thermo Scientific mzCloud network database, Thermo Scientific high-resolution mass spectrometry database for traditional Chinese medicine components, and relevant literature reports, identify the chemical components. Results Rapid analysis was conducted on the chemical components of Liubao tea, and a total of 36 kinds of compounds were identified, included 3 kinds of amino acid compounds, 3 kinds of phenolic compounds, 16 kinds of flavonoids, 1 kinds of lignin compound, 4 kinds of alkaloids, 1 kinds of terpenoid compound, 2 kinds of coumarins, 4 kinds of organic acid compounds, and 2 kinds of other compounds. Conclusion This study apply UPLC-Q-Orbitrap database to analyze the composition of Liubao tea, which can provide scientific basis for further study on the efficacy and flavor substance basis of Liubao tea.

六堡茶, 因原产于梧州市苍梧县六堡镇而得名[1], 作为一种传统的黑茶, 其历史可追溯超过1500年, 以其“红、浓、陈、醇”的特点而著称 [2-3]。六堡茶属于后发酵茶, 与其他5大茶类不同, 细菌与真菌在黑茶生产、陈化和存放过程中持续存在生理活动[4-5], 使六堡茶“越陈越好”, 因而六堡茶有“耐于久藏, 且越陈越香”之说[6], 而实质上, 不同六堡茶茶香是茶叶中各类香气成分和浓度组合而形成的特有香型, 市面上六堡茶有: 槟榔香、陈香、药香、蜜香、参香、枣香、菌香等16种香型[7]。传统上常用于消渴降暑、除腻去积、调整肠胃[8]等, 现代研究证实其具有良好的有降血糖、抗氧化、抗高血脂、缓解糖尿病等功效[9-12]
近些年来, 在当地政府的指导, 以及在茶农、茶厂、茶商的共同努力下, 六堡茶各方面都得到长足发展, 传统加工工艺被列入国家级非物质文化遗产名录[13-14]; 茶园面积、产量和产值都逐年攀升[15-17]; 也发布实施DB 45/T479— 2014《六堡茶加工技术规程》、GB/T 32719.4—2016《黑茶第4部分: 六堡茶》、DBS 45/057—2018《六堡茶(传统工艺)》等标准, 进一步规范了六堡茶加工工艺标准[18-19]。六堡茶发展至今也碰到困境与瓶颈, 潘凤等[20]对六堡茶面临的成本不断攀升, 产品安全问题, 推广力度不足等, 提出六堡茶产业要不断创新和探索, 积极应对挑战和不足, 提高知名度; 池云凤[21]通过深入分析总资产周转率、净利润率、总资产收益率和净资产收益率这4个关键财务指标下滑的原因, 提出茶企实行品牌年轻化战略部署的必要性和可行性, 并建议推出与鲜花结合的花风味调配茶; 吴燕[22]分别发明了山楂六堡茶、灵芝六堡茶[23]、补钙六堡茶[24]、姜糖六堡茶[25]则是对六堡茶多样化的有力补充。
相对于功效研究与产品研发, 六堡茶在产品质量控制方面的研究也颇为深入, 如: 对六堡茶香气成分[26-29]的研究, 对其他化学物质[30-31]的研究, 对有毒有害物质[32-34]的研究, 对元素[35-37]的研究均有较多报道, 但以超高效液相色谱-串联四极杆静电场轨道阱质谱法(ultra performance liquid chromatography-tandem quadrupole electrostatic field orbital trap mass spectrometry, UPLC-Q-Orbitrap)对六堡茶的研究却鲜有报道, 这对花风味调配茶、水果风味茶、保健功能茶的研发尤其不利, 这些风味已经改变了的产品, 其风味物质与原茶相比, 风味成分已经发生了较大变化, 需要对其品质进行质量控制。本研究拟运用UPLC-Q-Orbitrap技术, 对六堡茶中主要的化学成分进行快速分析, 期望为其他风味六堡茶开拓风味物质快速分析的方法。
U3000超高压液相色谱仪、Q-Exactive四极杆/静电场轨道阱高分辨质谱仪(美国Thermo Fisher Scientific有限公司); PS-50T型超声波清洗器(深圳市洁康洗净有限公司); BP211D电子分析天平(感量0.01 mg, 德国赛多利斯仪器公司); IQ 7000超纯水仪(德国默克Milli-Q公司); DFT-100A 100克手提式高速粉碎机(温岭市林大机械有限公司)。
六堡茶产地为梧州。
甲醇、乙腈(色谱级, 美国Thermo Fisher Scientific有限公司); 甲酸(色谱级, 国药集团有限公司); 水为自制超纯水, 符合GB/T 6682—2008《分析实验室用水规格和试验方法》规定的一级水。
取六堡茶适量, 以粉碎机粉碎成细粉。取样品细粉2.0 g, 精密称定, 置三角锥形瓶中, 精密加入20 mL甲醇, 称定重量, 超声提取(300 W, 60 KHz) 30 min; 放冷至室温, 补足重量, 取10 mL提取液, 6000 r/min离心5 min, 取上清液, 过0.22 μm微孔滤膜, 待测。
色谱柱: Thermo Scientific Hypersil GOLD AQ (2.1 mm× 100 mm, 1.9 μm); 柱温: 40 ℃; 流动相A: 0.1%甲酸-水; 流动相B: 0.1%甲酸-乙腈。梯度洗脱程序见表1
主要质谱参数见表2
通过TraceFinder4.1软件中的中药成分高分辨质谱数据库(Thermo Scientific OTCML)及Compound Discoverer3.0软件筛查网络数据库(Thermo Scientific mzCloud)与样品的保留时间、同位素情况、主要二级离子、二级质谱图相识度比对及相关文献报道等多种方式, 综合判断分析[38], 确定检出成分。
通过UPLC-Q-Orbitrap技术分析, 六堡茶鉴定出9类共36种化学成分, 3种氨基酸类, 3种酚类, 16种黄酮类, 1种木质素类, 4种生物碱类, 1种萜类, 2种香豆素类, 4种有机酸类, 其他2种; 有3种是通过监测模式(正负离子)能同时检测到的化学成分。各类化学成分归属见表3, 总离子流图(正负离子图)见图1
本研究鉴定出氨基酸3种, 以L-苯丙氨酸为例: 测定值[M+H]+ m/z 166.08589拟合化学式C9H11NO2, 理论值166.08626, 偏差2.228 ppm。高分辨二级离子谱图中, m/z 120.08068为母离子失去一分子-H2O和-CO后所生成的离子碎片, m/z 149.05949为母离子失去一分子-NH3后所生成的离子碎片, 而m/z 103.05439为m/z 149.05949失去一分子-H2O和-CO后生成的离子碎片, 根据二级离子特征, 及OTCML数据库筛选, 可推断该成分为L-苯丙氨酸。L-苯丙氨酸的高分辨二级离子质谱图见图2
本研究鉴定出酚类3种, 以原儿茶酸为例: 测定值[M-H]- m/z 153.01968, 拟合化学式C7H6O4, 理论值153.01933, 偏差-2.287 ppm。高分辨二级离子谱图中, m/z 109.02965为母离子失去-CO2后生成的离子碎片; m/z 91.01904为m/z 109.02965打开苯环并失去一分子-H2O后生成的离子碎片根据二级离子特征, 及OTCML数据库筛选, 可推断该成分为原儿茶酸。原儿茶酸的高分辨二级离子质谱图见图3
本研究鉴定出黄酮类16种, 以异牡荆素为例: 测定值[M+H]+433.11209, 拟合化学式C21H20O10, 理论值433.11292, 偏差1.916 ppm。高分辨二级离子质谱图中, m/z 287.05432为母离子失去一个-C9H6O2后生成的离子碎片; m/z 397.09098为母离子失去两个-H2O后生成的离子碎片; m/z 303.04953为m/z 397.09098更进一步失去-C6H6O后生成的离子碎片; m/z 163.03859为母离子失去的[C9H7O3]+产生的离子碎片; 根据二级离子特征, 及OTCML数据库筛选, 可推断该成分为异牡荆素。异牡荆素的高分辨二级离子质谱图见图4
本研究鉴定出香豆素类2种, 以7-羟基香豆素为例: 测定值[M+H]+ m/z 163.03894, 拟合化学式C9H6O3, 理论值163.03897, 偏差0.184 ppm。高分辨二级离子质谱图中, m/z 147.04376为母离子失去一个-O后生成的离子碎片; m/z 135.04379为母离子失去一个-CO后生成的离子碎片, m/z 107.04921为m/z 135.04379失去一个-CO后生成的离子碎片; 根据二级离子特征, 及OTCML数据库筛选, 可推断该成分为7-羟基香豆素。7-羟基香豆素的高分辨二级离子质谱图见图5
本试验鉴定出有机酸类4种, 以没食子酸为例: 测定值[M-H]- m/z 169.01477, 拟合化学式C7H6O5, 理论值169.01425, 偏差-3.077 ppm。高分辨二级离子质谱图中, m/z 125.02476为母离子失去一个-CO2后生成的离子碎片; m/z 123.00962为m/z 125.02476失去2个-H离子形成双键后生成的离子碎片。m/z 150.97131为母离子失去一个-OH后生成的离子碎片; 根据二级离子特征, 及OTCML数据库筛选, 可推断该成分为没食子酸。没食子酸的高分辨二级离子质谱图见图6
本研究采用UPLC-Q-Orbitrap技术, 通过高分辨质谱二级离子碎片信息、网络数据库(Thermo Scientific mzCloud)、中药成分高分辨质谱数据库(Thermo Scientific OTCML), 及相关文献报道[39-40], 对六堡茶化学成分进行快速分析, 共鉴定出36种, 包括3种氨基酸类, 3种酚类, 16种黄酮类, 1种木质素类, 4种生物碱类, 1种萜类, 2种香豆素类, 4种有机酸类, 其他2种。本研究可为六堡茶的工艺控制, 产品质量, 等级评定等提供参考。
在化学成分鉴定上, 该方法过程简单、快速, 可同时获得正负离子两种扫描模式的高分辨一级和二级质谱图[39-40], 一级离子质谱图具有较高的质量精度, 其质荷比偏差均小于±5 ppm, 根据测得质量数和二级碎片离子特征, 拟合出各离子碎片的分子式, 为进一步鉴定化合物成分及其成分的结构解析提供支持。
